Isolation of gene transcripts from desiccated leaf tissues of the resurrection grass, Sporobolus stapfianus, resulted in the identification of a gene, SDG8i, encoding a Group 1 glycosyltransferase (UGT). Here, we examine the effects of introducing this gene, under control of the CaMV35S promoter, into the model plant Arabidopsis thaliana. Results show that Arabidopsis plants constitutively over-expressing SDG8i exhibit enhanced growth, reduced senescence, cold tolerance and a substantial improvement in protoplasmic drought tolerance. We hypothesise that expression of SDG8i in Arabidopsis negatively affects the bioactivity of metabolite/s that mediate/s environmentally-induced repression of cell division and expansion, both during normal development and in response to stress. The phenotype of transgenic plants over-expressing SDG8i suggests modulation in activities of both growth- and stress-related hormones. Plants overexpressing the UGT show evidence of elevated auxin levels, with the enzyme acting downstream of ABA to reduce drought-induced senescence. Analysis of the in vitro activity of the UGT recombinant protein product demonstrates that SDG8i can glycosylate the synthetic strigolactone analogue GR24, evoking a link with strigolactone-related processes in vivo. The large improvements observed in survival of transgenic Arabidopsis plants under cold-, salt- and drought-stress, as well as the substantial increases in growth rate and seed yield under non-stress conditions, indicates that overexpression of SDG8i in crop plants may provide a novel means of increasing plant productivity.

Muscle fatty acid (FA) metabolism is impaired in obesity and insulin resistance, reflected by reduced rates of FA oxidation and accumulation of lipids. It has been suggested that interventions that increase FA oxidation may enhance insulin action by reducing these lipid pools. Here, we examined the effect of endurance training on rates of mitochondrial FA oxidation, the activity of carnitine palmitoyltransferase I (CPT I), and the lipid content in muscle of obese individuals and related these to measures of glucose tolerance. Nine obese subjects completed 8 wk of moderate-intensity endurance training, and muscle biopsies were obtained before and after training. Training significantly improved glucose tolerance, with a reduction in the area under the curve for glucose (P< 0.05) and insulin (P = 0.01) during an oral glucose tolerance test. CPT I activity increased 250% (P = 0.001) with training and became less sensitive to inhibition by malonyl-CoA. This was associated with an increase in mitochondrial FA oxidation (+120%, P < 0.001). Training had no effect on muscle triacylglycerol content; however, there was a trend for training to reduce both the total diacylglcyerol (DAG) content (−15%, P = 0.06) and the saturated DAG-FA species (−27%, P = 0.06). Training reduced both total ceramide content (−42%, P = 0.01) and the saturated ceramide species (−32%, P < 0.05). These findings suggest that the improved capacity for mitochondrial FA uptake and oxidation leads not only to a reduction in muscle lipid content but also a to change in the saturation status of lipids, which may, at least in part, provide a mechanism for the enhanced insulin action observed with endurance training in obese individuals.

Background:
Two small studies had evaluated the efficacy of rTMS in migraine. One tested high frequency rTMS over the dorsolateral prefrontal cortex while the other evaluated 1 Hz rTMS over the vertex.
Aim:
To test the feasibility of 10 Hz rTMS of motor cortex as an adjunctive therapy in patients with chronic migraine.
Materials and Methods:
We randomized (2:1 ratio) chronic migraine patients on medical preventive treatment to receive either rTMS or sham therapy for 10 sessions. rTMS (80% resting motor threshold, 10Hz, 20 trains, 5 secs/train, inter-train interval 1 min, total 1000 stimuli/session) was applied over the right motor cortex.
Result:
Nine patients were randomized. Six received rTMS
and three had sham therapy. Three patients in the rTMS arm withdrew from the study due to increased headache frequency and discomfort from the treatment. The remaining six cases (3 rTMS, 3 sham) completed the study. The study was prematurely stopped due to the significant worsening of headache from rTMS. No significant differences in outcome measures were found between real and sham rTMS.
Conclusion:
Although the study was terminated prematurely, the high dropout rate (50%) due to worsening headaches suggested that rTMS over the motor cortex is poorly tolerated in chronic migraine.

Issue addressed: To evaluate the effectiveness of a brief intervention using a pedometer and step-recording diary on promoting physical activity in people with type 2 diabetes or impaired glucose tolerance (IGT). Methods: People with type 2 diabetes or IGT who attended the Illawarra Diabetes Service were invited to participate. Participants in the intervention group received a pedometer and a diary to record their daily steps for a two-week period. Both the intervention and comparison group received advice on physical activity. Physical activity levels were measured using the Active Australia Survey at baseline, and at two and 20 weeks. Results: A total of 226 participants were recruited. At two-week follow-up the mean self-reported minutes of walking was significantly higher in the intervention group than the comparison group (223 minutes versus 164 minutes; p=0.01), as was the percentage of intervention participants achieving recommended levels of moderate-intensity physical activity (63.5% versus 41.8%, p=0.02) and the percentage of intervention participants achieving adequate levels of total physical activity (68.9% versus 48.0%, p=0.04). There were no differences between study groups for any physical activity measure at 20-week follow-up. Conclusions: A pedometer and a step-recording diary were useful tools to promote short-term increase in physical activity in people diagnosed with type 2 diabetes or IGT. Future studies need to examine whether a longer intervention, individualised physical activity counselling and support for achieving step goals could result in increasing physical activity over the long term.

In data gathering wireless sensor networks, data loss often happens due to external faults such as random link faults and hazard node faults, since sensor nodes have constrained resources and are often deployed in inhospitable environments. However, already known fault tolerance mechanisms often bring new internal faults (e.g. out-of-power faults and collisions on wireless bandwidth) to the original network and dissipate lots of extra energy and time to reduce data loss. Therefore, we propose a novel Dual Cluster Heads Cooperation (CoDuch) scheme to tolerate external faults while introducing less internal faults and dissipating less extra energy and time. In CoDuch scheme, dual cluster heads cooperate with each other to reduce extra costs by sending only one copy of sensed data to the Base Station; also, dual cluster heads check errors with each other during the collecting data process. Two algorithms are developed based on the CoDuch scheme: CoDuch-l for tolerating link faults and CoDuch-b for tolerating both link faults and node faults; theory and experimental study validate their effectiveness and efficiency. © 2010 The Author Published by Oxford University Press on behalf of The British Computer Society. The incidence of food allergies in western countries has increased dramatically in recent decades. Tolerance to food antigens relies on mucosal CD103(+) dendritic cells (DCs), which promote differentiation of regulatory T (Treg) cells. We show that high-fiber feeding in mice improved oral tolerance and protected from food allergy. High-fiber feeding reshaped gut microbial ecology and increased the release of short-chain fatty acids (SCFAs), particularly acetate and butyrate. High-fiber feeding enhanced oral tolerance and protected against food allergy by enhancing retinal dehydrogenase activity in CD103(+) DC. This protection depended on vitamin A in the diet. This feeding regimen also boosted IgA production and enhanced T follicular helper and mucosal germinal center responses. Mice lacking GPR43 or GPR109A, receptors for SCFAs, showed exacerbated food allergy and fewer CD103(+) DCs. Dietary elements, including fiber and vitamin A, therefore regulate numerous protective pathways in the gastrointestinal tract, necessary for immune non-responsiveness to food antigens.

The examination of links between a high degree of encephalisation (i.e., a large brain mass relative to body size) and the capacity of wildlife to inhabit anthropogenic habitats has formed the basis of several recent studies, although typically they have not uncovered any relationship. It, however, remains unclear whether encephalisation is directly related to a species' capacity to develop tolerance to human proximity (i.e., a reduction in response to approaching humans). It is also unknown whether such a relationship is related to the size of specific areas of the brain. Using published data on flight-initiation distance (FID), the distance at which animals flee from an approaching human, we estimate the degree of tolerance of human proximity for 42 bird species by comparing FIDs in urban and rural areas, with relatively high and low exposure to humans, respectively. We used a phylogenetic, comparative approach to analyse the relationship of degree of tolerance, and of FID in urban and rural populations more directly, to relative sizes of whole brains (42 species) and brain components (25 species) for the species, and examine the effect of the year that the bird species was first recorded in an urban area (year of urbanization). We demonstrate an interaction between bird habitat and year of urbanization on FIDs. Urban populations of species that have a longer history of inhabiting urban areas have lower FIDs (i.e., birds that were urbanized earlier are more tolerant), which may suggest local selection for birds with reduced responsiveness to humans in urban areas. The pattern is not seen in rural populations of the same species, providing additional evidence that it is greater exposure to humans that has resulted in this tolerance. While we found that forebrain mass and optic lobe mass are influential positive predictors of FID there was no indication that degree of tolerance itself was related to any brain size metric and hence no support for the idea that urban populations of species with larger brains are better able to habituate to human presence. This suggests that processes other than encephalisation explain the high degree of tolerance evident in urban-dwelling birds.

Wireless body area networks (WBANs), as a promising health-care system, can provide tremendous benefits for timely and continuous patient care and remote health monitoring. Owing to the restriction of communication, computation and power in WBANs, cloud-assisted WBANs, which offer more reliable, intelligent, and timely health-care services for mobile users and patients, are receiving increasing attention. However, how to aggregate the health data multifunctionally and efficiently is still an open issue to the cloud server (CS). In this paper, we propose a privacy-preserving and multifunctional health data aggregation (PPM-HDA) mechanism with fault tolerance for cloud-assisted WBANs. With PPM-HDA, the CS can compute multiple statistical functions of users' health data in a privacy-preserving way to offer various services. In particular, we first propose a multifunctional health data additive aggregation scheme (MHDA+) to support additive aggregate functions, such as average and variance. Then, we put forward MHDA as an extension of MHDA+ to support nonadditive aggregations, such as min/max, median, percentile, and histogram. The PPM-HDA can resist differential attacks, which most existing data aggregation schemes suffer from. The security analysis shows that the PPM-HDA can protect users' privacy against many threats. Performance evaluations illustrate that the computational overhead of MHDA+ is significantly reduced with the assistance of CSs. Our MHDA scheme is more efficient than previously reported min/max aggregation schemes in terms of communication overhead when the applications require large plaintext space and highly accurate data.
