Background: Uterine Leiomyomas (ULs) are the most common benign tumours affecting women of reproductive age. ULs represent a major problem in public health, as they are the main indication for hysterectomy. Approximately 40-50% of ULs have non-random cytogenetic abnormalities, and half of ULs may have copy number alterations (CNAs). Gene expression microarrays studies have demonstrated that cell proliferation genes act in response to growth factors and steroids. However, only a few genes mapping to CNAs regions were found to be associated with ULs. Methodology: We applied an integrative analysis using genomic and transcriptomic data to identify the pathways and molecular markers associated with ULs. Fifty-one fresh frozen specimens were evaluated by array CGH (JISTIC) and gene expression microarrays (SAM). The CONEXIC algorithm was applied to integrate the data. Principal Findings: The integrated analysis identified the top 30 significant genes (P<0.01), which comprised genes associated with cancer, whereas the protein-protein interaction analysis indicated a strong association between FANCA and BRCA1. Functional in silico analysis revealed target molecules for drugs involved in cell proliferation, including FGFR1 and IGFBP5. Transcriptional and protein analyses showed that FGFR1 (P = 0.006 and P<0.01, respectively) and IGFBP5 (P = 0.0002 and P = 0.006, respectively) were up-regulated in the tumours when compared with the adjacent normal myometrium. Conclusions: The integrative genomic and transcriptomic approach indicated that FGFR1 and IGFBP5 amplification, as well as the consequent up-regulation of the protein products, plays an important role in the aetiology of ULs and thus provides data for potential drug therapies development to target genes associated with cellular proliferation in ULs. © 2013 Cirilo et al.

Objectives: The objective of this study was to apply low-level laser therapy (LLLT) to accelerate the recovery process of a child patient with Bell's palsy (BP). Design: This was a prospective study. Subject: The subject was a three-year-old boy with a sudden onset of facial asymmetry due to an unknown cause. Materials and methods: The low-level laser source used was a gallium aluminum arsenide semiconductor diode laser device (660 nm and 780 nm). No steroids or other medications were given to the child. The laser beam with a 0.04-cm2 spot area, and an aperture with approximately 1-mm diameter, was applied in a continuous emission mode in direct contact with the facial area. The duration of a laser session was between 15 and 30 minutes, depending on the chosen points and the area being treated. Light was applied 10 seconds per point on a maximum number of 80 points, when the entire affected (right) side of the face was irradiated, based on the small laser beam spot size. According to the acupuncture literature, this treatment could also be carried out using 10-20 Chinese acupuncture points, located unilaterally on the face. In this case study, more points were used because the entire affected side of the face (a large area) was irradiated instead of using acupuncture points. Outcome measures: The House-Brackmann grading system was used to monitor the evolution of facial nerve motor function. Photographs were taken after every session, always using the same camera and the same magnitude. The three-year-old boy recovered completely from BP after 11 sessions of LLLT. There were 4 sessions a week for the first 2 weeks, and the total treatment time was 3 weeks. Results: The result of this study was the improvement of facial movement and facial symmetry, with complete reestablishment to normality. Conclusions: LLLT may be an alternative to speed up facial normality in pediatric BP. © Copyright 2013, Mary Ann Liebert, Inc. 2013.

The possible benefits of some bioactive flavones and xanthones present in plants of the genus Syngonanthus prompted us to screen them for estrogenic activity. However, scientific research has shown that such substances may have undesirable properties, such as mutagenicity, carcinogenicity and toxicity, which restrict their use as therapeutic agents. Hence, the aim of this study was to assess the estrogenicity and mutagenic and antimutagenic properties. We used recombinant yeast assay (RYA), with the strain BY4741 of Saccharomyces cerevisiae, and Ames test, with strains TA100, TA98, TA97a and TA102 of Salmonella typhimirium, to evaluate estrogenicity, mutagenicity and antimutagenicity of methanolic extracts of Syngonanthus dealbatus (S.d.), Syngonanthus macrolepsis (S.m.), Syngonanthus nitens (S.n.) and Syngonanthus suberosus (S.s.), and of 9 compounds isolated from them (1 = luteolin, 2 = mix of A-1,3,6-trihydroxy-2-methoxyxanthone and B-1,3,6-trihydroxy-2,5- dimethoxyxanthone, 3 = 1,5,7-trihydroxy-3,6-dimethoxyxanthone, 4 = 1,3,6,8-tetrahydroxy-2,5-dimethoxyxanthone, 5 = 1,3,6,8-tetrahydroxy-5- methoxyxanthone, 6 = 7-methoxyluteolin-8-C-β-glucopyranoside, 7 = 7-methoxyluteolin-6-C-β-glucopyranoside, 8 = 7,3′-dimethoxyluteolin- 6-C-β-glucopyranoside and 9 = 6-hydroxyluteolin). The results indicated the estrogenic potential of the S. nitens methanol extract and four of its isolated xanthones, which exhibited, respectively, 14.74 ± 1.63 nM; 19.54 ± 6.61; 7.20 ± 0.37; 6.71 ± 1.02 e 10.01 ± 4.26 nM of estradiol-equivalents (EEQ). None of the extracts or isolated compounds showed mutagenicity in any of the test strains and all of them showed antimutagenic potential, in particular preventing mutations caused by aflatoxin B1 (AFB1) and benzo[a]pyrene (B[a]P). The results show that the xanthones, only isolated from the methanol extract of S. nitens capitula, probably were the responsible for its estrogenic activity and could be useful as phytoestrogens, providing a new opportunity to develop hormonal agents. In addition, flavones and xanthones could also be used as a new antimutagenic agent. Since, the mutagens are involved in the initiation and promotion of several human diseases, including cancer, the significance of novel bioactive phytocompounds in counteracting these pro-mutagenic and carcinogenic effects is now gaining credence. © 2013 Elsevier Inc. This study provides histochemical data of the hepatopancreatic cells of adult female pink-shrimp (Farfantepenaeus brasiliensis) at two different developmental stages (those with developed gonads and those with exhausted gonads). The F. brasiliensis females were collected in seawater off the Guarapari coast, Espirito Santo, Brazil. Five cell types were identified in this digestive gland: B (vesicular), E (embryonic), F (fibrillar), M (basal) and R (resorptive). The digestive gland was stained with the following techniques: PAS/Alcian blue (for polysaccharides), bromophenol blue (for protein), von Kossa (for bound calcium) and Baker (for lipids). Acid glycoconjugates were found inside vacuoles in the R cells, while neutral polysaccharides were present in the B cells and near to the microvilli. In females with exhausted gonads polysaccharides were also seen in the intertubular spaces and inside the lumina of the tubules. The F and M cells were the most marked by the presence of large amounts of proteins observed in R cells and also inside the vacuoles of B cells. The bound calcium was mainly found in the F and M cells. The F cells showed strong positive staining for lipid while the R cell only stained weakly. The E cells did not react to any of the applied staining techniques. The similarities in the histochemical composition of these hepatopancreatic cells in females with developed gonads, compared to exhausted ones, is justified by the fact that transfer of these elements to the oocytes occurs, in significant quantity, only during the initial stages of gonadal development in F. brasiliensis. Also, they may be more related to the molt stage, as in the case of calcium salts. © 2013 Elsevier GmbH. All rights reserved.

The reproductive biology of a species includes factors beyond its sexual maturity, fecundity and reproductive period, and may extend to the differential distribution of individuals. The reproductive dynamics of the blue crab Callinectes ornatus was investigated through monthly collections over the course of 2 years in three bays on the southeastern coast of Brazil. For each bay, six transects were established, four of them parallel to the beach line (at depths of 5, 10, 15, and 20 m), one transect exposed to wave action, and another sheltered from waves. Females and males were classified according to the gonadal maturation stage, and were grouped as individuals with reproductive potential (mature gonads or breeding females) or not (rudimentary gonads or in development). Analyses using ordination techniques (PCA) and gradient analysis (CCA) showed that 82.13 % of environmental variations were explained by the transect arrangement, and these characteristics explained 86.70 % of the differential distribution of female crabs and 96.57 % of the distribution of males. These results indicate that females with reproductive potential were more abundant in deeper regions, while females with rudimentary or developed gonads were abundant in shallower habitats and areas sheltered from wave action. Thus, the distribution of C. ornatus in these bays was linked to their reproductive state, as part of the reproductive strategy of the population. © 2013 Senckenberg Gesellschaft für Naturforschung and Springer-Verlag Berlin Heidelberg.

Neural plasticity has been observed in the bed nucleus of the stria terminalis (BNST) following exposure to both cocaine and androgenic-anabolic steroids. Here we investigated the involvement of the BNST on changes in cardiovascular function and baroreflex activity following either single or combined administration of cocaine and testosterone for 10 consecutive days in rats. Single administration of testosterone increased values of arterial pressure, evoked rest bradycardia and reduced baroreflex-mediated bradycardia. These effects of testosterone were not affected by BNST inactivation caused by local bilateral microinjections of the nonselective synaptic blocker CoCl2. The single administration of cocaine as well as the combined treatment with testosterone and cocaine increased both bradycardiac and tachycardiac responses of the baroreflex. Cocaine-evoked baroreflex changes were totally reversed after BNST inactivation. However, BNST inhibition in animals subjected to combined treatment with cocaine and testosterone reversed only the increase in reflex tachycardia, whereas facilitation of reflex bradycardia was not affected by local BNST treatment with CoCl2. In conclusion, the present study provides the first direct evidence that the BNST play a role in cardiovascular changes associated with drug abuse. Our findings suggest that alterations in cardiovascular function following subchronic exposure to cocaine are mediated by neural plasticity in the BNST. The single treatment with cocaine and the combined administration of testosterone and cocaine had similar effects on baroreflex activity, however the association with testosterone inhibited cocaine-induced changes in the BNST control of reflex bradycardia. Testosterone-induced cardiovascular changes seem to be independent of the BNST. © 2013 IBRO.
