Calcineurin signaling is essential for successful muscle regeneration. Although calcineurin inhibition compromises muscle repair, it is not known whether calcineurin activation can enhance muscle repair after injury. Tibialis anterior (TA) muscles from adult wild-type (WT) and transgenic mice overexpressing the constitutively active calcineurin-Aα transgene under the control of the mitochondrial creatine kinase promoter (MCK-CnAα*) were injected with the myotoxic snake venom Notexin to destroy all muscle fibers. The TA muscle of the contralateral limb served as the uninjured control. Muscle structure was assessed at 5 and 9 days postinjury, and muscle function was tested in situ at 9 days postinjury. Calcineurin stimulation enhanced muscle regeneration and altered levels of myoregulatory factors (MRFs). Recovery of myofiber size and force-producing capacity was hastened in injured muscles of MCK-CnAα* mice compared with control. Myogenin levels were greater 5 days postinjury and myocyte enhancer factor 2a (MEF2a) expression was greater 9 days postinjury in muscles of MCK-CnAα* mice compared with WT mice. Higher MEF2a expression in regenerating muscles of MCK-CnAα* mice 9 days postinjury may be related to an increase of slow fiber genes. Calcineurin activation in uninjured and injured TA muscles slowed muscle contractile properties, reduced fatigability, and enhanced force recovery after 4 min of intermittent maximal stimulation. Therefore, calcineurin activation can confer structural and functional benefits to regenerating skeletal muscles, which may be mediated in part by differential expression of MRFs.

We examine a mathematical model of non-destructive testing of planar waveguides, based on numerical solution of a nonlinear integral equation. Such problem is ill-posed, and the method of Tikhonov regularization is applied. To minimize Tikhonov functional, and find the parameters of the waveguide, we use two new optimization methods: the cutting angle method of global optimization, and the discrete gradient method of nonsmooth local optimization. We examine how the noise in the experimental data influences the solution, and how the regularization parameter has to be chosen. We show that even with significant noise in the data, the numerical solution is of high accuracy, and the method can be used to process real experimental da.ta..

Objective: Patellar tendon injury, defined by tendon abnormality (TA) on imaging and by pain, is common among volleyball players, but little is known about change in this injury over a volleyball season. Increased activity in the season compared with the off season may result in the development of TA and/or pain. This study investigated the behaviour of TA and pain over a competitive volleyball season.

Methods: Tendon abnormality and pain were measured in 101 volleyball players at the beginning and end of a season. Pain was measured with the single leg decline squat test, which loads the patellar tendon, and TA was detected with ultrasound imaging. Hours of weekly activity were measured and compared during the season and the off season. The proportion of tendons that underwent development and resolution in TA and/or pain over the season was investigated.

Results: Hours of weekly activity was greater during the season than in the off season. Most of the tendons investigated (66.3%) did not undergo a change in TA or pain over the season. Tendon abnormality and/or pain developed in 16.6% of tendons and resolved in 11.2%.

Conclusions: The tendons of volleyball players respond variably to the increased load over the season. Change in TA and pain does not appear to be entirely dependent upon load.

Clubroot, caused by Plasmodiophora brassicae, is the most devastating soil-borne disease of vegetable brassicas. It occurs all over the world and is responsible for crop losses of up to 10% every year. In Australia, the disease is being managed effectively with chemicals and cultural practices, but ideally control can be improved in the long term by the introduction of resistant cultivars. The life cycle ofP. brassicae and mode of action of plant resistance has not been fully elucidated because of the technical difficulties of working with an obligate, soil-borne plant pathogen. However, Arabidopsis thaliana, which is a host ofP. brassicae, has great potential as a model system for studying the life cycle, the infection process and development of resistance. We have developed a sand-liquid-culture system for growing Arabidopsis that allows easy observation of all life stages and, most importantly, the primary plasmodial stages within the root hair. The method was first optimised for observations of the lifecycle of the pathogen in a susceptible Arabidopsis ecotype (Col-3) where all stages of the lifecycle have now been observed and characterised. Further screening of Arabidopsis ecotypes for disease resistance has utilised one of the most virulent Australian pathotypes of brassica (ECD number 16/19/31). To date, Arabidopsis ecotype Ta-0 has shown a level of tolerance to the disease even though the roots get infected. It has been reported earlier that resistance toP. brassicae in Arabidopsis is due to one or a small number of genes. To examine changes in gene expression during the early, critical stages of infection, RNA was extracted from the susceptible and resistant ecotypes at two time points, 4 days and 17 days after inoculation. Microarray analysis will be used to investigate genome wide changes in gene expression during infection but also to identify candidate genes that may confer resistance to Australian isolates of the pathogen.

This paper explores the enhancement of teaching and learning perfonnance outcomes from the perspective of educators from different cultural backgrounds and a very large mix of international students. The research milieu focuses on social connectivity, peer-ta-peer networking and peer-ta-peer meDtaring amongst teachers, tutors and students; particularly aspects of trust and cultural awareness between teacher and learner.  The key findings of this research are that the social capital dimensions of trust and reciprocity, and the notion of academic guanxi can assist academics and institutions to better understand ways in which to optimize teaching and learning. The result is a pool of educators who have become change-agents by inspiring a network of their peers in using components of a collective cultural toolkit as a culture reference point.

A modified sand–liquid culture method facilitated easy visualisation of the primary life cycle stages of Plasmodiophora brassicae within clean root hairs of the Arabidopsis host. Pathogen penetration occurred from day 4 onwards and then primary plasmodia developed within the host root. Several Arabidopsis ecotypes tested in varying growth conditions showed differences in disease expression. Defined growth cabinet conditions were found most suitable for studying disease progression in the ecotypes and for achieving uniform infection and disease development. Arabidopsis ecotypes Ta-0 and Tsu-0 known to be partially resistant to a German single-spore isolate of P. brassicae were susceptible to an Australian (Victorian) field population of P. brassicae. The European clubroot differential test was used to confirm virulence and describe the pathotype of the Victorian field population. Knowledge of the interaction of an Australian population of P. brassicae with its host will provide valuable information on a disease which is very difficult to control.

An Al70Ni10Ti10Zr5Ta5 amorphous alloy powder was fabricated by mechanical alloying. The phase structure and characteristic temperatures of the alloy were determined by X-ray diffraction, transmission electron microscopy and differential scanning calorimetry. The glass transition behavior and crystallization kinetics were analyzed using Lasocka and Kissinger functions. The results show that the alloy has a higher crystallization temperature, a higher effective activation energy of crystallization and a wider supercooled liquid region than the previously reported values, suggesting a high thermal stability and promising applications.

Silk fibroin films are promising materials for a range of biomedical applications. To understand the effects of casting solvents on film properties, we used water (W), formic acid (FA), and trifluoroacetic acid (TFA) as solvents. We characterized molecular weight, secondary structure, mechanical properties, and degradation behavior of cast films. Significant degradation of fibroin was observed for TFA-based film compared to W and TA-based films when analyzed by SDS-PAGE. Fibroin degradation resulted in a significant reduction in tensile strength and modulus of TFA-based films. Compared to water, TFA-based films demonstrated lower water solubility (19.6% vs. 62.5% in 12 h) despite having only a marginal increase in their ß-sheet content (26.9% vs. 23.7%). On the other hand, FA-based films with 34.3% ß-sheet were virtually water insoluble. Following solubility treatment, ß-sheet content in FA-based films increased to 50.9%. On exposure to protease XIV, water-annealed FA-based films lost 74% mass in 22 days compared to only 30% mass loss by ethanol annealed FA films. This study demonstrated that a small variation in the ß-sheet percentage and random coil conformations resulted in a significant change in the rates of enzymatic degradation without alteration to their tensile properties. The film surface roughness changed with the extent of enzymatic hydrolysis.

Aims New Zealand has a high incidence of cryptosporidiosis compared to other developed countries. This study aimed to describe the epidemiology of this disease in detail and to identify potential risk factors.

Methods We analysed anonymous cryptosporidiosis notification (1997–2006) and hospitalisation data (1996–2006). Cases were designated as “urban” or “rural” and assigned a deprivation level based on their home address. Association between disease rates and animal density was studied using a simple linear regression model, at the territorial authority level.

Results Over the 10-year period 1997–2006, the average annual rate of notified cryptosporidiosis was 22.0 cases per 100,000 population. The number of hospitalisations was equivalent to 3.6% of the notified cases. There was only 1 reported fatality. The annual incidence of infection appeared fairly stable, but showed marked seasonality with a peak rate in spring (September–November in New Zealand). The highest rates were among Europeans, children 0–9 years of age, and those living in low deprivation areas. Notification rates showed large geographic variations, with rates in rural areas 2.8 times higher than in urban areas, and with rural areas also experiencing the most pronounced spring peak. At the territorial authority (TA) level, rates were also correlated with farm animal density.

Conclusions Most transmission of Cryptosporidium in New Zealand appears to be zoonotic: from farm animals to humans. Prevention should focus on reducing transmission in rural setting, though more research is needed to identify which strategies are likely to be most effective in that environment.

Aims New Zealand has a higher incidence rate of giardiasis than other developed countries. This study aimed to describe the epidemiology of this disease in detail and to identify potential risk factors.

Methods We analysed anonymous giardiasis notification (1997–2006) and hospitalisation data (1990–2006). Cases were designated as urban or rural and assigned a deprivation level based on their home address. Association between disease rates and animal density was studied using a simple linear regression model, at the territorial authority (TA) level.

Results Over the 10-year period 1997–2006 the average annual rate of notified giardiasis was 44.1 cases per 100,000 population. The number of hospitalisations was equivalent to 1.7% of the notified cases. There were 2 reported fatalities. The annual incidence of notified cases declined over this period whereas hospitalisations remained fairly constant. Giardiasis showed little seasonality. The highest rates were among children 0–9 years old, those 30–39 years old, Europeans, and those living in low deprivation areas. Notification rates were slightly higher in rural areas. The correlation between giardiasis and farm animal density was not significant at the TA level.

Conclusions The public health importance of giardiasis to New Zealand mainly comes from its relatively high rates in this country. The distribution of cases is consistent with largely anthroponotic (human) reservoirs, with a relatively small contribution from zoonotic sources in rural environments and a modest contribution from overseas travel. Prevention efforts could include continuing efforts to improve hand washing, nappy handling, and other hygiene measures and travel health advice relating to enteric infections.

Room temperature synchrotron x-ray diffraction experiments were performed on nanocrystal Ta2N in a diamond anvil cell to a pressure of 55.48 GPa. This nitride is a well-known kind of high-hardness material, and it is found to be highly incompressible. The structure is stable with no phase transitions observed in this pressure range. The zero-pressure bulk modulus and its pressure derivative at ambient pressure are B0 = 360 ± 3 GPa, B0' = 4, and in room conditions, the a and c parameters are 3.054 Å, 4.996 Å, respectively. The bulk modulus of Ta2N is greater than those of TaC, ε-TaN, Cr2N and MoN. The differences in bulk moduli might be due to the differences in structure and the cohesive energy among these phases.

In this paper, a hybrid neural classifier combining the auto-encoder neural network and the Lattice Vector Quantization (LVQ) model is described. The auto-encoder network is used for dimensionality reduction by projecting high dimensional data into the 2D space. The LVQ model is used for data visualization by forming and adapting the granularity of a data map. The mapped data are employed to predict the target classes of new data samples. To improve classification accuracy, a majority voting scheme is adopted by the hybrid classifier. To demonstrate the applicability of the hybrid classifier, a series of experiments using simulated and real fault data from induction motors is conducted. The results show that the hybrid classifier is able to outperform the Multi-Layer Perceptron neural network, and to produce very good classification accuracy rates for various fault conditions of induction motors.

Triplicate groups of 20 European sea bass (35 g) were fed five diets in which the added lipid was 100% fish oil (FO), 40% (CSO40), 60% (CSO60), 80% (CSO80) and 100% (CSO100) refined cottonseed oil (CSO), for a period of 120 days. Overall fish growth, feed conversion ratio and protein utilization were unaffected by dietary treatment, but hepatosomatic and visceral fat indexes increased with increasing dietary CSO. Fillet fatty acid composition of total lipids reflected the fatty acids in the test diets. The monounsaturated fatty acids were significantly higher in fillet of fish fed diet FO, CSO40 and CSO60 compared to other treatments while saturated and polyunsaturated fatty acids (PUFA) were not affected by the dietary treatment. Some fatty acids (18:0, 18:1n-9, 20:5n-3 and 22:6n-3) were present in higher concentration in fillet lipid than in the CSO100 dietary lipid indicating accumulation in fillet relative to test diets. Retention of n-3 LC-PUFA within the fillet was increasingly inefficient among fish fed increasing levels of FO. Thus, this study suggests that CSO can be considered as a relatively effective substitute for fish oil in European sea bass (35 g) in terms of growth performances and feed efficiency as far as fish meal is present in the diet.

The purpose of the present study was to determine if there is an acclimation effect when unacclimatized males exercise in the heat at weekly intervals. Five subjects performed four exercise bouts, each lasting 1 h at 55% VO2max. The first trial was in moderate conditions (mean(s.d.) temperature (Ta) = 22.0(0.8)degrees C; mean(s.d.) relative humidity (rh) = 67(6)%) and the subsequent three trials were carried out at weekly intervals in the heat (mean(s.d.) Ta = 34.6(0.6)degrees C; mean(s.d.) rh = 60(7)%). There were no significant differences between trials in the heat for heart rate, rectal temperature, skin temperature or VO2 (repeated measures analysis of variance), and total sweat loss (one-way analysis of variance). As changes in these variables are seen with heat acclimation it was concluded that there was no heat acclimation effect and separating exercise bouts by 1 week was a valid method for comparing the effects of different treatments on unacclimatized males during exercise in the heat.
