Metabolic flux analysis (MFA) is a powerful tool for analyzing cellular metabolism. In order to control the growth conditions of a specific organism, it is important to have a complete understanding of its MFA. This would allowed us to improve the processes for obtaining products of interest to human and also to understand how to manipulate the genome of a cell, allowing optimization process for genetic engineering. Streptomyces olindensis ICB20 is a promising producer of the antibiotic cosmomycin, a powerful antitumor drug. Several Brazilian researchers groups have been developing studies in order to optimize cosmomycin production in bioreactors. However, to the best of our knowledge, nothing has been done on metabolic fluxes analysis field. Therefore, the aim of this work is to identify several factors that can affect the metabolism of Streptomyces olindensis ICB20, through the metabolic flux analysis. As a result, the production of the secondary metabolite, cosmomycin, can be increased. To achieve this goal, a metabolic model was developed which simulates a distribution of internal cellular fluxes based on the knowledge of metabolic pathways, its interconnections, as well as the constraints of microorganism under study. The validity of the proposed model was verified by comparing the computational data obtained by the model with the experimental data obtained from the literature. Based on the analysis of intracellular fluxes, obtained by the model, an optimal culture medium was proposed. In addition, some key points of the metabolism of Streptomyces olindensis were identified, aiming to direct its metabolism to a greater cosmomycin production. In this sense it was found that by increasing the concentration of yeast extract, the culture medium could be optimized. Furthermore, the inhibition of the biosynthesis of fatty acids was found to be a interesting strategy for genetic manipulation. Based on the metabolic model, one of the optimized medium conditions was experimentally tested in order to demonstrate in vitro what was obtained in silico. It was found that by increasing the concentration of yeast extract in the culture medium would induce to an increase of the cosmomycin production

In this work we will analyze the function of the city of Currais Novos-RN front of four cities in its hinterland in the process of globalization, now set. Such cities are, Acari, São Vicente, Lagoa Nova and Cerro Corá. We take the theoretical and territory division of labor that result while providing the articulation of cities in urban networks, and the tertiary sector, which holds the means for the articulation. Beyond the historical analysis and secondary database, the analysis of flows of materials and people between the cities above would, lay a greater diagnostic functions of the regional center of Currais Novos, and escape of the search as much of the spread of products

Experiments were performed to study the effect of surface properties of a vertical channel heated by a source of thermal radiation to induce air flow through convection. Two channels (solar chimney prototype) were built with glass plates, forming a structure of truncated pyramidal geometry. We considered two surface finishes: transparent and opaque. Each stack was mounted on a base of thermal energy absorber with a central opening for passage of air, and subjected to heating by a radiant source comprises a bank of incandescent bulbs and were performed field tests. Thermocouples were fixed on the bases and on the walls of chimneys and then connected to a data acquisition system in computer. The air flow within the chimney, the speed and temperature were measured using a hot wire anemometer. Five experiments were performed for each stack in which convective flows were recorded with values ranging from 17 m³ / h and 22 m³ / h and air flow velocities ranging from 0.38 m / s and 0.56 m / s for the laboratory tests and air velocities between 0.6 m/s and 1.1m/s and convective airflows between 650 m³/h and 1150 m³/h for the field tests. The test data were compared to those obtained by semi-empirical equations, which are valid for air flow induced into channels and simulated data from 1st Thermodynamics equation. It was found that the chimney with transparent walls induced more intense convective flows than the chimney with matte finish. Based on the results obtained can be proposed for the implementation of prototype to exhaust fumes, mists, gases, vapors, mists and dusts in industrial environments, to help promote ventilation and air renewal in built environments and for drying materials, fruits and seeds

The Exception Handling (EH) is a widely used mechanism for building robust systems. In Software Product Line (SPL) context it is not different. As EH mechanisms are embedded in most of mainstream programming languages (like Java, C# and C++), we can find exception signalers and handlers spread over code assets associated to common and variable SPL features. When exception signalers and handlers are added to an SPL in an unplanned way, one of the possible consequences is the generation of faulty family instances (i.e., instances on which common or variable features signal exceptions that are mistakenly caught inside the system). In this context, some questions arise: How exceptions flow between the optional and alternative features an LPS? Aiming at providing answers to these questions, this master thesis conducted an exploratory study, based on code inspection and static analysis code, whose goal was to categorize the main ways which exceptions flow in LPSs. To support the study, we developed an static analysis tool called PLEA (Product Line Exception Analyzer) that calculates the exceptional flows of LPSs, and categorize these flows according to the features associated with handlers and signalers. Preliminary results showed that some types of exceptional flows have more potential to yield failures in exceptional behavior of SLPs

This thesis analyzes another side of Potiguar tourism , the unplanned side, neglected and kept out of touristic activities: excursionism, a leisure practice enjoyed by tourists with low consumer power, and who are commonly referred by the pejorative term farofeiros (picnic lovers). The geographic research sites considered for this study include Arituba, Boágua and Carcará lakes in Nísia Florest, Rio Grande do Norte, where on Sundays and holidays the arrival of hundreds of excursionists, from around the metropolitan region of Natal, from surrounding municipalities, and neighboring States, such as Paraíba and Pernambuco, can be observed. The objective of this study is to analyze the appropriation of the physical site by the practice of excursionism, focusing on its relation to other social agents that also appropriate a designated touristic area. The theoretical discussion considers the use of the space by the touristic leisure practice and the appropriation by distinct social agents, using categories of analysis, such as, production of the space, territory and leisure. The field work was completed with interviews and questionnaires administered to excursionists, excursion organizers, local merchants, representatives of the public setor from the municipalities, and professional dune buggy drivers; besides this, photos, informal dialogue and field observations were important methodological instruments used. From the data, statistical analysis and the development of thematic maps demonstrating the established flux between excursionists and the segregated activity were done. With this research, one can affirm that the practice of excursionism is neglected by the public sector, contrary to the intention of the hegemonic agent‟s intentionality present in this touristic territory which aim at the development of a lucrative activity, geared toward tourists with greater spending power. This ignored and neglected faction of Potiguar tourism is considered poor or dirty , and generate conflicts among the distinct social agents: tourists, the market and the public sector, simultaneously peaking interest, which is then appropriated by the informal sector and formal economy. Excursionism is an expressive phenomenon, a socially relevant practice, enjoyed by citizens of the working class who, in order to have a day of leisure, use alternative consumer practices and subvert various strategies of segregation that are imposed within these tourist areas, behavior that, in part, justifies the nickname, picnic lovers , given to these tourists

In recent decades the production of irrigated fruits destined to supply the foreign market reveals itself as an expression of the thriving agricultural modernization, consisting an important and dynamic segment of Brazilian agribusiness, especially in the Northeast. The Rio Grande do Norte contributes to this scenario through the production of several tropical fruit, especially melon production in the municipalities of Baraúna/RN and Mossoró/ RN. Accordingly, the overall goal of the study is to understand the agricultural use of the territory of Rio Grande do Norte by irrigated orcharding from the circuit productive space configuration of melon. The outlook presented in the work on the irrigated agriculture developed in Rio Grande do Norte reveals that it falls within an agricultural division of labor, through productive specialization of places that have natural and technical conditions favorable to the exploitation and reproduction of capitals. The land use caused by circuit productive space melon enables flows of tangible and intangible consolidate a extraverted network of production and consumption in order to meet the requirements of foreign markets
