The characteristics of log smoldering after an Amazonian deforestation fire are described. The experiment was carried out in 2001 at the Caiabi farm, near the city of Alta Floresta, state of Mato Grosso, Brazil, as part of a set of tests that have been performed in the same area since 1997. A 200 x 200m(2) test area was slashed in the beginning of June and burned on 20 August. The area contained 507 logs with diameter at breast height (DBH) higher than 10 cm, per hectare. In the day following the main burn 59 logs were found to remain smoldering, a number that corresponds to 2.9% of the total in the area. We chose 11 of the 59 logs to have their smoldering process monitored. Their diameter, moisture content and CHN dry biomass composition after the plot burn and before smoldering were determined. Other parameters such as temperature distribution while smoldering, porosity, density and mass volatilized during thermogravimetric test were also determined. Average smoldering speeds were in the range from 0.8 to 1.5 cm h(-1) for logs that smoldered without transition to the flaming regime. The average speed increased to 2.1 cm h(-1) for those logs that oscillated between smoldering and flaming. The speeds were lower overnight as compared to values determined during daytime for the same log. Higher log moisture contents were found to produce decreased speeds. Micro-porous biomass was not observed in the set of the 11 selected logs. Smoldering was observed to occur at substantial intensity in crossing of logs, with no longitudinal propagation.

Lippia alba, family Verbenaceae, is widely spread in Central and South American. It's a shurb with a quadrangular branch reaching 1,7m tall. The leaves are membranaceous, petiolate, pubescent with a strong flavor. It's limbs have variable forms with pointed apex, cuneiform or decumbent base, and serrated or crenated hordes. It was determined the best harvest season to biomass production, essential oil content and chemical composition, on different plant parts (apical, medium, basal). It has been observed that, both apical and medium parts represented around 80 % of the fresh leaf mass. The foliar biomass yields are about 5 ton/ha in four harvests during an year. The average yield of essential oil considering the three plant parts were 0.15%, 0.47%, 0.46%, 0.55% and 0.61% for summer/98, autumn/98, winter/98, spring/98 and summer/99, respectively. Essential oils showed similar chemical composition either in relation to seasonality, neral, geranial and t-cariofilene were the majority compounds.

The influence of water level variation (flood pulse) on the biomass and chemical composition of the aquatic macrophyte Eichhornia azurea, was investigated in a tropical oxbow lake of the Rio Mogi-Guacu, State of São Paulo, Brazil. The flood pulse causes an increase in total nitrogen content from 0.67 to 1.35 mg/L and total phosphorus content from 10.5 to 101.0 mu g/L of the water. This fertilization, associated with other factors, determines a typical seasonal variation in the biomass and chemical composition of the macrophyte.

The nutritional value of zooplankton (lipid and phosphorus contents) was analyzed in outdoor, plankton-culture tanks, to monitor the effects of diurnal variation and fertilization. Total lipid contents were significantly higher (average values of about 14% DW) for the treatment fertilized during the sampling week. A significantly highher total lipid concentrations generally coincided with high biomass of Moina sp. (Cladocera) and Thermocyclops sp. nauplii (Copepoda), which is probably related to zooplankton filtration rates. The values tended to be higher during the afternoon and the night. Phosphorus contents in the zooplanktonic population varied from 0.15 to 0.91% DW, with significant differences among the tanks. The zooplankton nutritional quality was greatly influenced by fertilization, which also determined the species composition in the tanks.

The family Piperaceae contains nearly 2000 species, most of them distributed in two genera, Piper and Peperomia. In Brazil circa 170 Piper species are found, mainly in tropical areas Their use ranges from flavoring and culinary to raw material for pharmaceutical and cosmetic industry. One of these species, Piper callosum, is used in folk medicine in the Amazon area. The objective of this study was to evaluate the production of biomass (aerial parts) as well as yield and composition of the essential oil from the leaves, according to different spacing between cultivated plants at Embrapa Western Amazon, in Manaus, State of Amazonas, Brazil. The experimental design was randomized blocks, with four treatments and seven replicates with six plants. Biomass production was inversely proportional to the spatial arrangements, with the greatest biomass production (1034.93 kg/ha) in the shortest spacing (E1), although no statistical difference was verified between E3 and E4. The same response was observed for the production of essential oil. The chemical composition of the oil was not affected by spacing, and major compounds found were safrole (59.1%), beta-pinene (8.3%), alpha-pinene (6.5%), methyl eugenol (6.3%) and 1,8-cineole (4.1).

The extracellular glycerol kinase gene from Saccharomyces cerevisiae (GUT]) was cloned into the expression vector pPICZ alpha. A and integrated into the genome of the methylotrophic yeast Pichia pastoris X-33. The presence of the GUT1 insert was confirmed by PCR analysis. Four clones were selected and the functionality of the recombinant enzyme was assayed. Among the tested clones, one exhibited glycerol kinase activity of 0.32 U/mL, with specific activity of 0.025 U/mg of protein. A medium optimized for maximum biomass production by recombinant Pichia pastoris in shaker cultures was initially explored, using 2.31 % (by volume) glycerol as the carbon source. Optimization was carried out by response surface methodology (RSM). In preliminary experiments, following a Plackett-Burman design, glycerol volume fraction (phi(Gly)) and growth time (t) were selected as the most important factors in biomass production. Therefore, subsequent experiments, carried out to optimize biomass production, followed a central composite rotatable design as a function of phi(Gly) and time. Glycerol volume fraction proved to have a significant positive linear effect on biomass production. Also, time was a significant factor (at linear positive and quadratic levels) in biomass production. Experimental data were well fitted by a convex surface representing a second order polynomial model, in which biomass is a function of both factors (R(2)=0.946). Yield and specific activity of glycerol kinase were mainly affected by the additions of glycerol and methanol to the medium. The optimized medium composition for enzyme production was: 1 % yeast extract, 1 % peptone, 100 mM potassium phosphate buffer, pH=6.0, 1.34 % yeast nitrogen base (YNB), 4.10(-5) % biotin, 1 %, methanol and 1 %, glycerol, reaching 0.89 U/mL of glycerol kinase activity and 14.55 g/L of total protein in the medium after 48 h of growth.

The ichthyofauna of 24 stretches of streams, all of 100 m length and of fifth or lower order and most of second and third order, were sampled along four left bank tributaries (Rio do Peixe, Rio Aguapei, Rio Sao Jose dos Dourados, lower Rio Tiete of the main channel of the Rio Parana in the state of São Paulo, southeastern Brazil. Sampling of the fish fauna at each of the six sites in the four basins incorporated a standardized fish collecting methodology and a standardized documentation of environmental data serving as the basis for a comparative analysis of the collecting locations. The 8,189 fish specimens collected represented six orders, 18 families, 42 genera, and 56 species, with a total biomass of 28.8 kg. Approximately 52% of the collected species were characiforms, 28% siluriforms, 9% gymnotiforms, 5% cyprinodontiforms, 4% perciforms, and 2% synbranchiforms. The most abundant of the species were the characiforms Astyanax altiparanae (15% of total) and Knodus moenkhausii (12% of total). The two species with the largest overall biomasses were A. altiparanae (34% of total biomass) and the siluriform Hypostomus sp. (8% of total biomass). Analysis of the trophic structure of the studied ichthyofauna indicated that the 10 numerically dominant species across the 24 sampled streams can be grouped into five guilds that are in decreasing order of numerical importance: omnivores, insectivores, insectivores/invertivores, periphytivores, and algivores. Species richness in the sampled stream stretches varied from six to 20 species with an average richness of 14. The species richness estimated by extrapolation for all 24 sampled stream stretches was 67 species. The Characidae are predominant among the collected specimens with approximately 50% of both individuals and biomass, a fact hypothesized to be a function of several attributes typical of the family. Six of the 56 collected species were new to science and six other species are of indefinite taxonomic status and require further analysis in order to determine their identity.

The size distributed composition of ambient aerosols is used to explore seasonal differences in particle chemistry and to show that dry deposition fluxes of soluble species, including important plant nutrients, increase during periods of biomass (sugar cane trash) burning in São Paulo State, Brazil. Measurements were made at a single site centrally located in the State's sugar cane growing region but away from the immediate vicinity of burns, so that the air sampled was representative of the regional background. Calculation of ion equivalent balances showed that during burning periods smaller particles (Aitken and accumulation modes) were more acidic, containing higher concentrations of SO(4)(2-), oxalate, NO(3)(-), HCOO(-), CH(3)COO(-), and Cl(-), but insufficient NH(4)(+) and K(+) to achieve neutrality. Larger particles showed an anion deficit due to the presence of unmeasured ions and comprised resuspended dusts modified by accumulation of nitrate, chloride, and organic anions. Increases of resuspended particles during the burning season were attributed to release of earlier deposits from the surfaces of burning vegetation as well as increased vehicle movement on unsurfaced roads. During winter months the relative contribution of combined emissions from road transport and industry diminished due to increased emissions from biomass combustion and other activities specifically associated with the harvest period. Positive increments in annual particulate dry deposition fluxes due to higher fluxes during the sugar cane harvest were 44.3% (NH(4)(+)), 42.1 % (K(+)), 31.8% (Mg(2+)), 30.4% (HCOO(-)), 12.8% (Cl(-)), 6.6% (CH(3)COO(-)), 5.2% (Ca(2+)), 3.8% (SO(4)(2-)), and 2.3% (NO(3)(-)). Na(+) and oxalate fluxes were seasonally invariant. Annual aerosol dry deposition fluxes (kg ha(-1)) were 0.5 (Na(+)), 0.25 (NH(4)(+)), 0.39 (K(+)), 0.51 (Mg(2+)), 3.19 (Ca(2+)), 1.34 (Cl(-)), 4.47 (NO(3)(-)), 3.59 (SO(4)(2-)), 0.58 (oxalate), 0.71 (HCOO(-)), and 1.38 (CH(3)COO(-)). Contributions of this mechanism to combined aerosol dry deposition and precipitation scavenging (inorganic species, excluding gaseous dry deposition) were 31% (Na(+)), 8% (NH(4)(+)), 26% (K(+)), 63% (Mg(2+)), 66% (Ca(2+)), 32% (Cl(-)), 33% (NO(3)(-)), and 36% (SO(4)(2-)).

Seasonal variability in the major soluble ion composition of atmospheric particulate matter in the principal sugar cane growing region of central São Paulo State indicates that pre-harvest burning of sugar cane plants is an important influence on the regional scale aerosol chemistry. Samples of particulate matter were collected between April 1999 and February 2001 in coarse (> 3.5 mum) and fine (< 3.5 mum) fractions, and analysed for HCOO-, CH3COO-, C2C42-, SO42- . Results indicated that the principal sources of the aerosols investigated NO3-, Cl-, Na+, K+, NH4+, Mg2+ and Ca2+ were local or regional in nature (scale of tens to a few hundreds of km), and that differences between air masses of varying origins were small. Fine particles were typically acidic, containing secondary nitrates, sulphates and organic species. Coarse fraction concentrations were mainly influenced by physical parameters (wind speed, movement of vehicles and surface condition) affecting rates of re-suspension, although secondary nitrate and sulphate were also present in the larger particles.Concentrations of all measured species except sodium and chloride were higher during the burning season. Although concentrations were lower than often found in polluted urban environments, the massive increases during much of the year, due to a single anthropogenic activity (sugar cane burning) are indicative of a very large perturbation of the lower troposphere in the region relative to the natural condition. These aerosols are suspected of promoting respiratory disease. They also represent an important mechanism for the tropospheric transport of species relevant to surface acidification (sulphates, nitrates, ammonium and organic acids) and soil nutrient status (potassium, nitrogen, ammonium, calcium), so their impact on fragile natural ecosystems (following deposition) needs to be considered.
