Stingrays are elasmobranchs found along the seacoast and in some rivers of Brazil. Pain is the most conspicuous symptom observed in patients wounded by the bilaterally retroserrate stingers located in the tail, which are covered by glandular and integument tissues. In addition, cutaneous necrosis is commonly observed in injuries caused by freshwater stingrays. The aim of this work was to characterize and compare certain properties of tissue extracts obtained from the glandular tissues covering the stinger apparatus of Potamotrygon falkneri and Dasyatis guttata stingrays. By sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), tissue extracts have similar bands above 80kDa, but most differences were observed below this molecular mass. Lethal, dermonecrotic and myotoxic activities were detected only in P. falkneri tissue extract. Edematogenic activity was similar and dose dependent in both tissue extracts. Nociceptive activity was verified in both tissue extracts, but P. falkneri presented a two-fold higher activity than D. guttata tissue extract. No direct hemolysis, phospholipase A(2) and coagulant activities were observed in both tissue extracts. Antigenic cross-reactivity was noticed by ELISA and Western blotting, using antisera raised in rabbits. Species-specific sera reacted with several components of both tissue extracts, noticeably above 22 kDa. Both tissue extracts presented gelatinolytic, caseinolytic and fibrinogenolytic activities, which were not caused by the action of metalloproteinases. Hyaluronidase activity was detected only in P. falkneri tissue extract. Our experimental observations suggest that P. falkneri tissue extract is more toxic than D. guttata tissue extract. These results may explain why injuries caused by freshwater stingrays are more severe in human accidents. (c) 2007 Elsevier Ltd. All rights reserved.

Realizou-se uma investigação das mudanças histológicas e ultra-estruturais das células de Sertoli durante o ciclo reprodutivo de machos de Piaractus mesopotamicus. Os resultados mostraram que o desenvolvimento das células de Sertoli está estritamente relacionado à maturação das células gaméticas. Portanto, as células de Sertoli têm algum papel na maturação das células germinativas durante o ciclo reprodutivo dessa espécie, talvez formando um tecido de sustentação para os cistos espermatogênicos em desenvolvimento, ajudando a reorganização testicular para um novo ciclo reprodutivo, além de outras possíveis funções discutidas no texto.

Some aspects of both the nucleolar organizer activity and meiosis were studied in the testes of Triatoma vitticeps (Heteroptera, Triatominae). The techniques used included squashing followed by lacto-acetic orcein staining, silver-ion impregnation, fluorescent banding (CMA(3), Quinacrine mustard and DAPI) and fluorescent in situ hybridization (FISH). A close relationship between heterochromatin and nucleolus in testicular cells was observed. During meiosis, the silver-ion impregnation pattern varied. At metaphase plate, a small body appeared apart from the chromosomes. In the spermatids this small body was seen in preparations stained with orcein and silver- ion impregnation but not with fluorochromes or FISH. These characteristics combined suggest that these corpuscles represent a source of ribonucleoproteins (RNP) - RNA and specific nucleolar proteins. Silver-ion impregnation and (FISH) revealed nucleolar organizer activity in two metaphase sex chromosomes (X). These results indicate that, in these species, nucleolar organizer regions (NORs) are located in the sex chromosomes, X chromosomes were were CMA(3)(+) and Y chromosome was DAPI(+).

We studied the karyotype, spermatogenesis and nucleolar activity at spermatogenesis in five species of Heteropera: Hyalymenus sp and Neomegalotomus pallescens, Alydidae; Catorhintha guttula and Hypselonotus fulvus, Coreidae; and Niesthrea sidae, Rhopalidae. They showed a red (Alydidae) or orange (Coreidae and Rhopalidae) membrane covering the testes, which consisted of seven testicular lobes, except in N. pallescens, which had only five. All the species had m-chromosomes, an X0 sex chromosome system and 10 (Hyalymenus sp, N. pallescens, and N. sidae), 16 (H. fulvus) or 22 (C. guttula) autosomes. Similar to the other species described to date, all these species showed holocentric chromosomes, interstitial chiasmata in most autosomes, and autosomes dividing reductionally in the first meiotic division and equationally in the second, while sex chromosomes, divided equationally and reductionally in the first and second meiotic divisions, respectively. In addition, we observed that the sex chromosome is heteropycnotic at prophase and that heteropycnotic chromosomal material is found in the nuclei at spermatogenesis; variation in size, shape and location of the nucleolar material occurs during spermatogenesis, denoting a variable degree of activity in the different stages.

Em Corydoradinae, a presença de espermátides junto com espermatozóides no lúmen dos túbulos testiculares sugere uma espermatogênese do tipo semicística, enquanto que em Callichthyinae a produção do esperma ocorre inteiramente dentro dos espermatocistos no epitélio germinativo, caracterizando a espermatogênese cística. A espermiogênese em Callichthyinae é caracterizada por um desenvolvimento inicial lateral do flagelo, pela presença de rotação nuclear em diferentes graus, formação de uma fossa nuclear excêntrica ou medial, formação de um canal citoplasmático, e presença de migração centriolar, sendo mais similar à espermiogênese do tipo I. em Corydoradinae, a espermiogênese é caracterizada pelo desenvolvimento excêntrico do flagelo, ausência de rotação nuclear, fossa nuclear excêntrica, formação de um canal citoplasmático, e ausência de migração centriolar, diferindo dos tipos descritos previamente. O processo de espermatogênese e espermiogênese em Corydoradinae e Callichthyinae revelaram caracteres únicos para cada subfamília, corroborando a hipótese de que as mesmas constituem grupos monofiléticos. em relação à ultraestrutura do esperma, a análise comparativa das espécies de Callichthyidae mostra que as características gerais encontradas nos espermatozóides foram similares, reforçando a hipótese de monofilia da família.
