The opportunistic bacterium Proteus mirabilis secretes a metalloprotease, ZapA, considered to be one of its virulence factors due to its IgA-degrading activity. However, the substrate specificity of this enzyme has not yet been fully characterized. In the present study we used fluorescent peptides derived from bioactive peptides and the oxidized ß-chain of insulin to determine the enzyme specificity. The bradykinin- and dynorphin-derived peptides were cleaved at the single bonds Phe-Ser and Phe-Leu, with catalytic efficiencies of 291 and 13 mM/s, respectively. Besides confirming already published cleavage sites, a novel cleavage site was determined for the ß-chain of insulin (Val-Asn). Both the natural and the recombinant enzyme displayed the same broad specificity, demonstrated by the presence of hydrophobic, hydrophilic, charged and uncharged amino acid residues at the scissile bonds. Native IgA, however, was resistant to hydrolysis by ZapA.

In this article we investigated the platelet aggregating activity of whole crotoxin and its subunits isolated from Crotalus durissus cascavella venom. During the purification protocols of the venom, using HPLC molecular exclusion, we detected the presence of two different serine protease activities in the gyroxin fraction, and another in the crotoxin fraction, which induced strong and irreversible platelet aggregation, in addition to blood coagulation. From crotoxin, we isolated PLA(2), crotapotin (both fractions corresponding approximately 85% of whole crotoxin) and another minor fraction (F20) that exhibited serine protease activity. After a new fractionation on reverse phase HPLC chromatography, we obtained three other fractions named as F201, F202 and F203. F202 was obtained with high degree of molecular homogeneity with molecular mass of approximately 28 kDa and a high content of acidic amino residues, such as aspartic acid and glutamic acid. Other important amino acids were histidine, cysteine and lysine. This protein exhibited a high specificity for BApNA, a Michaelis-Menten behavior with Vmax estimated in 5.64 mu M/min and a Km value of 0.58 mM for this substrate. In this work, we investigated the ability of F202 to degrade fibrinogen and observed alpha and beta chain cleavage. Enzymatic as well as the platelet aggregation activities were strongly inhibited when incubated with TLCK and PMSF, specific inhibitors of serine protease. Also, F202 induced platelet aggregation in washed and platelet-rich plasma, and in both cases, TLCK inhibited its activity. The N-terminal amino acid sequence of F202 presented a high amino acid sequence homology with other thrombin-like proteins, but it was significantly different from gyroxin. These results showed that crotoxin is a highly heterogeneous protein composed of PLA(2), thrombin-like and other fractions that might explain the diversity of physiological and pharmacological activities of this protein.

Our findings revealed two distinct patterns of substrate preparation: the pattern of leaf-cutting ants foraging on dicotyledons is marked by highly fragmented substrate resulting in a more advanced initial decomposition. The pattern of leaf-cutting ants harvesting grasses is characterized by large pieces of substrate, resulting in little initial decomposition. Ants foraging on both types of plants are apparently intermediary between the two patterns, although more similar to the patterns of those foraging on dicotyledons. Also, the behavior of scraping the substrate was described for the first time, it is very important for the removal of the epicuticular wax layer of the leaves helping the growth of the symbiotic fungus.

Biosurfactants are bioactive agents that can be produced by many different microorganisms. Among those, special attention is given to yeasts, since they can produce many types of biosurfactants in large scale, using several kinds of substrates, justifying its use for industrial production of those products. For this production to be economically viable, the use of residual carbon sources is recommended. The present study isolated yeasts from soil contaminated with petroleum oil hydrocarbons and assessed their capacity for producing biosurfactants in low cost substrates. From a microbial consortium enriched, seven yeasts were isolated, all showing potential for producing biosurfactants in soybean oil. The isolate LBPF 3, characterized as Candida antarctica, obtained the highest levels of production - with a final production of 13.86 g/L. The isolate LBPF 9, using glycerol carbon source, obtained the highest reduction in surface tension in the growth medium: approximately 43% of reduction after 24 hours of incubation. The products obtained by the isolates presented surfactant activity, which reduced water surface tension to values that varied from 34 mN/m, obtained from the product of isolates LBPF 3 and 16 LBPF 7 (respectively characterized as Candida antarctica and Candida albicans) to 43 mN/m from the isolate LPPF 9, using glycerol as substrate. The assessed isolates all showed potential for the production of biosurfactants in conventional sources of carbon as well as in agroindustrial residue, especially in glycerol.

Pseudomonas aeruginosa LBI produced surface active rhamnolipids when cultivated on waste from the sunflower-oil process under different conditions. These biosurfactants, which reduce the superficial and interfacial tensions between fluids, offer advantages over their chemical counterparts, especially because of their ecological acceptability. These molecules can be used in fields as diverse as chemical, pharmaceutical and petrochemical industries. In this work, we present the effect of C/N ratio on growth and production yield. The best production yields (Y-P/S) were achieved for C/N ratios (in g/g) of 8/1 (0.22) and 6.4/1 (0.23). The product concentration was very satisfactory (7.3 g/L) at C/N ratio of 8/1, especially when considering that the substrate was basically composed of wastes that would otherwise constitute an environmental disposal problem. The slow and uneven germination of Syagrus oleracea seeds represents a problem for its propagation, especially for commercial production. This work aimed to study the effects of substrate and mechanical scarification on the germination of S. oleracea seeds. Germination (% G), emergence rate (ER), leaf area (cm(2)), root length (cm), leaf dry mass (g) and root dry mass (g) were also determined. Non-scarified seeds sown in vermiculite germinated faster and showed higher germination (65%), as well as greater root length (16.55 cm). leaf dry mass (5.07 g) and root dry mass (12.41 g), when compared to the scarified seeds.

Dypsis leptocheilos is highly valued as an ornamental palm. Its propagation is done by seeds; however, there is little information about this process. The objective of this work was to study the substrate and temperature effects on the germination of D. leptocheilos seeds. The experiments were carried out at FCAV/UNESP, Campus of Jaboticabal, São Paulo State, Brazil. In order to study the substrate effects, four treatments were arranged (coconut fiber, sand, vermiculite and Plantmax (R)) at 30 degrees C. For the temperature effects, six treatments were performed (temperature at laboratory conditions (21.5 degrees C and 72% RU), 25 degrees C, 30 degrees C, 35 degrees C, 20-30 degrees C and 25-35 degrees C), having the coconut fiber as the substrate. The experimental design was completely randomized, with four replicates of 25 disseminules per plot (seeds with attached endocarp). Water replacement was managed to maintain 100% of the retention capacity of the substrates. The germination tests were observed every two days, and conducted over 148 days for the substrate effects, and over 152 days for the temperature effects, when no more seed germination was noted. In order to determine the germination percentage (% G) and the Germination Speed Index (GSI), the seed was considered germinated when the germinative button appeared. At the end of the experiments, leaf area (cm(2)), root and shoot length (cm), root and shoot dry mass (g) were also obtained. Statistical analysis was performed and means were compared by the Tukey test. Germination rate and speed in coconut fiber was higher at 25 and 30 degrees C. However, when other substrates were tested at 30 degrees C, the highest germination percentage was observed in vermiculite, which also sustained better results for the seed germination and for the characteristics related to shoots and roots.

A lichia é considerada em todo o mundo como a rainha das frutas, por sua aparência e sabor delicado. Atualmente, a cultura vem despertando interesse, principalmente no Estado de São Paulo, onde, em virtude dos problemas enfrentados pelos agricultores, destacando-se os de laranja e cana-de-açúcar, a procura por alternativas agrícolas vem se intensificando ainda mais. A propagação através de sementes não é recomendada comercialmente, pois as plantas de pé-franco são geneticamente muito desuniformes e apresentam período juvenil muito longo, demorando até 10 anos para entrar em produção; no entanto, a semente pode ser utilizada para a obtenção de porta-enxertos. Observou-se que a germinação de sementes de lichia ocorre rapidamente, reforçando a importância do uso de um substrato adequado. Pela análise dos resultados obtidos, verifica-se que o substrato que aparentemente permitiu as melhores combinações de água e disponibilidade de oxigênio para as sementes - casca de arroz carbonizada e areia lavada, foi o que proporcionou valores de germinação mais rápidos e mais altos.

Com a procura cada vez maior de alternativas por parte dos produtores rurais e de frutas exóticas pelos consumidores, o mercado de frutíferas tem crescido consideravelmente. Neste contexto, a pitaya vem sendo procurada não apenas pelo exotismo de sua aparência, como também por suas características organoléticas. O presente trabalho foi realizado objetivando avaliar a influência do volume de substrato no desenvolvimento de mudas de pitaya vermelha, utilizando-se bandejas de poliestireno expandido com diferentes volumes de células: 10, 15, 30 e 65 mL. As avaliações, realizadas aos 90 dias, foram quanto: porcentagem de sobrevivência; ao número e altura dos cladódios (cm), ao comprimento da maior raiz (cm); às massas fresca e seca dos cladódios e das raízes (gramas). O delineamento experimental foi inteiramente casualizado. Os dados de porcentagem de sobrevivência foram transformados em arc sen para fins de análise estatística e as médias comparadas pelo teste de Tukey, a 5% de probabilidade. A sobrevivência e o desenvolvimento vegetativo das mudas de pitaya vermelha através de sementes foram diretamente proporcionais ao volume de substrato usado no experimento. Para formação de mudas de pitaya vermelha através de sementes é recomendado o uso de containeres com capacidade de 65 mL.

The present research describes an efficient procedure to obtain high levels of trypsinogen and chymotrypsinogen by using a simple, rapid, and easily reproducible method. The extraction process and the time-course of activation of zymogens can be carried out in a single laboratory period, without sophisticated equipment. The main objective was to prepare a laboratory class that would stimulate student interest in enzyme regulation, exploring the fact that the catalytic activity of some enzymes is regulated by different mechanisms. The regulation of proteolytic enzymes requires the synthesis of an inactive zymogen and its being irreversibly switched on by specific proteolytic cleavage.

P>The present work evaluates of harvested mushroom and viability of Agaricus bisporus growth in several casing materials based on spent mushroom substrate. The experiment consisted of eight casing layer, which six were made with spent mushroom substrate. The results confirm the usefulness of reincorporating the spent substrate in new cultivation cycles as an ingredient of casing mixtures. In general, biological efficiency was high, three of the SMS based-casings surpassing the threshold value of 100 kg 100 kg-1 of compost. The high electrical conductivity of mixtures containing a large proportion of spent substrate limits the extent to which it can be used, although mixing it with other materials (such as peat) reduces these values to acceptable levels. In short, it makes economic and environmental sense to reuse spent mushroom substrate as an ingredient of alternative casing materials.
