Symptoms of huanglongbing (HLB) were reported in São Paulo State (SPS), Brazil, in March 2004. In Asia, HLB is caused by 'Candidatus Liberibacterasiaticus'and in Africa by 'Candidatus Liberibacter africanus'. Detection of the liberibacters is based on PCR amplification of their 16S rRNA gene with specific primers. Leaves with blotchy mottle symptoms characteristic of HLB were sampled in several farms of SIPS and tested for the presence of liberibacters. 'Ca. L. asiaticus' was detected in a small number of samples but most samples gave negative PCR results. Therefore, a new HLB pathogen was suspected. Evidence for an SPS-HLB bacterium in symptomatic leaves was obtained by PCR amplification with universal primers for prokaryotic 16S rRNA gene sequences. The amplified 16S rRNA gene was cloned and sequenced. Sequence analysis and phylogeny studies showed that the 16S rRNA gene possessed the oligonucleotide signatures and the secondary loop structure characteristic of the alpha-Proteobacteria, including the liberibacters. The 16S rRNA gene sequence phylogenetic tree showed that the SPS-HLB bacterium clustered within the a-Proteobacteria, the liberibacters being its closest relatives. For these reasons, the SPS-HLB bacterium is considered a member of the genus 'Ca. Liberibacter'. However, while the 16S rRNA gene sequences of 'Ca. L. asiaticus' and 'Ca. L. africanus' had 98-4% similarity, the 16S rRNA gene sequence of the SPS-HLB liberibacter had only 96(.)0% similarity with the 16S rRNA gene sequences of 'Ca. L. asiaticus'or'Ca. L. africanus'. This lower similarity was reflected in the phylogenetic tree, where the SPS-HLB liberibacter did not cluster within the 'Ca. L asiaticus'/'Ca. L. africanus group', but as a separate branch. Within the genus 'Candidatus Liberibacter' and for a given species, the 16S/23S intergenic region does not vary greatly. The intergenic regions of three strains of 'Ca. L. asiaticus', from India, the People's Republic of China and Japan, were found to have identical or almost identical sequences. In contrast, the intergenic regions of the SPS-HLB liberibacter, 'Ca. L. asiaticus' and 'Ca. L. africanus' had quite different sequences, with similarity between 66(.)0 and 79(.)5%. These results confirm that the SPS-HLB liberibacter is a novel species for which the name 'Candidatus Liberibacter americanus' is proposed. Like the African and the Asian liberibacters, the 'American' liberibacter is restricted to the sieve tubes of the citrus host. The liberibacter could also be detected by PCR amplification of the 16S rRNA gene in Diaphorina citri, the psyllid vector of 'Ca. L. asiaticus', suggesting that this psyllid is also a vector of 'Ca. L. americanus' in SPS. 'Ca. L. americanus' was detected in 216 of 218 symptomatic leaf samples from 47 farms in 35 municipalities, while 'Ca. L. asiaticus' was detected in only 4 of the 218 samples, indicating that 'Ca. L. americanus' is the major cause of HLB in SIPS.

Soil columns were produced by filling PVC tubes with a Dark Red Latosol (Acrortox, 22% of clay). A compacted layer was established at the depth of 15 cm in the columns. In the compacted layer, soil was packed to 1.13, 1.32, 1.48, and 1.82 Mg kg(-1), resulting in cone resistances of 0.18, 0.43, 1.20, and 2.50 MPa. Cotton was cropped for 30 days. Lime was applied to raise base saturation to 40, 52, and 67%. The highest base saturation caused a decrease in phosphorus (P) and zinc (Zn) concentrations in the plants. A decrease in root dry matter, length and surface area was also observed. This could be a consequence of lime induced Zn deficiency. Root growth was decreased in the compacted layer, and complete inhibition was noticed at 2.50 MPa. Once the roots got through the compacted layer, there was a growth recovery in the bottom layer of the pots. The increase in base saturation up 52% was effective in preventing a decrease in cotton root length at soil resistances to 1.20 MPa. Where the roots were shorter, there was an increase in nutrient uptake per unit of root surface area, which kept the plants well nourished, except for P.

Soil compaction has a negative effect and Ca was shown to enhance root growth. The effects of soil subsurface compaction and liming on root growth and nutrient uptake by soybean were studied at the Department of Agriculture and Plant Breeding, São Paulo State University, Brazil. A Dark Red Latosol, sandy loam (Haplortox) was limed to raise base saturations to 40.1, 52.4 and 66.7%. The experimental pots were made of PVC tubes with 100 mm of diameter. Three rings with 150, 35 and 150 mm long were fixed one on the top of the other. In the central ring of 35 mm, the soil was compacted to bulk densities of 1.06, 1.25, 1.43 and 1.71 g.cm(-3). There was no effect of base saturation on soybean root and shoot growth and nutrition. Subsurface compaction led to an increase in root growth in the superficial layer of the pots with a correspondent quadratic decrease in the compacted layer. There was no effect of subsoil compaction on total root length and surface, soybean growth and nutrition. Soybean root growth was decreased by 10% and 50% when the soil penetrometer resistances were 0.52 MPa (bulk density of 1.45 g.cm(-1)) and 1.45 MPa (bulk density of 1.69 g.cm(-3)), respectively. In spite of the poor root growth in the compacted layer, once it nas overcome the root system showed an almost complete recovery.

The aim of this study was to evaluate the effect of 1% sodium hypochlorite and five intracanals medications on Candida albicans harvested inside root canals. The contaminated canals were irrigated with sterile saline solution and then treated as follows: (i) filled with Calen paste (calcium hydroxide/glycol polyethylene paste); (ii) filled with camphorated paramonochloro phenol (CPMC); (iii) filled with 2% iodine-iodate solution; (iv) filled with tricresol formalin; (v) filled with Calen and CPMC pastes; (vi) irrigation with 1% sodium hypochlorite and filled with no intracanal medication; and (vii) no intracanal medication was used. Canal access and the apical foramen were then sealed with Cavit and the roots were stored in a humid chamber at 37 +/- 1 degreesC for 14 days. The canals were reinstrumented and irrigated with sterile saline solution. Sterile paper points were used to transfer the root canal contents to test tubes containing sterile saline solution. Part of the suspension was harvested in Sabouraud dextrose agar with chloramphenicol and incubated at 37 +/- 1 degreesC for 48 h, CPMC was effective in 100% of the samples followed in decreasing order of effectiveness by calcium hydroxide with CPMC (70% effective), 1% sodium hypochlorite (70% effective) (p < 0.05), tricresol formalin (60% effective), 2% iodine-iodate solution (50% effective), calcium hydroxide paste (30% effective), and saline + no intracanal medication.

The rheological behavior of Brazilian orange juice with different water content (0.34-0.73 w/w) was studied at a wide range of temperatures (0.5-62 degrees C) using a concentric cylinder viscometer. The results indicated that the juices behave as pseudoplastic fluids with yield stress, being represented by the Herschel-Bulkley model. The rheological parameters were fitted as functions of both temperature and water content in the tested range. Based on dimensional analysis it was proposed a modified Reynolds number (Re-M), which includes the Herschel-Bulkley parameters. Experimental data of friction factors during heating and cooling processes of orange juice in laminar flow through circular tubes could be well correlated as a function of Re-M. In this work, Eu(III) and Eu(II) doped gadolinium silicates has been obtained as compact tubes starting from spherical gadolinium hydroxide carbonate using the pores of silica matrix as support and its surface as reagent. Eu(III) doped gadolinium silicate with hexagonal phase shows an interesting visible shifted charge transfer band when compared to disilicate with orthorhombic phase that was also obtained. Eu(II) gadolinium silicate has been prepared using CO atmosphere presenting an intense blue emission. The divalent europium system shows a potential application as an UV-blue converter. Statement of problem. Acrylic resin denture teeth soften upon immersion in water, and the heating generated during microwave sterilization may enhance this process.Purpose. Six brands of acrylic resin denture teeth were investigated with respect to the effect of microwave sterilization and water immersion on Vickers hardness (VHN).Material and Methods. The acrylic resin denture teeth (Dentron [D], Vipi Dent Plus [V], Postaris [P], Biolux [B], Trilux [T], and Artiplus [A]) were embedded in heat-polymerized acrylic resin within polyvinylchloride tubes. For each brand, the occlusal surfaces of 32 identical acrylic resin denture posterior teeth were ground flat with 1500-grit silicon carbide paper and polished on a wet polishing wheel with a slurry of tin oxide. Hardness tests were performed after polishing (control group, C) after polishing followed by 2 cycles of microwave sterilization at 650 W for 6 minutes (MwS group), after polishing followed by 90-day immersion in water (90-day Wim group), and after polishing followed by 90-day storage in water and 2 cycles of microwave sterilization (90-day Wim + MwS group). For each specimen, 8 hardness measurements were made and the mean was calculated. Data were analyzed with a 2-way analysis of variance followed by the Bonferroni procedure to determine any significance between pairs of mean values (alpha=.01).Results: Mircrowave sterilization of specimens significantly decreased (P <.001) the hardness of the acrylic resin denture tooth specimens P (17.8 to 16.6 VHN, V (18.3 to 15.8 VHN), T (17.4 to 15.3 VHN), B (16.8 to 15.7 VHN), and A (17.3 to 15.7 VHN). For all acrylic resin denture teeth, no significant differences in hardness were found between the groups Mws, 90-day Wim, and 90-day Wim + MwS, with the exception of the 90-day Wim + MwS tooth A specimens (14.4 VHN), which demonstrated significant lower mean values (P <.001) than the 90-day Wim (15.8 VHN) and MwS (15.7 VHN) specimens.Conclusions. For specimens immersed in water for 90 days, 2 cycles of microwave sterilization had no effect on the hardness of most of the acrylic resin denture teeth.

Arsenic and germanium have been evaluated as internal standards to minimize matrix effects on the direct determination of selenium in milk by graphite furnace atomic absorption spectrometry (GFAAS) using tubes with integrated platform, pre-treated with W together with I'd as chemical modifier. The efficiency of As and Ge as internal standards for 25 mu g L-1 Se plus 500 mu g (L)-1 As or Ge in diluted (1 + 9 v/v) milk plus 1.0% (v/v) HNO3 was evaluated by means of correlation graphs plotted from the normalized absorbance signals (n = 20) of internal standard (axis gamma) versus analyte (axis x). The equations that describe the linear regression were: A(As)= - 0.004 +/- 0.019 +/- 1.02 + 0.019 A(Se) (r=0.9967 +/- 0.005); A(Ge)= - 0.0 17 +/- 0.015 + 1.01 +/- 0.015 A(Se) (r = 0.9978 +/- 0.004). Samples and reference solutions were automatically spiked with 500 mu g L-1 Ge or As and 1.0% (v/v) HNO3 by the autosampler. For 20 mu L of aqueous standard solutions, analytical curves in the 5.00-40.0 mu g L-1 Se range were established using the ratio of Se absorbance to internal standard absorbance (A(Se)A(IS)) versus analyte concentration, and good linear correlations were obtained. The characteristic mass was 40 pg Se. Limits of detection were 0.55 and 0.40 mu g L-1 with As and Ge as the internal standard, respectively. Relative standard deviations (RSD) for a sample containing 25 mu g L-1 Se were 1.2% and 1.0% (n = 12) using As and Ge, respectively. The RSD without internal standardization was about 6%. The accuracy of the proposed method was evaluated by an addition-recovery experiment and all recovered values were in the 99-105% range with IS and in the 70-80% range without IS. Using Ge as the internal standard, results of analysis of standard reference materials were in agreement with certified values at a 95% confidence level. The aim of this work is the evaluation of four different heat exchangers used for myocardium during cardioplegic system in cardiac surgeries. Four types of shell and tube heat exchangers made of different exchange elements were constructed, as follows: stainless steel tubes, aluminium tubes, polypropylene hollow fiber, and bellows type. The evaluation was performed by in vitro tests of parameters such as heat transfer, pressure drop, and hemolysis tendency. The result has shown that all four systems tested were able to achieve the heat performance, and to offer low resistance to flow, and safety, as well as have low tendency to hemolysis. However, we can emphasize that the bellows type heat exchanger has a significant difference with regard to the other three types.

Development of intraxylary phloem and internal cambium in Ipomoea hederifolia (Convolvulaceae). J. Torrey Bot. Soc. 136: 423-432. 2009-In Ipomoea hederifolia L. (Convolvulaceae), internal/intraxylary phloem originated as isolated strands from the procambially derived cells after the formation of protoxylem and protophloem. Bands of internal phloem were apparent in the sixth internode after the development of metacambium. In the relatively thick stems several small arcs/segments of internal cambium ensues from the parenchyma cells between the protoxylem and internal protophloem. Though all the segments were active, some of them (two of them located opposite to each other) were relatively more active. Bidirectional differentiation of these segments gave rise to secondary xylem centrifugally and secondary phloem centripetally, resulting inverted vascular bundles. Rest of the internal cambium segments were unidirectional and formed only secondary phloem centripetally. Like external vascular cambium, the internal cambium was non-storied. Structurally, secondary xylem and phloem was composed of axial and radial system in which rays were mostly uni- to biseriate. Secondary xylem produced by the internal cambium was more or less similar to the xylem formed by the external successive cambia. Secondary phloem produced by the internal cambium was composed of sieve tubes, companion cells, axial and ray parenchyma cells. Simple sieve plates of internal phloem were mostly arranged on transverse end walls in contrast to compound and obliquely placed sieve plates of external phloem formed by the successive cambia.

Introduction: Recently, a new sealer composed of Portland cement named Endo-CPM-Sealer was developed. The aim of this study was to investigate the effects of Endo-CPM-Sealer (EGEO SRL, Buenos Aires, Argentina), Sealapex (Sybron Endo, Glendora, CA), and Angelus MTA (Angelus, Londrina, Brazil) on cell viability and cytokine (interleukin [IL]-1 beta and IL-6) production by mouse fibroblasts. Methods: Millipore culture plate inserts with polyethylene tubes filled with materials were placed into 24-well cell culture plates with mouse fibroblasts. Cells cultured with only empty polyethylene tubes were used as the control. After 24 hours, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay was used to evaluate the cell viability. For cytokine assay, mouse fibroblasts were incubated in 24-well flat-bottom plates with set material disks at the bottom. Cells cultured without the material disks served as the negative control. After 24 hours of incubation, culture media were collected for cytokine evaluation by using an enzyme-linked immunosorbent assay. The data were statistically analyzed by analysis of variance and Bonferroni correction. Results: Endo-CPM-Sealer, Sealapex, and Angelus MTA did not inhibit the cell viability. All materials induced IL-6 releasing, but the amount was not statistically significant compared with the control group. Angelus MTA induced IL-1 beta releasing significantly more than the control. Conclusions: All materials were not considered cytotoxic in fibroblast culture. (J Endod 2009;35:1577-1579)

Different kinds of modifiers and coatings on the integrated platform of transversely heated graphite atomizer (THGA) have been tested for the simultaneous determination of two group of elements: the first, the more volatile, formed by arsenic, bismuth, lead, antimony and selenium; the second, the less volatile, formed by cobalt, chromium, cupper, iron and manganese in milk by electrothermal atomic absorption spectrometry. Different Rh-modifiers were studied, such as Rh-coated platforms (Rh), carbide plus rhodium coated platforms (W-Rh, Zr-Rh), carbide-coated platforms (W and Zr) with co-injection of RhCl3, solutions and uncoated platforms with injection of solutions of Pd(NO3)(2), Mg(NO3)(2), and RhCl3. Milk samples were diluted 1:10 in 1.0% HNO3 and injected into the tube. The mass of modifier deposited and co-injected in the tube and the use of end capped tubes were also evaluated in order to improve the electrothermal behavior of analytes. Integrated platform pretreated with W plus co-injection RhCl3 for first group and pretreated with W-Rh for second group were elected. For 20 mu L injected samples the analytical curves in the 5.0-20.0 mu g L-1 concentration range have good linear correlation coefficients (r > 0.998). Relative standard deviations (n = 12) are < 6% and the calculated characteristic masses are between 5 pg and 62 pg.

Eight taxa of marine invertebrates, including two new bivalve species, are described from the Low Head Member of the Polonez Cove Formation (latest early Oligocene) cropping out in the Vaureal Peak area, King George Island, West Antarctica. The fossil assemblage includes representatives of Brachiopoda (genera Neothyris sp. and Liothyrella sp.), Bivalvia (Adamussium auristriatum sp. nov., ?Adamussium cf. A. alanbeui Jonkers, and Limatula (Antarctolima) ferraziana sp. nov.), Bryozoa, Polychaeta (serpulid tubes) and Echinodermata. Specimens occur in debris flows deposits of the Low Head Member, as part of a fan delta setting in a high energy, shallow marine environment. Liothyrella sp., Adamussium auristriatum sp. nov. and Limatula ferraziana sp. nov. are among the oldest records for these genera in King George Island. In spite of their restrict number and diversification, bivalves and brachiopods from this study display an overall dispersal pattern that roughly fits in the clockwise circulation of marine currents around Antarctica accomplished in two steps. The first followed the opening of the Tasmanian Gateway at the Eocene/Oligocene boundary, along the eastern margin of Antarctica, and the second took place in post-Palaeogene time, following the Drake Passage opening between Antarctic Peninsula and South America, along the western margin of Antarctica.

The lacewings (Ceraeochrysa cincta) are important predators found associated to many pests in several agroecosystems. Eggs of Sitotroga cerealella and Anagasta kuehniella are used traditionally for massal rearing of these predators. Other foods can be used in substitution to the traditional ones, but little is known about the biology of lacewings and other alternative foods. So, this work was carried out to evaluate the effect of different diets on the post-embryonic development of C. cincta in laboratory. After been individualized in glass tubes, each larva was fed with S. cereallella and Diatraea saccharallis. It was evaluated the larvae duration, weight and viability in each instar, the duration, weight and viability of the pupa and pre-pupa stages and the duration of the post-embryonic period. It was adopted a completely randomized design with ten replications per treatment, each consisting of five eggs or larvae of C. cincta. Larvae were weighed 48 hours after hatching and 48 hours after each molt, and pre-pupae and pupae, 48 hours after starting these stages. There was no effect of the kind of food provided on the parameters evaluated. Therefore, D. saccharallis eggs can be used to feed C. cincta.
