A Fe-2.8%Si single crystal was scratched in order to randomise the texture in the neighbourhood of the notch. Annealing resulted in recrystallization and grain growth starting from the deformed zone. Misorientations between the single crystal matrix and the grown grains were gathered and were studied in order to investigate the possibility for selective growth based on a specific misorientation. However, instead of studying the misorientation angle or axis profiles separately in a 1D or 2D projection a full misorientation analysis was carried out in the 3-dimensional Rodrigues-Frank misorientation space, which offers an unambiguous interpretation of the data because no features are hidden or masked by a projection. It is concluded that the selective growth phenomenon following the <110>26.5deg misorientation relationship is strongly supported by the gathered orientation data, after appropriately normalizing these data with respect to a random misorientation distribution.

Context: The mitochondrial uncoupling protein-3 (UCP3) has been implicated in the protection of the mitochondrial matrix against lipid-induced mitochondrial damage. Recent evidence points toward mitochondrial aberrations as a major contributor to the development of insulin resistance and diabetes, and UCP3 is reduced in diabetes.
Objective: We compared skeletal muscle UCP3 protein levels in prediabetic subjects [i.e. impaired glucose tolerance (IGT)], diabetic patients, and healthy controls and examined whether rosiglitazone treatment was able to restore UCP3.
Patients, Design, Intervention: Ten middle-aged obese men with type 2 diabetes mellitus [age, 61.4 ± 3.1 yr; body mass index (BMI), 29.8 ± 2.9 kg/m2], nine IGT subjects (age, 59.0 ± 6.6 yr; BMI, 29.7 ± 3.0 kg/m2), and 10 age- and BMI-matched healthy controls (age, 57.3 ± 7.4 yr; BMI, 30.1 ± 3.9 kg/m2) participated in this study. After baseline comparisons, diabetic patients received rosiglitazone (2 x 4 mg/d) for 8 wk.
Main Outcome Measures: Muscle biopsies were sampled to determine UCP3 and mitochondrial protein (complex I–V) content.
Results: UCP3 protein content was significantly lower in prediabetic IGT subjects and in diabetic patients compared with healthy controls (39.0 ± 28.5, 47.2 ± 24.7, and 72.0 ± 23.7 arbitrary units, respectively; P < 0.05), whereas the levels of the mitochondrial protein complex I–V were similar between groups. Rosiglitazone treatment for 8 wk significantly increased insulin sensitivity and muscle UCP3 content (from 53.2 ± 29.9 to 66.3 ± 30.9 arbitrary units; P < 0.05).
Conclusion: We show that UCP3 protein content is reduced in prediabetic subjects and type 2 diabetic patients. Eight weeks of rosiglitazone treatment restores skeletal muscle UCP3 protein in diabetic patients.

Muscle invasive transitional cell carcinoma (TCC) of the bladder is associated with a high frequency of metastasis, resulting in poor prognosis for patients presenting with this disease. Models that capture and demonstrate step-wise enhancement of elements of the human metastatic cascade on a similar genetic background are useful research tools. We have utilized the transitional cell carcinoma cell line TSU-Pr1 to develop an in vivo experimental model of bladder TCC metastasis. TSU-Pr1 cells were inoculated into the left cardiac ventricle of SCID mice and the development of bone metastases was monitored using high resolution X-ray. Tumor tissue from a single bone lesion was excised and cultured in vitro to generate the TSU-Pr1-B1 subline. This cycle was repeated with the TSU-Pr1-B1 cells to generate the successive subline TSU-Pr1-B2. DNA profiling and karyotype analysis confirmed the genetic relationship of these three cell lines. In vitro, the growth rate of these cell lines was not significantly different. However, following intracardiac inoculation TSU-Pr1, TSU-Pr1-B1 and TSU-Pr1-B2 exhibited increasing metastatic potential with a concomitant decrease in time to the onset of radiologically detectable metastatic bone lesions. Significant elevations in the levels of mRNA expression of the matrix metalloproteases (MMPs) membrane type 1-MMP (MT1-MMP), MT2-MMP and MMP-9, and their inhibitor, tissue inhibitor of metalloprotease-2 (TIMP-2), across the progressively metastatic cell lines, were detected by quantitative PCR. Given the role of MT1-MMP and TIMP-2 in MMP-2 activation, and the upregulation of MMP-9, these data suggest an important role for matrix remodeling, particularly basement membrane, in this progression. The TSU-Pr1-B1/B2 model holds promise for further identification of important molecules.

The research was a detailed investigation into a challenging analytical chemistry problem for the alumina industry. The successful outcomes were derived through innovative reagent chemistry and novel instrumental development. The resultant methodology and instrumentation deployed on this most demanding sample matrix is more robust, reliable and less expensive than anything currently used in this industry worldwide.

The influence of the mixing parameters on the synthesis of Al–SiCp reinforced metal matrix composites (MMCs) by the stir casting technique is investigated through a water model. The effects of some important mixing parameters such as impeller blade angle, rotating speed, direction of impeller rotation and effect of baffles are investigated and optimized. The results have shown that the axial concentration variation of natural graphite during stirring in the presence of four vertical baffles is 1.0 wt% against in the absence of baffles it is increased to 2.3 wt%. The variations observed in natural graphite concentration in water during mixing are in close agreement with the earlier modeling and limited experimental studies reported on the real molten aluminum–SiC system. Semi-empirical correlations arrived at between the dimensionless numbers for stirred water – natural graphite slurries are Po = Re−0.0545 Fr−1.099 and Po = Re−0.0219 Fr−1.0382 for clockwise and counter clockwise rotation respectively.

The formation of Al2O3, MgAl2O4, and MgO has been widely studied in different Al base metal matrix composites, but the studies on thermodynamic aspects of the Al2O3/ MgAl2O4/MgO phase equilibria have been limited to few systems such as Al/Al2O3 and Al/SiC. The present study analyzes the Al2O3/MgAl2O4 and MgAl2O4/MgO equilibria with respect to the temperature and the Mg content in Al/SiO2 system using an extended Miedema model. There is a linear and parabolic variation in Mg with respect to the temperature for MgAl2O4/MgO and Al2O3/MgAl2O4 equilibria, respectively, and the influence of Si and Cu in the two equilibria is not appreciable. The experimental verification has been limited to MgAl2O4/MgO equilibria due to the high Mg content (≥0.5 wt pct) required for composite processing. The study has been carried out on two varieties of Al/SiO2 composites, i.e., Al/Silica gel and Al/Micro silica processed by liquid metallurgy route (stir casting route). MgO is found to be more stable compared to MgAl2O4 at Mg levels ≥5 and 1 wt pct in Al/Silica gel and Al/Micro silica composites, respectively, at 1073 K. MgO is also found to be more stable at lower Mg content (3 wt pct) in Al/Silica gel composite with decreasing particle size of silica gel from 180 micron to submicron and nanolevels. The MgO to MgAl2O4 transformation has taken place through a series of transition phases influenced by the different thermodynamic and kinetic parameters such as holding temperature, Mg concentration in the alloy, holding time, and silica particle size.

When using linguistic approaches to solve decision problems, we need the techniques for computing with words (CW). Together with the 2-tuple fuzzy linguistic representation models (i.e., the Herrera and Mart´ınez model and the Wang and Hao model), some computational techniques for CW are also developed. In this paper, we define the concept of numerical scale and extend the 2-tuple fuzzy linguistic representation models under the numerical scale.We find that the key of computational techniques
based on linguistic 2-tuples is to set suitable numerical scale with
the purpose of making transformations between linguistic 2-tuples
and numerical values. By defining the concept of the transitive
calibration matrix and its consistent index, this paper develops an optimization model to compute the numerical scale of the linguistic term set. The desired properties of the optimization model are also presented. Furthermore, we discuss how to construct the transitive calibration matrix for decision problems using linguistic preference relations and analyze the linkage between the consistent index of the transitive calibration matrix and one of the linguistic preference relations. The results in this paper are pretty helpful to complete the fuzzy 2-tuple representation models for CW.

Carbon black (CB) fillers were used to study the feasibility of achieving multiple percolation using an immiscible (polar) polymer blend matrix. By tailoring the morphology of the insulating dual phase matrix it has been shown that the percolation threshold (Фc) can be reduced over single-phase matrices. Cocontinuity in the polymer matrix is important in reducing Фc by either preferentially isolating the conducting filler at the interface of the two phases or within one particular continuous phase of the matrix thereby forming a continuous conducting network within a continuous network (multiple percolation). Actual melt processing time has been found to influence the dispersion of the fillers and hence Фc. Polarity of the matrix as well as the processing method has also been found to influence the dispersion of the filler within the host polymer.

Allosteric regulation is a fundamental mechanism of biological control. Here, we investigated the allosteric mechanism by which GTP inhibits cross-linking activity of transglutaminase 2 (TG2), a multifunctional protein, with postulated roles in receptor signaling, extracellular matrix assembly, and apoptosis. Our findings indicate that at least two components are involved in functionally coupling the allosteric site and active center of TG2, namely (i) GTP binding to mask a conformationally destabilizing switch residue, Arg-579, and to facilitate interdomain interactions that promote adoption of a compact, catalytically inactive conformation and (ii) stabilization of the inactive conformation by an uncommon H bond between a cysteine (Cys-277, an active center residue) and a tyrosine (Tyr-516, a residue located on a loop of the p-barrel 1 domain that harborst he GTP-bindings ite). Although not essential for GTP-mediated inhibition of cross-linking, this H bond enhances the rate of formation of the inactive conformer.

A MoO3-carbon nanocomposite was synthesized from a mixture of MoO3 and graphite by a controlled ball milling procedure. The as-prepared product consists of nanosized MoO3 particles (2-180 nm) homogeneously distributed in carbon matrix. The nanocomposite acts as a high capacity anode material for lithium-ion batteries and exhibits good cyclic behavior. Its initial capacity exceeds the theoretical capacity of 745 mA h g-1 in a mixture of MoO3 and graphite (1:1 by weight), and the stable capacity of 700 mA h g-1 (94% of the theoretical capacity) is still retained after 120 cycles. The electrode performance is linked with the unique nanoarchitecture of the composite and is compared with the performance of MoO3-based anode materials reported in the literature previously (nanoparticles, ball milled powders, and carbon-coated nanobelts). The high value of capacity and good cyclic stability of MoO3-carbon nanocomposite are attractive in respect to those of the reported MoO3 electrodes.

In this manuscript, we investigate the importance that must be placed on the selection of standard compounds when undertaking studies to optimize the performance of 2-D-HPLC separations. A geometric approach to factor analysis and a measure of peak density across the separation space were applied to assess localized measures of component distributions within the 2-D separation plane. The results of this analysis of data showed that the measure of separation quality varied markedly, depending on the elution zone for which the test was undertaken. The study concluded that if standards cannot be obtained that adequately describe the entire sample matrix, the sample itself should be used, and also, the separation should be optimized for regions of interest, not necessarily the separation as a whole.

Nonnegative matrix factorization (NMF) is widely used in signal separation and image compression. Motivated by its successful applications, we propose a new cryptosystem based on NMF, where the nonlinear mixing (NLM) model with a strong noise is introduced for encryption and NMF is used for decryption. The security of the cryptosystem relies on following two facts: 1) the constructed multivariable nonlinear function is not invertible; 2) the process of NMF is unilateral, if the inverse matrix of the constructed linear mixing matrix is not nonnegative. Comparing with Lin's method (2006) that is a theoretical scheme using one-time padding in the cryptosystem, our cipher can be used repeatedly for the practical request, i.e., multitme padding is used in our cryptosystem. Also, there is no restriction on statistical characteristics of the ciphers and the plaintexts. Thus, more signals can be processed (successfully encrypted and decrypted), no matter they are correlative, sparse, or Gaussian. Furthermore, instead of the number of zero-crossing-based method that is often unstable in encryption and decryption, an improved method based on the kurtosis of the signals is introduced to solve permutation ambiguities in waveform reconstruction. Simulations are given to illustrate security and availability of our cryptosystem.

Skeletal muscle development and regeneration requires the fusion of myoblasts into multinucleated myotubes. Because the enzymatic proteolysis of a hyaluronan and versican-rich matrix by ADAMTS versicanases is required for developmental morphogenesis, we hypothesized that the clearance of versican may facilitate the fusion of myoblasts during myogenesis. Here, we used transgenic mice and an in vitro model of myoblast fusion, C2C12 cells, to determine a potential role for ADAMTS versicanases. Versican processing was observed during in vivo myogenesis at the time when myoblasts were fusing to form multinucleated myotubes. Relevant ADAMTS genes, chief among them Adamts5 and Adamts15, were expressed both in developing embryonic muscle and differentiating C2C12 cells. Reducing the levels of Adamts5 mRNA in vitro impaired myoblast fusion, which could be rescued with catalytically active but not the inactive forms of ADAMTS5 or ADAMTS15. The addition of inactive ADAMTS5, ADAMTS15, or full-length V1 versican effectively impaired myoblast fusion. Finally, the expansion of a hyaluronan and versican-rich matrix was observed upon reducing the levels of Adamts5 mRNA in myoblasts. These data indicate that these ADAMTS proteinases contribute to the formation of multinucleated myotubes such as is necessary for both skeletal muscle development and during regeneration, by remodeling a versican-rich pericellular matrix of myoblasts. Our study identifies a possible pathway to target for the improvement of myogenesis in a plethora of diseases including cancer cachexia, sarcopenia, and muscular dystrophy.

Due to the nature of wireless transmission, communication in wireless mesh networks (WMNs) is vulnerable to many adversarial activities including eavesdropping. Pairwise key establishment is one of the fundamental issues in securing WMNs. This paper presents a new matrix-based pairwise key establishment scheme for mesh clients. Our design is motivated by the fact that in WMNs, mesh routers are more powerful than mesh clients, both in computation and communication. By exploiting this heterogeneity, expensive operations can be delegated to mesh routers, which help alleviate the overhead of mesh clients during key establishment. The new scheme possesses two desirable features: (1) Neighbor mesh clients can directly establish pairwise keys; and (2) Communication and storage costs at mesh clients are significantly reduced.

Peptide toxins found in a wide array of venoms block K+ channels, causing profound physiological and pathological effects. Here we describe the first functional K+ channel-blocking toxin domain in a mammalian protein. MMP23 (matrix metalloprotease 23) contains a domain (MMP23TxD) that is evolutionarily related to peptide toxins from sea anemones. MMP23TxD shows close structural similarity to the sea anemone toxins BgK and ShK. Moreover, this domain blocks K+ channels in the nanomolar to low micromolar range (Kv1.6 > Kv1.3 > Kv1.1 = Kv3.2 > Kv1.4, in decreasing order of potency) while sparing other K+ channels (Kv1.2, Kv1.5, Kv1.7, and KCa3.1). Full-length MMP23 suppresses K+ channels by co-localizing with and trapping MMP23TxD-sensitive channels in the ER. Our results provide clues to the structure and function of the vast family of proteins that contain domains related to sea anemone toxins. Evolutionary pressure to maintain a channel-modulatory function may contribute to the conservation of this domain throughout the plant and animal kingdoms.
