Infertility is directly related to chromosomal abnormalities in germ cells. Among them, the aneuploidies are the most frequent chromosomal abnormalities and responsible for embryo implantation failures, miscarriages, fetal losses and newborns with congenital malformations, mental disability and neuropsychomotor developmental delay. Male patients with normal somatic karyotype may present different rates of aneuploidies in sperm, resulting in abnormal embryos. This study aimed to correlate the frequency of chromosomal aneuploidies in spermatozoa with embryo implantation rate in couples undergoing assisted reproductive techniques. The methodology has included chromosomal analysis by GTG banding and molecular cytogenetic study using Fluorescent In Situ Hybridization technique for evaluation of chromosomes 9, X and Y in germ cells of 22 patients referred to the Human Reproduction Service of the Clinical Hospital FMRP-USP. Embryo implantation rates were determined by hormonal evaluation in maternal peripheral blood and ultrasound confirmation. Two patients presented abnormal karyotype, characterized by polymorphism of the heterochromatic region of the long arm of chromosome 9 and a satellite in the short arm of chromosome 22. Both alterations, usually considered variants of normality, have been related to infertility phenotype and miscarriages. Significant differences were detected between couples who presented pregnancy (group 1) and couples with embryo implantation failure (group 2), with higher frequency of aneusomy and diploidy of chromosome 9, as well as total aneuploidy in sperm of group 2 patients. Our results suggest a correlation between aneuploidy and embryo implantation rates, since the infertile group with reproductive failure has showed higher frequency of aneuploidy. Screening for aneuploidies detection in male germ cells should be included in order to decrease embryo implantation failures, miscarriages and fetuses with chromosomal ...

In the present work we performed molecular cytogenetic studies in two different populations of Astyanax bockmanni from streams of Botucatu, SP: one from Capivara river, on Tietê river basin and one from Água da Madalena river, on Paranapanema river basin. The results showed that the population of Astyanax bockmanni from Capivara river have a diploid number of 50 chromosomes, with karyotype consisting of 8 methacentric, 14 submethacentric, 12 subtelocentric and 16 acrocentric, while the individuals of the population from Água da Madalena river have 50 diploid chromosomes but with karyotype organized in 8 metacentric, 14 submetacentric, 16 subtelocentric and 12 acrocentric. Also, the rDNA 18s sequences are widely dispersed throughout the genome of two populations, with intra and interindividual variations. On the other hand, the sequences for rDNA 5S and Histone H1 remained chromosomally conserved in these two samples and sites located in pairs 2 and 19 (rDNA 5S) and the pairs 2 and 15 (Histone H1). The low dispersion of structural genes and a functional dynamic independence between sequences of rDNA 5S and rDNA 18S may be related to the process of karyotype maintenance and differentiation in these populations of Astyanax bockmanni

Compounds released into the environment can induce genetic alterations in living organisms. A group of chemicals that shows proven toxicity is the pesticides, and the insecticides are the most harmful. The insecticides of the family phenylpyrazole have wide application both in agriculture and in homes. Fipronil, an insecticide of this chemical group, is widely used in various cultures and in homes, mainly for fighting fleas and ticks on dogs and cats. The use of fipronil may represent a risk to man and the environmental health, since this pesticide can potentially induce cell death, regardless of cell type. Fipronil, when in contact with the environment, can undergo various degradation processes, including photodegradation. The toxic effect of one of its metabolites derived from photodegradation, sulfone-fipronil, is approximately 20 fold as great as fipronil itself. The A. cepa test system was used to evaluate cytotoxic, genotoxicity and mutagenic effects of fipronil before and after phptodegradation. Seeds of Allium cepa were subjected to solutions of fipronil, pre-exposed or not exposed to degradation by sunlight. The germination tests were conducted both under the effect of light and in the dark. We evaluated the cumulative potential of this insecticide using 48 and 72-hours recovery tests. The results showed that when fipronil was previously exposed to the sun, it presented a greater genotoxic and mutagenic potential, showing that the metabolites formed by photodegradation can show more harmfull effects

Several studies use only male rats in their experiments, even with all the evidence of differences between the sexes, not only in the reproductive behavior but also in the development of diseases. This absence of studies using female rats is due in part to the estrous cycle. It is known that one estrous cycle lasts four to 5 days and consists of four phases: proestrus, estrus, metestrus and diestrus. Ramos et al. (1999) identified and mapped a QTL in an F2 population derived from interbreeding of strains of SHR (spontaneously hypertensive rats) and Lewis (LEW) rats and found, on chromosome 4, the first QTL of the world related to anxiety in rats. After several backcrosses between SHR and LEW strains, it was developed a congenic strain called SLA16, to refine the study of this QTL. This QTL corresponds to a large genomic region, approximately 75 million basepairs (Mb), and was first called Ofil1 (inner open field locomotion 1). Currently it is called Anxrr16 (anxiety-related response region 16) according to the rat genome database (RGD). Izidio et al. (2011) suggested that the natural fluctuation of the hormones in the estrous cycle of female rats could influence the effects of this locus on behavior. That is, females in DP (diestrus-proestrus) present the QTL in the same genomic region that males, while females in EM (estrus-metestrus) do not present it. The effects of the estrous cycle in the SLA16 strain and its isogenic control SHR were analyzed on the elevated plus maze (EPM), black and white box and on the open field test (OF). In the open field test, the SLA16 strain had higher central locomotion (F=5.0, p <0.05) and peripheral locomotion (F=12.6, p <0.01) compared to the SHR strain. In the elevated plus maze test, the SLA16 strain had the higher number of entries (F=9.5, p <0.01) and time spent in the open arms (F=10.3, p <0.01) compared to the SHR strain. Micronuclei (MN) originate from chromosome fragments or whole chromosomes that lag behind at anaphase during cell division can occur due to excessive exposure to harmful agents, defects in mitosis and / or DNA repair process. The study evaluated the frequency of micronuclei in cytological samples after daily use of alcohol -based mouthwash mouth in smokers and non-smokers. We assessed 30 subjects, aged between 17-42 years and established as exclusion criteria: alcohol consumption; periodontal disease; systemic diseases that may interfere with the integrity of the oral mucosa; individuals with medical or pharmacological history that affect the performance of the study, family history of malignancies. For smokers the inclusion criterion was a minimum consumption of 20 cigarettes / day, and the contrast required for non-smokers. The sample was divided into 02 experimental groups: Group 1 Smokers and Group 2 Non-Smokers, both groups performed mouthwash (alcohol 21,6%) with Listerine® Defense of Teeth and Gums for 04 weeks cytological collection were performed on buccal mucosa and portion median border of the tongue, one for initial, week 0 control, and subsequent three weeks with continued use of mouthwash. The collected material was stained by specific staining Feulgen / Fast Green. The analysis of 3000 cells / individual was performed by light microscopy statistics used Kruskal- Wallis and Mann-Whitney tests for frequency MN. The frequency of micronuclei in buccal mucosa was 1.6 ± 1.75 and 1.46 ± 2.06 for the site language to Group 1 Smokers of showing a slight increase compared with Group 2 Non-smoking, which showed a variation of 1.2 ± 1.53 for the buccal mucosa site and 0.86 ± 1.41 for the site language. However, this difference was not statistically significant. The results suggest that alcohol-based mouthwash tested is not an inducer of genotoxic changes, thus not changing the frequency...
