A presente invenção se refere à síntese química e à aplicação como composto orgânico paramagnético do tipo marcador de spin, do produto denominado ácido 2,2,5,5-tetramentilpirrolidina-1-oxi-3-[n-(fluorenilmetiloxicarbonil)-amino-4-carboxílico, abrevias peptídicas ou de determinados sistemas. Possibilita-se, ainda, a ligação do composto apenas pelo seu radical poac, com a remoção do protetor fmoc, em meio básico orgânico, e, da mesma forma, quando se deseje prolongar a seqüência peptídica até o ponto desejado. Por conter um anel do tipo pirrolidina, a sua introdução no meio de uma cadeia peptídica irá provocar conformações diferenciadas das de <244>-aminoácidos comuns, sendo portanto um composto valioso para se estudar a relação estrutura-atividade biológica de diversos peptídeos de importância. Pelo mesmo processo de síntese viabiliza-se o uso de fmoc-poac na marcação do peptídeo angiotensina ii (poac^ 7^ -aii).

Background: The use of all by-products of bovine slaughter is of high economic importance for the industries of products of animal origin. Among these products, fat has an important role, once fat rendering may generate several different products, such as protein material that may be used in the manufacture of meat products. However, in spite of the importance that the use of all by-products has for the economic balance of the industry, there are no reports on their use in Brazil, or studies that supply data on microbiological and physical-chemical local standards for this protein. Thus, the objective of this study was to evaluate microbiological and physical-chemical characteristics of protein material obtained from fat rendering, as well as to provide support for companies to use fat rendering to generate protein material, adding value to industrialized meat products.Materials, Methods & Results: The experimental production of edible protein obtained of fat rendering was conducted in slaughterhouse with supervision of the Brazilian Ministry of Agriculture, Livestock and Food Supply. Protein material was obtained in a continuous, humid heat system at high temperatures. Fat scraps containing protein were ground and cooked at high temperature (85 degrees C), and placed in a three phase decanter centrifuge. After centrifugation, protein material was ground again and packed. Samples were collected from 15 batches of protein material, and the following microbiological analyses were carried out: counts of aerobic mesophilic and psychrotrophic microorganisms, coliforms at 35 degrees C, Escherichia coli, sulfite-reducing Clostridium, and Staphylococcus aureus, besides presence or absence of Salmonella and Listeria monocytogens. The following physical-chemical analyses were also carried out: protein, total lipid, moisture, ash, carbohydrate, and energy content. Mean counts of mesophiles, psychrotrophs, and coliforms at 35 degrees C were 4.17; 3.69 and 1.87 (log CFU/g), respectively. Levels of protein, total lipids, moisture, ashes and carbohydrates were 27.50; 7.83; 63.88%; 0.24%; and 0.55%, respectively, and energy content was 182.63 kcal/100g.Discussion: Results of microbiological analyses demonstrated that, although low, the final product showed to be contaminated. Contamination that occurred during the second grinding procedure may be an explanation for these bacterial counts. Also, the temperature used for fat fusion was not enough to eliminate thermoduric microorganisms. However, even with the presence of indicator microorganisms in the samples, none was contaminated by E. coli, sulfite-reducing Clostridium, S. aureus, Salmonella or L. monocytogenes. Physical-chemical analyses showed that the product had adequate nutritional quality. Based on these results, it was possible to conclude that protein material obtained in fat rendering showed characteristics that enable the use of this product as raw material for processed meat products. Besides, the present study was the first one to present scientific results in relation to edible by-products obtained in fat rendering, supplying important information for slaughterhouses and meat-processing plants. The study also produced relevant data on the innocuousness of the product, which may be used to guide decision-making of health inspectors.

A taurina (ácido 2-etanoaminossulfônico) apresenta diversas funções fisiológicas, como atuação no neurodesenvolvimento, ação no coração contra arritimias, ação nos músculos esqueléticos contra atrofia, faz parte da bile na forma de um composto conjugado, constutuindo um dos sais biliares, e anti-inflamatória, inibindo mediadores pró-inflamatórios. Neste projeto a ênfase foi voltada para a ação anti-oxidante da taurina, que forma um complexo com espécies como o ácido hipocloroso (HOCl) e ânions clorito (OCl-) que potencialmente possuem ações danosas à célula. Esse complexo, a taurina cloramina (Tau-Cl), possui também ações contra mediadores inflamatórios, como o fator de necrose tumoral alfa (TNF-α) e o óxido nítrico (NO). Por ser uma molécula com alta polaridade e hidrossolubilidade, a taurina não consegue penetrar na pele pelo extrato córneo intacto, a camada mais externa da pele, que constitui sua barreira contra perda ou absorção de água e eletrólitos. Foi feita então a síntese de um pró-fármaco derivado da taurina, formado por seu acoplamento com uma cadeia carbônica longa, aumentando significativamente a lipossolubilidade em relação a taurina pura, e podendo assim, essa molécula, transpor o extrato córneo intacto e exercer sua ação anti-oxidante, podendo ser, portanto, caracterizado como um pró-ativo. Este pró-fármaco, o ácido 2-palmitamidoetanossulfônico, foi sintetizado por meio de uma reação entre um cloreto de acila (cloreto de palmitoíla) e taurina, utilizando trietilamina como base, por meio de um método conhecido por Schotten-Baumann. O produto formado foi analisado por cromatografia em camada delgada (CCD), faixa de fusão, espectroscopia de absorção no infravermelho (IV) e espectometria de massa (EM), sendo que todas as análises confirmaram que a síntese foi bem sucedida

When a bolted joint is loaded in tension with dynamically, part of this load is absorbed by the bolt and rest is absorbed by the joint material. What determines the portion that is to absorbed by the bolt is the joint stiffness factor. This factor influences the tension which corresponds to pre-load and the safety factor for fatigue failure, thus being an important factor in the design of bolted joints. In this work, three methods of calculating the stiffness factor are compared through a spreadsheet in Excel software. The ratio of initial pre-load and the safety factor for fatigue failure depending on the stiffness factor graph is generated. The calculations for each method show results with a small difference. It is therefore recommended that each project case is analyzed, and depending on its conditions and the range of stiffness values, the more or less rigid method about the safety factor for fatigue failure is chosen. In general, the approximation method provides consistent results and can be easily calculated

This study aimed to evaluate the dynamic factor applied to the results obtained by dynamic resistance formulas Janbu and Hiley, would lead to the results obtained by static strength analysis CAPWAPC ®. The evaluation was done by backcalculation dynamic loading tests (ECD), taking into consideration the type of soil in which the cutting base precast concrete were settled, and using the information on the effective energy transmitted to the stakes by hammers, obtained by CAPWAPC ®. The results are shown in tables and graphs, and showed that the use of these formulations and their dynamic factors can become an efficient and economical field, assisting the engineer in making decisions regarding the staking of the work

This paper presents a brief review of theoretical studies on the stimulation and brain development. We also addressed questions about its plasticity, its influence on learning and behavior of the individual in interaction with the environment. Studies clearly show that the brain does not go into decline as we age. In this sense, a new discovery in neuroscience has revealed that the brain maintains the ability to grow and change the pattern of its connections to death. Findings such as these form the basis of neurobics. Demonstrating the importance of a relevant application of appropriate stimuli to change the morphology and function of neurons, the technique can be applied in different ways, ie, clinical, behavioral and neurological. Furthermore it can be applied during all phases of human development

The Brazilian livestock stands out for having the world largest commercial herd of cattle and leads meat exportation and production of bovine embryos. The in vitro production (IVP) of embryos is considered an effective option to overcome problems such as infertility in cows with high economic value and also for genetic improvement of cattle. The in vitro oocyte maturation is an essential step to the success of IVP, but is still considered poor when compared to in vivo maturation. Recent studies have suggeested an important role of Fibroblast Growth Factor 10 (FGF10) on the in vitro maturation of oocytes, which favored the expression of genes related to oocyte maturation and cumulus cell expansion. Aware that maturity stage influences the final production of blastocysts, we aimed study to verify if the addition of FGF10 into the maturation medium is able to affect positively the IVP of bovine embryos. Hence, FGF10 was added to maturation in five different concentrations: 0.5 ng/mL (group 0.5), 2.5 ng/mL (group 2.5), 5 ng/mL (group 5), 10 ng/mL (group 10) and 50 ng/mL (group 50). Additionally, two other maturation groups were used, group BSA (Bovine Serum Albumin, 4 mg/mL) and group FCS (Fetal Calf Serum, 10%). The rates of cleavage, morula and blastocyst were analyzed by Analysis of Variance (ANOVA), differences of P<0.05 were considered significant. Cleavage rates did not differ between the seven groups. On the other hand, morula rate on FCS group was higher than groups BSA, 0.5, 10 and 50 (P<0.05), but did not differ among groups treated with intermediate doses of FGF10 (2.5 and 5). FCS group presented higher blastocyst rate compared to all other groups that were well below the FCS group (P<0.0001). Therefore, the use of FGF10 during oocyte maturation did not affect positively embryo development on the IVP of bovine embryos

Preeclampsia is a specific disorder of pregnancy, characterized by arterial hypertension and proteinuria detected after 20 weeks of gestation. This pathology is associated with hyperuricemia, higher levels of pro-inflammatory cytokines, enhanced leukocyte activation and oxidative stress. Adenosine deaminase (ADA) is an enzyme present in all human tissues and, it is involved with the maturation of the immune system. Although its function is not fully understood, ADA is considered an indicator of cellular inflammation and, its increased serum concentration is observed in inflammatory diseases, such as tuberculosis and rheumatoid arthritis. This study aimed to assess serum ADA levels in preeclamptic patients (PE) compared with normotensive pregnant (NT) and non-pregnant women (NP), and to correlate these values with TNF-α and IL-1β production. Ninety pregnant women were included: 60 were pre-eclamptic and 30 were normotensive matched for gestational age. As control group 20 healthy non-pregnant women matched with pregnant for age were included. Peripheral blood mononuclear cells (PMMC) obtained from the three groups studied were cultured with or without lipopolysaccharide (LPS) for 18h at 37oC, and TNF-α and IL-1β production was assessed in the supernatant of cultures by enzyme immunoassay (ELISA). ADA plasmatic concentration was determined by colorimetric method. The results show that ADA plasma levels were significantly higher in PE group compared with NT and NP groups. A positive correlation between ADA and uric acid levels was detected in preeclamptic women. There was no significant difference in relation to ADA levels when PE patients were classified in early and late-onset PE. The endogenous production of IL-1β and TNF-α by PBMC was significantly higher in PE group than in NT and NP women, showing the activation state of these cells in PE.
