Na busca de novos recursos genéticos capazes de produzir enzimas celulolíticas a baixas/médias temperaturas, o continente Antártico vem demonstrando ser um ambiente bastante promissor. Neste contexto, o objetivo do presente trabalho foi avaliar a influência de diferentes fatores na produção de celulases por fungos filamentosos isolados de amostras da Antártica visando otimização do processo possível aplicação das mesmas na produção de etanol de segunda geração. Foram utilizados os fungos L1-1 e E5B da Central de Recursos Microbianos da UNESP (CRM-UNESP) os quais foram previamente selecionados devido ao potencial celulolítico. O delineamento experimental foi utilizado para analisar a influência de variáveis independentes na produção enzimática. A quantificação da celulase foi realizada pelo método do ácido dinitrosalicílico (ADNS). Antes de iniciar à aplicação dos planejamentos experimentais, foram adotadas estratégias para tentar minimizar e otimizar ao máximo o potencial dos isolados, as quais resultaram no estabelecimento da melhor agitação e a temperatura para a produção de celulase em 150 rpm e 20°C, para os dois isolados estudados. Inicialmente os fungos E5B e L1-1 apresentavam suas produções enzimáticas em 0,233U/mL e 0,342U/mL, respectivamente (antes da aplicação dos desenhos experimentais). Durante a condução do planejamento experimental do tipo Plackett&Burman(PB), foi verificada a preferência dos isolados pela fonte de carbono glicose, com efeito significativo na produção de celulases para os dois isolados. Tendo em vista o seu elevado custo comercial, foram realizados estudos com a sacarose, uma fonte de carbono alternativa e mais barata, bem como indutores enzimáticos. This work deals with the synthesis, spectroscopic characterization of mono and polynuclear Pd(II) compounds obtained from the reaction between [PdCl2(MeCN)2] precursor and imidazolidine-2-thione (L1) and benzimidazoline-2-thione (L2). The interaction between [PdCl2(MeCN)2] and imidazolidine-2-thione, in the 1:2 molar ratio, using MeOH as solvent, yielded compound [Pd2Cl4(L1)4]Cl2 (1) whereas the complex [Pd2Br4(L1)4]Br2 (2) was obtained from the reaction between 1 and Br- ions. Binuclear compounds of general formulae [Pd2(L2)4]X23/2 MeOH {X= Cl-(3), Br-(4)} were synthesized using benzimidazoline-2-thione as ligand, employing the same procedure used in the synthesis of 1 and 2, respectively. The new complexes 1-4 are crystalline, air stable and soluble in methanol and dmso. The compounds were characterized by elemental analysis, IR spectroscopy, ESI/MS mass spectrometry and thermogravimetry. Coordination of the ligands L1 and L2 via sulphur atom was evidenced by the shift of the CS band (~499 cm-1, 1 and 2; ~620 cm-1, 3 and 4) to lower frequencies in comparison with those found in the free ligands (510 cm-1, L1; 660 cm-1, L2), indicating the weakening of the CS bom after coordination. ESI/EM mass spectra (positive mode) of the complexes 1-4 allowed atribute the molecular formulae [Pd2X4(L1)4]X2 {X= Cl-(1), Br-(2)} and [Pd2(L2)4]X2 {X= Cl-(3), Br-(4)} by the appearance of the peaks at m/z 794,61 [(1) + MeOH]+, m/z 836,77 [(2) - 2H - L1]+, m/z 810 [(3) - 2Cl]+, m/z 810 [(4) - 2Br]+. The TG-DTA curves showed that the complexes 1 and 2 are thermally stable up to 212 and 169º C, respectively, and further decompose to metallic palladium at 527º C (1, obt. 28,46 %; calcd. 27,88%) and 895º C (2, obt. 22,85 %; calcd. 22,62%). Several studies have required Haematobia irritans (L.) raising in laboratory. The present study assessed two methods of inoculating immature forms of H. irritans to obtain adults. In 2007, 15 Nellore steers (Bos indicus) (L.) were used for the collection of feces free of anthelmintic treatment and flies to produce for eggs and larva. For method I, 30 eggs were incubated in square filter paper (5 × 5 cm) and deposited on bovine feces (500 g) where they were kept until hatching and spontaneous penetration of larvae (L1) into the fecal mass. After 24 h, eggs were analyzed under a stereoscope microscope (40×) for the number of larvae that instinctively penetrated the feces. In method II, larvae were obtained only by natural egg hatching. At birth, 30 larvae were collected and individually inoculated, directly onto the fecal plate by employing a moistened brush. The tests were carried out at controlled temperature (28˚C ± 2˚C) and saturated humidity (80%) until the emergence of flies with both methods. The number of emerged flies was considered in the result. Using method I, 276 (76.7%) flies emerged from 360 inoculated eggs, while using method II, 283 (78.6%) flies emerged from 360 inoculated larvae. There was no significant difference (P = 0.7821) between methods for the number of flies; however, the proportion between males and females by means of larva inoculation was different from 1:1 (P = 0.0146). Results indicated that both methods led to a satisfactory production of flies and egg inoculation provided an easier establishment.

Background and Aims Bone metabolism involves understanding many factors, especially during puberty, when bone turnover is significant and the bone mass peak must be achieved as a protective factor of future bone health. The objective was to evaluate the behavior of formation and resorption bone biomarkers (BB) in function of biological maturation in female adolescents.Methods Evaluation of formation and resorption BB, osteocalcin (OC), bone alkaline phosphatase (BAP) and carboxyterminal telopeptide (S-CTx) by correlating them with bone mineralization, bone age and pubertal development in healthy female adolescents. Seventy-two volunteers were subdivided into groups according to chronological age/bone age (BA): 10 11 years (n=12), 12 13 years (n=16), 14 15 years (n=15) and 16 19 years (n=29). The following were evaluated: weight (kg), height (m), BMI (kg/m2), calcium intake (3-day 24h food recalls (mg/day), puberty events (Tanner stages), serum OC (ng/mL), BAP (U/L), S-CTx (ng/mL) and bone mineral density (BMD) as calculated by DXA (g/cm2) in the spine (L1-L4), proximal femur and whole body. The project was approved by the UNESP Ethics Committee.Results BB showed similar behaviors, with higher mean values for 10 12 years and when adolescents were in the B2-B3 Pubertal Maturation Stage (B2: BAP=110.16 U/L, OC=33.81ng/mL, S-CTx=1.66 ng/mL and B3: BAP=136.50 U/L, OC=39.15ng/mL and S-CTx=1.88 ng/mL; p<0.001). Mean BB values decreased with advancing BA and pubertal maturity.Conclusions BB values showed parallelism with peak height velocity and significant negative correlation with BMD in the different evaluated sites, with chronological and BA ; higher BMD values correlated with lower bone biomarker values.

Objective: To examine the effect of different doses of triiodothyronine (T3) on mRNA levels of thyroid hormone receptors, TRα and TRβ, at different times. Materials and methods: 3T3-L1 adipocytes were incubated with T3 (physiological dose: F; supraphysiological doses: SI or SII), or without T3 (control, C) for 0.5, 1, 6, or 24h. TRα and TRβ mRNA was detected using real-time polymerase chain reaction. Results: F increased TRβ mRNA levels at 0.5h. After 1h, TRα levels increased with F and SI and TRβ levels decreased with SII compared with C, F, and SI. After 6h, both genes were suppressed at all concentrations. In 24h, TRα and TRβ levels were similar to those of C group. Conclusions: T3 action with F began at 1h for TRα and at 0.5h for TRβ. These results suggest the importance of knowing the times and doses that activate T3 receptors in adipocytes.

Aiming to evaluate the methodologies of surveys related with the specifications of Geo-referencing Rural Parcels in Brazil, established by INCRA (National Institute of Colonization and Agrarian Reform), in attendance to the Law 10.267/01, and considering aspects of precision and accuracy, a test area was set up in the Assentamento Florestan Fernandes, in the county of Presidente Bernardes, SP. This area was subdivided in three sub-areas in order to simulate the existence of three contiguous rural parcels. The first stage of work consisted in the implantation and determination of the control points coordinates in their respective areas. These control points were determined by the process of direct transport, using dual frequency (L1/L2) GPS receivers and through the process of traverse with baselines of up to 20 km, using single frequency receivers (L1). The coordinates of the points of the perimeter of the three sub-areas were determined using single frequency GPS receivers, from two survey methods, both using static relative positioning. The first one, so called traverse, each point that delimits the property was occupied successively, starting from the control point and closing at another. In other, denominated double irradiation, each point of the property was irradiated from two control points. These procedures were accomplished with the intention to define and to implement a mistake control strategy, to realize redundancy measurements and to use an adjustment method correctly, to obtain trustworthily values in the patterns demanded in each kind of survey. With the data collected and processed some analyses could be accomplished from the discrepancies between the coordinates obtained by different methodologies. The results show that this test area may be used to validate other methodologies and equipments. The results were satisfactory and attended the specifications of Geo-referencing Rural Parcel.
