As bromélias apresentam formas exóticas com uma grande diversidade de cores e de flores, constituindo importantes espécies para uso em paisagismo e floricultura. em conseqüência disto, são muito comercializadas. Porém, parte das plantas que se encontram no mercado, ainda são provenientes do extrativismo. Esta situação é também reflexo do pequeno número de informações sobre técnicas de propagação e cultivo. Uma das limitações é o desconhecimento do tipo de substrato e adubação adequados ao cultivo destas espécies. Assim, o objetivo deste trabalho foi avaliar o crescimento de bromélias do Neoregelia cruenta, cultivadas em diferentes substratos e adubações foliares através da altura, do número de folhas, da matéria fresca e seca, da parte aérea e das raízes. As mudas utilizadas foram provenientes de cultura de tecidos. Após um período de pré-aclimatização, foram transplantadas para estufa sem nebulização. Os substratos foram constituídos da mistura de diferentes proporções de solo, areia, casca de arroz carbonizada e por um substrato comercial à base de vermiculita. Aos substratos foi aplicada adubação foliar, combinando uréia e sacarose, em intervalos de quinze dias. Os resultados obtidos mostraram que as interações entre os substratos, dose de sacarose e de uréia, não afetaram a altura das mudas e o número de folhas. O uso de sacarose também não influenciou o desenvolvimento das mudas. O substrato comercial à base de vermiculita, independente da aplicação de adubação foliar, proporcionou maior altura das mudas e número de folhas. A aplicação de uréia apresentou efeito linear crescente durante o período avaliado.

A durabilidade e a qualidade pós-colheita de flores de corte são atributos fundamentais na sua valoração ao longo da cadeia produtiva e na satisfação dos consumidores. Objetivou-se, nesta pesquisa, verificar o efeito do tiossulfato de prata, associado ou não à sacarose, na manutenção da qualidade pós-colheita de hastes de crisântemos (Chrysanthemum morifolium Ramat cv. Dragon) . O experimento foi conduzido sob delineamento inteiramente casualizado, em esquema fatorial testando soluções de manutenção com tiossulfato de prata (STS), sob cinco níveis (Água destilada; STS a 0,2 mM; STS a 0,2 mM + sacarose a 50 g L-1; STS a 0,4 mM; STS a 0,4 mM + sacarose a 50 g L-1); e data de amostragem, por três níveis (0; 3; 6 dias). Utilizaram-se três repetições com duas hastes florais em cada tratamento. Foram feitas avaliações físicas: coloração, massa fresca e conteúdo relativo de água (CRA); avaliações químicas: açúcares redutores e pigmentos; e avaliações qualitativas: turgescência, cor das flores, e número de botões, flores entreabertas e abertas. O tratamento com 0,2 mM de STS possibilitou melhor manutenção da massa fresca das hastes. A concentração de pigmentos e carboidratos redutores foi maior naqueles tratamentos em que a sacarose foi associada. A coloração e o conteúdo relativo de água foram favorecidos nos tratamentos STS a 0,2 e 0,4mM. A concentração de 0,2 mM de STS foi a que obteve os melhores resultados, prolongando a vida de vaso das hastes. A qualidade das hastes foi superior, tendo as melhores avaliações de conteúdo de água, cor e turgescência.

O desenvolvimento e uso de técnicas que prolonguem a durabilidade das flores, mantendo a qualidade do produto, é imprescindível para redução de perdas pós-colheita. Objetivou-se, neste trabalho, avaliar soluções de manutenção, associadas ou não a sacarose, na manutenção da qualidade pós-colheita de hastes de crisântemos. O experimento foi conduzido sob delineamento inteiramente casualizado, em esquema fatorial, com cinco tratamentos e três repetições, com 2 hastes florais cada uma. Os tratamentos utilizaram água destilada; 8-HQC a 100 mg L-1; 8-HQC a 100 mg L-1 + sacarose a 50 g L-1, 8-HQC a 200 mg L-1; 8-HQC a 200 mg L-1 + sacarose a 50 g L-1. Foram feitas avaliações físicas: coloração, massa fresca e conteúdo relativo de água (CRA); avaliações químicas: açúcares redutores e pigmentos; e avaliações qualitativas: turgescência, cor das flores, e número de botões, flores entreabertas e abertas. A combinação de 8-HQC 200 mg L-1 + sacarose a 50 g L-1 foi a que apresentou melhor desempenho para a manutenção da qualidade das hastes florais, favorecendo a abertura de botões e a turgescência das lígulas. A sacarose contribuiu para uma melhor manutenção de substâncias de reserva na haste, que tiveram a sua vida de vaso estendida.

This work investigated the effects of increasing temperature from 30 degrees C to 47 degrees C on the physiological and genetic characteristics of Saccharomyces cerevisiae strain 63M after continuous fermentation with cell recycling in a system of five reactors in series. Steady state was attained at 30 degrees C, and then the temperature of the system was raised so it ranged from 35 degrees C in the last reactor to 43 degrees C in the first reactor or feeding reactor with a 2 degrees C difference between reactors. After 15 days at steady state, the temperature was raised from 37 degrees C to 45 degrees C for 25 days at steady state, then from 39 degrees C to 47 degrees C for 20 days at steady state. Starter strain 63M was a hybrid strain constructed to have a MAT a/alpha, LYS/lys, URA/ura genotype. This hybrid yeast showed vigorous growth on plates at 40 degrees C, weak growth at 41 degrees C, positive assimilation of melibiose, positive fermentation of galactose, raffinose and sucrose. of 156 isolates obtained from this system at the end of the fermentation process, only 17.3% showed the same characteristics as starter strain 63M. Alterations in mating type reaction and in utilization of raffinose, melibiose, and sucrose were identified. Only 1.9% of the isolates lost the ability to grow at 40 degrees C. Isolates showing requirements for lysine and uracil were also obtained. In addition, cell survival was observed at 39-47 degrees C, but no isolates showing growth above 41 degrees C were obtained.

Curtobacterium wilt has become an important disease of beans in several localities in the country. Its causal agent, Curtobacterium flaccumfacciens pv. flaccumfaciens (Cff), survives and is disseminated through seeds. To date, few studies have been conducted with the objective of developing an effective and low-cost culture medium to isolate this bacterium from bean seeds, for health analysis purposes. Usually, the culture media employed for coryneform bacteria isolation contain specific carbon sources and antimicrobial products not available in the Brazilian market. A culture medium known as MSCFF was developed (peptone - 5 g, meat extract - 3 g, sucrose - 5 g, agar 15 g, skim milk powder* - 5 g. Congo red* - 0.05 g-, chlorothalonil* - 0.01 g, thiophanate methyl* - 0.01 g, nalidixic acid* - 0.01 g, nitrofurantoin* - 0.01 g. oxacillin* 0.001 g, sodium azide* - 0.001 g and distilled water q.s. 1L; *added after autoclaving the basal medium), which has the ability to inhibit growth of a large amount of saprophytic bacteria, but with low supressivity to Cff isolates. The MSCFF medium was highly effective for Cff isolation from naturally infected bean seeds and could be used for routine detection of this bacterium in bean seeds.

This work describes fructose oligosaccharide (FOS) production by the immobilized mycelia (IM) of a strain of Aspergillus japonicus, isolated from soil. The microorganism was inoculated into 50 mi of medium composed of sugar cane molasses (5.0% of total sugars); yeast powder; 2.0%; K2HPO4, 0.5%; NaNO3, 0.2%; MgSO4. 7H(2)O, 0.05%; KCl, 0.05%, final pH 5.0, and the flasks were agitated in an orbital shaker at 200 rpm for 60 h, at 30 degrees C. The beta-fructofuranosidase activity (Uf), transfructosylating activity (Ut), hydrolyzing activity (Uh), and FOS production were analyzed by high performance liquid chromatography. FOS production was performed in a batch process in a 2-l jar fermenter by IM in calcium alginate beads. The optimum pH and temperature were 5.0-5.6 and 55 degrees C, respectively No loss of activity was observed when the mycelium was maintaned at 60 degrees C for 60 min. Maximum production was obtained using 5.75% (cellular weight/volume) of mycelia (122.4 Ut g(-1)) and 65% sucrose solution (w:v) for 4 h of reaction when the final product reached 61.28% of fetal FOS containing GF(2) (30.56%), GF(3) (26.45%), GF(4) (4.27%), sucrose (9.6%) and glucose (29.10%). In the assay conditions, 23 batches were performed without loss of activity of the IM, showing that the microorganism and the process utilized have potential for industrial applications. The effect of intraperitoneal injection of clonidine (9-72 mu g/kg) on need-free 1.5% NaCl intake and on performance (defined as percent of a complete trial) in the rotarod test, was studied in normovolemic adult male rats. Clonidine (18 and 36 mu g/kg) inhibited the 1.5% NaCl intake in a 2-h test at doses that did not alter the performance in the rotarod test. The dose of 36 mu g/kg did not inhibit 10% sucrose intake. Only the highest dose (72 mu g/kg) of clonidine inhibited the 1.5% NaCl intake and the performance in the rotarod test, and produced signs of sedation. Sedation was determined either by change in posture (immobility or lack of postural tonus) of the animals during the ingestive test or by their performance in the rotarod test. The results suggest that sedation is not a determinant effect on the inhibition of 1.5% NaCl intake induced by clonidine. (C) 1999 Elsevier B.V.

Different concentrations of sucrose (3-25% w/v) and peptone (2-5% w/v) were studied in the formulation of media during the cultivation of Aspergillus japonicus-FCL 119T and Aspergillus niger ATCC 20611. Moreover, cane molasses (3.5-17.5% w/v total sugar) and yeast powder (1.5-5% w/v) were used as alternative nutrients for both strains' cultivation. These media were formulated for analysis of cellular growth, P-Fructosyltransferase and Fructooligosaccharides (FOS) production. Transfructosylating activity (U-t) and FOS production were analyzed by HPLC. The highest enzyme production by both the strains was 3% (w/v) sucrose and 3% (w/v) peptone, or 3.5% (w/v) total sugars present in cane molasses and 1.5% (w/v) yeast powder. Cane molasses and yeast powder were as good as sucrose and peptone in the enzyme and FOS (around 60% w/w) production by studied strains.

The apparent diffusion coefficients for sucrose, NaCl and water during osmotic dehydration of tomatoes in ternary solutions were determined. Long time experiments (up to 60 h) were carried out in order to determine equilibrium concentrations inside tomatoes, whereas short time experiments (up to 4 h) were performed to provide detailed information on kinetics of water loss and solids gain at the beginning of osmotic treatment. The mass transfer rates for water and solutes showed to be dependent of NaCl and sucrose concentrations in osmotic solution and simple regression models as functions of solutes concentration were determined for diffusion coefficients. Salt and sucrose diffusivities showed to be interdependent, with increasing NaCl concentration causing the enhancement of water loss, at the same time that higher sucrose contents hindered the excessive salt penetration. THE INFLUENCE OF CHEMICAL PLAQUE CONTROL, using topically applied 0.12% chlorhexidine, on the severity of cyclosporin A (CsA)-induced gingival overgrowth (GO) was evaluated. Forty Holtzman rats were divided into four groups: 1) control; 2) cyclosporin A: a 10mg/kg/day subcutaneous dose of CsA; 3) chlorhexidine: 0.12% chlorhexidine (CHX) was applied to the buccal surface of the right mandibular molars; and 4) cyclosporin A/chlorhexidine: a combination of the treatment described for cyclosporin A and chlorhexidine groups. The animals were fed a high sucrose diet during the experiment and were sacrificed after 14 and 21 days. The histometric analysis revealed a significant increase in buccal gingival area in the cyclosporin A group compared to other groups (P < 0.01) after 21 days. The epithelium thickness of the buccal gingiva was significantly increased in the cyclosporin A group, compared to the control group (P < 0.05). The cyclosporin A/chlorhexidine group exhibited statistically significantly lower gingival overgrowth than the cyclosporin A group. These findings, if replicated in human studies, suggest that topically applied 0.12% chlorhexidine may be a valuable measure in the management of cyclosporin-induced gingival overgrowth.

Glass transition temperatures of freeze-dried tomato conditioned at various water activities at 25 C were determined by differential scanning calorimetry (DSC). Air-dried tomato with and without osmotic pre-treatment in sucrose/NaCl solutions was also analyzed. Thermograms corresponding to the low water activity domain (0.11 less than or equal to a(w) less than or equal to 0.75) revealed the existence of two glass transitions, which were attributed to separated phases formed by sugars and water and other natural macromolecules present in the vegetable. Both transitions were plasticized by water and experimental data could be well correlated by the Gordon-Taylor equation in the low-temperature domain, and by the Kwei model in the high-temperature domain. For higher water activities, the low-temperature glass transition curve exhibited a discontinuity, with suddenly increased glass transition temperatures approaching a constant value that corresponds to the T-g of the maximally freeze-concentrated amorphous matrix. The unfreezable water content was determined through the melting enthalpy dependence on the moisture content. (C) 2002 Elsevier B.V. Ltd. All rights reserved.

The effect of deacetylated xanthan gum, additives (sucrose, soybean oil, sodium phosphate and propylene glycol) and pH modifications on mechanical properties, hydrophilicity and water activity of cassava starch-xanthan gum films has been studied. Sucrose addition resulted in the highest effect observed on cassava starch films elongation at break. The deacetylated xanthan gum had higher effect on elongation at break when comparing to the acetylated gum, although both gums presented an inferior effect in relation to the obtained with sucrose. However, when comparing to the control and PVC films, lower tensile strength resistance values were observed when adding sucrose. Increased water activity was observed for films added with sucrose, thus, increasing the material biodegradation. Sucrose and deacetylated xanthan gum addition resulted in a slight hydrophilicity increase. (C) 2004 Elsevier Ltd. All rights reserved.

Cephalosporin C production process optimization was studied based on four experiments carried out in an agitated and aerated tank fermenter operated as a fed-batch reactor. The microorganism Cephalosporium acremonium ATCC 48272 (C-10) was cultivated in a synthetic medium containing glucose as major carbon and energy source. The additional medium contained a hydrolyzed sucrose solution as the main carbon and energy source and it was added after the glucose depletion. By manipulating the supplementary feed rate, it was possible to increase antibiotic production. A mathematical model to represent the fed-batch production process was developed. It was observed that the model was applicable under different operation conditions, showing that optimization studies can be made based on this model. (C) 1999 Elsevier B.V. Ltd. All rights reserved.
