α-l-Rhamnosidase (EC 3.2.1.40) is an enzyme that catalyzes the cleavage of terminal rhamnoside groups from naringin to prunin and rhamnose. In this study, a His-tag was genetically attached to the rhamnosidase gene ramA from Clostridium stercorarium to facilitate its purification from Escherichia coli BL21 (DE3) cells containing the pET-21d/ramA plasmid. Immobilized metal-chelate affinity chromatography (IMAC) resulted in one-step purification of N-terminally His-tagged recombinant rhamnosidase (N-His-CsRamA) which was immobilized in Ca2+ alginate (3%) beads. The optimum pH levels of the free and immobilized recombinant rhamnosidase were found to be 6.0 and 7.5, and the optimum temperature 55 and 60 °C respectively. At 50 °C, the free enzyme was relatively stable and exhibited a less than 50% reduction in residual activity after 180 min of incubation. The free and immobilized enzymes achieved 76% and 67% hydrolysis of the naringin in Kinnow juice respectively. Immobilization of recombinant rhamnosidase enabled its reutilization up to 9 hydrolysis batches without an appreciable loss in activity. This result indicated that the His-tagged thermostable rhamnosidase could be prepared as described and may serve to illustrate an economical and commercially viable process for industrial application.

Cell-envelope proteinases (CEPs) are a class of proteolytic enzymes produced by lactic acid bacteria and have several industrially relevant applications. However, soluble CEPs are economically unfavorable for such applications due to their poor stability and lack of reusability. In a quest to prepare stable biocatalysts with improved performance, CEP from Lactobacillus delbrueckii subsp. lactis 313 and trypsin (as a model enzyme) were immobilized onto nonwoven polyester fabrics in a three-step protocol including ethylenediamine activation and glutaraldehyde crosslinking. Immobilization gave protein loading yields of 21.9% (CEP) and 67.7% (trypsin) while residual activity yields were 85.6% (CEP) and 4.1% (trypsin). The activity of the immobilized enzymes was dependent on pH, but was retained at elevated temperatures (40-70°C). An increase in Km values was observed for both enzymes after immobilization. After 70 days of storage, the immobilized CEP retained ca. 62% and 96% of initial activity when the samples were stored in a lyophilized form at -20°C or in a buffer at 4°C, respectively. Both immobilized CEP and trypsin were able to hydrolyze proteins such as casein, skimmed milk proteins and bovine serum albumin. This immobilization protocol can be used to prepare immobilized biocatalyst for various protein degradation processes.

In recent years, there has been an increasing focus on the study and assessment of animal welfare in captive settings, such as zoological gardens and aquaria. Canada lynx (Lynx canadensis) are a relatively common species in zoos, yet are known to exhibit frequent reproductive problems in captive environments. We provide an exploratory analysis of housing and husbandry factors that are associated with patterns of adrenocortical activity in lynx. Adrenocortical activity was assessed using the non-invasive technique of monitoring faecal glucocorticoid metabolites (FGM). First, we calculated baseline FGM values for each individual and controlled for sex, age class, and reproductive status. The residual values were used to determine how levels of adrenocortical activity correlated with institutional husbandry practices. Second, we compared the occurrence of FGM peaks to events and disturbances recorded by keepers. Our results highlighted that adrenocortical activity is strongly correlated with: (i) the size of the enclosure, (ii) the number of hiding locations available, and (iii) the social environment. Based on our findings, we recommend that lynx should generally be housed alone (unless with dependant offspring or temporarily paired up for mating purposes), in larger enclosures and with the provision of several species-appropriate hiding locations.
