The full-length human immunodeficiency virus type 1 (HIV-1) mRNA encodes two precursor polyproteins, Gag and GagProPol. An infrequent ribosomal frameshifting event allows these proteins to be synthesized from the same mRNA in a predetermined ratio of 20 Gag proteins for each GagProPol. The RNA frameshift signal consists of a slippery sequence and a hairpin stem-loop whose thermodynamic stability has been shown in in vitro translation systems to be critical to frameshifting efficiency. In this study we examined the frameshift region of HIV-1, investigating the effects of altering stem-loop stability in the context of the complete viral genome and assessing the role of the Gag spacer peptide p1 and the GagProPol transframe (TF) protein that are encoded in this region. By creating a series of frameshift region mutants that systematically altered the stability of the frameshift stem-loop and the protein sequences of the p1 spacer peptide and TF protein, we have demonstrated the importance of stem-loop thermodynamic stability in frameshifting efficiency and viral infectivity. Multiple changes to the amino acid sequence of p1 resulted in altered protein processing, reduced genomic RNA dimer stability, and abolished viral infectivity. The role of the two highly conserved proline residues in p1 (position 7 and 13) was also investigated. Replacement of the two proline residues by leucines resulted in mutants with altered protein processing and reduced genomic RNA dimer stability that were also noninfectious. The unique ability of proline to confer conformational constraints on a peptide suggests that the correct folding of p1 may be important for viral function.

Traditional practical classes in many countries are being rationalised to reduce costs. The challenge for university educators is to provide students with the opportunity to reinforce theoretical concepts by running something other than a traditional practical program. One alternative is to replace wet labs with comparable computer simulations. These virtual experiments involve no harm to animals and require little ongoing expenditure. This study documents second-year physiology students' perceptions of and attitudes to simulations by incorporating several computer simulations into the practical program. Computer simulations met the conceptual and, to some extent, the motivational goals of university practical programs. While students enjoyed both wet labs and computer-simulated exercises, overwhelmingly the wet lab provided the more memorable and stimulating learning experience. Based on this study, students suggested that computer simulations could be effectively used to complement rather than replace practical classes where students gain laboratory skills.

Information technology outsourcing has become a pervasive and important phenomenon in business organizations and there is substantial evidence about its benefits and pitfalls. Initially, firms used outsourcing as a way to lower costs, gain access to expertise and focus on core activities. Recently, there is a shift in focus and more firms are outsourcing to attain innovative products and services. However, current research is still unclear about how innovation can be achieved through outsourcing. Drawing predominantly from the dynamic capability theory, the objective of this paper is to explore how absorptive capacity unfolds as a process within and between firms when client firms outsource their information technology services with expectations of innovation generation. In this paper, we propose a research model that links absorptive capacity to innovation generation. We draw on three case studies to focus on how absorptive capacity, as a process, impacts innovation generation. Results show that assimilation and transformation stages are critical in generating radical innovation while acquisition and exploitation play a key role in incremental innovation. The implications of these findings for both researchers and practitioners are discussed.

IF steel sheets were processed by conventional symmetric and asymmetric rolling (ASR) at ambient temperature. The asymmetry was introduced in a geometric way using differential roll diameters with a number of different ratios. The material strength was measured by tensile testing and the microstructure was analyzed by optical and transmission electron microscopy as well as electron backscatter diffraction (EBSD) analysis. Texture was also successfully measured by EBSD using large surface areas. Finite element (FE) simulations were carried out for multiple passes to obtain the strain distribution after rolling. From the FE results, the velocity gradient along selected flow lines was extracted and the evolution of the texture was simulated using polycrystal plasticity modeling. The best mechanical properties were obtained after ASR using a roll diameter ratio of 2. The textures appeared to be tilted up to 12 deg around the transverse direction, which were simulated with the FE-combined polycrystal plasticity modeling in good agreement with measurements. The simulation work revealed that the shear component introduced by ASR was about the same magnitude as the normal component of the rolling strain tensor.

There is currently no universally recommended and accepted method of data processing within the science of indirect calorimetry for either mixing chamber or breath-by-breath systems of expired gas analysis. Exercise physiologists were first surveyed to determine methods used to process oxygen consumption ([OV0312]O 2) data, and current attitudes to data processing within the science of indirect calorimetry. Breath-by-breath datasets obtained from indirect calorimetry during incremental exercise were then used to demonstrate the consequences of commonly used time, breath and digital filter post-acquisition data processing strategies. Assessment of the variability in breath-by-breath data was determined using multiple regression based on the independent variables ventilation (VE), and the expired gas fractions for oxygen and carbon dioxide, FEO 2 and FECO2, respectively. Based on the results of explanation of variance of the breath-by-breath [OV0312]O2 data, methods of processing to remove variability were proposed for time-averaged, breath-averaged and digital filter applications. Among exercise physiologists, the strategy used to remove the variability in sequential [OV0312]O2 measurements varied widely, and consisted of time averages (30 sec [38%], 60 sec [18%], 20 sec [11%], 15 sec [8%]), a moving average of five to 11 breaths (10%), and the middle five of seven breaths (7%). Most respondents indicated that they used multiple criteria to establish maximum [OV0312]O 2 ([OV0312]O2max) including: the attainment of age-predicted maximum heart rate (HRmax) [53%], respiratory exchange ratio (RER) >1.10 (49%) or RER >1.15 (27%) and a rating of perceived exertion (RPE) of >17, 18 or 19 (20%). The reasons stated for these strategies included their own beliefs (32%), what they were taught (26%), what they read in research articles (22%), tradition (13%) and the influence of their colleagues (7%). The combination of VE, FEO 2 and FECO2 removed 96-98% of [OV0312]O2 breath-by-breath variability in incremental and steady-state exercise [OV0312]O2 data sets, respectively. Correction of residual error in [OV0312]O2 datasets to 10% of the raw variability results from application of a 30-second time average, 15-breath running average, or a 0.04 Hz low cut-off digital filter. Thus, we recommend that once these data processing strategies are used, the peak or maximal value becomes the highest processed datapoint. Exercise physiologists need to agree on, and continually refine through empirical research, a consistent process for analysing data from indirect calorimetry.

Older adults with congestive heart failure [CHF] are likely to experience multiple readmissions to hospital. There have been several studies conducted on hospital readmissions; however, generalising the findings is problematic due to the use of variable definitions of what constitutes a readmission. This paper addresses the absence of Australian research comparing groups of older patients with CHF who are readmitted to hospital with those who are not readmitted. It also adopts one of the more frequently used definitions of readmission to aid in future comparability of research. Using a comparative cohort design, a multivariate logistic regression model was used to compare readmitted patients with non-readmitted patients and identify risk factors associated with readmission. Significant risk factors identified were male gender, numerous diagnoses, lengths of stay of 3 days or longer and admission from acute, subacute or aged care facilities. The increased likelihood of readmission among patients from acute, subacute and aged care services warrants further investigation.

The Macquarie perch (Macquaria australasica) is a threatened fish species that inhabits rivers and impoundments in south-eastern Australia. Previous studies have shown that Macquarie perch in impoundments exhibit synchronised upstream spawning migrations to shallow, fast-flowing habitats in the lower reaches of inflowing streams. There has been little study of movement behaviours of entirely riverine populations of Macquarie perch despite this being the species’ natural habitat. Here, radio-telemetry is used to test the hypothesis that riverine populations exhibit synchronised migrations during the spawning season. Thirty Macquarie perch in the Yarra River, Victoria, a translocated population outside of the species’ natural range, were radio-tagged before the late spring–early summer spawning season and their movements followed over a 10-month period (May 2011 to February 2012). Tagged fish typically occupied restricted reaches of stream (<450 m). Sixteen of the fish undertook occasional upstream or downstream movements (~250–1000 m) away from their usual locations, particularly associated with large flow variations during the spawning season. There was no evidence of synchronised migratory behaviour or movement of multiple fish to specific locations or habitats during the spawning season. Whilst further research over more years is needed to comprehensively document the spawning-related behaviours of riverine Macquarie perch, our study demonstrates that management of riverine populations of this threatened species cannot necessarily be based on the model of spawning behaviour developed for lacustrine populations.

The present paper uses a dynamic open-economy model with wage indexation to examine the impact of tourism on employment and welfare. Both short-run and long-run situations are analysed. It is well known that tourism converts non-traded goods into tradable goods. An increase in the demand for a non-traded good raises its relative price, which results in an expansion of the non-traded sector at the expense of the traded goods sector. This output shift raises labour employment in the short run. However, in the long run, the higher relative price leads to higher wages, resulting in a negative impact on labour employment. If the output effect is dominant, the expansion in tourism raises employment and welfare. However, under realistic conditions tourism may lower both labour employment and welfare due to rising costs. These results are demonstrated by simulating a dynamic model for the case of Hong Kong.

 Kinect has been increasingly applied in rehabilitation as a motion capture device. However, the inherent limitations significantly hinder its further development in this important area. Although a number of Kinect fusion approaches have been proposed, only a few of them was actually considered for rehabilitation. In this paper, we propose to fuse information from multiple Kinects to achieve this. Given the specific scenario of users suffering from limited range of movements, we propose to calibrate depth cameras in multiple Kinects with 3D positions of joints on a human body rather than in a checkerboard pattern, so that patients are able to calibrate Kinects without extra support. Kalman filter is applied for skeleton-wise Kinect fusion since skeleton data (3D positions of joints) and its derivatives are preferred by physiotherapists to evaluate the exercise performance of patients. Various preliminary experiments were conducted to illustrate the accuracy of proposed calibration and fusion approach by comparing with a commercial Vicon system®, confirming the practical use of the system in rehabilitation exercise monitoring.

The narrative of William Wallace holds a prominent position in the current conception of England as a negative referent for Scotland’s national identity—its binary “Other”, against which Wallace valiantly fought. This article considers a contrasting understanding of Scottish national identity from the late-nineteenth century, and explores the events surrounding the unveiling of a statue of William Wallace in Australia during the year of 1889. It illuminates how settlers interpreted this national hero in such a way that demonstrated loyalty to the Union and Empire, and accommodated a convergence of English, Scottish, Irish, and Welsh migrants in a British colonial city. The article highlights how statues, the ceremonies surrounding them, and their public reception help us to investigate the symbolic, ritualistic, and performative dimensions of identity formulation. It considers how public monuments, providing a sense of authority to particular groups, can marginalise others by acting to settle cultural competition, and will reflect on competing interpretations of the statue at its unveiling.

BACKGROUND: In developed countries, individuals experiencing socioeconomic disadvantage - whether a low education level, low income, low-status occupation, or living in a socioeconomically disadvantaged neighbourhood - are less likely than those more advantaged to engage in eating and physical activity behaviours conducive to optimal health. These socioeconomic inequities in nutrition and physical activity (and some sedentary) behaviours are graded, persistent, and evident across multiple populations and studies. They are concerning in that they mirror socioeconomic inequities in obesity and in health outcomes. Yet there remains a dearth of evidence of the most effective means of addressing these inequities. People experiencing disadvantage face multiple challenges to healthy behaviours that can appear insurmountable. With increasing recognition of the role of underlying structural and societal factors as determinants of nutrition and physical activity behaviours and inequities in these behaviours, and the limited success of behaviour change approaches in addressing these inequities, we might wonder whether there remains a role for behavioural scientists to tackle these challenges. DISCUSSION: This debate piece argues that behavioural scientists can play an important role in addressing socioeconomic inequities in nutrition, physical activity and sedentary behaviours, and that this will involve challenging myths and taking on new perspectives. There are successful models for doing so from which we can learn. Addressing socioeconomic inequities in eating, physical activity and sedentary behaviours is challenging. However, successful examples demonstrate that overcoming such challenges is possible, and provide guidance for doing so. Given the disproportionate burden of ill health carried by people experiencing socioeconomic disadvantage, all our nutrition and physical activity interventions, programs and policies should be designed to reach and positively impact these individuals at greatest need.

 BACKGROUND: Interactions between wildlife and humans are increasing. Urban animals are often less wary of humans than their non-urban counterparts, which could be explained by habituation, adaptation or local site selection. Under local site selection, individuals that are less tolerant of humans are less likely to settle in urban areas. However, there is little evidence for such temperament-based site selection, and even less is known about its underlying genetic basis. We tested whether site selection in urban and non-urban habitats by black swans (Cygnus atratus) was associated with polymorphisms in two genes linked to fear in animals, the dopamine receptor D4 (DRD4) and serotonin transporter (SERT) genes.

RESULTS: Wariness in swans was highly repeatable between disturbance events (repeatability = 0.61) and non-urban swans initiated escape from humans earlier than urban swans. We found no inter-individual variation in the SERT gene, but identified five DRD4 genotypes and an association between DRD4 genotype and wariness. Individuals possessing the most common DRD4 genotype were less wary than individuals possessing rarer genotypes. As predicted by the local site selection hypothesis, genotypes associated with wary behaviour were over three times more frequent at the non-urban site. This resulted in moderate population differentiation at DRD4 (FST = 0.080), despite the sites being separated by only 30 km, a short distance for this highly-mobile species. Low population differentiation at neutrally-selected microsatellite loci and the likely occasional migration of swans between the populations reduces the likelihood of local site adaptations.

CONCLUSION: Our results suggest that wariness in swans is partly genetically-determined and that wary swans settle in less-disturbed areas. More generally, our findings suggest that site-specific management strategies may be necessary that consider the temperament of local animals.

Better managing diabetes has become a global priority, especially given the exponential increase in the number of diabetes patients and the financial implications of treating this silent epidemic. In this paper, we focus on how it might be possible to use a mobile technology solution to support and enable superior diabetes monitoring and management. To test this solution, we examined the context of gestational diabetes and adopted a non-blinded randomized control trial with two-arm cross over applied to a private hospital in Victoria, Australia. Further, we use an accountable care system as the theoretical lens and, from this, develop a conceptual framework to bridge evidence-based management with technologies. Theoretically, we unpack McCleallan, McKethan, Lewis, Roski, and Fisher’s (2010) study with our conceptual framework that comprises providers for information (evidence-based management) and technology (smartphone). We enhance Muhlestein, Croshaw, Merrill, Pena, and James’ (2013) accountable care paradigm with three concepts: 1) quality of life, 2) evidence-based management, and 3) affordable care. From the perspective of practice, far-reaching implications have arisen particularly for hospital management pertaining to the cost and quality of care issues. In particular, it appears that adapting mobile technology solutions such as smartphones to support various aspects of care and patient-clinician interactions is a prudent choice to minimize costs and yet provide highquality care.

Abstract—
After a decade of extensive research on application-specific wireless sensor networks (WSNs), the recent development of information and communication technologies makes it practical to realize the software-defined sensor networks (SDSNs), which are able to adapt to various application requirements and to fully explore the resources of WSNs. A sensor node in SDSN is able to conduct multiple tasks with different sensing targets simultaneously. A given sensing task usually involves multiple sensors to achieve a certain quality-of-sensing, e.g., coverage ratio. It is significant to design an energy-efficient sensor scheduling and management strategy with guaranteed quality-of-sensing for all tasks. To this end, three issues are investigated in this paper: 1) the subset of sensor nodes that shall be activated, i.e., sensor activation, 2) the task that each sensor node shall be assigned, i.e., task mapping, and 3) the sampling rate on a sensor for a target, i.e., sensing scheduling. They are jointly considered and formulated as a mixed-integer with quadratic constraints programming (MIQP) problem, which is then reformulated into a mixed-integer linear programming (MILP) formulation with low computation complexity via linearization. To deal with dynamic events such as sensor node participation and departure, during SDSN operations, an efficient online algorithm using local optimization is developed. Simulation results show that our proposed online algorithm approaches the globally optimized network energy efficiency with much lower rescheduling time and control overhead.

Face recognition with multiple views is a challenging research problem. Most of the existing works have focused on extracting shared information among multiple views to improve recognition. However, when the pose variation is too large or missing, 'shared information' may not be properly extracted, leading to poor recognition results. In this paper, we propose a novel method for face recognition with multiple view images to overcome the large pose variation and missing pose issue. By introducing a novel mixed norm, the proposed method automatically selects candidates from the gallery to best represent a group of highly correlated face images in a query set to improve classification accuracy. This mixed norm combines the advantages of both sparse representation based classification (SRC) and joint sparse representation based classification (JSRC). A trade off between the ℓ1-norm from SRC and ℓ2,1-norm from JSRC is introduced to achieve this goal. Due to this property, the proposed method decreases the influence when a face image is unseen and has large pose variation in the recognition process. And when some face images with a certain degree of unseen pose variation appear, this mixed norm will find an optimal representation for these query images based on the shared information induced from multiple views. Moreover, we also address an open problem in robust sparse representation and classification which is using ℓ1-norm on the loss function to achieve a robust solution. To solve this formulation, we derive a simple, yet provably convergent algorithm based on the powerful alternative directions method of multipliers (ADMM) framework. We provide extensive comparisons which demonstrate that our method outperforms other state-of-the-arts algorithms on CMU-PIE, Yale B and Multi-PIE databases for multi-view face recognition.
