The presence of nitric oxide synthase (NOS) and role of nitric oxide (NO) in vascular regulation was investigated in the Australian lungfish, Neoceratodus forsteri. No evidence was found for NOS in the endothelium of large and small blood vessels following processing for NADPH-diaphorase histochemistry. However, both NADPH-diaphorase histochemistry and neural NOS immunohistochemistry demonstrated a sparse network of nitrergic nerves in the dorsal aorta, hepatic artery, and branchial arteries, but there were no nitrergic nerves in small blood vessels in tissues. In contrast, nitrergic nerves were found in non-vascular tissues of the lung, gut and kidney. Dual-wire myography was used to determine if NO signalling occurred in the branchial artery of N. forsteri. Both SNP and SIN-1 had no effect on the pre-constricted branchial artery, but the particulate guanylyl cyclase (GC) activator, C-type natriuretic peptide, always caused vasodilation. Nicotine mediated a dilation that was not inhibited by the soluble GC inhibitor, ODQ, or the NOS inhibitor, L-NNA, but was blocked by the cyclooxygenase inhibitor, indomethacin. These data suggest that NO control of the branchial artery is lacking, but that prostaglandins could be endothelial relaxing factors in the vasculature of lungfish.

Objective. To examine associations between family physical activity and sedentary environment and changes in body mass index (BMI) z-scores among 10-12-year-old children over three years.
Method. Design. Longitudinal (three-year follow-up). Subjects. In total, 152 boys and 192 girls aged 10-12 years at baseline.
Measurements. Measured height and weight at baseline and follow-up (weight status, BMI z-scores); aspects of the family physical activity and sedentary environment (parental and sibling modelling, reinforcement, social support, family-related barriers, rules/restrictions, home physical environment) measured with a questionnaire completed by parents at baseline.
Results. At baseline, 29.6% of boys and 21.9% of girls were overweight or obese, and mean (standard deviation, SD) BMI z-scores were 0.44 (0.99) and 0.28 (0.89), respectively. There was a significant change in BMI z-score among girls (mean change=0.19, SD=0.55, p<0.001), but not boys. Among boys, the number of items at home able to be used for sedentary behaviour (B=0.11, p=0.037) was associated with relatively greater increases in BMI z-score. Among girls, sibling engagement in physical activity at least three times/wk (B=-0.17, p=0.010) and the number of physical activity equipment items at home (B=-0.05, p=0.018) were associated with relatively greater decreases in BMI z-score.
Conclusion. Sibling physical activity and environmental stimuli for sedentary behaviours and physical activity within the home may be important targets for prevention of weight gain during the transition from childhood to adolescence.

Introduction: The purpose of the study was to compare the exercise-induced changes in bone mass and geometry between boys and girls.

Methods: Eighty competitive tennis players (43 boys, 37 girls) aged 7–19 years participated. Pubertal status was self-assessed using Tanner stages (TS 1–5). The dominant and nondominant humeri were compared for DXA-derived bone mass (BMC) and MRI-derived bone geometry [total bone area (TA), medullary area (MA) and cortical bone area (CA)].

Results/Discussion: Exercise-induced side-to-side differences in BMC, TA and CA were significant from TS1 to 5 in boys and girls (p < 0.06). Pre-pubertal (TS1) girls and boys show similar side-to-side difference in BMC after adjustment for training volume (19% vs. 15%, ns). Similar findings were found forTA and CA. In contrast, during puberty (TS2-4) boys displayed greater side-to-side differences than girls for BMC (27% vs. 18%, p < 0.05), TA (13–15% vs. 8%, p < 0.05) and CA (32% vs. 20%, p < 0.01), even after adjustment for tennis history. Girls partly compensated for the lack of an exercise-induced increase in bone size by a reduction of the medullary cavity on the dominant side (−5.5 to −13%, p < 0.05). In post-puberty (TS 5 or postmenarche), the size of the medullary cavity remained smaller on the dominant side in girls (−5% to −9%, p = 0.1–0.05??) whereas no such reduction was observed in boys.

Conclusion: Regular exercise initiated before puberty and maintained throughout puberty leads to greater skeletal benefits in peri-pubertal boys than girls for bone mass and bone size, two of the major determinants of bone strength.

This study examined the level of long chain omega-3 and omega-6 polyunsaturated fats, the ratio of polyunsaturated fat to saturated fat (PUFA/SFA) and the ratio of omega-6 to omega-3 (n-6/n-3) fat in sheep grown under grazing conditions in Australia. The sheep genotypes used were Poll Dorsetgrowth × Border Leicester Merino (PDg × BLM), Poll Dorsetgrowth × Merino (PDg × M), Poll Dorsetmuscling × Merino (PDm × M), Border Leicester × Merino (BL × M) and Merino × Merino (M × M). Loin muscles (Longissimus lumborum) collected from 40 ewe and wether sheep slaughtered at 14 months of age were processed for fatty acid determination. After frozen storage, 20 g samples were minced and a 7 g homogenate was processed for muscle lipid extraction using a chloroform:methanol (2:1) procedure. There was an increase in PUFA/SFA as the proportion of Merino genetics increased in the progeny (second-cross < first-cross < Merino), but this was not shown in the n-6/n-3 ratio. The PUFA/SFA trend appeared to be associated with an increase in the level of total polyunsaturated fats, but not a decrease in the level of total saturated fats. The results demonstrate that there is a need to improve the PUFA/SFA content in first- and second-cross animals which are mainly used for meat production in Australia so as to maintain the healthy lipids in meat. Nutritional manipulation through feeding systems or selection of sires for greater heritability of omega-3 fat deposition may be suitable pathways to elevate the ratio of polyunsaturated fatty acids, and in particular omega-3.

Background: This article aims to examine the relative contribution of occupational activity to English adults’ meeting of government recommendations for physical activity (PA).

Methods: Data were extracted from a cross-sectional survey of householders in the UK via the Health Survey for England.1 In total, 14,018 adult participants were included in the analysis. Multivariate logistic regression was used to examine the odds of achieving PA recommendations with and without including occupational activity and to examine the contribution of gender and social and demographic characteristics.

Results: When occupational PA was included, 36% of men and 25% of women were active at the recommended level. Once occupational PA was removed, these proportions were 23% and 19%, respectively. These results were socially patterned, most notably by age and gender.

Conclusions: Occupational PA provides a substantial contribution to those meeting the government target for PA.

Peritoneal dissemination of ovarian carcinoma is mediated by epithelial–mesenchymal interconversions leading to the disruption of cell–cell contact and modulation of cell–extracellular matrix (ECM) interactions. The present study was designed to evaluate the effects of epidermal growth factor (EGF) as a modulator of Janus kinase 2 (JAK2) and signal transducer and activator of transcription 3 (STAT3) signalling and changes in integrin expression during the process similar to EMT. A fibroblastic morphology with reduced intercellular cell contacts and increased cell motility was observed in ovarian cancer cell lines in response to EGF and was concomitant with the up regulation of EMT-associated N-cadherin and vimentin expression. These changes were accompanied by an increase in α2, α6 and β1 integrin subunits and activation of JAK2 and STAT3 signalling which was suppressed by a specific JAK2 inhibitor. Consistent with the suppression of STAT3 activity, N-cadherin and vimentin expression were abrogated and was coherent with the loss of cell motility and the expression of α6 and β1 integrin subunits. Neutralizing antibodies against α6 and β1 subunits inhibited cancer cell migration. A strong correlation between the expression of N-cadherin, vimentin and JAK2/STAT3 levels were detected in high-grade ovarian tumors and was consistent with the previously reported enhanced expression of α6 integrin subunit in advanced tumors [Ahmed N, Riley C, Oliva K, Rice G, Quinn M. Ascites induces modulation of α6β1 integrin and urokinase plasminogen activator receptor expression and associated functions in ovarian carcinoma. British Journal of Cancer 2005;92:1475–85]. Our data incorporating the clinical samples and the cancer cell lines is the first to demonstrate that JAK2/STAT3 pathway may be one of the downstream events in EMT-like process and α6β1 integrin-mediated signalling in ovarian carcinomas.

The performance of a solar-boosted heat pump water heater (HPWH) operating under full load and part load conditions was determined in an outdoor experimental study. The system utilised flat unglazed aluminium solar evaporator panels to absorb solar and ambient energy. Absorbed energy was transferred to the water tank by means of the heat pump and a wrap around condenser coil on the outside of the tank. The system COP was found to be in the range of 5–7 under clear daytime conditions and 3–5 under clear night-time conditions. Using part load testing of the HPWH system it was found that concentrating the coils in the lower portion of the tank could increase the efficiency of the condenser coil. It was also shown that there exists a generalised linear relationship that can be used to describe the system COP in terms of the temperature difference between the water in the storage tank and the ambient air.

Fiji is a small open island economy dependent on energy for its growth and development; hence, the relationship between energy consumption and economic growth is crucial for Fiji's development. In this paper, we investigate the nexus between electricity consumption and economic growth for Fiji within a multivariate framework through including the labour force variable. We use the bounds testing approach to cointegration and find that electricity consumption, GDP and labour force are only cointegrated when GDP is the endogenous variable. We use the Granger causality F-test and find that in the long-run causality runs from electricity consumption and labour force to GDP, implying that Fiji is an energy dependent country and thus energy conservation policies will have an adverse effect on Fiji's economic growth.

This article applies recently developed panel unit root and panel cointegration techniques to estimate the long-run and short-run income and price elasticities for residential demand for electricity in G7 countries. The panel results indicate that in the long-run residential demand for electricity is price elastic and income inelastic. The study concludes that from an environmental perspective there is potential to use pricing policies in the G7 countries to curtail residential electricity demand, and thus curb carbon emissions, in the long run.

Background

Polyethyleneterephthalate (PET) and polytetrafluoroethylene (PTFE) are polymers successfully used as large diameter arterial grafts for peripheral vascular surgery. However, these prosthetic grafts are rarely used for coronary bypass surgery because of their low patency rates. Endothelialisation of the lumenal surface of these materials may improve their patency. This study aimed to compare the endothelialisation of PET, PTFE and pericardium by examining their seeding efficiency over time and the effect of various shear stresses on retention of endothelial cells.

Methods


Ovine endothelial cells at 4 × 105 cells/cm2 were seeded onto PET, PTFE and pericardium, and cultured for 1–168 hours. Cell coverage was determined via en face immunocytochemistry and cell retention was quantified after being subjected to shear stresses ranging from 0.018 to 0.037 N/m2 for 15, 30 and 60 minutes.

Results

Endothelial cells adhered to all of the materials one hour post-seeding. PET exhibited better cell retention rate, ranging from 66.9 ± 5.6% at 0.018 N/m2 for 15 min to 44.7 ± 1.9% at 0.037 N/m2 for 60 minutes, when compared to PTFE and pericardium (p < 0.0001, three-way ANOVA).

Conclusion

PET shows superior retention of endothelial cells during shear stress compare to PTFE and pericardium.

The potential effects of early environmental conditions on adult female mate choice have been largely neglected in studies of sexual selection. Our study tested whether developmental stress affects the mate choice behaviour of female zebra finches, Taeniopygia guttata, when choosing between potential mates. In an experiment manipulating developmental condition, female zebra finches were raised under nutritional stress or control conditions. In adulthood, female preferences were assessed using extensive four-stimulus mate choice trials. Nutritional stress affected growth rates during the period of stress, with experimentally stressed females lighter than controls. During mate choice trials stressed females were almost three times less active than controls and made fewer sampling visits to the stimulus males, although we found no evidence of a direct effect of developmental experience on which males were preferred. Thus, developmental experience had a clear effect on behavioural patterns in a mate choice context. To test whether this effect is specific to a mate choice context, we also investigated the effect of developmental stress on female activity rates in three social contexts: isolation, contact with a conspecific male (a potential mate) and contact with a conspecific female. Here, female activity did not differ between the experimental treatments in any of the social situations. Overall, our findings suggest that environmental conditions during early development can have long-term context-dependent consequences for adult female mate choice behaviour, mediated by changes in activity rates.
