Hyperglycemia is a defining feature of Type 1 and 2 diabetes. Hyperglycemia also causes insulin resistance, and our group (Kraegen EW, Saha AK, Preston E, Wilks D, Hoy AJ, Cooney GJ, Ruderman NB. Am J Physiol Endocrinol Metab Endocrinol Metab 290: E471–E479, 2006) has recently demonstrated that hyperglycemia generated by glucose infusion results in insulin resistance after 5 h but not after 3 h. The aim of this study was to investigate possible mechanism(s) by which glucose infusion causes insulin resistance in skeletal muscle and in particular to examine whether this was associated with changes in insulin signaling. Hyperglycemia (∼10 mM) was produced in cannulated male Wistar rats for up to 5 h. The glucose infusion rate required to maintain this hyperglycemia progressively lessened over 5 h (by 25%, P < 0.0001 at 5 h) without any alteration in plasma insulin levels consistent with the development of insulin resistance. Muscle glucose uptake in vivo (44%; P < 0.05) and glycogen synthesis rate (52%; P < 0.001) were reduced after 5 h compared with after 3 h of infusion. Despite these changes, there was no decrease in the phosphorylation state of multiple insulin signaling intermediates [insulin receptor, Akt, AS160 (Akt substrate of 160 kDa), glycogen synthase kinase-3β] over the same time course. In isolated soleus strips taken from control or 1- or 5-h glucose-infused animals, insulin-stimulated 2-deoxyglucose transport was similar, but glycogen synthesis was significantly reduced in the 5-h muscle sample (68% vs. 1-h sample; P < 0.001). These results suggest that the reduced muscle glucose uptake in rats after 5 h of acute hyperglycemia is due more to the metabolic effects of excess glycogen storage than to a defect in insulin signaling or glucose transport.

A key regulatory point in the control of fatty acid (FA) oxidation is thought to be transport of FAs across the mitochondrial membrane by carnitine palmitoyltransferase I (CPT I). To investigate the role of CPT I in FA metabolism, we used in vivo electrotransfer (IVE) to locally overexpress CPT I in muscle of rodents. A vector expressing the human muscle isoform of CPT I was electrotransferred into the right lateral muscles of the distal hindlimb [tibialis cranialis (TC) and extensor digitorum longus (EDL)] of rats, and a control vector expressing GFP was electrotransferred into the left muscles. Initial studies showed that CPT I protein expression peaked 7 days after IVE (+104%, P < 0.01). This was associated with an increase in maximal CPT I activity (+30%, P < 0.001) and a similar increase in palmitoyl-CoA oxidation (+24%; P < 0.001) in isolated mitochondria from the TC. Importantly, oxidation of the medium-chain FA octanoyl-CoA and CPT I sensitivity to inhibition by malonyl-CoA were not altered by CPT I overexpression. FA oxidation in isolated EDL muscle strips was increased with CPT I overexpression (+28%, P < 0.01), whereas FA incorporation into the muscle triacylglycerol (TAG) pool was reduced (−17%, P < 0.01). As a result, intramyocellular TAG content was decreased with CPT I overexpression in both the TC (−25%, P < 0.05) and the EDL (−45%, P < 0.05). These studies demonstrate that acute overexpression of CPT I in muscle leads to a repartitioning of FAs away from esterification and toward oxidation and highlight the importance of CPT I in regulating muscle FA metabolism.

A novel hierarchical MnO2/carbon strip (MnO2/C) microsphere is synthesized via galvanostatic charge-discharge of a MnO@C matrix precursor where the carbon is from a low-cost citric acid. This hierarchical structure is composed of manganese oxides nanoflakes and inlaid carbon strips. The ultrathin nanoflakes assemble to form porous microspheres with a rippled surface superstructure. Due to its improved conductivity and remarkable increased phase contact area, this novel structure exhibits an excellent electrochemical performance with a specific capacitance of 485.6 F g -1 at a current density of 0.5 A g-1 and an area capacitance as high as 4.23 F cm-2 at a mass loading of 8.7 mg cm-2. It also shows an excellent cycling stability with 88.9% capacity retention after 1000 cycles. It is speculated that the present low-cost novel hierarchical porous microspheres can serve as a promising electrode material for pseudocapacitors. © 2014 American Chemical Society.

Herein, we demonstrate that a flexible, air-permeable, thermoelectric (TE) power generator can be prepared by applying a TE polymer (e.g. poly(3,4-ethylenedioxythiophene):poly(4-styrenesulfonate)) coated commercial fabric and subsequently by linking the coated strips with a conductive connection (e.g. using fine metal wires). The poly(3,4-ethylenedioxythiophene):poly(4-styrenesulfonate) coated fabric shows very stable TE properties from 300 K to 390 K. The fabric device can generate a TE voltage output (V) of 4.3 mV at a temperature difference (ΔT) of 75.2 K. The potential for using fabric TE devices to harvest body temperature energy has been discussed. Fabric-based TE devices may be useful for the development of new power generating clothing and self-powered wearable electronics.

Roll forming is increasingly used in the automotive industry to form High Strength Steel (HSS) and Advanced High Strength Steel (AHSS) for structural components. Because of the large variety of applications of roll forming in the industry, Finite Element Analysis (FEA) is increasingly utilized for roll forming process design. Bending is the dominant deformation mode in roll forming and sheet materials used in the process are often temper rolled (skin passed), roller- or tension-levelled. These processes introduce residual stresses into the material, and recent studies have shown that those affect the material behaviour in bending. A thickness reduction rolling process available at Deakin that leads to material deformation similar to an industrial temper rolling operation was used in this study to introduce residual stresses into a dual phase, DP780, steel strip. The initial and thickness reduced strips were then used in a 5-stand experimental V-section roll forming set-up to identify the effect of residual stress on the final shape. The influence of residual stress and the effect of plastic deformation on the material behaviour in roll forming are separately determined in numerical simulation. The results show that the thickness reduction rolling process decreases the maximum bow height while the springback angle and end flare increase. Comparison with experimental results shows that using material data from the conventional tensile test in a numerical simulation does not allow for the accurate prediction of shape defects in a roll forming process if a residual stress profile exists in the material. On the other hand including the residual stress information leads to improved model accuracy.

Australia’s inner cities experienced an upheaval in the 1960s and 70s which left them changed forever. People from all walks of life who valued their suburbs – places like Balmain, Battery Point, Carlton, Indooropilly, North Adelaide or Subiaco – resisted large-scale development projects for freeways, ‘slum clearance’ and mass-produced high-rise. Unlikely alliances of post-war migrants, university students and staff, construction workers and their unions, long-term residents and city workers,challenged land-grabs and inappropriate development.When the dust settled, Australian cities were different. Many suburbs kept their village qualities. Shopping strips were revived and cultures celebrated. While areas like Fitzroy or Redcliff were derided as ‘Trendyville’, the fate many American cities suffered – a ‘hollow core’ – had been avoided. In the process, heritage conservation, party politics, and Australian assumptions about domestic life, education and lifestyle had all been transformed. This book is an in-depth examination of the causes and consequences of urban protest in a democracy. It shows how it changed the built environment as well as its participants, and resonated in many of our institutions including politics, media and multiculturalism.

Habitat restoration has become an important part of biodiversity conservation in the face of extensive habitat loss and fragmentation, especially in agricultural landscapes. Study of invertebrates such as beetles (Coleoptera) may be important to assess the effectiveness of restoration techniques in maintaining native fauna, because they provide a variety of trophic roles and ecosystem services. In this study we examined the conservation value for beetles of revegetation in linear strips and alongside remnant patches compared with remnant vegetation and cleared roadsides. We also assessed how habitat variables structured beetle community composition. Beetle species richness and abundance did not substantially differ between revegetated, remnant and cleared areas, and was not substantially influenced by vegetation type and structure. Herbivorous beetles and the family Curculionidae were more species rich in cleared linear strips. Beetle fauna in these agricultural landscapes may be a robust subset of the pre-clearing beetle community, possibly due to the widespread degradation of remnant areas and the ground layer habitats within them. One beetle species had slightly higher abundance in remnant linear strips, suggesting that remnant habitats may be important for some beetle species. Importantly, environmental variables strongly influenced beetle community composition, signifying that beetle communities are still responding to factors such as soil type and native vegetation, rather than variables mainly associated with land management. The restoration practices currently being undertaken in agricultural areas may not maintain beetle species that require specific habitat variables to survive. Ground-layer attributes need to be included in future revegetation projects, and translocation of specialist species of beetles may be required to restore communities.

Using a model of birth asphyxia, we previously reported significant structural and functional deficits in the diaphragm muscle in spiny mice, deficits that are prevented by supplementing the maternal diet with 5% creatine from mid-pregnancy. The long-term effects of this exposure are unknown. Pregnant spiny mice were fed control or 5% creatine-supplemented diet for the second half of pregnancy, and fetuses were delivered by caesarean section with or without 7.5 min of in-utero asphyxia. Surviving pups were raised by a cross-foster dam until 33±2 days of age when they were euthanized to obtain the diaphragm muscle for ex-vivo study of twitch tension and muscle fatigue, and for structural and enzymatic analyses. Functional analysis of the diaphragm revealed no differences in single twitch contractile parameters between any groups. However, muscle fatigue, induced by stimulation of diaphragm strips with a train of pulses (330 ms train/sec, 40 Hz) for 300 sec, was significantly greater for asphyxia pups compared with controls (p<0.05), and this did not occur in diaphragms of creatine + asphyxia pups. Birth asphyxia resulted in a significant increase in the proportion of glycolytic, fast-twitch fibres and a reduction in oxidative capacity of Type I and IIb fibres in male offspring, as well as reduced cross-sectional area of all muscle fibre types (Type I, IIa, IIb/d) in both males and females at 33 days of age. None of these changes were observed in creatine + asphyxia animals. Thus, the changes in diaphragm fatigue and structure induced by birth asphyxia persist long-term but are prevented by maternal creatine supplementation.

In agricultural regions worldwide, linear networks of vegetation such as hedges, fencerows and live fences provide habitat for plant and animal species in heavily modified landscapes. In Australia, networks of remnant native vegetation along roadsides are a distinctive feature of many rural landscapes. Here, we investigated the richness and composition of woodland-dependent bird communities in networks of eucalypt woodland vegetation along roadsides, in an agricultural region in which >80% of native woodland and forest vegetation has been cleared. We stratified sites in a) cross sections and b) linear strips of roadside vegetation, to test the influence on woodland birds of site location and configuration in the linear network (the 'intersection effect'). We also examined the influence of tree size at the site, the amount of wooded vegetation surrounding the site, and the abundance of an aggressive native species, the noisy miner Manorina melanocephala. Birds were surveyed at 26 pairs of sites (cross section or linear strip) on four occasions. A total of 66 species was recorded, including 35 woodland species. The richness of woodland bird species was influenced by site configuration, with more species present at cross sections, particularly those with larger trees (>30 cm diameter). However, the strongest influence on species richness was the relative abundance of the noisy miner. The richness of woodland birds at sites where noisy miners were abundant was ~20% of that where miners were absent. These results recognise the value of networks of roadside vegetation as habitat for woodland birds in depleted agricultural landscapes; but highlight that this value is not realised for much of this vast vegetation network because of the dominance of the noisy miner. Nevertheless, roadside vegetation is particularly important where the configuration of networks create nodes that facilitate movement. Globally, the protection, conservation and restoration of such linear networks has an important influence on the persistence of biota within human-dominated landscapes.

My abstract moving image making has provided a foundation for my practice since I first started processing and solarizing my own 16mm film in one of those LOMO Russian processing tanks in 1973. Feyers, Zoomfilm (1976) and Running (1976) rework some of those early strips of black and white film. Whenever funding dried up I always fell back on my abstract direct on film work. It was cheap. Like knitting, it gave me a space to process the dilemmas and incongruities of daily life and to escape its clutches. I also began to understand that these forces were still there, embedded implicitly in the work. Now, more than ever, I understand this as a survival response to corporate doublespeak. I would never throw anything away. New scratching, painting, taping or bleaching strategies could be added later. Intensive cluster editing of single frames became an obsession. The translated difference between what you saw over a light-box and what was projected drew me in. Like the migrant position I was allocated from childhood I survived in the space between these two territories. As well as an archive of images and movement I collect optical effects. The flash frame. The trail of afterimages resulting from flickering between positive and negative images. At their liveliest these images float above the screen. Now the digital allows me to amplify the material presence of 16m and 35mm film and a whole new world opens up before me. I cobble together found footage films from my own archive of discarded data and unfinished sentences.

Tackling the global threat of habitat fragmentation on biodiversity requires knowledge of how species move within agricultural landscapes. However, the specific mechanisms influencing dispersal within such landscapes remain poorly understood. The objective of our study was to assess how matrix type (improved pasture, native pasture or crop) and structure (grass height) influence fine-scale reptile movement, as well as influences of crop sowing direction and setting-sun position. In an agricultural region of south-eastern Australia, we first released 20 individuals of an arboreal gecko (Christinus marmoratus) at set distances from trees to determine the distance at which they could perceive their tree habitat (perceptual range). We then translocated 36 individuals into six matrix environments within their perceptual range of isolated trees to examine how gecko movement was modified by the type and structure of the matrix. We also recorded crop sowing direction and setting-sun position and examined all recorded tracks using angular statistics. We found that geckos exhibited a perceptual range of 40–80m. Short matrix environments promoted direct movements towards trees, irrespective of matrix type. Furthermore, movements were significantly affected by crop sowing direction with individuals following the planted lines. Our study has three significant implications: (i) restoring mature tree spacing to 80 m apart will assist gecko movements, (ii) targeted management for low pasture height, such as by maintaining directional narrow strips of low vegetation among taller pastures, might assist movement and facilitate increased connectivity, (iii) directional sowing of crops between habitat patches presents a simple but potentially effective tool for reconnecting fragmented landscapes.
