The expansion of cultivated areas with genetically modified crops (GM) is a worldwide phenomenon, stimulating regulatory authorities to implement strict procedures to monitor and verify the presence of GM varieties in agricultural crops. With the constant growing of plant cultivating areas all over the world, consumption of aflatoxin-contaminated food also increased. Aflatoxins correspond to a class of highly toxic contaminants found in agricultural products that can have harmful effects on human and animal health. Therefore, the safety and quality evaluation of agricultural products are important issues for consumers. Lateral flow tests (strip tests) is a promising method for the detection both proteins expressed in GM crops and aflatoxins-contaminated food samples. The advantages of this technique include its simplicity, rapidity and cost-effective when compared to the conventional methods. In this study, two novel and sensitive strip tests assay were developed for the identification of: (i) Cry1Ac and Cry8Ka5 proteins expressed in GM cotton crops and; (ii) aflatoxins from agricultural products. The first strip test was developed using a sandwhich format, while the second one was developed using a competitive format. Gold colloidal nanoparticles were used as detector reagent when coated with monoclonal antibodies. An anti-species specific antibody was sprayed at the nitrocellulose membrane to be used as a control line. To validate the first strip test, GM (Bollgard I® e Planta 50- EMBRAPA) and non-GM cotton leaf (Cooker 312) were used. The results showed that the strip containing antibodies for the identification of Cry1Ac and Cry8Ka5 proteins was capable of correctly distinguishing between GM samples (positive result) and non-GM samples (negative result), in a high sensitivity manner. To validate the second strip test, artificially contaminated soybean with Aspergillus flavus (aflatoxin-producing fungus) was employed. Food samples, such as milk and soybean, were also evaluated for the presence of aflatoxins. The strip test was capable to distinguish between samples with and without aflatoxins samples, at a sensitivity concentration of 0,5 μg/Kg. Therefore, these results suggest that the strip tests developed in this study can be a potential tool as a rapid and cost-effective method for detection of insect resistant GM crops expressing Cry1Ac and Cry8Ka5 and aflatoxins from food samples.

Eutrophication is the most common cause of water quality degradation in the world. This process occurs by excessive nutrients inputs, nitrogen and phosphorus, to the aquatic systems resulting in algal and cyanobacterial blooms. In shallow lakes these effects are pronounced due to the higher interaction of the lake with watershed, air and sediment. There are innumerous restoration techniques of eutrophied lakes with a range of successful results but there is only one case of successful lake restoration in Brazil: Paranoá Lake in Brasília city. The Brazilian semiarid region has many artificial lakes, named açudes, which are mostly eutrophic and shallow lakes. The eutrophication in these lakes is reported and the phytoplankton community is dominated by potentially toxic cyanobacteria species, mainly Cylindrospermopsis raciborskii. The aim of this thesis is to test techniques for water quality management which can be easily applied in Brazilian semiarid lakes. Results from a laboratory experiment suggest that the addition of a phosphorus sorbent clay associated with an aluminium based coagulant is an effective technique in removing soluble reactive phosphorus and reducing C. raciborskii growth rate – cyanobacteria potentially toxic dominant in reservoirs of Brazilian semiarid – but this effect is dependent on the biomass in the application moment. Results from a field experiment in mesocosm in a eutrophied lake showed that the addition of aluminium based coagulant and removal of benthivorous fish is more efficient in removing total phosphorus and chlorophyll-a from water column than the isolated application of one of the techniques. Lastly, laboratory tests showed that aluminium based coagulant exhibited good performance in removing turbidity and total phosphorus from water of six reservoirs but the efficiency was reduced by algal biomass and pH. The results of this study showed that the improvement in water quality of eutrophied reservoirs in semiarid region is possible through internal loading control by phosphorus precipitation and inactivation in sediments or inhibition of phosphorus release by benthivorous fishes, and also that these results show are additives in water quality improvement.

Introduction: This study aimed to investigate the effects of the two peptide NOP partial agonists (UFP-113 and [F/G]N/OFQ(1-13)NH2) and the non peptide NOP partial agonist (AT-090) in the mouse emotional behavior as well as in the intracellular transduction pathways following the receptor binding. Methods: Male Swiss or CD-1 mice were used in this study together with NOP(+/+) and NOP(-/-) mice. The elevated plus maze (EPM) was used to evaluate the effects of compounds on anxiety-like behaviors. Diazepam and the NOP agonists, N/OFQ and Ro 65-6570, were used as positive controls in the EPM. NOP(+/+) and NOP(-/-) mice were used to evaluate the selectivity of those compounds that induced anxiolytic-like behaviors. The forced swim test (FST) was used to evaluate the effects of compounds on depressive-like behaviors. Nortriptyline and the NOP antagonists, UFP-101 and SB-612111, were used as positive controls in the FST. The effects of N/OFQ, UFP-101, SB-612111, UFP-113, [F/G]N/OFQ(1-13)NH2, and AT-090 were assessed in the methylphenidate-induced hyperlocomotion (MIH) test; in this assay valproate was used as positive control. The G protein and β-arrestin 2 transduction pathways of NOP receptor agonists (N/OFQ and Ro 65-6570), antagonist (UFP-101), and partial agonists (UFP-113, [F/G]N/OFQ(1-13)NH2, and AT-090) were also evaluated using an innovative assay that measures a bioluminescence resonance energy transfer process. For this, cell lines permanently co-expressing the NOP receptor coupled to luciferase (energy donor), and green fluorescent protein (energy acceptor) coupled to one of the effector proteins (G protein or β-arrestin 2) were used. Results: Diazepam (1 mg/kg), N/OFQ (1 nmol), Ro 65-6570 (0.1 mg/kg), and AT-090 (0.01 mg/kg) induced anxiolytic-like effect in mice in the EPM. The effects of Ro 65-6570 and AT-090 were selective to NOP receptor. UFP-113 (0.01-1 nmol) and [F/G]N/OFQ(1-13)NH2 (0.1-3 nmol) were inactive in the EPM. In the FST, nortriptyline (30 mg/kg), UFP-101 (10 nmol), SB-612111 (10 mg/kg), UFP-113 (0.01 and 0.1 nmol), and [F/G]N/OFQ(1-13)NH2 (0.3 and 1 nmol) induced antidepressant-like effects, while AT-090 (0.001-0.1 mg/kg) was inactive in this assay. The effects of UFP-113 and [F/G]N/OFQ(1-13)NH2 were selective to NOP receptor. Valproate (400 mg/kg) counteracted methylphenidate (MPH, 10 mg/kg)-induced hyperlocomotion in mice in the open field. N/OFQ (1 nmol), UFP-113 (0.01-0.1 nmol), and [F/G]N/OFQ(1-13)NH2 (1 nmol) were also able to reduce the MPH-induced hyperlocomotion, without changing the locomotor activity per se. The effect of UFP-113 was selective to NOP receptor. The UFP-101 (10 nmol), SB-612111 (10 mg/kg), and AT-090 (0.001-0.03 mg/kg) did not change the hyperlocomotor effect of methylphenidate. In vitro, N/OFQ and Ro 65-6570 behaved as NOP full agonists for G-protein and β-arrestin 2 pathways. AT-090 behaved as NOP receptor partial agonist for both transduction pathways, while UFP-113 and [F/G]N/OFQ(1-13)NH2 behaved as partial agonists and antagonists of NOP receptor for NOP/G protein and NOP/β-arrestin 2, respectively. UFP-101 behaved as NOP receptor antagonist for both transduction pathways. Conclusion: NOP ligands producing same effects on NOP/G protein interaction (partial agonism), but with opposite effects on β-arrestin 2 recruitment (partial agonism vs antagonism), can promote different in vivo effects on anxiety and mood as it was observed in the behavioral tests. This work corroborates the potential of NOP receptor as an innovative pharmacological target for the treatment of emotional disorders.

Sugarcane is an important culture for Brazil that holds almost half of all worldwide productivity. Plants face many challenges, because of biotic and abiotic stresses presents in the production field, which could prevent plants from reaching their genetic potential. As consequence, those stresses can generate Reactive Oxygen Species – ROS – that can cause damages on DNA. Another consequence of stress is the early-flowering process, which contributes for a reduction on yield. In this context, the aim of this work is to characterize ScMUTM1 and ScMUTM2, two DNA glycosylases belonging to base excision repair pathway; and identify genes potentially related to stress and DNA repair in two sugarcane cultivars with contrasting flowering phenotypes. The characterization of the DNA glycosylases included the construction of vector to over express the recombinant proteins ScMUTM1 and ScMUTM2; they will be used in a near future to purification of these proteins and use in enzymatic assays. It was also made a phylogenetic reconstruction of this gene in plants and analysis of its promoter. With the phylogenetic analysis, it is possible to observe the presence of these genes grouped inside a branch with monocots and another one with dicots. This suggests that the duplication of this gene probably occurred after the separation of these two groups. The analysis of the promotor of MUTM shows of the presence of stress-related regulatory motifs at ScMUTM2 promoter, when compared with ScMUTM1. This may suggests that ScMUTM1 might be suffering sub functionalization process. After the analysis of microarrays data, it is observed an up-regulation from some stress-related genes in one of the conditions analyzed, related to early flowering process.

Omnivory is a predominant feeding strategy among tropical fishes, but knowledge about its causes and consequences of this pattern is scarce. In this study we hypothesized that tropical fish feed lower in food web as a way to compensate a higher energetic demand, which increases with increasing water temperature and body size. Information about 8172 freshwater and marine fish species from whole world, from tropical and temperate ecosystems, showed that the trophic position of non-carnivore fish decreases with increasing body size in tropical but not in temperate ecosystems. This result indicates that the higher energetic demand of large-bodied tropical fish should exert a selective force in favor of omnivory. As a consequence, trophic dynamics in tropical freshwater ecosystems should have different patterns comparing to temperate ones, with major implications for water management and restoration of eutrophic aquatic ecosystems. Another hypothesis of this work was that effects of tropical omnivorous planktivorous fish on planktonic communities depend of primary producers stoichiometric composition, which depends of light availability relative to nutrients ratios. A mesocosm experiment, manipulating light availability and planktivorous fish presence, confirmed our hypothesis indicating that resource stoichiometric composition (consequently nutritional quality), determine trophic structure of pelagic food webs in tropical lakes. Finally another mesocosm experiment indicated that the removal of omnivorous benthivorous fish should be more efficient than removal of omnivorous planktivorus fish, as a way to improve water quality in tropical lakes and reservoirs. This last experiment showed that omnivorous planktivorous fish increase phytoplankton biomass due to trophic cascade interactions, without increasing nutrient concentrations in the water column. On the other hand, omnivorous benthivorous fish, feeding on detritus and other benthonic food sources and excreting nutrients in the water column, are responsible for translocate nutrient from sediments to the water column, increasing phosphorus pool and phytoplankton biomass. Thereby, internal phosphorus supply should be reduced and water quality of eutrophicated lakes could be improved by removing omnivorous benthivorous fish.

The Implicit Association Test (IAT) has succeeded in accessing mental phenomena hardly exposed through self-report and explicit measures well known in the traditional studies on psychology. Although this measurement technique is among the most consistent in international publications, the few articles in Brazil have not deepened on theoretical issues that underlie the practice. This created a gap between the spread of this measure in the country and the production seen in international laboratories, both at quantitative and qualitative terms. In this context, the online labs implemented in several countries, including Brazil, created unique opportunities to overcome these disparities. Our work sought theoretical and conceptual clarification, contextualized to the historical development of the IAT and its online version, displaying affordably an unprecedented presentation of the virtual tool adapted to the Brazilian public. In a second step, we investigated empirically the data obtained by the Brazilian electronic portal "Countries", analyzing the degree of implicit and explicit nationalism of 2271 Brazilian subjects collected from early 2009 to late 2014. Our goal was to determine whether the data results obtained in a time of major sporting confrontation (FIFA World Cup 2010 and 2014) differ from periods when the tournament did not happen. Analyzes showed differences on the eve of the sporting confrontation, when the increase in the implicit nationalism was clearly superior, even with no effect by self-reports. In an independent analysis of a cultural context, there is an oscillation of explicit nationalism over the years, but not for the implicit action. In addition, it was found in women greater degree of implicit and explicit nationalism than in men, with both sexes presenting their preferences towards Brazil. In an end section, we suggest that nationalism may be a by-product of universal mental mechanisms that evolved to identify of the belonging group categorization, corroborating with the group favoritism. We propose that the intensification of the group preference on the eve of the competition has as its ancestral function strengthen the cohesion of the group in preparation to confrontational situations. We pointed out the need for studies on the differences between the sexes in matters relating to group membership belonging. It was expected largest nationalist attitudes in men reflecting the maintenance of cohesive groups in ancient societies. Thus, it appears a singular importance impart the traditional TAI studies with its online version for future investigations of human behavior in various areas of knowledge.

In this context, the online labs implemented in several countries, including Brazil, created unique opportunities to overcome these disparities. Our work sought theoretical and conceptual clarification, contextualized to the historical development of the IAT and its online version, displaying affordably an unprecedented presentation of the virtual tool adapted to the Brazilian public. In a second step, we investigated empirically the data obtained by the Brazilian electronic portal "Countries", analyzing the degree of implicit and explicit nationalism of 2271 Brazilian subjects collected from early 2009 to late 2014. Our goal was to determine whether the data results obtained in a time of major sporting confrontation (FIFA World Cup 2010 and 2014) differ from periods when the tournament did not happen. Analyzes showed differences on the eve of the sporting confrontation, when the increase in the implicit nationalism was clearly superior, even with no effect by self-reports. In an independent analysis of a cultural context, there is an oscillation of explicit nationalism over the years, but not for the implicit action. In addition, it was found in women greater degree of implicit and explicit nationalism than in men, with both sexes presenting their preferences towards Brazil. In an end section, we suggest that nationalism may be a by-product of universal mental mechanisms that evolved to identify of the belonging group categorization, corroborating with the group favoritism. We propose that the intensification of the group preference on the eve of the competition has as its ancestral function strengthen the cohesion of the group in preparation to confrontational situations. We pointed out the need for studies on the differences between the sexes in matters relating to group membership belonging. It was expected largest nationalist attitudes in men reflecting the maintenance of cohesive groups in ancient societies. Thus, it appears a singular importance impart the traditional TAI studies with its online version for future investigations of human behavior in various areas of knowledge.

Cada indivíduo apresenta uma respostas comportamental diferente diante de um desafio ambiental, muitas vezes mesmo quando comparado a outro indivíduo da mesma espécie, sexo e idade, essas diferenças são chamadas tipos comportamentais e quando consistentes ao longo do tempo caracterizam uma síndrome comportamental ou personalidade animal. Evidências apontam que a personalidade animal pode interferir na aptidão reprodutiva do indivíduo. Tendo isso em vista, no presente estudo machos e fêmeas adultos de Betta splendens foram observados em três contextos (exploratório, social e avaliação de risco), para identificar a existência de tipos comportamentais e diferenças entre os sexos nos tipos comportamentais nessa espécie. Os indivíduos foram classificados em três níveis em cada contexto: contexto social (social; intermediário e não-social), contexto exploratório (explorador, intermediário e não-explorador) e avaliação de risco (bold, intermediário e shy). Após essa classificação, machos sociais e nãosociais, exploradores e não-exploradores de coloração vermelha e azul foram utilizados no experimento de escolha de parceiro pelas fêmeas. Nossos resultados apontam que a maioria dos indivíduos testados possui fenótipos intermediários para os três contextos avaliados, e uma minoria de machos e fêmeas apresentaram tipos comportamentais. E a análise de correlação mostra que machos e fêmeas se comportaram de modo diferente em relação aos contextos. No experimento de escolha de parceiro, os resultados mostram que os tipos comportamentais da fêmea e do macho influenciam no processo de escolha de parceiro e que, mesmo na presença de pistas visuais (coloração corporal dos machos), o tipo comportamental continua sendo importante no processo de escolha.

Substâncias psicoestimulantes vêm sendo utilizadas de forma indiscriminada há muitos anos, e pouco se sabe os efeitos que elas causam a curto e longo prazo no comportamento geral, na aprendizagem e na memória. Essas substâncias são bastante usadas por jovens e adultos e elas possuem efeitos diferentes. Essas substâncias são dose dependente, caso consumidas em baixa quantidade agem como estimulante, aumentando a atividade locomotora, caso consumidas em alta quantidade, causam efeito depressor, diminuindo a atividade locomotora e/ou causando ansiedade. Poucos estudos vêm investigando os efeitos dessas substâncias na atividade locomotora, aprendizagem e memória e grande parte desses estudos são realizados em roedores. Peixe paulistinha é um modelo animal promissor para estudos comportamentais, cognitivos, ontogenéticos, dentre outros. Nossos objetivos foram determinar os efeitos do álcool, cafeína e de seu uso combinado com álcool, na atividade locomotora desses animais, usando para isso doses crônicas durante 27 dias e doses agudas durante um dia. Visto que pouco se sabe sobre os efeitos dessa exposição prolongada. Também investigamos os efeitos das substâncias em teste de reconhecimento de objetos, que se baseia na memória de único evento. Essas memórias são mais vulneráveis que memórias baseadas em várias repetições de eventos. Sendo assim, um teste adequado para utilizar com uso de substâncias psicoativas. Observamos que o uso crônico de cafeína provoca alteração na atividade locomotora dos animais, do mesmo modo, abstinência de álcool combinada com cafeína em dose aguda (dose média) provoca aumento de atividade locomotora. Quando submetidos a testes de memória, os animais exposto a doses altas agudas de álcool e em abstinência dessa droga têm prejuízo na formação e/ou resgate da memória. No entanto, tratamento com cafeína não prejudica a formação de memória. Animais expostos a tratamento com dose crônica moderada de álcool e dose aguda moderada de cafeína tem melhor desempenho na tarefa, indicando que dose aguda moderada de cafeína pode evitar os efeitos deletérios ocasionados pela abstinência do álcool. Em termos do comportamento geral, doses agudas de cafeína aumentam a locomoção, enquanto doses elevadas e a abstinência de cafeína induzem a comportamentos tipo-ansioso. A combinação álcool crônico e cafeína aguda induzem a alto comportamento tipo-ansiedade, enquanto a combinação cafeína crônica e álcool agudo diminuem tanto a locomoção quanto a ansiedade.
