Análise da expressão do gene CaPox que codifica uma peroxidase de classe III envolvida na interação do cafeeiro com nematóides

A caracterização e isolamento do gene com expressão específica em raiz de café (Coffea arabica) que codifica uma peroxidase (CaPOX) e suas respectivas regiões promotoras, permitiu realizar a caracterização da expressão desse gene em reposta a estresse biótico (infecção por nematóides) assim como a análise funcional do seu promotor. Promotores tecido-específicos responsáveis pela regulação de genes responsivos a estresses bióticos tornam-se fundamentais em programas biotecnológicos que visam o aumento da resistência e tolerância vegetal. Partindo desse princípio, realizou-se a quantificação da expressão relativa do gene CaPOX em raízes de café utilizando plantas de Coffea arabica de um cultivar susceptível (Mundo Novo) e de outro cultivar resistente (IAC 388-17-1) a nematóides, respectivamente. Em paralelo utilizou- se plantas transgênicas de tabaco (Nicotiana tabacum SR1) contendo a versão completa do promotor do gene CaPOX em fusão transcricional ao gene repórter uidA (que codifica a β-glucuronidase; GUS) . A partir disso, pode-se observar que o gene CaPOX tem sua expressão aumentada em resposta a infecção por nematóides, sendo que a indução observada ocorre nos tempos iniciais pós-inoculação. Da mesma maneira, o promotor do gene CaPOX é responsivo a infecção por nematóides, sendo ativado nos tempos iniciais pós-inoculação

Genotipagem de HLA de classe I e II para seleção de células precursoras dendríticas para estudo da imunobiologia do vírus associado ao Sarcoma de Kaposi (KSHV)

The Kaposi-associated Herpesvirus (KSHV) also known as Human Herpesvirus 8 (HHV-8) is associated with the development of Kaposi’s sarcoma (KS) and others limphoprolipheratives diseases such as Primary Effusion Lymphoma (PEL) and Multicentric Castleman Disease (MCD). Even though the virus is considered lymphotropic, it is able to infect others cell types such as macrophages, dendritic cells, endothelial cells, monocytes and fibroblasts. After infection, KSHV be latent expressing essential viral genes to its maintenance in a infected cell. However, in some circumstances may occur the reactivation of lytic cycle producing new viral particles. K1 protein of KSHV interferes in the cellular signaling inducing proliferation and supporting cellular transformation. K1 is encoded by viral ORF-K1, which shows high variability between different genotypes of KSHV. So far, it is not clear whether different isoforms of K1 have specific immunobiological features. The KSHV latency is maintained under strict control by the immune system supported by an adequate antigen presentation involving Human Leucocyte Antigen (HLA) class I and II. Polymorphisms of HLA class I and II genes confer an enormous variability in molecules that recognize a large amount of antigens, but also can increase the susceptibility to autoimmune diseases. Therefore, the present study aims to genotype HLA class I (A and B) and class II (DR and DQ) from volunteers to identify haplotypes that can provide better response to K1 epitopes of different KSHV genotypes. First of all, 20 volunteers were selected to genotype HLA genes. In our results we observed prevalence of certain HLA class I haplotypes as HLAA1, HLA-A2, HLA-A24, HLA-A26, HLA-B8, HLA-B18 e HLA-B44. O conto de Rubens Figueiredo em As palavras secretas: conto contemporâneo

We work with the storybook, As Palavras Secretas, written by carioca Rubens Figueiredo. We treat, then, the Brazilian contemporary tale. For we study the writer’s fiction, first we met a critical bibliography about the narrative and a set of diverse texts: reports, articles, press releases, interviews and essays about the author’s work. We discourse characteristics and the compositional elements of each story and noticed that they are full of symbolism and various elements that they are recurrent. Some of these narratives deal with fragmentation of the man who, indeed, is a very striking feature in the actual Brazilian tale. We meet, then, with characters who feel the need to simulate an identity and that, therefore, coexist with a strong inner conflict. Three of the tales include the unlikelihood of that work, and present, therefore, elements that escape from reality. Finally, Rubens Figueiredo, in As Palavras Secretas, presented us narratives rather elongated, unlike what was ordinary in the category of the tale, which it usually consists of a short extension narrative

Estudo do efeito da classe de furação na resistência à fadiga em junta alumínio – fibra de carbono

This work performs a comparative study of fatigue life of riveted lap joints involving classes of drilling which adjustment is made with interference or clearance. For this study, representative specimens of this joints were manufactured with four rivets distributed in two rows. In this context, are presented the test matrix, the methodology employed in performing of the tests, the used mathematical modeling, and that methods that are the basis for the latter are described through the theoretical foundation. Next, are present the results obtained in fatigue tests and images of the region of failure of the specimens. Finally, are present some comments and conclusions related to the results obtained

Sistema de recomendação de produtos online baseado em palavras-chave

This project aims to explore the many methods used for the development of recommendation systems to user ’ s items and apply the content - based recommendation method on a prototype system whose purpose is to recommend books to users. This paper exposes the most popular methods for creating systems capable of providing items (products) according to user preferences, such as collaborat ive filtering and content - based. It also point different techniques that can be applied to calculate the similarity between two entities, for items or users, as the Pearson ’s method, calculating the cosine of vectors and more recently, a proposal to use a Bayesian system under a Dirichlet distribution. In addition, this work has the purpose to go through various points on the design of an online application, or a website, dealing not only oriented algorithms issues, but also the definition of development to ols and techniques to improve the user’s experience. The tools used for the development of the page are listed, and a topic about web design is also discussed in order to emphasize the importance of the layout of the application. At the end, some examples of recommender systems are presented for curiosity , learning and research purposes

Projeto elétrico básico de uma subestação industrial típica na classe 15 kV

The electrical substations have the goal to transmit, distribute and change the characteristics of electrical energy transmission networks of dealers to the end user with a high rate of reliability and continuously. For this it’s necessary to accomplish a gathering of datas from the local dealers where will be installed the substation to make a good electric project. This work has the objective to gather all the contents relatives to electric project of substation that are scattered in the literature, in order to organize and develop a basic guide that presents a methodology to dimension electrical equipments that belong to an industrial substation at 15 kV. In addition to elaborate a basic guide, this work aids to specify electrical equipments installed in the substation, bringing information and comparisons to know and determinate the kind of equipments that will be used, in a correct and coherent way with the national and international rules. Ultimately, after determined, got and specified all the equipments that belongs to an industrial substation, the work mention in global manner how to calculate the dimension of substation physical arrangement, determining all the least and required dimensions of each cubicle

Hipersegmentação de palavras: análise de aspectos prosódicos e discursivos

This paper deals with hypersegmentation of words that are characterized by the unconventional employment of a graphical boundary (using a white space or hyphen) within the limits of the word, as in "em bora" (“although”, using a white space), and "chama-da" (“called”, using the hyphen). In a study conducted on these data, we identified motivations arising not only from their literate nature but also from the morphosyntactic and prosodic information. We showed that there are linguistic features recurrent in these registers of word boundaries, based on the analysis of texts produced by the students who attended the last four years of elementary school in a public school in São Paulo. In this paper, we advance on this study by selecting data whose characteristics do not match those which were recurrent. We will argue that the unconventional presence of boundary within the written word limits may be interpreted as representing prosodic configurations (of intonation and rhythm nature) which contribute to the construction of the relation of meanings in the text.

No poema Em silêncio descobri essa cidade no mapa, inserido em A máquina lírica (1964), Herberto Helder desenha um contra mapa desnorteante com as palavras, que pulalam na travessia da escrita, deslocando-se incessantemente e ocupando diferentes posições nos versos. A proliferação dos vocábulos que se cruzam em novas combinações dão origem aos fios cambiantes que se dobram, redobram e desdobram-se no tecido da linguagem que objetivamos desfiar neste trabalho.

A maldição das palavras e o sujeito dilacerado de Marcelo Mirisola (2009)

There is an area in the Brazilian Contemporary Literature which is articulated according to the first person narrator who presents the problematic of the own creational action, the expression of a conflictual subjectivity divided between the vertiginous dive into itself – what would imply in a renounce of the world and the reality, where the authors’ work are their only matter – and the social participation - not as an awareness, but as the recognition of the limits and impasses which the real representation imposes on the creative writing. Therefore, this article proposes a reading on the novel O Azul do Filho Morto (2002) by Marcelo Mirisola, to try to situate him into the recent Brazilian literary production, as well as to reflect on his narrative the complex notion of authorship which is called into question on his novels, since they produce an intrincate imbrication between the fictional subject and the empirical image of the writer himself, breaking and confusing the limits between the fictional discourse and the extralinguistic reality which is evoked, manipulated, distorted and made unstable on his representational process.

A segmentação não-convencional de palavras em textos do ciclo II do ensino fundamental

This paper deals with unconventional segmentations of words in texts produced by students of the last four years of elementary school. The main hypothesis is that these data allow us to observe the characteristics of written and spoken utterances. Through analysis of data on prosodic constituents, we argue that students deal with (conflicting) hypotheses on the organization of unstressed syllables into prosodic constituents: metric feet, prosodic word and clitic group. We found evidence that unconventional spellings have their main motivation in the difficulty of students to assign the status of written word to grammatical items that are prosodic clitics.
