The invasive thistle Carduus nutans has been reported to be allelopathic, yet no allelochemicals have been identified from the species. In a search for allelochemicals from C. nutans and the closely related invasive species C. acanthoides, bioassay-guided fractionation of roots and leaves of each species were conducted. Only dichloromethane extracts of the roots of both species contained a phytotoxin (aplotaxene, (Z,Z,Z)-heptadeca-1,8,11,14-tetraene) with sufficient total activity to potentially act as an allelochemical. Aplotaxene made up 0.44 % of the weight of greenhouse-grown C. acanthoides roots (ca. 20 mM in the plant) and was not found in leaves of either species. It inhibited growth of lettuce 50%(I-50) in soil at a concentration of ca. 0.5 mg g(-1) of dry soil (ca. 6.5 mM in soil moisture). These values gave a total activity in soil value (molar concentration in the plant divided by the molarity required for 50 % growth inhibition in soil = 3.08) similar to those of some established allelochemicals. The aplotaxene I-50 for duckweed (Lemna paucicostata) in nutrient solution was less than 0.333 mM, and the compound caused cellular leakage of cucumber cotyledon discs in darkness and light at similar concentrations. Soil in which C. acanthoides had grown contained aplotaxene at a lower concentration than necessary for biological activity in our short-term soil bioassays, but these levels might have activity over longer periods of time and might be an underestimate of concentrations in undisturbed and/or rhizosphere soil.

A quantificação do impacto das práticas de preparo sobre as perdas de carbono do solo é dependente da habilidade de se descrever a variabilidade temporal da emissão de CO2 do solo após preparo. Tem sido sugerido que as grandes quantidades de CO2 emitido após o preparo do solo podem servir como um indicador das modificações nos estoques de carbono do solo em longo termo. Neste trabalho é apresentado um modelo de duas partes baseado na temperatura e na umidade do solo e que inclui um termo exponencial decrescente do tempo que é eficiente no ajuste das emissões intermediárias após preparo: arado de disco seguido de uma passagem com a grade niveladora (convencional) e escarificador de arrasto seguido da passagem com rolo destorroador (reduzido). As emissões após o preparo do solo são descritas utilizando-se estimativa não linear com um coeficiente de determinação (R²) tão alto quanto 0.98 após preparo reduzido. Os resultados indicam que nas previsões da emissão de CO2 após o preparo do solo é importante considerar um termo exponencial decrescente no tempo após preparo.

The strain used in this work was a Salmonella enterica subsp enterica serovar Kottbus (6,8:e,h:1,5) isolated from imported day-old ducklings in Laboratório Nacional Agropecuário (LANAGRO/SP) of the Ministry of Agriculture of Brazil (MAPA). In view of the lack of information available about this Salmonella isolate and also because it was detected in day-old imported birds, this study was carried out to investigate the dissemination of S. Kottbus among newly hatched chicks. The birds were placed in three groups: one group of 20 birds received 0.1 mL of S. Kottbus culture containing 1.2 x 10(8) CFU/mL, the second group of 20 birds was inoculated with 1.2 x 10(5) CFU/mL and the third group of 10 birds was untreated (control group). Results were similar for both infected groups. The bacterium was recovered from cloacal swabs collected from the first day following the experimental infection until the end of the trial (42 days post-inoculation). At 15 and 42 days post-inoculation (dpi), half of the birds of each group were killed for bacteriological examination of cecal contents, liver and spleen. At 15 dpi, viable cell counts of S. Kottbus were obtained in all kinds of samples. At 42 dpi, Salmonella was present in the liver and spleen of few birds, but in large amounts in the cecal contents of almost all birds.

O manejo das plantas de cobertura visa obter maiores benefícios com a sua introdução nos sistemas agrícolas. O uso de espécies forrageiras como as do gênero Brachiaria, devido à grande quantidade de biomassa e ao fato de persistirem por mais tempo sobre o solo, vem se tornando uma boa opção. Quando se almeja maior rendimento operacional no plantio, maior disponibilidade de nutrientes, maior presença de palha na superfície do solo e menor liberação de possíveis substâncias alelopáticas, um ponto importante a ser observado é o intervalo de tempo entre a dessecação da cobertura com glifosato e a semeadura do arroz. O objetivo deste trabalho foi verificar o efeito do manejo de B. ruziziensis, com ou sem herbicida, antecedendo a semeadura da cultura, nos componentes de produção e na produtividade do arroz. O ensaio foi conduzido em casa de vegetação e consistiu em três tipos de manejo de B. ruziziensis antes da semeadura do arroz. Essa forrageira apresentou crescimento linear, e a presença de grande quantidade de sua matéria seca na superfície do solo foi prejudicial ao arroz. Quando manejada próximo do plantio do arroz, B. ruziziensis causou redução significativa na produtividade dessa cultura; o manejo dessa forrageira com herbicida deve ser realizado com mais de 20 dias antes do plantio do arroz; o manejo dela com retirada da parte aérea, com ou sem herbicida, deve ser realizado com 10 ou mais dias antes do plantio do arroz; e o manejo no momento adequado proporcionou aumento significativo na produtividade do arroz.

Growing corn mixed with forage crops can be an alternative for pasture and Mulch production during relatively dry winters in tropical areas, making no-till feasible in some regions. However, little is known about nutrient dynamics in this cropping system. The objective of the present work was to evaluate K dynamics in a production system in which corn was either grown as a sole crop or mixed with Brachiaria brizantha. In the second year of the experiment, nitrogen rates ranging from 0 to 200 kg ha(-1) were applied to the system. Dry matter yields and potassium contents in the soil, as well as residues and plants were determined at corn planting and harvest. Potassium balance in the system was calculated. Corn grain yield in mixed crop responded up to 200 kg ha(-1) N. The introduction of brachiaria in the system resulted in higher amounts of straw on the soil Surface and higher K recycling. Soil exchangeable K balance showed an excess K for both N rates only in the mixed system, however, when non-exchangeable K was also included in calculations, excess K was found in both mixed and sole corn systems. Large amounts of non-exchangeable K were taken up in the system involving brachiaria, which showed a considerable capacity in recycling K, increasing its contents in the surface soil layer. Em alguns solos com baixos teores de potássio trocável, formas não trocáveis participam do suprimento às plantas. Há evidências de que a disponibilização do K não trocável depende mais da demanda das plantas pelo nutriente do que das propriedades do solo. O objetivo deste trabalho foi avaliar o suprimento e a exaustão de formas de K em um Latossolo Vermelho do Cerrado em decorrência da adubação potássica residual e do cultivo de Brachiaria ruziziensis (Syn. Urochloa ruziziensis). Amostras de solos foram coletadas na camada de 0-20 cm, em parcelas de um experimento de campo em que a soja vinha sendo adubada anualmente, por 10 anos, com 0, 60, 120 e 180 kg ha-1 de K2O. em casa de vegetação, as amostras de solo receberam a aplicação de 0, 150 e 300 mg dm-3 de K e foram cultivadas com B. ruziziensis, com cinco cortes sucessivos. O suprimento de K às plantas dependeu mais do fertilizante recém-adicionado do que do efeito residual de adubações anteriores. O K não trocável foi responsável, ao longo do tempo, pela manutenção dos teores de K trocáveis e, na ausência de adubação potássica, constituiu a principal fonte de K para as plantas de B. ruziziensis. As plantas de B. ruziziensis possuem capacidade de extrair quantidade considerável de K do solo, confirmando sua importância como cultura de cobertura, na ciclagem do nutriente no solo.

Uma das grandes limitações do plantio direto é a produção e manutenção da cobertura vegetal. Por esse motivo, o milheto tem-se constituído em boa opção de cultivo no inverno, gerando palhada com decomposição mais lenta viabilizando esse sistema de produção. O objetivo do experimento foi estudar a cultura do milheto em três épocas de semeadura (5/3, 25/3 e 19/4/99), sob condições de sequeiro, e a ceifa da parte aérea (ceifa a cada florescimento e retirada do resíduo vegetal; ceifa a cada florescimento e permanência do resíduo vegetal; ceifa no florescimento e retirada do resíduo vegetal; ceifa no florescimento e permanência do resíduo vegetal e livre crescimento, sem ceifar), bem como seu efeito na produção da soja cultivada na seqüência. Pode-se concluir que o milheto semeado em março e submetido à ceifa na época de cada emissão da panícula proporcionou as maiores produções de matéria seca dessa espécie. Tal cultura demonstrou grande capacidade na produção de matéria seca e, quando semeada em 5 de março e 19 de abril, propiciou as maiores produtividades da soja cultivada em sucessão.

We investigated the effects of treatments with the enzymes pepsin and trypsin on the in vitro immunological reactivity of the major globulins found in the seeds of sweet lupin, chickpea, and lentil. Polyclonal major globulin-specific antiserum was obtained by immunization of rabbits with a solution of the 11 S globulin of each legume. The globulins were hydrolyzed with pepsin and trypsin for 1, 5, 15, and 30 min. The native globulins and their hydrolysates were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and immunoblotting to identify the polypeptide bands with antigenic activity, and the hypoantigenicity of the hydrolysates was analyzed by enzyme-linked immunosorbent assay. Our results show that enzymatic treatment of the major storage protein (11 S globulin) of sweet lupin, chickpea, and lentil with pepsin or trypsin lead to the formation of large amounts of short peptides and free amino acids that do not allow antibody binding, resulting in a weakened immunoreactivity.

The yeast Kluyveromyces marxianus var. bulgaricus produced large amounts of extracellular inulinase activity when grown on inulin, sucrose, fructose and glucose as carbon source, This protein has been purified to homogeneity by using successive DEAE-Trisacryl Plus and Superose 6 HR 10/30 columns. The purified enzyme showed a relative molecular weight of 57 kDa by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and 77 kDa by gel filtration in Superose 6 HR 10/30, Analysis by SDS-PAGE showed a unique polypeptide band with Coomassie Blue stain and nondenaturing PAGE of the purified enzyme obtained from media with different carbon sources showed the band, too, when stained for glucose oxidase activity, the optimal hydrolysis temperature for sucrose, raffinose and inulin was 55 degrees C and the optimal pH for sucrose was 4.75, the apparent K-m values for sucrose, raffinose and inulin are 4.58, 7.41 and 86.9 mg/ml, respectively, Thin layer chromatography showed that inulinase from K. marxianus var. bulgaricus was capable of hydrolyzing different substrates (sucrose, raffinose and inulin), releasing monosaccharides and oligosaccharides, the results obtained suggest the hypothesis that enzyme production was constitutive.

The fruit bat Artibeus lituratus absorbs large amounts of glucose in short periods of time and maintains normoglycemia even after a prolonged starvation period. Based on these data, we aimed to investigate various aspects related with glucose homeostasis analyzing: blood glucose and insulin levels, intraperitoneal glucose and insulin tolerance tests (ipGTT and ipITT), glucose-stimulated insulin secretion (2.8, 5.6 or 8.3 mmol/L glucose) in pancreas fragments, cellular distribution of beta cells, and the amount of pAkt/Akt in the pectoral muscle and liver. Blood glucose levels were higher in fed bats (6.88 +/- 0.5 mmol/L) than fasted bats (4.0 +/- 0.8 mmol/L), whereas insulin levels were similar in both conditions. The values of the area-under-the curve obtained from ipGTT were significantly higher when bats received 2 (5.5-fold) or 3 g/kg glucose (7.5-fold) b.w compared to control (saline). These bats also exhibited a significant decrease of blood glucose values after insulin administration during the iplTT. Insulin secretion from fragments of pancreas under physiological concentrations of glucose (5.6 or 8.3 mmol/L) was similar but higher than in 2.8 mmol/L glucose 1.8- and 2.0-fold, respectively. These bats showed a marked beta-cell distribution along the pancreas, and the pancreatic beta cells are not exclusively located at the central part of the islet. The insulin-induced Akt phosphorylation was more pronounced in the pectoral muscle, compared to liver. The high sensitivity to glucose and insulin, the proper insulin response to glucose, and the presence of an apparent large beta-cell population could represent benefits for the management of high influx of glucose from a carbohydrate-rich meal, which permits appropriate glucose utilization.
