Caging and a mark–recapture design were used to estimate the growth rate of the brittle, infaunal bivalve Soletellina alba in the Hopkins River estuary. The growth of both caged and uncaged individuals was monitored at three sites near the mouth of the estuary over 180 days. Growth rates did not differ for caged and uncaged bivalves, or for bivalves subject to different amounts of handling, or between sites. Growth did differ between consecutive time intervals, which was attributable to negligible growth occurring during the colder months of autumn/winter. Comparisons of the condition (as indicated by total mass for length3) of S. alba were inconsistent between sites for caged and uncaged bivalves and for those subject to different amounts of handling. Soletellina alba is a rapidly growing bivalve with mean growth rates for the three time intervals being 0.04±0.002 mm day−1 in summer, 0.02±0.001 mm day−1 in autumn and 0.03±0.001 mm day−1 from summer to winter. Using existing literature, it was shown that a significant relationship exists between maximum shell length and onset of sexual maturity in bivalve molluscs. This relationship predicts that S. alba should reach the onset of sexual maturity at 15.8 mm length. Therefore, it appears that it may be possible for juvenile S. alba (<1 mm) to grow, reach sexual maturity and reproduce in between annual mass-mortality events caused by winter flooding.

STUDY DESIGN: Prospective longitudinal study. OBJECTIVE: To evaluate the recovery of the lumbar intervertebral discs after bed rest. SUMMARY OF BACKGROUND DATA: Prolonged bed rest is a useful model to understand the modeling and remodeling of tissues due to disuse and reloading, yet this process in the lumbar intervertebral discs has not been examined in detail. METHODS: A total of 24 male subjects completed 60 days of head-down tilt bed rest as part of the 2nd Berlin BedRest Study and returned for magnetic resonance scanning 180 days (n = 22) and 2 years (n = 21) after bed rest. Lumbar disc volume, anterior and posterior disc height, disc signal intensity, intervertebral length, and lordosis were measured on sagittal plane magnetic resonance images. RESULTS.: Compared with prior to bed rest, increases in disc volume, disc height, and intervertebral length persisted 180 days (P ≤ 0.0004) and 720 days (P ≤ 0.024) after bed rest. Disc signal intensity remained increased 180 days (P = 0.034) after bed rest but was then decreased (P = 0.018) compared with baseline at the next measurement date. CONCLUSION: The recovery of the lumbar intervertebral discs after 60-day bed rest is a prolonged process and incomplete within 2 years.

Objective: Psycho-neuro-immune research suggests an association between cancer outcomes and psychosocial distress. Objective criteria to determine patients’ levels of distress are important to establish potential links to disease outcomes. Methods: We compared three patient-reported with one doctor-reported measures of psychooncologic distress frequently used in routine cancer care and investigated associations with standard disease severity parameters in melanoma patients. We enrolled n = 361 patients, successively seen at two outpatient university clinics in Germany. In the naturalistic study, n = 222 patients had been diagnosed <180 days and were seen for the first time (Group I); n = 139 had been diagnosed >180 days and were in after-care (Group II). Results: Across groups, only moderate associations were seen between patient- reported and doctorreported measures. Regarding clinical variables, disease severity and perceived need of psychooncologic support reported by patients or doctors showed hardly any association. After subgroup stratification, in patients of Group II, patient-reported and doctor-reported instruments showed some small associations with disease parameters commonly linked to more rapid cancer progression in patients who are in cancer after-care. Conclusions: Overall, the few and low associations suggest that need of psycho-oncologic support and clinical variables were largely independent of each other and doctors’ perception may not reflect the patient’s view. Therefore, the assessment of the patient perspective is indispensable to ensure that melanoma patients receive appropriate support, as such need cannot be derived from other disease parameters or proxy report. More research is needed applying psychometrically robust instruments that are ideally combined with sensitive biomarkers to disentangle psycho-neuro-immune implications in melanoma patients.

This study examined whether providing preschool children with simple groundrules (the importance of being complete, saying „I don‟t know‟, correcting the interviewer and not guessing) would reduce false details in their recall of a staged event. Forty-nine preschool children participated in an event that consisted of two activities. One or two days later they were given a biasing interview that included false suggestions about one of the experienced activities as well as a non-experienced activity. For the other activity, no suggestions were made. Eight, 15, and 22 days after the event, the children were required to recall all three activities in their own words. Immediately prior to their recall, half of the children were provided with the groundrules while the remaining children were not. The children in the control group also participated in a fifth interview in which they received the groundrule instructions. The results revealed that the provision of the groundrules had negligible impact on the accuracy of information provided irrespective of the context or order of the interview or the activity being recalled. The implications of these results are discussed and suggestions for future research are offered.

Two experiments were conducted to assess the water stability of a practical research diet manufactured with various binders and differing levels of moisture. In the first experiment the binders – agar, gelatine, carrageenan, and carboxymethylcellulose (CMC) were included at both 3 and 5% of total ingredient weight. All binders were tested with equal ingredient weight to water volume, and additionally carrageenan was tested in a diet with double the water volume. The dry matter remaining following immersion for up to 180 min was calculated and the rate of pellet decay was modelled using the Weibull distribution. The analysis revealed that the rate of dry matter loss decreased with time, and that carrageenan and CMC binders were significantly better (P < 0.001) binders than the agar and gelatine. The 5% binder concentration slowed the decay rate by as much as 62% as compared with the 3% binder concentration. The second experiment compared the binding performance of carrageenan and sodium alginate in both 50% moisture and 10% moisture pellets. The same analysis revealed that 10% moisture alginate-bound pellets were more water stable than the others. A discussion of the use of moist diets for crayfish research is included.

The purpose of the present study was to determine in human skeletal muscle whether a single exercise bout and 7 days of consecutive endurance (cycling) training 1) increased insulin-stimulated Akt pSer473and 2) altered the abundance of the protein tyrosine phosphatases (PTPases), PTP1B and SHP2. In healthy, untrained men (n = 8; 24 ± 1 yr), glucose infusion rate during a hyperinsulinemic euglycemic clamp, when compared with untrained values, was not improved 24 h following a single 60-min bout of endurance cycling but was significantly increased (~30%; P < 0.05) 24 h following completion of 7 days of exercise training. Insulin-stimulated Akt pSer473was ~50% higher (P < 0.05) 24 h following the acute bout of exercise, with this effect remaining after 7 days of training (P < 0.05). Insulin-stimulated insulin receptor and insulin receptor substrate-1 tyrosine phosphorylation were not altered 24 h after acute exercise and short-term training. Insulin did not acutely regulate the localization of the PTPases, PTP1B or SHP2, although cytosolic protein abundance of SHP2 was increased (P < 0.05; main effect) 24 h following acute exercise and short-term training. In conclusion, insulin-sensitive Akt pSer473and cytosolic SHP2 protein abundance are higher after acute exercise and short-term training, and this effect appears largely due to the residual effects of the last bout of prior exercise. The significance of exercise-induced alterations in cytosolic SHP2 and insulin-stimulated Akt pSer473on the improvement in insulin sensitivity requires further elucidation.

Ganopoly is an aqueous polysaccharide fraction extracted from G. lucidum by patented biochemical technique and has been marketed as an over-the-counter product for chronic diseases including cancer and hepatopathy in many Asian countries. This study was undertaken to explore the anti-tumour effect and the underlying mechanisms of Ganopoly in mice and human tumor cell lines. The maximum tolerateddose (MTD) of Ganopoly in mice was estimated to be 100 mg/kg from a pilot study. Treatment of mice with oral Ganopoly for 10 days significantly reduced the tumour weight of sarcoma-180 in a dose-dependent manner, with inhibition rates of 32.3, 48.2 and 84.9% and growth delays of 1.5, 3.5, and 13.1 days at 20, 50, and 100 mg/kg, respectively. Incubation of Ganopoly at 0.05-1.0 mg/ml for 48 hours showed little or negligible cytotoxicity against human tumor CaSki, SiHa, Hep3B, HepG2, HCT116, HT29, and MCF7 cells in vitro. In contrast, 10 mg/ml of Ganopoly caused significant cytotoxicity in all tumour cells tested except MCF7, with marked apoptotic effects observed in CaSki, HepG2, and HCT116 cells, as indicated by nuclear staining and DNA fragmentation. In addition, Ganopoly enhanced concanavalin A-stimulated proliferation of murine splenocytes by 35.3% at 10 mg/ml, and stimulated the production of nitric oxide in thioglycollate-primed murine peritoneal macrophages in a concentration-dependent manner over 0.05-10 mg/ml. Addition of Ganopoly at 1 mg/ml to murine peritoneal macrophages also potentiated lipopolysaccharide-induced nitric oxide production by 64.2%. Treatment of healthy mice or mice bearing sarsoma-180 with oral Ganopoly over 20-100 mg/kg for 7 day significantly increased the expression of both TNF-α and IFN-γ (at both mRNA and protein levels) in splenocytes in a dose-dependent manner. Moreover, treatment of Ganopoly over 20-100 mg/kg significantly increased cytotoxic T lymphocyte cytotoxicity and NK activity in mice. The overall findings indicated that Ganopoly had antitumor activity with a broad spectrum of immuno-modulating activities and may represent a novel promising immunotherapeutic agent in cancer treatment.

Consumption of hot chips is a convenience food in most countries. Unfortunately, these are high in fat and contribute to fat-related diseases in societies with a high fat consumption. There is substantial scope through best-practice deep-frying techniques for producing lower fat, high-quality chips. From a review of the literature, the main factors associated with a lower-fat content of chips are thick (>12 mm), straight cut chips; cryogenic freezing methods; low moisture content of potatoes (specific gravity >1.1); frying fat: chip volume ratio of 6:1; frying at optimal temperature (180 to 185°C) during cooking and turning the temperature down (∼140°C) and covering the vats during slack periods; vigorously shaking the basket and hanging it over the deep fryer to drain after frying; maintaining the quality of the frying fat by regularly skimming the cracklings, filtering the fat, and topping up the fryer with fresh fat; keeping the fat turnover <5 days; regular cleaning of frying equipment. It is important that all deep frying operators are adequately trained in these techniques. It is also important that the frying medium is low in saturated and trans fatty acids (<20%) because of their effects on blood lipids and low in linolenic acid (<3%) because it is readily degraded. The widespread implementation of best-practice deep-frying would reduce fat content of hot chips and thus lower overall fat consumption.

Christian survivors have frequently likened their experiences of sexual abuse to crucifixion. While it is understandable that many cannot reconcile their experiences within their faith tradition, the question of how it is possible to move forward within a Christian framework which accommodates the experience of sexual abuse has tended to be overlooked. In doing so, this paper considers why identifying with the crucified Christ may be an important step in the process of moving forward, and why the terminology ‘survivor’ may be preferable to ‘victim’. The challenge to move forward may require churches and church personnel to consider whether they need to do some things differently. Above all, this requires integrity for the individuals and communities involved rather than a one-size-fits-all approach.
