With the salmonid industry currently exploiting the vast majority of globally available fish oil, there is the need to optimise fish oil utilisation by increasing its efficiency in terms of transferring the health-promoting long chain omega-3 fatty acids (n−3 LC-PUFA) into farmed fish flesh. The aim of this study was to evaluate if dietary fatty acid deposition is affected by the time of feeding, and hence identify possible innovative feeding strategies towardsmore efficient use of dietary fish oil. Over a period of 12 weeks, three diets with different lipid sources, canola oil (CO), fish oil (FO) or a 50/50 blend of the two oils (Mix), were alternated daily and fed to rainbow trout (Oncorhynchus mykiss). Six treatments were administered to fish, reference treatment (REF, continuously fed FO), control treatment (CT, continuously fed Mix), am canola oil ration (amCOR), pm canola oil ration (pmCOR), am canola oil satiation (amCOS) and pm canola oil satiation (pmCOS). Fish received either the CO diet in the am or pm feeds and received the FO diet at the opposite time. A significant increase in growth and feed consumption was noted in the pmCOS treatment. Fillet fatty acid profile was modified by associated feeding schedules and was generally reflective of dietary fatty acid profile. No significant increases in n−3 LCPUFA deposition were observed. However, both linoleic acid (18:2n−6) and α-linolenic acid (18:3n−3) contents were significantly higher in pmCOR compared to amCOR and CT. The results of the present study suggest the existence of cyclical circadian patterns in fatty acid deposition in rainbow trout.

A study of the cholesterol content and fatty acid composition of fresh retail Australian pork was undertaken to determine whether new breeding, feeding and processing methods had resulted in any compositional changes in fresh pork in the market place since surveys undertaken in previous decades. Samples of 13 popular pork cuts were purchased from randomly selected supermarkets and butchers’ stores in urban areas across the socioeconomic scale in three States of Australia, and analysed, separable fat and separable lean, in late 2005 and early 2006. Variability was low across States for saturated and monounsaturated fatty acids, but more pronounced for polyunsaturated acids. The separable lean portions of all pork cuts contained levels of n-3 fatty acids and conjugated linoleic acid (C18:1c9t11) in measurable but not nutritionally claimable amounts, whilst total trans fatty acid levels were very low. There appeared to be some differences in fatty acid composition across States that may have resulted from feeding method. Cholesterol contents were similar to levels in the 80s and 90s for separable lean pork tissue, but presently are lower for separable fat tissue than for separable lean.

Triplicate groups of 20 European sea bass (35 g) were fed five diets in which the added lipid was 100% fish oil (FO), 40% (CSO40), 60% (CSO60), 80% (CSO80) and 100% (CSO100) refined cottonseed oil (CSO), for a period of 120 days. Overall fish growth, feed conversion ratio and protein utilization were unaffected by dietary treatment, but hepatosomatic and visceral fat indexes increased with increasing dietary CSO. Fillet fatty acid composition of total lipids reflected the fatty acids in the test diets. The monounsaturated fatty acids were significantly higher in fillet of fish fed diet FO, CSO40 and CSO60 compared to other treatments while saturated and polyunsaturated fatty acids (PUFA) were not affected by the dietary treatment. Some fatty acids (18:0, 18:1n-9, 20:5n-3 and 22:6n-3) were present in higher concentration in fillet lipid than in the CSO100 dietary lipid indicating accumulation in fillet relative to test diets. Retention of n-3 LC-PUFA within the fillet was increasingly inefficient among fish fed increasing levels of FO. Thus, this study suggests that CSO can be considered as a relatively effective substitute for fish oil in European sea bass (35 g) in terms of growth performances and feed efficiency as far as fish meal is present in the diet.

Macrophage apoptosis, a key process in atherogenesis, is regulated by oxidation products, including hydroxyoctadecadienoic acids (HODEs). These stable oxidation products of linoleic acid (LA) are abundant in atherosclerotic plaque and activate PPARγ and GPR132. We investigated the mechanisms through which HODEs regulate apoptosis. The effect of HODEs on THP-1 monocytes and adherent THP-1 cells were compared with other C18 fatty acids, LA and α-linolenic acid (ALA). The number of cells was reduced within 24 hours following treatment with 9-HODE (p < 0.01, 30 μM) and 13 HODE (p < 0.01, 30 μM), and the equivalent cell viability was also decreased (p < 0.001). Both 9-HODE and 13-HODE (but not LA or ALA) markedly increased caspase-3/7 activity (p < 0.001) in both monocytes and adherent THP-1 cells, with 9-HODE the more potent. In addition, 9-HODE and 13-HODE both increased Annexin-V labelling of cells (p < 0.001). There was no effect of LA, ALA, or the PPARγ agonist rosiglitazone (1μM), but the effect of HODEs was replicated with apoptosis-inducer camptothecin (10μM). Only 9-HODE increased DNA fragmentation. The pro-apoptotic effect of HODEs was blocked by the caspase inhibitor DEVD-CHO. The PPARγ antagonist T0070907 further increased apoptosis, suggestive of the PPARγ-regulated apoptotic effects induced by 9-HODE. The use of siRNA for GPR132 showed no evidence that the effect of HODEs was mediated through this receptor. 9-HODE and 13-HODE are potent—and specific—regulators of apoptosis in THP-1 cells. Their action is PPARγ-dependent and independent of GPR132. Further studies to identify the signalling pathways through which HODEs increase apoptosis in macrophages may reveal novel therapeutic targets for atherosclerosis.

Inflammatory bowel disease (IBD) is a poorly understood chronic immune disorder for which there is no medical cure. Milk and colostrum are rich sources of bioactives with immunomodulatory properties. Here we compared the therapeutic effects of oral delivery of bovine milk-derived iron-saturated lactoferrin (Fe-bLF), angiogenin, osteopontin (OPN), colostrum whey protein, Modulen IBD (Nestle Healthsciences, Rhodes, Australia), and cis-9,trans-11 conjugated linoleic acid (CLA)-enriched milk fat in a mouse model of dextran sulfate-induced colitis. The CLA-enriched milk fat significantly increased mouse body weights after 24d of treatment, reduced epithelium damage, and downregulated the expression of proinflammatory cytokines and nitrous oxide. Modulen IBD most effectively decreased the clinical score at d 12, and Modulen IBD and OPN most effectively lowered the inflammatory score. Myeloperoxidase activity that denotes neutrophil infiltration was significantly lower in mice fed Modulen IBD, OPN, angiogenin, and Fe-bLF. A significant decrease in the numbers of T cells, natural killer cells, dendritic cells, and a significant decrease in cytokine expression were observed in mice fed the treatment diets compared with dextran sulfate administered mice. The Fe-bLF, CLA-enriched milk fat, and Modulen IBD inhibited intestinal angiogenesis. In summary, each of the milk components attenuated IBD in mice, but with differing effectiveness against specific disease parameters.

The objective of this study was to investigate the incorporation and metabolism of punicic acid (PA, cis9,trans11,cis13-18:3) in healthy young humans. The study was a randomized controlled trial. After 7 days adaptation with sunflower seed kernels supplementation, 30 subjects were then divided into the control and test group (n = 15). The test group was supplemented with Trichosanthes kirilowii (TK) seed kernels containing 3 g of PA per day in the form of triacylglycerols for 28 days. The control group was provided with sunflower seed kernels. After consumption of TK seeds containing 3 g PA per day for 28 days, the proportion of PA was increased from 0.00 to 0.47% in plasma and 0.00 to 0.37% in red blood cell membranes (RBCM), respectively. The proportion of cis9,trans11-18:2 was increased from 0.05 to 0.23% in plasma and 0.03 to 0.17% in RBCM after 28 days of intervention, respectively. Our results suggest that PA can be effectively incorporated into human plasma and RBCM, and is also associated with the increasing proportion of cis9,trans11-18:2 in humans, presumably as a result of metabolism by a saturation reaction. Edible TK seeds could be a potential dietary source of conjugated linoleic acids.
