Sibutramine is a drug recommended clinically for the treatment of obesity, but there are women that use the drug for maintenance of body weight. Many time this use occur associate to habit of tabagism, being the nicotine the main toxic compound in the cigarette. The goal of study was to evaluate the side effects promoted by sibutramine, associated or not to nicotine, in the reproductive tissue of adult female rats. Wistar animals (n = 30), were distributed in the groups: a) Control A (0.3 mL of distilled water; oral); b) Sibutramine (15 mg/kg of body weight; oral); c) Control B (0.3 mL of saline solution; intraperitoneal); d) Nicotine (4.0 mg/kg of body weight; intraperitoneal); e) sibutramine + nicotine. The treatments were conducted during 30 consecutive days (single dose, daily). Sibutramine, associated or not to nicotine, affected the folliculogenesis and luteogenesis. There were significant alterations (p<0.05) in the thickness of uterine layers, considerate each treatment. In conclusion, the administration isolated of sibutramine or nicotine promoted deleterious effects in the reproductive tissues of female rats and these effects were potential in the group that received both drugs.

Considering that alcohol has been identified, in the literature, as an evident risk factor for osteoporosis development, inducing loss of bone mass, the aim of this study was to evaluate the effect of the alcohol on the femur of male and female rats. For this purpose, 35 male and 35 female rats, divided into seven groups of five animals per genera were included. The groups were: control group that received food and water as much as they wished; alcohol group in the concentrations of 10, 20 and 30% and the isocalorie: groups corresponding to the concentration of alcohol. After 8 weeks, the animals were sacrificed and evaluated regarding to the body weight and nutritional conditions. Unilateral femurs were removed and analyzed in relation to the length, width, and thickness of the bone cortical, trabeculae percentage and optical density. Data obtained were analyzed by ANOVA (Tukey; p<0.05). Regarding to the weight, loss of weight was observed in the animals of the group alcohol 20% among females and 30% among the males. In relation to the nutritional conditions, the females with the dosages of 10, 20 and 30% of alcohol swallowed a mean of 25, 39 and 53% of Kcall and the males 22, 36 and 45 Kcal Jday coming from the alcohol. Alcohol reduced the length of the femur only in the group MG6. Mesio-distal width was higher in the group FG6 although the antero-posterior width was not modified. The cortical thickness among males and females was not altered however the percentage of trabeculae bone was reduced among females alcohol 30% and among males with the three alcoholic concentrations. Optical density was reduced in the concentration of 30% among the males and females. Under the experimental conditions, it could be concluded that the effect of alcohol was more evident among the males and in the trabeculae bone, and that the alcoholic concentration of 30% was the most harmful for the bone tissue, ....

Experimental models are necessary to elucidate pathophysiological mechanisms not yet understood in humans. To evaluate the repercussions of the diabetes, considering two methodologies, on the pregnancy of Wistar rats and on the development of their offspring. In the 1st induction, female offspring were distributed into two experimental groups: Group streptozotocin (STZ, n=67): received the β-cytotoxic agent (100mg STZ/kg body weight - sc) on the 1st day of the life; and Non-diabetic Group (ND, n=14): received the vehicle in a similar time period. In the adult life, the animals were mated. After a positive diagnosis of pregnancy (0), female rats from group STZ presenting with lower glycemia than 120 mg/dL received more 20 mg STZ/kg (ip) at day 7 of pregnancy (2nd induction). The female rats with glycemia higher than 120mg/dL were discarded because they reproduced results already found in the literature. In the mornings of days 0, 7, 14 and 21 of the pregnancy glycemia was determined. At day 21 of pregnancy (at term), the female rats were anesthetized and killed for maternal reproductive performance and fetal development analysis. The data were analyzed using Student-Newman-Keuls, Chi-square and Zero-inflated Poisson (ZIP) Tests (p<0.05). STZ rats presented with increased rates of pre (STZ=22.0%; ND=5.1%) and post-implantation losses (STZ=26.1%; ND=5.7%), reduced rates of fetuses with appropriate weight for gestational age (STZ=66%; ND=93%) and reduced degree of development (ossification sites). Conclusion: Mild diabetes led a negative impact on maternal reproductive performance and caused intrauterine growth restriction and impaired fetal development

Plumbism is considered the oldest occupational disease. Among the pathophysiological effects associated with lead (Pb) are cardiovascular disorders. Many diseases that develop later in life are determined during the early stages of life, under the influence of exposure and preferred diet of the mother. Still, one should consider that many environmental contaminants at levels not harmful can determine pathophysiological processes if physical or chemical stressors and/or pathological conditions are present. In this context, the intrauterine malnutrition may represent an additional risk factor in exposure to Pb during pregnancy and lactation. For these reasons, the objective of this study was to evaluate the cardiovascular risk of weaned rats that have suffered perinatal exposure to Pb and intrauterine malnutrition, alone or in combination. After mating, female rats were divided into control (ctrl, ad libitum), food restriction (RA, the same diet 50% of consumption in the control group during pregnancy), exposed to Pb (500 ppm Pb in drinking water during pregnancy and lactation) and association (As, received the last two procedures in combination). Cumulative concentration-effect curves (CECs) to CaCl2 and noradrenaline (NA) were obtained in rings with and without endothelium of the same thoracic aorta from male weaned rats (23-25 days old). Maternal weight, litter weight, weight and number of pups at birth, anogenital distance, arterial blood pressure (ABP) and weight of tissues (kidney, liver, aorta, left ventricle) were evaluated. Changes in vascular reactivity were assessed by the maximum response (MR) and 50% effective concentration (EC50). Data were presented as mean ± SEM. Statistical analysis was performed by multifactorial analysis of variance and Tukey's post test. Maternal smoking during pregnancy causes heart changes, impaired fetal growth, spontaneous abortion and low birth weight. Even in pregnant women exposed to cigarette smoke these changes can be observed. But the exercise is to answer the increased fetal weight, besides the improvement of cardiorespiratory fitness of the pregnant and the fetus. The objective of this study was to analyze the ventricular muscle of young rats subjected to passive smoking associated with aquatic exercise. For the experiments used 24 female rats were divided into: GC (control), GE (exercise protocol in the water), GF(exposed to cigarette smoke) and GEF (exercise protocol on water and exposed to cigarette smoke). Several studies use only male rats in their experiments, even with all the evidence of differences between the sexes, not only in the reproductive behavior but also in the development of diseases. This absence of studies using female rats is due in part to the estrous cycle. It is known that one estrous cycle lasts four to 5 days and consists of four phases: proestrus, estrus, metestrus and diestrus. Ramos et al. (1999) identified and mapped a QTL in an F2 population derived from interbreeding of strains of SHR (spontaneously hypertensive rats) and Lewis (LEW) rats and found, on chromosome 4, the first QTL of the world related to anxiety in rats. After several backcrosses between SHR and LEW strains, it was developed a congenic strain called SLA16, to refine the study of this QTL. This QTL corresponds to a large genomic region, approximately 75 million basepairs (Mb), and was first called Ofil1 (inner open field locomotion 1). Currently it is called Anxrr16 (anxiety-related response region 16) according to the rat genome database (RGD). Izidio et al. (2011) suggested that the natural fluctuation of the hormones in the estrous cycle of female rats could influence the effects of this locus on behavior. That is, females in DP (diestrus-proestrus) present the QTL in the same genomic region that males, while females in EM (estrus-metestrus) do not present it. The effects of the estrous cycle in the SLA16 strain and its isogenic control SHR were analyzed on the elevated plus maze (EPM), black and white box and on the open field test (OF). In the open field test, the SLA16 strain had higher central locomotion (F=5.0, p <0.05) and peripheral locomotion (F=12.6, p <0.01) compared to the SHR strain. In the elevated plus maze test, the SLA16 strain had the higher number of entries (F=9.5, p <0.01) and time spent in the open arms (F=10.3, p <0.01) compared to the SHR strain. Purpose: The objective of this article is to describe a method to construct an intraoralacrylic device that permits a reline material to be added to the inner surface of thepalatal plate.Materials and Methods: Fifteen 60-day-old adult female rats (Rattus NorvegicusAlbinus Wistar), weighing 150 to 250 g were used for this study and allocated to threegroups (n = 5): G1, animals wearing a heat-polymerized acrylic resin palatal plate(Lucitone 550) for 14 days; G2, animals wearing a heat-polymerized acrylic resinpalatal plate (Lucitone 550) relined with Tokuyama Rebase II for 14 days; and G3,animals maintained under the same conditions as the experimental groups, withoutwearing palatal plates for 14 days. The manipulation of the animals followed theguidelines of the Brazilian College of Animal Experimentation, under the approval ofthe animal ethics committee of the State University of Ponta Grossa. The palatal platescovered the whole palate, were fixed in the molar region with light-cured resin, andwere kept there for 14 days. The animals received a paste diet and water ad libitum.Before and after the trial period, the rats were weighed individually on a precisionscale. Statistical analysis was performed using a two-way analysis of variance (α =0.05) test for comparison of the animals’ weight (g) at time 0 and after 14 days ofusing the palatal plate.Results: No statistical differences were observed regarding the weight of the animalsamong the experimental groups in the study.Conclusions: The individual master impressions, the molar teeth coverage, and themethod of cementation with nonadhesive composite resin provided good stability forthe palatal plate showed in this study, not disturbing the eating habits and nutritionof the animals. This model seems reproducible, offering adequate histopathologicalevaluation. Differences in tissue morphology exist between the animals that used thepalatal plate and the animals that did not use this device. Use of these palatal platescould clarify how prostheses bring changes in the palatal mucosa of users.
