Theta rhythm consists of an electrophysiological hippocampal oscillation present in mammalian species (4-12 Hz with variations across species). This oscillation is present during active waking and is also prevalent in local field potentials (LFP) during rapid eye movement sleep (REM sleep). Several studies have shown that theta rhythm is important in cognitive tasks and that the medial septum is a key region for its occurrence. The septum sends cholinergic, GABAergic and glutamatergic projections to the hippocampus, which in turn projects axons to the septum. Besides the septum, other regions are involved in regulating theta rhythm, forming a complex network of interactions among brain areas that result in theta rhythm. Optogenetics is a recently developed method that has been widely used in various research areas. It allows us to manipulate the electrical activity of neurons through light stimulation. One of the existing techniques consists in using a viral vector to induce the neuronal expression of ion channels associated with the light-sensitive molecule rhodopsin (e.g. ChR2). Once infected, the neurons become sensitive to light of a particular wavelength. The present M. Sc. research aimed to perform luminous stimulation of the brain in anesthetized and freely behaving animals using chronically implanted electrodes and optical fibers in animals infected with a viral vector for ChR2 expression. Surgical viral injections were performed in the medial septum; histological results confirmed the expression of ChR2 by way of the presence of the eYFP reporter protein in the septum and also in hippocampal processes. Moreover, we performed acute experiments with luminous stimulation of the medial septum and LFP recordings of the septum and hippocampus of anesthetized animals. Action potentials were recorded in the septum. In these experiments we observed a significant increase in the firing rates of septal neurons during luminous stimulation (n = 300 trials). Furthermore, we found an early light-evoked response in the hippocampal LFP. Chronic experiments with luminous stimulation of the medial septum and hippocampus in freely behaving animals were also performed in combination with LFP recordings. We found that the luminous stimulation of the septum is able to induce theta rhythm in the hippocampus. Together, the results demonstrate that the luminous stimulation of the medial septum in optogenetically-modified animals causes relevant electrophysiological changes in the septum and the hippocampus.

Nicotine administration in humans and rodents enhances memory and attention, and also has a positive effect in Alzheimer's Disease. The Medial Septum / Diagonal Band of Broca complex (MS/DBB) – a main cholinergic system – massively projects to the hippocampus through the fimbria-fornix, and this pathway is called the septohippocampal pathway. It has been demonstrated that the MS/DBB acts directly on the local field potential (LFP) rhythmic organization of the hippocampus, especially in the rhythmogenesis of Theta (4-8Hz) – an oscillation intrinsically linked to hippocampus mnemonic function. In vitro experiments gave evidence that nicotine applied to the MS/DBB generates a local network Theta rhythm within the MS/DBB. Thus, the present study proposes to elucidate the function of nicotine in the MS/DBB on the septo-hippocampal pathway. In vivo experiments compared the effect of MS/DBB microinfusion of saline (n=5) and nicotine (n=8) on Ketamine/Xylazine anaesthetized mice. We observed power spectrum density in the Gamma range (35 to 55 Hz) increasing in both structures (Wilcoxon Rank-Sum test, p=0.038) but with no change in coherence between these structures in the same range (Wilcoxon Rank-Sum test, p=0.60). There was also a decrease in power of the ketamineinduced Delta oscillation (1 to 3 Hz). We also performed in vitro experiments on the effect of nicotine on membrane voltage and action potential. We patch-clamped 22 neurons in current-clamp mode; 12 neurons were responsive to nicotine, half of them increased firing rate and other 6 decreased, and they significantly differed in action potential threshold (-47.3±0.9 mV vs. -41±1.9 mV, respectively, p=0.007) and halfwidth time (1.6±0.08 ms vs. 2±0.12 ms, respectively, p=0.01). Furthermore, we performed another set of in vitro experiments concerning the connectivity of the three major neuronal populations of MS/DBB that use acetylcholine, GABA or glutamate as neurotransmitter. Paired patch-clamp recordings found that glutamatergic and GABAergic neurons realize intra-septal connections that produce sizable currents in MS/DBB postsynaptic neurons. The probability of connectivity between different neuronal populations gave rise to a MS/DBB topology that was implemented in a realistic model, which corroborates that the network is highly sensitive to the generation of Gamma rhythm. Together, the data available in the full set of experiments suggests that nicotine may act as a cognitive enhancer, by inducing gamma oscillation in the local circuitry of the MS/DBB.

A presente pesquisa visa entender os fatores que explicam a manutenção dos aspectos artesanais no extrativismo da cera de carnaúba em Limoeiro do Norte. Justificamos a exploração desse tema pelo fascínio do autor pelo tema e por ser esse recurso ainda uma importante fonte de renda para produtores e trabalhadores rurais no respectivo município. Objetivamos nessa pesquisa identificar as razões pelas quais a atividade mantém seu padrão tecnológico intacto. Primeiro objetivamos uma descrição do processo produtivo da cera, tanto no passado, quanto no presente, comparando estes momentos e ressaltando o caráter artesanal da atividade, além de analisar a importância atual da atividade para aqueles que ainda lidam com esse tipo de produção, bem como procuramos revelar estrutura histórica e institucional que permeia a lógica e as escolhas destes produtores, a qual pode explicar a preservação destes aspectos artesanais. Utilizando o referencial teórico do institucionalismo, explicamos que a preservação destes aspectos artesanais, ocorre em virtude da prevalência de determinadas instituições, como a memória coletiva cultivada entre os rendeiros idosos e resistentes, a concentração fundiária, responsável pelo arrendamento e também as restritas possibilidades de investimento e poupança no processo produtivo, em face do baixo nível de renda de produtores, os quais com um produto pouco competitivo no mercado, pouco podem fazer para inovar tecnologicamente.

Epilepsies are neurological disorders characterized by recurrent and spontaneous seizures due to an abnormal electric activity in a brain network. The mesial temporal lobe epilepsy (MTLE) is the most prevalent type of epilepsy in adulthood, and it occurs frequently in association with hippocampal sclerosis. Unfortunately, not all patients benefit from pharmacological treatment (drug-resistant patients), and therefore become candidates for surgery, a procedure of high complexity and cost. Nowadays, the most common surgery is the anterior temporal lobectomy with selective amygdalohippocampectomy, a procedure standardized by anatomical markers. However, part of patients still present seizure after the procedure. Then, to increase the efficiency of this kind of procedure, it is fundamental to know the epileptic human brain in order to create new tools for auxiliary an individualized surgery procedure. The aim of this work was to identify and quantify the occurrence of epilepticform activity -such as interictal spikes (IS) and high frequency oscillations (HFO) - in electrocorticographic (ECoG) signals acutely recorded during the surgery procedure in drug-resistant patients with MTLE. The ECoG recording (32 channels at sample rate of 1 kHz) was performed in the surface of temporal lobe in three moments: without any cortical resection, after anterior temporal lobectomy and after amygdalohippocampectomy (mean duration of each record: 10 min; N = 17 patients; ethic approval #1038/03 in Research Ethic Committee of Federal University of São Paulo). The occurrence of IS and HFO was quantified automatically by MATLAB routines and validated manually. The events rate (number of events/channels) in each recording time was correlated with seizure control outcome. In 8 hours and 40 minutes of record, we identified 36,858 IS and 1.756 HFO. We observed that seizure-free outcome patients had more HFO rate before the resection than non-seizure free, however do not differentiate in relation of frequency, morphology and distribution of IS. The HFO rate in the first record was better than IS rate on prediction of seizure-free patients (IS: AUC = 57%, Sens = 70%, Spec = 71% vs HFO: AUC = 77%, Sens = 100%, Spec = 70%). We observed the same for the difference of the rate of pre and post-resection (IS: AUC = 54%, Sens = 60%, Spec = 71%; vs HFO: AUC = 84%, Sens = 100%, Spec = 80%). In this case, the algorithm identifies all seizure-free patients (N = 7) with two false positives. To conclude, we observed that the IS and HFO can be found in intra-operative ECoG record, despite the anesthesia and the short time of record. The possibility to classify the patients before any cortical resection suggest that ECoG can be important to decide the use of adjuvant pharmacological treatment or to change for tailored resection procedure. The mechanism responsible for this effect is still unknown, thus more studies are necessary to clarify the processes related to it

In the present work, we investigated behavioral changes associated with the increase in Zif268 protein expression within telencephalic areas of the tropical lizard Tropidurus hispidus that correspond to the mammalian hippocampus (HC). We used 13 male individuals of this species, collected at the Federal Agrotechnical School of Rio Grande do Norte, under SISBIO license number 19561-1. Four animals had their brains removed and were submitted to a Western blot with antibodies for the Zif268 protein. The remaining animals were separated in two different groups: a control group (n=4) and an exploration group (n=5). Animals from the exploration group were exposed to an enriched environment with many sensory cues novel to them. Control group animals stayed in the environment they were already habituated to. After 90 min from the onset of exposure to the new environment, animals from both groups were submitted to intracardiac perfusion with fixative, and the brains were removed, cryoprotected and frozen. After that, brains were sectioned at 20 μm and the sections were subjected to immunohistochemistry for the Zif268 protein. We verified that the Zif268 protein is likely conserved in the brain of T. hispidus, which showed antigenicity for the antibody anti-Zif268 made in mammals. In animals from the exploration group, we detected an increase of the Zif268 protein in the Septum, Striatum, Dorsoventricular Area and in cortical areas corresponding to the HC. This increase was proportional to the amount of environmental exploration, with maximum positive correlation in the hippocampal subareas Medial Cortex (R = 0.94 and p = 0.004) and Dorsomedial Cortex (R = 0.92 and p = 0.006). The data corroborate the notion that the reptilian hippocampus, as well as the mammalian HC, plays an important role in spatial exploration.

Ce travail a comme objectif analyser la structure et l'efficacité d'un système symbolique, qui est configuré à travers qui le philosophe Dufour (2000), à partir du linguiste Benveniste (1995) et du philosophe Lyotard (1998), dans la manutention et la sustentation des récits et d'histoires, appelle de trinité naturel des la langue. C'est sous cet appareil, inscrit à travers des pronomes « moi », « toi » et « lui » qui nous étudierons les suivantes questions: 1. comme s'il établit, synchronement, la mise à jour du mythe religieux Santa Menina présente dans la ville de Florânia, gêné dans le coeur broussard du Rio Grande do Norte? 2. qui manière, diachronement, les récits sur mentionné mythe s´il récupère? Le culte religieux à la Santa Menina est marqué discoursivement à travers d´un univers à facettes multiples qui se soutient à travers les voix de floranienses sur l'histoire d'une fille, dont le corps est trouvé intact et qui, sacralitée par la population, est connu comme la Santa Menina. Une histoire qui, notamment, avait/a répercussion et résonance, en participant, efficacement, des la réalité culturelle des la population. Théoriquement, notre travail est nourri par des savoirs arrivés, des la Linguistique, et des études sur la Mémoire et l'Histoire Verbale

Taking into account the Environmental Restorative Theory (ERT) , created by Fre derick Law Olmsted in mid XIX ce ntury , according which, urban parks can contribute to solve problems arisen from crowding , particularly urban stress, we analize how the ERT arrived at XXI century, having as approach the evaluation of New York Central Park (CP). Considering that the CP and the cinema were born around at the same, we question if the North American cinema produced between 1960 and 2013 show the ideals, which engendered the CP. By answering this question we defend the hypothesis that, even though has existed adjusts and modificati ons in the CP plan through time, it kept reasonably faithful to the ERT premises, propitiating to the XX and XXI centuries cinema identify and bring forth the presence of the Olmsted’s Ideals in the present days. The thesis main objective was nonetheless u nderstand similarities and/or differences between the XIX century ideals (that gave birth to CP) and the way the cinema represents the present uses of the place, taking into account that the Olmstedian ERT proposal have survived to the context changes (soc ial, economic, political and cultural). Methodologically , we drew upon bibliographical and documental analysis to build the first chapters and to the cinema as analytical lenses to investigate the ERT. The results point that although the CP plan has kept r elatively intact and faithful to the ERT – with the presence of natural elements in the films (notably vegetation and water) – many of contemporary behaviors were not foreseen previously, especially in relation to sports practice, the massive feminine pres ence, as well as criminality.
