Analisa as prticas de mediação desenvolvidas nas bibliotecas universitrias pelos bibliotecrios diante das tecnologias digitais. Para tanto estabelece como objetivo geral analisar de forma comparativa, o impacto e mediação das tecnologias digitais no funcionamento de bibliotecas Universitrias de Portugal e da regio Nordeste do Brasil. Integraram esta pesquisa 10 universidades federais brasileiras e 12 universidades pblicas portuguesas, com um total geral de 115 bibliotecrios, que so os sujeitos participantes. uma pesquisa qualitativa que adota o mtodo quadripolar recomendado para os trabalhos desenvolvidos no mbito das Cincias Sociais Aplicadas, e em especial na rea de Cincia da Informao. Atravs da interao entre os polos: epistemolgico, terico, tcnico e morfolgico, que fundamentam este mtodo, houve o fortalecimento e a fluidez das questes estudadas. Os resultados dos questionrios aplicados aos bibliotecrios, bem como da anlise dos stios das bibliotecas pesquisadas, foram interpretados atravs de um alicerce terico baseado em trs pontos principais: as questes paradigmticas que envolvem a rea de Cincia da Informao, a anlise da mediação ps-custodial informacional e cientfica e as Tecnologias de Informao e Comunicao presentes nas bibliotecas. Como principais resultados vemos que o impacto das tecnologias digitais nas bibliotecas universitrias considerado pelos bibliotecrios brasileiros e portugueses como positivo, com nfase em dois pontos: a inovao dos suportes de informao e a autossuficincia dos utilizadores. A maior diferena se percebe em relao ao aspecto social, atravs de uma maior preocupao entre os bibliotecrios brasileiros com as barreiras informacionais causadas por questes econmica, social e educacional e sentido com menos intensidade pelos bibliotecrios portugueses, que ascendem as tecnologias digitais com mais facilidade. De forma conclusiva, a anlise do impacto e a mediação das tecnologias digitais nas bibliotecas pesquisadas, apontam para uma evoluo nas prticas mediadoras das bibliotecas universitrias de Portugal e do Nordeste do Brasil e uma convergncia laboral entre os bibliotecrios portugueses e brasileiros.

A pesquisa aborda o uso das Tecnologias de Informao e Comunicao, que vem revolucionando as atividades e ocasionando muitas mudanas relacionadas ao acesso e uso de informaes. O objetivo foi analisar o grau de utilizao do conhecimento cientfico produzido pelos Programas de Ps-Graduao das Universidades Pblicas Brasileiras, atravs da BDTD, pelos mestrandos dos referidos programas. Nos procedimentos metodolgicos realizados, procurou-se inicialmente analisar o amplo espectro da populao do corpus da pesquisa. Em razo da impossibilidade de trabalhar com os Programas de Ps-Graduao como um todo, optou-se por fazer um recorte, elegendo os cursos de Ps-Graduao em Cincia da Informao, vez que estes representam o principal segmento social de interesse da pesquisa. Foi utilizado o mtodo de estudo de usurios, onde se optou por adotar o grupo, estudos orientados aos usurios, que identifica as necessidades e comportamento de acesso e uso da informao. Para coletar os dados, elaborou-se um questionrio semi-estruturado com 25 questes, que versavam sobre o uso, dificuldades de acesso e recuperao da informao, bem como a satisfao na utilizao dessa fonte informacional. Dentre os vrios resultados obtidos, podemos destacar o fato de que a maioria dos mestrandos (71,8%) s teve contato com a BDTD somente no momento em que se encontrava cursando o mestrado e, somente 24,3%, tiveram contato durante a graduao. Estes resultados representam um problema, que pode afetar o bom desempenho do projeto BDTD, o qual consiste em disseminar e divulgar a produo cientfica dos Programas de Ps-Graduao das Universidades Pblicas Brasileiras para a sua comunidade. Foi observado tambm, que os mestrandos oriundos do curso de Biblioteconomia tende a ter contato com a BDTD bem mais cedo do que mestrandos de outros cursos de graduao. A fim de minimizar o problema detectado, prope-se uma divulgao mais eficaz na graduao atravs de dois procedimentos: Primeiro, o docente deve fazer uma divulgao mais eficaz da BDTD junto aos discentes de todos os cursos de graduao; segundo: dever ser feita a divulgao na mdia eletrnica, atravs da insero de cones da BDTD, nos portais dos Departamentos dos Cursos de Graduao das Universidades Pblicas Brasileiras. Acredita-se que com estes procedimentos seja possvel aperfeioar o uso dessa fonte de informao cientfica.

Apresenta uma breve evoluo da comunicao cientfica ao longo do tempo; discute os impactos causados pela Internet na comunicao, disponibilizao e acessibilidade de informaes tcnico-cientficas; e caracteriza BDTD destacando os desafios a serem superados. Dentre eles esto, a questo dos direitos autorais que ocasionam a baixa adeso dos autores ao projeto piloto do IBICT para disponibilizar teses e dissertaes no meio eletrnico. Para este desafio, so apontadas sugestes.

As bibliotecas de teses e dissertaes tm um papel fundamental para o desenvolvimento cientfico e cultural de um pas. Nesse sentido, buscou-se, de modo geral, analisar o panorama brasileiro das bibliotecas digitais de teses e dissertaes. Especificamente, objetivou-se caracterizar a sociedade da informao enquanto contexto das BDTD; caracterizar os vrios tipos de bibliotecas surgidos a partir da insero das novas tecnologias de informao e comunicao; identificar as diretrizes norteadoras da implantao de BDTD; e levantar as BDTD existentes no Brasil. Para tato, alm de levantamento bibliogrfico, realizou-se uma pesquisa nos sites de instituies de ensino superior brasileiras que j dispe de bibliotecas digitais de teses e dissertaes em funcionamento. A anlise e interpretao destes dados nos permite considerar que no Brasil as BDTD encontram-se num estgio embrionrio, sendo relevante pesquisar a histrica implantao e configurao desse novo modelo de biblioteca.

Galactans are polysaccharides sulfated present in the cell wall of red algae. Carrageenans are galactans well known in the food industry as gelling polysaccharides and for induce inflammatory process in rodents as animal model. The extraction of polysaccharides from A. multifida has been carried out by proteolysis and precipitation in different volumes of acetone, which produced three fractions (F1, F2, and FT). Chemical and physical analyses revealed that these fractions are sulfated galactan predominantly. Results of the antioxidant activity assays showed that all of these fractions have antioxidant activity and that was associated with sulfate content of the analysis of reducing power and total antioxidant capacity. However, these fractions were not effective against lipid peroxidation. The fraction FT presented higher activity on the APTT test at 200 μg (> 240 s). The assessment of the hemolytic activity showed that the FT fraction has the best activity, increasing lyses by the complement system to 42.3% (50 μg) (p< 0,001). The fraction FT showed the best yield, anticoagulant and hemolytic activity between the three fractions and therefore it was choose for the in vivo studies. The Inflammation assessment using the FT fraction (50 mg / kg MB) showed that the cellular migration and the IL-6 production increased 670.1% (p< 0,001) and 531.8% (p< 0,001), respectively. These results confirmed its use as an inflammation inducer in animal model. Cytotoxicity assay results showed that all fractions have toxic effects on 3T3 and HeLa cells after exposition of 48 hours, except when 100 μg for both F1 and FT were used. These results arise the discussion whether these polysaccharides it should be used as additive in foods, cosmetics and medicines.

Cancer is a term used to represent a set of more than 100 diseases, including malignant tumors from different locations. The malignancies are the second leading cause of death in the population, representing approximately 17% of deaths of known cause. Strategies that induce differentiation have had limited success in the treatment of established cancers. In this work, a lectin purified from the marine sponge Cinachyrella apion (CaL) was evaluated due to its hemolytic, cytotoxic and antiproliferative properties, besides the ability to induce cell death via apoptosis in tumor cells. The antiproliferative activity of CaL was tested against cell lines, with the highest inhibition of tumor growth for HeLa, reducing cell growth at a dose dependent manner, with a concentration of 10 g/mL. The hemolytic activity and toxicity against peripheral blood cells were tested using the concentration of IC50 for both trials and twice the IC50 for analysis in flow cytometry, indicating that CaL is not toxic to these cells. To assess the mechanism of cell death caused by CaL in HeLa cells, we performed flow cytometry and western blotting. The results showed the lectin probably induces cell death by apoptosis activation by pro-apoptotic protein Bax, promoting mitochondrial membrane permeabilization, cell cycle arrest in S phase, with accumulation of cells of approximately 57% in this phase, and acting as both dependent and/or independent of caspases pathway. These results suggest that CaL has the potential to be used as drug treatment against cancer.

Heparin is a pharmaceutical animal widely used in medicine due to its potent anticoagulant effect. Furthermore, it has the ability to inhibit the proliferation, invasion and adhesion of cancer cells to vascular endothelium. However, its clinical applicability can be compromised by side effects such as bleeding. Thus, the search for natural compounds with low bleeding risk and possible therapeutic applicability has been targeted by several research groups. From this perspective, this study aims to evaluate the hemorrhagic and anticoagulant activities and citotoxic effect for different tumor cell lines (HeLa, B16-F10, HepG2, HS-5,) and fibroblast cells (3T3) of the Heparin-like from the crab Chaceon fenneri (HEP-like). The HEP-like was purified after proteolysis, ion-exchange chromatography, fractionation with acetone and characterized by electrophoresis (agarose gel) and enzymatic degradation. Hep-like showed eletroforetic behavior similar to mammalian heparin, and high trisulfated /Nacetylated disaccharides ratio. In addition, HEP-like presented low in vitro anticoagulant activity using aPTT and a minor hemorrhagic effect when compared to mammalian heparin. Furthermore, the HEP-like showed significant cytotoxic effect (p<0.001) on HeLa, HepG2 and B16-F10 tumor cells with IC50 values of 1000 ug/mL, after incubation for 72 hours. To assess the influence of heparin-like on the cell cycle in HeLa cells, analysis was performed by flow cytometry. The results of this analysis showed that HEP-like influence on the cell cycle increasing S phase and decreasing phase G2. Thus, these properties of HEP-like make these compounds potential therapeutic agents

This work addresses the production of lightweight concrete building elements, such as plates, prefabricated slabs for pre-molded and panels of fencing, presenting a singular concrete: the Lightweight Concrete, with special properties such low density and good strength, by means of the joint use of industrial waste of thermosetting unsaturated polyesters and biodegradable foaming agent, named Polymeric Lightweight Concrete. This study covered various features of the materials used in the composition of the Polymeric Lightweight Concrete, using a planning of factorial design 23, aiming at studying of the strength, production, dosage processes, characterization of mechanical properties and microstructural analysis of the transition zone between the light artificial aggregate and the matrix of cement. The results of the mechanical strength tests were analyzed using a computational statistics tool (Statistica software) to understand the behavior and obtain the ideal quantity of each material used in the formula of the Polymeric Lightweight Concrete. The definition of the ideal formula has the purpose of obtaining a material with the lowest possible dry density and resistance to compression in accordance with NBR 12.646/92 (≥ 2.5 MPa after 28 days). In the microstructural characterization by scanning electron microscopy it was observed an influence of the materials in the process of cement hydration, showing good interaction between the wrinkled face of the residue of unsaturated polyesters thermosetting and putty and, consequently, the final strength. The attaining of an ideal formula, given the Brazilian standards, the experimental results obtained in the characterization and comparison of these results with conventional materials, confirmed that the developed Polymeric Lightweight Concrete is suitable for the production of building elements that are advantageous for construction

Seaweeds are organisms known to exhibit a variety of biomolecules with pharmacological properties. The coast of Rio Grande do Norte has over 100 species of seaweeds, most of them not yet explored for their pharmacological potential. Sugars and phenolic compounds are the most studied of these being assigned a range of biological properties, such as anticoagulant , antiinflammatory, antitumor and antioxidant activities. In this work, we obtained methanolic extracts from thirteen seaweeds of the coast of Rio Grande do Norte (Dictyota cervicornis; Dictiopterys delicatula; Dictyota menstruallis; D. mertensis; Sargassum filipendula; Spatoglossum schrederi; Acanthophora specifera; Botryocladia occidentalis; Caulerpa cupresoides; C. racemosa; C. prolifera; C. sertularioides e Codium isthmocladum). They were evaluated as anticoagulant and antioxidant drugs, as well as antiproliferative drugs against the tumor cell line HeLa. None of the methanolic extracts showed anticoagulant activity, but when they were evaluated as antioxidant drugs all of extracts showed antioxidant activity in all tests performed (total antioxidant capacity, sequestration of superoxide and hydroxyl radicals, ferric chelation and reductase activity), especially the algae D. mentrualis, D. cilliolata and C. prolifera, who had the greatest potential to donate electrons.In addition, the ability of iron ions chelation appears as the main antioxidant mechanism of the methanolic extracts of these seaweeds mainly for the extract of the C. racemosa seaweed, which reached almost 100% activity. In the MTT assay, all extracts showed inhibitory activity at different levels againts HeLa cells. Moreover, D. cilliolata (MEDC) and D. menstrualis (MEDM) extracts showed specific activity to this cell line, not inhibiting the viability of 3T3 normal cell line, so they were chosen for detailing the antiproliferative mechanism of action. Using flow cytometry, fluorescence microscopy and in vitro assays we demonstrated that MEDC and MEDM induced apoptosis in HeLa cells by activation of caspases 3 and 9 and yet, MEDC induces cell cycle arrest in S phase. Together, these results showed that the methanolic extracts of brown seaweed D. menstrualis and D. cilliolata may contain agents with potential use in combatting cells from human uterine adenocarcinoma. This study also points to the need for more in-depth research on phytochemical and biological context to enable the purification of biologically active products of these extracts

Bacterial cellulose (BC) has a wide range of potential applications, namely as temporary substitute skin in the treatment of skin wounds, such as burns, ulcers and grafts. Surface properties determine the functional response of cells, an important factor for the successful development of biomaterials. This work evaluates the influence of bacterial cellulose surface treatment by plasma (BCP) on the cellular behavior and its genotoxicity potential. The modified surface was produced by plasma discharge in N2 and O2 atmosphere, and the roughness produced by ion bombardment characterized by scanning electron microscopy (SEM) and atomic force microscopy (AFM). Cell adhesion, viability and proliferation on BCP were analysed using crystal violet staining and the 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl-tetrazolium (MTT) method. Genotoxicity was evaluated using the comet and cytokinesis block micronucleus assay. The results show that the plasma treatment changed surface roughness, producing an ideal cell attachment, evidenced by more elongated cell morphology and improved proliferation. The excellent biocompatibility of BCP was confirmed by genotoxicity tests, which showed no significant DNA damage. The BCP has therefore great potential as a new artificial implant

Visceral leishmaniasis (VL) is a disease caused by protozoa of the Leishmania donovani complex, whose infection has clinical spectrum ranging from asymptomatic infection to active disease characterized by fever, cachexia, hepatosplenomegaly, and immunosuppression. The healing or protective immunity require an antigen-specific type 1. The Montenegro skin test (DTH) has been interpreted as a marker of protective immunity. However, there is no known correlation between the DTH response to type 1 and DTH and immunity of type 1 are maintained in the long term. Thus, a longitudinal study of 8 years, nested in a cohort family held in Brazil, documented the status of DTH and cytokine production by peripheral blood mononuclear cells in response to antigen-specific stimulation. This study was the interdisciplinary approach of physicians, biochemists, nutritionists, veterinary medicine, biology and statistics. The results show that 46.2% of subjects were analyzed DTH positive at baseline. The prevalence of positive and DTH induration size increased with age (p = 0.0021). 15.7% of individuals positive DTH "retro-converted" the negative and 50.4% (64) of individuals negative DTH became positive. The size of DTH induration was correlated significantly with the antigen-induced production of IFN-γ (r = 0.6186, p = 0.0001). IL-6 was secreted at higher levels in peripheral blood mononuclear cells of individuals who "retro-converted" DTH positive to negative than individuals who remained stable DTH status (p = 0.005). Thus, IFN-γ produced by peripheral blood mononuclear cells, may be a surrogate marker for protective immunity instead of the DTH response. In addition, differences in innate immune response may determine whether individuals maintain or eliminate the infection by Leishmania infantum chagasi in asymptomatic patients

A resistncia microbiana a antimicrobianos tem favorecido a busca por substncias bioativas provenientes de plantas usadas na medicina popular, com o intuito de se obter novos frmacos com atividade antimicrobiana. Neste estudo, foi proposta a investigao da atividade antibacteriana do leo-resina de Copaifera duckei e de diferentes extratos da casca de Pseudobombax marginatum, e seus possveis mecanismos de ao. O potencial inibitrio antibacteriano foi avaliado utilizando-se os mtodos de difuso e diluio em gar, e a bioautografia. O mecanismo de ao foi analisado por microscopia eletrnica, no qual se observou alteraes na ultraestrutura bacteriana, e por eletroforese em SDS-PAGE, que determinou ao sobre as protenas das superfcies celulares. A anlise qumica foi realizada pelas tcnicas de Espectrometria de massas acoplada ao Cromatgrafo a gs- EM/CG (C. duckei) e Cromatografia Lquida de Alta Eficincia- CLAE (P. marginatum). Entre as bactrias estudadas, B. cereus foi a mais suscetvel s plantas em estudo, com concentraes inibitrias mnimas (CIMs) correspondentes a 0,3125 mg/mL para o leo-resina de copaba, e 0,5 mg/mL para extrato hidroalcolico (1:1) e 0,512 mg/mL para a frao butanlica da casca P. marginatum, nos quais pde-se observar alteraes na parede celular do B. cereus, com remoo da camada S, espessamento da parede celular e formao de diversos septos nos centros de diviso celular. A anlise qumica por EM/CG mostrou compostos terpnicos no leo-resina de C. duckei, tendo como composto majoritrio o β-bisaboleno, e a anlise por CLAE mostrou a presena de compostos derivados da catequina na casca do P. marginatum. Desta forma, as plantas em estudo mostram um potencial antibacteriano considervel, podendo contribuir tanto na terapia antimicrobiana como na rea de alimentos, tendo como um de seus provveis stios de ao a parede celular bacteriana
