Crops have different levels of dependence on pollinators; this holds true even for cultivars of the same species, as in the case of cucumber (Cucumis sativus). The aim of this research was to assess the attractiveness of flowers of three Japanese parthenocarpic cucumber cultivars and evaluate the importance of Africanized bees (Apis mellifera), and the Brazilian native stingless bees, Jata (Tetragonisca angustula) and Ira (Nannotrigona testaceicornis) on fruit production. Several parameters, including frequency of bee visits to flowers as well as duration of nectar collection and fruit set were examined; additionally, fruit weight, length and diameter were evaluated. Three greenhouses located in Ribeirao Preto, SP, were used for planting three cucumber cultivars (Hokushin, Yoshinari and Soudai). The female flowers were more attractive than male flowers; however, Jata bees were not observed visiting the flowers. The Africanized and the Ira bees collected only nectar, with a visitation peak between 10 and 12h. Visits to female flowers had a longer duration than visits to male flower visits in all three cultivars. Africanized bee colonies declined due to loss of bees while in the greenhouse; the native stingless bee colonies did not suffer these losses. When bees were excluded, fruit set was 78%; however, when bees had access to the flowers, fruit set was significantly (19.2%) higher. Fruit size and weight did not differ with and without bees. This demonstrates that even in parthenocarpic cucumber cultivars, which do not require pollination in order to from fruits, fruit production is significantly increased by bee pollination.

A análise econômica complementa a avaliação da eficiência dos cultivos consorciados, considerando além da produção física das culturas, o preço dos produtos segundo sua classificação qualitativa e época do ano. Avaliou-se economicamente consórcios de alface crespa e pepino, em duas populações de plantas, no inverno-primavera, em casa de vegetação, em Jaboticabal, SP, Brasil. Foram calculados o custo operacional total (COT), a receita bruta (RB) e o lucro operacional (LO) das culturas do pepino e alface em monocultura e em consórcio. A mão-de-obra foi a componente com maior participação no COT das culturas em consórcio e monocultura. Maiores RB e LO foram observadas nos cultivos consorciados estabelecidos com o transplante da alface e do pepino no mesmo dia, independente da densidade populacional. Considerando-se o lucro operacional e o índice de uso eficiente da área, recomenda-se o cultivo consorciado com transplante da alface até 10 dias após o transplante (DAT) do pepino, com duas linhas, e ao 0 DAT do pepino cultivado com uma linha no canteiro

O pepino (Cucumis sativus L.) é, dentre os produtos hortícolas, um dos mais visados para o cultivo em estufas no Brasil devido ao ciclo vegetativo curto e ao elevado valor econômico no período da entressafra. Estudou-se, em condições de ambiente protegido, a influência de diferentes cores de cobertura plástica de solo no florescimento e na produção de pepino híbrido 'Yoshinari' enxertado ou não sobre abóbora híbrida 'Ikky'. Os tratamentos de cobertura foram: preto, preto pintado de branco, verde, e sem cobertura. Os tratamentos cobertos com plástico e sem enxertia apresentaram florescimento com distribuição mais uniforme na planta. O número de flores foi superior no tratamento preto/branco enxertado. Para as plantas não enxertadas, todas as coberturas favoreceram o florescimento. A enxertia favoreceu o florescimento somente para os tratamentos com plástico preto ou verde. A fixação dos frutos aumentou pelo uso de cobertura plástica, mas a enxertia não teve influência. A distribuição uniforme do florescimento manteve-se na frutificação apenas para os tratamentos com plantas enxertadas e cobertura do solo com plástico preto ou verde. Tanto o uso de cobertura plástica quanto a enxertia favoreceram a colheita precoce. Os frutos de melhor qualidade e as maiores produções, em número de frutos, foram obtidos nos tratamentos preto e preto/branco, não enxertados. A enxertia 'Yoshinari'/'Ikky' provocou o aparecimento de frutos mais grossos e menores, fora do padrão de comercialização.

A endogamia reduz o vigor em plantas pela diminuição da proporção de loci heterozigotos. Entretanto, a influência da endogamia é diferente entre as espécies. O objetivo deste trabalho foi avaliar a depressão causada por endogamia em uma população de pepino do tipo japonês. A partir do intercruzamento entre plantas do híbrido Natsu suzumi foi obtida a geração F2, considerada como população S0. Obtiveram-se progênies S1, S2, S3, S4 e S5, através de autofecundações sucessivas pelo método do SSD ('Single Seed Descent'). Foram sete tratamentos (híbrido Natsu suzumi, populações S0 a S5) e o delineamento experimental foi em blocos ao acaso, com seis repetições e cinco plantas por parcela cultivadas em ambiente protegido de 21/08/2002 à 29/11/2002. Foram avaliados o número de folhas, semanalmente, o número e a massa de frutos, total e comercial, número de nós e porcentagem de nós com brotações laterais. Na comparação entre as populações S0 a S5 não foram observadas diferenças para todas as características avaliadas demonstrando não haver perda de vigor por endogamia nesta população.

Avaliou-se a capacidade produtiva, partenocarpia e expressão sexual de linhagens de pepino caipira sob cultivo em ambiente protegido. A capacidade produtiva foi avaliada em experimento em blocos ao acaso com 18 tratamentos (16 linhagens e dois híbridos do tipo caipira, Safira e Guarani), quatro repetições e cinco plantas por parcela, em estufa agrícola com as laterais abertas possibilitando a entrada de insetos polinizadores. A expressão sexual e a partenocarpia foram avaliadas em delineamento semelhante, utilizando 16 linhagens da população SHS (F2RC1 do cruzamento entre os híbridos Safira e Hatem (tipo holandês)) e 19 da população GH (F2 do cruzamento entre os híbridos Guarani e Hatem), porém este experimento foi conduzido em estufa agrícola fechada, com tela anti-afídeo, evitando- se a entrada de insetos polinizadores. A partenocarpia foi avaliada verificando o pegamento de frutos em cada linhagem. Várias linhagens foram estatisticamente similares aos híbridos em termos de produção, mas nenhuma foi superior. Os híbridos Safira e Guarani produziram respectivamente 23,1 e 19,1 frutos planta-1, sendo, respectivamente, 21,9 e 17,5 frutos comerciais. As massas total e comercial de frutos por planta foram respectivamente 3,8 e 3,6 kg para o híbrido Safira e 2,9 e 2,7 kg para o híbrido Guarani. A linhagem SHS-2 destacou-se pela boa produção, similar aos dois híbridos, apresentando frutos do tipo caipira claro, além de ser partenocárpica e possuir plantas ginóicas. Todas as linhagens da população SHS apresentaram frutos do tipo caipira, mas apenas cinco foram partenocárpicas. Na população GH, nove linhagens foram partenocárpicas e dez apresentaram frutos do tipo caipira. Duas linhagens da população SHS apresentaram apenas plantas monóicas, com as demais segregando para plantas ginóicas e monóicas. Na população GH, todas as plantas foram ginóicas. A população SHS é mais interessante para a obtenção de linhagens principalmente devido ao tipo de fruto caipira mais próximo dos frutos comerciais.

Studies were carried out in Brazil to study the inheritance of tolerance to Zucchini yellow mosaic virus (ZYMV) in cucumber cv. Formosa. This cultivar was individually crossed with two cucumber lines from different varietal types (L(b) from a Brazilian type, and L(j) from a Japanese type), both susceptible to the virus. Two experiments, one for each line, were separately carried out, where 6 treatments (parents, generations F1, F2 and F1BC1 for both parents) were evaluated in a randomized block design with 5 repetitions. Cotyledons of 2-week-old cucumber seedlings were inoculated with ZYMV. Only the plants that did not show symptoms up to 63 days post inoculation were considered as tolerant. A chi-square (chi(2)) analysis for assessing segregation from F2 and both F1BC1, led to the conclusion that the tolerance found in cv. Formosa is determined by a recessive gene.

Grafting is an alternative technique often recommended for cucumber culture in protected environments. This study aimed to study yield of cucumber grafted on different rootstocks compared to non-grafted plants in non-infected soil. Five treatments (Japanese cucumber hybrid 'Tsuyataro' without grafting, grafted on squash hybrids 'Shelper', 'Excitte Ikki', 'Daiguy' and 'Keiji') were evaluated in a completely randomized blocks design, with four repetitions and six plants per plot. Lesser fruit yield was obtained in cucumber grafted on 'Keiji', while cucumber grafted on other rootstocks did not differ from non-grafted plants. So, in the absence of soil borne diseases, grafting did not increase cucumber fruit yield, and, depending on rootstock, grafting can decrease production compared with non grafted plants.

Electron microscopy and immunolabelling with antiserum specific to cucumber mosaic virus coat protein were used to examine tobacco leaf cells infected by cucumber mosaic virus isolated from Catharanthus roseus (CMV-Cr). Crystalline and amorphous inclusions in the vacuoles were the most obvious cytological modifications seen. Immunogold labelling indicated that the crystalline inclusion was made up of virus particles and amorphous inclusions contained coat protein. Rows of CMV-Cr particles were found between membranes of dictyosomes, but membranous bodies and tonoplast-associated vesicles were not evident. Virus particles and/or free coat protein were easily detected in the cytoplasm by immunolabelling. No gold labelling was found within nuclei, chloroplasts and mitochondria.

Symptoms of Cucumber mosaic virus (CMV) on yellow passion flower (Passiflora edulis f. flavicarpa) are characterized by bright yellow mottling on leaves, starting at random points on the vine and diminishing in intensity towards the tip, which becomes symptomless as it grows. To determine whether symptomless portions of vines are CMV-free or represent latent infection, leaves with and without symptoms were collected from infected vines in the field. Biological, serological (plate-trapped antigen enzyme-linked immunosorbent assay, PTA-ELISA), Western blot and dot-blot hybridization assays showed that portions of the vines without symptoms were CMV-free. Vegetatively propagated vines with symptoms showed remission of symptoms on newly developed leaves. One year later, no CMV was detected in the upper leaves of these plants. Mechanically inoculated passion flower seedlings behaved similarly; symptoms were shown by few leaves after inoculation. Afterwards, plants became symptomless and CMV was not detected in the upper leaves or root system, 40 or 85 days after inoculation. The mechanism responsible for remission of symptoms accompanied by CMV disappearance is not known.

Shikimate dehydrogenase (SDH, EC 1.1.1.25) extracted from cucumber pulp (Cucumis sativus L.) was purified 7-fold by precipitation with ammonium sulfate and elution from columns of Sephadex G-25, DEAE-cellulose, and hydroxyapatite. Two activity bands were detected on polyacrylamide gel electrophoresis at the last purification step. pH optimum was 8.7, and molecular weight of 45 000 was estimated on a Sephadex G-100 column. SDH was inhibited competitively by protocatechuic acid with a K(i) value of 2 x 10-4 M. K(m) values of 6 x 10-5 and 1 x 10-5 M were determined for shikimic acid and NADP+, respectively. The enzyme was completely inhibited by HgCl2 and p-(chloromercuri)benzoate (PCMB). NaCl and KCl showed partial protection against inhibition by PCMB. Heat inactivation between 50 and 55-degrees-C was biphasic, and the enzyme was completely inactivated after 10 min at 60-degrees-C. Incubation of SDH with either NADP+ or shikimic acid protected the enzyme against heat inactivation.

Calcium binding and charge distribution on a fucosylated chondroitin sulfate and a standard chondroitin 6-sulfate have been studied using a metallochromic indicator and conductimetric titrations. The fucosylated chondroitin sulfate has a similar to 5-fold greater affinity for calcium ions than the standard chondroitin 6-sulfate. Possibly, this increased affinity for calcium ions is due to the branches on the fucosylated chondroitin sulfate, since the calcium affinity of an unbranched, sulfated fucan is similar to that of the standard chondroitin 6-sulfate. More charged groups per disaccharide unit (and a shorter distance between these groups) also distinguish the fucosylated chondroitin sulfate from standard chondroitin 6-sulfate. Comparison between native and chemically modified (desulfated or carboxyl-reduced) polysaccharides suggests that the sulfate esters are responsible for the increased charge density of the fucosylated chondroitin sulfate and that the presence of the fucose branches does not alter the length of the repetitive units which compose the central core of chondroitin from sea cucumber. These results are consistent with the chemical studies of these two polysaccharides.

We evaluated the phytotoxic effects that the hexane, ethyl acetate and methanol extracts from leaves and branches of the species Turnera ulmifolia L. and Turnera diffusa Willd. ex Schult. (Turneraceae), at concentrations of 1.25, 2.50, 3.75 and 5.00 mg ml-1, have on seed germination and seedling development in cucumber (Cucumis sativus L.). None of the extracts tested prevented germination, although the ethyl acetate extracts of T. diffusa, at 3.75 and 5.00 mg ml-1, reduced the mean germination speed and time to germination. Hexane extracts of both species reduced the main root length, number of secondary roots and hypocotyl length. In the ethyl acetate and methanol extracts, the number of secondary roots and hypocotyl length varied by species and concentration. Ethyl acetate extracts of T. ulmifolia at ≤ 2.50 mg ml-1stimulated growth of the main root and hypocotyl, as did T. ulmifolia methanol extracts at 1.25 and 3.75 mg ml-1; all other extract/concentration combinations had an inhibitory effect on those parameters. The hexane and ethyl acetate extracts of T. diffusa inhibited the formation of secondary roots and of the main root, although significant inhibition of hypocotyl growth was observed only at ≥ 2.50 mg ml-1. Comparatively, T. diffusa extracts inhibited development to a greater degree, thus presenting greater allelopathic potential, than did T. ulmifolia extracts.

Weeds can act as important reservoirs for viruses. Solanum americanum (Black nightshade) is a common weed in Brazil and samples showing mosaic were collected from sweet pepper crops to verify the presence of viruses. One sample showed mixed infection between Cucumber mosaic virus (CMV) and Potato virus Y (PVY) and one sample showed simple infection by PVY. Both virus species were transmitted by plant extract and caused mosaic in tomato (Solanum lycopersicum cv. Santa Clara), sweet pepper (Capsicum annuum cv. Magda), Nicotiana benthamiana and N. tabaccum TNN, and local lesions on Chenopodium quinoa, C. murale and C. amaranticolor. The coat protein sequences for CMV and PVY found in S. americanum are phylogenetically more related to isolates from tomato. We conclude that S. americanum can act as a reservoir for different viruses during and between sweet pepper crop seasons.
