Natriuretic peptide receptors mediate the physiological response of&nbsp; natriuretic peptide hormones. One of the natriuretic peptide receptor types is the particulate guanylyl cyclase receptors, of which there are two identified: NPR-A and NPR-B. In fishes, these have been sequenced and characterized in eels, medaka, and dogfish shark (NPR-B only). The euryhaline rainbow trout provides an opportunity to further pursue examination of the system in teleosts. In this study, partial rainbow trout NPR-A-like and NPR-B-like mRNA sequences were identified via PCR and cloning. The sequence information was used in real-time PCR to examine mRNA expression in a variety of tissues of freshwater rainbow trout and rainbow trout acclimated to 35 parts per thousand seawater for a period of 10 days. In the excretory kidney and posterior intestine, real-time PCR analysis showed greater expression of NPR-B in freshwater fish than in those adapted to seawater; otherwise, there was no difference in the expression of the individual receptors in fresh water or seawater. In general, the expression of the NPR-A and NPR-B type receptors was quite low. These findings indicate that NPR-A and NPR-B mRNA expression is minimally altered under the experimental regime used in this study.<br />

Amyloid deposition and reduced &beta;-cell mass are pathological hallmarks of the pancreatic islet in type 2 diabetes; however, whether the extent of amyloid deposition is associated with decreased &beta;-cell mass is debated. We investigated the possible relationship and, for the first time, determined whether increased islet amyloid and/or decreased &beta;-cell area quantified on histological sections is correlated with increased &beta;-cell apoptosis. Formalin-fixed, paraffin-embedded human pancreas sections from subjects with (n = 29) and without (n = 39) diabetes were obtained at autopsy (64 ± 2 and 70 ± 4 islets/subject, respectively). Amyloid and &beta; cells were visualized by thioflavin S and insulin immunolabeling. Apoptotic &beta; cells were detected by colabeling for insulin and by TUNEL. Diabetes was associated with increased amyloid deposition, decreased -cell area, and increased &beta;-cell &beta;apoptosis, as expected. There was a strong inverse correlation between &beta;-cell area and amyloid deposition (r=0.42, P &lt; 0.001). &beta;-Cell area was selectively reduced in individual amyloid-containing islets from diabetic subjects, compared with control subjects, but amyloid-free islets had &beta;-cell area equivalent to islets from control subjects. Increased amyloid deposition was associated with &beta;-cell apoptosis (r= 0.56, P &lt; 0.01). Thus, islet amyloid is associated with decreased &beta;-cell area and increased &beta;-cell apoptosis, suggesting that islet myloid deposition contributes to the decreased &beta;-cell mass that characterizes type 2 diabetes. <br />

The aim of the study was to assess whether omega-3 polyunsaturated fatty acid supplementation alone or in combination with folic acid and B-group vitamins is effective in lowering homocysteine. The Medline Ovid, Embase and Cochrane databases were searched for randomized-controlled trial studies that intervened with omega-3 supplementation (with or without folic acid) and measured changes in homocysteine concentration. Studies were pooled using a random effects model for meta-analysis. Three different models were analyzed: all trials combined, omega-3 polyunsaturated fatty acid trials, and omega-3 polyunsaturated fatty acids with folic acid and B-group vitamin trials. Nineteen studies were included, consisting of 3267 participants completing 21 trials. Studies were heterogeneous; varying by dose, duration and participant health conditions. Across all trials, omega-3 supplementation was effective in lowering homocysteine by an average of 1.18μmol/L (95%CI: (-1.89, -0.48), P=.001). The average homocysteine-lowering effect was greater when omega-3 supplementation was combined with folic acid and B-group vitamins (-1.37μmol/L, 95%CI: (-2.38, -0.36), P&lt;.01) compared to omega-3 supplementation alone (-1.09μmol/L 95%CI: (-2.04, -0.13), P=.03). Omega-3 polyunsaturated fatty acid supplementation was associated with a modest reduction in homocysteine. For the purposes of reducing homocysteine, a combination of omega-3s (0.2-6g/day), folic acid (150 - 2500μg/day) and vitamins B6 and B12 may be more effective than omega-3 supplementation alone.

<b>Background and objective:b> Natural killer (NK) and natural killer T (NKT)-like cells represent a small but important proportion of effector lymphocytes that we have previously shown to be major sources of pro-inflammatory cytokines and granzymes. We hypothesized that these cells would be increased in the airway in chronic obstructive pulmonary disease (COPD), accompanied by reduced expression of the inhibitory receptor CD94 (Kp43) and increased expression of cytotoxic mediators granzyme B and perforin.<br /><b>Methods:b> We measured NK and NKT-like cells and their expression of CD94 in the blood of COPD patients (n = 71; 30 current and 41 ex-smokers), smokers (16) and healthy controls (25), and bronchoalveolar lavage fluid (BALF) from a cohort of subjects (19 controls, 12 smokers, 33 COPD). Activation was assessed by measuring CD69 in blood and the cytotoxic potential of NK cells by measuring granzymes A and B, and using a cytotoxicity assay in blood and BALF.<br /><b>Results:b> In blood in COPD, there were no significant changes in the proportion of NK or NKT-like cells or expression of granzyme A or NK cytotoxic potential versus controls. There was, however, increased expression of granzyme B and decreased expression of CD94 by both cell types versus controls. The proportion of NK and NKT-like cells were increased in BALF in COPD, associated with increased NK cytotoxicity, increased expression of granzyme B and decreased expression of the inhibitory receptor CD94 by both cell types.<br /><b>Conclusions:b> Treatment strategies that target NK and NKT-like cells, their cytotoxicity and production of inflammatory mediators in the airway may improve COPD morbidity.

Introduction : While consumption of omega-3 long-chain polyunsaturated fatty acids (n-3 LCPUFA) has been recommended for those at risk of inflammatory disease such as rheumatoid arthritis, the mechanism of their anti-inflammatory effect remains to be clearly defined, particularly in relation to the dose and type of n-3 LCPUFA. The objective of this study was to determine whether varying the levels of n-3 LCPUFA in erythrocyte membrane lipids, following dietary supplementation, is associated with altered numbers and function of circulating leukocytes conducive to protection against inflammation. Methods : In a double-blind and placebo-controlled study, 44 healthy subjects aged 23 to 63 years consumed either standard or n-3 LCPUFA-enriched versions of typical processed foods, the latter allowing a target daily consumption of 1 gram n-3 LCPUFA. After six months, peripheral blood leukocyte and subpopulation proportions and numbers were assessed by flow cytometry. Leukocytes were also examined for lymphoproliferation and cytokine production, neutrophil chemotaxis, chemokinesis, bactericidal, adherence and iodination activity. Erythrocytes were analyzed for fatty-acid content. Results : Erythrocyte n-3 LCPUFA levels were higher and absolute leukocyte and lymphocyte numbers were lower in subjects consuming n-3 enriched foods than in controls. There were no changes in the number of neutrophils, monocytes, T cells (CD3+), T-cell subsets (CD4+, CD8+) and B cells (CD19+). However, natural killer (NK) (CD3-CD16+CD56+) cell numbers were lower in n-3 supplemented subjects than in controls and were inversely related to the amount of eicosapentaenoic acid or docosahexaenoic acid in erythrocytes. No significant correlations were found with respect to lymphocyte lymphoproliferation and production of IFN-γ and IL-2, but lymphotoxin production was higher with greater n-3 LCPUFA membrane content. Similarly, neutrophil chemotaxis, chemokinesis, bactericidal activity and adherence did not vary with changes in erythrocyte n-3 LCPUFA levels, but the iodination reaction was reduced with higher n-3 LCPUFA content. Conclusion : The data show that regular long-term consumption of n-3 enriched foods leads to lower numbers of NK cells and neutrophil iodination activity but higher lymphotoxin production by lymphocytes. These changes are consistent with decreased inflammatory reaction and tissue damage seen in patients with inflammatory disorders receiving n-3 LCPUFA supplementation.

Apart from its classical function in bone and calcium metabolism, vitamin D is also involved in immune regulation and has been linked to various cancers, immune disorders and allergic diseases. Within the innate and adaptive immune systems, the vitamin D receptor and enzymes in monocytes, dendritic cells, epithelial cells, T lymphocytes and B lymphocytes mediate the immune modulatory actions of vitamin D. Vitamin D insufficiency/deficiency early in life has been identified as one of the risk factors for food allergy. Several studies have observed an association between increasing latitude and food allergy prevalence, plausibly linked to lower ultraviolet radiation (UVR) exposure and vitamin D synthesis in the skin. Along with mounting epidemiological evidence of a link between vitamin D status and food allergy, mice and human studies have shed light on the modulatory properties of vitamin D on the innate and adaptive immune systems. This review will summarize the literature on the metabolism and immune modulatory properties of vitamin D, with particular reference to food allergy.

The reaction of diorganotin sulfides, cyclo-(Rb>2b>SnS)b>nb> (R=Me, n-Bu; n=3; R=t-Bu; n=2) with the corresponding diorganotin dichlorides, Rb>2b>SnClb>2b>, provided the tetraorganodistannathianes, (Rb>2b>ClSn)b>2b>S (<b>1b>, R=Me; <b>2b>, R=n-Bu; <b>3b>, R=t-Bu). 1H-, 13C-, and 119Sn-NMR studies indicate that these compounds are kinetically labile and in equilibrium with the starting materials. Addition of equimolar amounts of [(Phb>3b>P)b>2b>N]Cl to the reaction mixtures gave the chloride complexes [(Phb>3b>P)b>2b>N]+[S(SnRb>2b>Cl)b>2b>Cl] (<b>4b>, R=Me; <b>5b>, R=n-Bu; <b>6b>, R=t-Bu). Single-crystal X-ray diffraction studies revealed the tin atoms in both <b>4b> and <b>6b> to adopt distorted trigonal bipyramidal configurations with the chlorine atoms occupying the axial positions.<br /><br /><br />

Terpene alcohols (−)-menthol and [(1S)-endo]-(−)-borneol react with SiClb>4b> in the presence of base to give (MenO)b>3b>SiCl (<b>1b>) and (BorO)b>3b>SiCl (<b>2b>) in high yields. Hydrolysis of <b>1b> yields (MenO)b>3b>SiOH (<b>4b>) and (MenO)b>4b>Si (<b>3b>). Hydrolysis of <b>2b> yields only (BorO)b>3b>SiOH (<b>5b>). The crystal structures of <b>3b> and <b>5b> are reported.<br /><br /><br />

The syntheses of cyclo-[Rb>2b>Sn(OPPhb>2b>O)b>2b>SnRb>2b>](Ob>3b>SCFb>3b>)b>2b> (R = Me (<b>1b>), t-Bu (<b>2b>)) by the consecutive reaction of Rb>2b>SnO (R = Me, t-Bu) with triflic acid and diphenylphosphinic acid are presented. In the solid state, <b>1b> and <b>2b> were investigated by 119Sn MAS and 31P MAS NMR spectroscopy as well as X-ray crystallography and appear to exist as ion pairs of cyclo-[Rb>2b>Sn(OPPhb>2b>O)b>2b>SnRb>2b>]2+ dications and triflate anions. In solution, <b>1b> and <b>2b> are involved in extensive equilibria processes featuring cationic diorganotin(IV) species with Sn-O-P linkages, as evidenced by 119Sn and 31P NMR spectroscopy, electrospray mass spectrometry, and conductivity measurements. <br />

The synthesis of the α,ω-bis[dichloro(trimethylsilylmethyl)stannyl]alkanes, (Meb>3b>SiCHb>2b>)C1b>2b>Sn(CHb>2b>)b>nb>SnClb>2b>(CHb>2b>SiMeb>3b>) (<b>13b>, n=5; <b>14b>, n=6; <b>15b>, n=7; <b>16b>, n=8; <b>17b>, n=10; <b>18b>, n=12) and the corresponding oligomethylene-bridged diorganotin oxides [(Meb>3b>SiCHb>2b>)(O)Sn(CHb>2b>)b>nb>Sn(O)(CHb>2b>SiMeb>3b>)]b>mb> (<b>19b>, n=5; <b>20b>, n=6; <b>21b>, n=7; <b>22b>, n=8; <b>23b>, n=10; <b>24b>; n=12) is reported. The reaction of the diorganodichlorostannanes <b>13–18b> with the corresponding diorganotin oxides <b>19–24b> provided the spacer-bridged tetraorganodistannoxanes {[(Meb>3b>SiCHb>2b>)ClSn(CHb>2b>)b>nb>SnCl(CHb>2b>SiMeb>3b>)]O}b>4b> (<b>25b>, n=5; <b>26b>, n=6; <b>27b>, n=7; <b>28b>, n=8; <b>29b>, n=10; <b>30b>, n=12). Compounds <b>13–30b> have been identified by elemental analyses and multinuclear NMR spectroscopy. Compounds <b>25, 27, 29b> and <b>30b> have also been characterised by single crystal X-ray diffraction analysis and electrospray mass spectrometry. For the latter the essential double ladder motif is maintained for all n in the solid state, but subtle changes in alignment of the ladder planes occur. Separation between the two layers of the double ladder ranges from approx. 8.7&nbsp; Å (for <b>25b>, n=5) to approx. 15 Å (for <b>30b>, n=12). In solution there is some dissociation of the double ladders into the corresponding dimers. The degree of dissociation is favoured by increasing oligomethylene chain length n.<br /><br /><br />

The condensation of [Phb>2b>(OH)Sn(CHb>2b>)b>nb>Sn(OH)Phb>2b>] (<b>1-3b>; n = 1-3) with HOb>3b>SCFb>3b> and HOb>2b>PPhb>2b> provided [Phb>2b>Sn(CHb>2b>)b>nb>SnPhb>2b>(OH)](Ob>3b>SCFb>3b>) (<b>4-6b>; n = 1-3) and [Phb>2b>(Ob>2b>PPhb>2b>)Sn(CHb>2b>)b>nb>Sn(Ob>2b>PPhb>2b>)Phb>2b>] (<b>10-12b>; n = 1-3), respectively. The reaction of [Phb>2b>Sn(CHb>2b>)b>nb>SnPhb>2b>(OH)](Ob>3b>SCFb>3b>) (<b>4-6b>; n = 1-3) with HOb>2b>PPhb>2b> and NaOb>2b>PPhb>2b> gave rise to the formation of [Phb>2b>Sn(CHb>2b>)b>nb>SnPhb>2b>(Ob>2b>PPhb>2b>)](Ob>3b>SCFb>3b>) (<b>7-9b>; n = 1-3) and [Phb>2b>(OH)Sn(CHb>2b>)b>nb>Sn(Ob>2b>PPhb>2b>)Phb>2b>] (<b>13-15b>; b>nb> = 1-3), respectively. In the solid state, compounds <b>4-9b> comprise ion pairs of cationic cyclo-[Phb>2b>SnCHb>2b>SnPhb>2b>(OH)]b>2b>2+, cyclo-[Phb>2b>Sn(CHb>2b>)b>nb>SnPhb>2b>(OH)]+ (n = 2, 3), and cyclo-[Phb>2b>Sn(CHb>2b>)b>nb>SnPhb>2b>(Ob>2b>PPhb>2b>)]+ (n = 1-3) and triflate anions. In MeCN, the eight-membered-ring system cyclo-[Phb>2b>SnCHb>2b>SnPhb>2b>(OH)]b>2b>2+ appears to be in equilibrium with the four-membered-ring system cyclo-[Phb>2b>SnCHb>2b>SnPhb>2b>(OH)]+. In contrast, compounds <b>10-15b> show no ionic character. Compounds <b>1-15b> were characterized by multinuclear NMR spectroscopy in solution and in the solid state, IR spectroscopy, conductivity measurements, electrospray mass spectrometry, osmometric molecular weight determinations, and X-ray crystallography (<b>4, 5, 7,b> and <b>12b>). <br /><br />

Fire-related accidents often result in injuries and sometimes death, which can be prevented through fire safety training. To estimate the extent to which fire safety training should be provided, it is essential to assess the current level of fire safety knowledge within the general community. Thus the objectives of the present study were to explore: (a) the level of fire safety knowledge among people of different age groups and investigate its relationship to the level of fire safety training, and (b) the manner in which people from different age groups would respond to a fire based on their fire safety training. Data from 158 participants aged between 18 and 80 years showed that fire safety training increases: (a) the level of fire safety knowledge and, (b) the accuracy of response to a fire. The results also show that middle-aged individuals would respond more accurately to a fire than younger and older adults. The findings demonstrate the importance of fire safety training in enhancing people's fire safety knowledge and their response in the event of a fire which could lead to a reduction in the rate of fire casualties. There are implications for incorporating fire safety training as part of health improvement programs to reduce the number of fire-related injuries and fatalities.<br />

This paper argues that the nature of IS research that deals with indigenous culture must be informed as much by context as it is by culture models, which has been the focus of such research in the past. This is considered important because it better reflects the meaning of the data collected for the researcher. To appreciate the importance of context this papers also argues that research subjects from designated individualist societies will inform the researcher in different ways from those subjects located in collectivist societies. To illustrate the practical implications of this argument the paper reports three separate case studies in IS research where the researchers reflect on the impact that a collectivist view has had on the research findings. The paper suggests that (1) similar ethnicity and appearance are significant in gaining the trust of subjects in a collectivist society; that is the researcher is part of the in-group as they belong to the same culture or ethnic group; that (2) who introduced the researcher to the subject is significant in that trust is best reflected when a member of the group/collective plays an important role in the research process itself; and that (3) an ability to (a) communicate in the natural language and (b) understand the implicit body language and (c) cultural codes is important in gaining significant and more meaningful research outcomes. This is enabled via the implicit meanings embedded in members of the collectivist society.<br />

The government sector in Australia has seen the introduction of accrual accounting principles in recent years. However, this process has been complicated by the presence of two alternative financial reporting frameworks in the form of a) the Government Finance Statistics (OFS) uniform framework and b) the accrual accounting rules specified in Australian professional accounting standards, principally AAS 31. While a variety of cash and accrual based measurements are available pursuant to these frameworks, there has been no prescription of the manner in which the alternative measures should be presented. This paper presents the findings from a case study of the 2005-06 annual budgets prepared by the Australian Commonwealth government and the governments of the six Australian States and the two Australian Territories. The study examined the headlined financial outcome (general government sector budget surplus or deficit) announced in the budget papers of the nine governments. Findings indicate the adoption of varying measurement bases and a consequent lack of inter-government comparability. A number of variations to the measurements prescribed in the accounting frameworks were also observed. The paper analyses these government budget surplus and deficit numbers in the context of fiscal responsibility, the Commonwealth government's Charter of Budget Honesty and the AASB's current GAAPIGFS Convergence project.<br />
