Palaeoclimates across Europe for 6000 y BP were estimated from pollen data using the modern pollen analogue technique constrained with lake-level data. The constraint consists of restricting the set of modern pollen samples considered as analogues of the fossil samples to those locations where the implied change in annual precipitation minus evapotranspiration (P–E) is consistent with the regional change in moisture balance as indicated by lakes. An artificial neural network was used for the spatial interpolation of lake-level changes to the pollen sites, and for mapping palaeoclimate anomalies. The climate variables reconstructed were mean temperature of the coldest month (T c ), growing degree days above 5  °C (GDD), moisture availability expressed as the ratio of actual to equilibrium evapotranspiration (α), and P–E. The constraint improved the spatial coherency of the reconstructed palaeoclimate anomalies, especially for P–E. The reconstructions indicate clear spatial and seasonal patterns of Holocene climate change, which can provide a quantitative benchmark for the evaluation of palaeoclimate model simulations. Winter temperatures (T c ) were 1–3 K greater than present in the far N and NE of Europe, but 2–4 K less than present in the Mediterranean region. Summer warmth (GDD) was greater than present in NW Europe (by 400–800 K day at the highest elevations) and in the Alps, but >400 K day less than present at lower elevations in S Europe. P–E was 50–250 mm less than present in NW Europe and the Alps, but α was 10–15% greater than present in S Europe and P–E was 50–200 mm greater than present in S and E Europe.

Energy use intensity (EUI) and climate have a well documented correlation, which is generally applied in building energy management. Green buildings have sought to greatly reduce energy consumption and a number of examples are documented in the literature. A sample of high performance buildings constructed in a variety of global locations is analyzed here, and provides evidence that measures to reduce energy consumption have reduced EUI to the point where its correlation with heating degree days is no longer apparent. This result suggests that end-user behaviour is the next major hurdle in lowering the energy consumption of greener buildings.

Changes to the behaviour of subseasonal precipitation extremes and active-break cycles of the Indian summer monsoon are assessed in this study using pre-industrial and 2 × CO2 integrations of the Hadley Centre coupled model HadCM3, which is able to simulate the monsoon seasonal cycle reasonably. At 2 × CO2, mean summer rainfall increases slightly, especially over central and northern India. The mean intensity of daily precipitation during the monsoon is found to increase, consistent with fewer wet days, and there are increases to heavy rain events beyond changes in the mean alone. The chance of reaching particular thresholds of heavy rainfall is found to approximately double over northern India, increasing the likelihood of damaging floods on a seasonal basis. The local distribution of such projections is uncertain, however, given the large spread in mean monsoon rainfall change and associated extremes amongst even the most recent coupled climate models. The measured increase of the heaviest precipitation events over India is found to be broadly in line with the degree of atmospheric warming and associated increases in specific humidity, lending a degree of predictability to changes in rainfall extremes. Active-break cycles of the Indian summer monsoon, important particularly due to their effect on agricultural output, are shown to be reasonably represented in HadCM3, in particular with some degree of northward propagation. We note an intensification of both active and break events, particularly when measured against the annual cycle, although there is no suggestion of any change to the duration or likelihood of monsoon breaks. Entomopathogenic nematodes, Steinernema carpocapsae, S. feltiae (Steinernematids) Heterorhabditis indica and H. bacteriophora (Heterorhabditids) were studied to control nymphs of desert locust Schistocerca gregaria. Results of all experiments showed a significant difference in mortality percentage among all isolates. All nematodes were found more effective when exposure time was increased up to 10 days. On the other hand, both Heterorhabditids caused maximum mortality as compared to Steinernematids at 30 degree C. When different moisture levels were tested in the sand arena, a medium level of moisture (1%) caused maximum insect mortality in all isolates. However, highest concentration of each isolate (200 IJs per ml) proved to be most appropriate for maximum insect death. Similarly, both Heterorhabditis nematodes when orally applied to insects killed maximum nymphs as compared to other two Steinernematids. A similar response was observed in infectivity test when maximum percentage of IJs of both isolates of Heterorhabditis successfully penetrated into the body of locust nymphs. This research suggests some useful basic findings in developing biocides with suitable virulent of entomopathogenic nematode for controlling nymphs of desert locust.

Our objective was to determine the effect of feeding rumen-inert fats differing in their degree of saturation on dry matter intake (DMI), milk production, and plasma concentrations of insulin, glucagon-like peptide 1 (7-36) amide (GLP-1), glucose-dependent insulinotropic polypeptide (GIP), and cholecystokinin (CCK) in lactating dairy cows. Four midlactation, primiparous Holstein cows were used in a 4 x 4 Latin square experiment with 2-wk periods. Cows were fed a control mixed ration ad libitum, and treatments were the dietary addition (3.5% of ration dry matter) of 3 rumen-inert fats as sources of mostly saturated fatty acids (SFA), monounsaturated fatty acids (MUFA), or polyunsaturated fatty acids (PUFA). Daily DMI, milk yield, and composition were measured on the last 4 d of each period. Jugular vein blood was collected every 30 min over a 7-h period on d 12 and 14 of each period for analysis of plasma concentrations of hormones, glucose, and nonesterified fatty acids. Feeding fat decreased DMI, and the decrease tended to be greater for MUFA and PUFA compared with SFA. Plasma concentration of GLP-1 increased when fat was fed and was greater for MUFA and PUFA. Feeding fat increased plasma glucose-dependent insulinotropic polypeptide and CCK concentrations and decreased plasma insulin concentration. Plasma CCK concentration was greater for MUFA and PUFA than for SFA and was greater for MUFA than PUFA. Decreases in DMI in cows fed fat were associated with increased plasma concentrations of GLP-1 and CCK and a decreased insulin concentration. The role of these peptides in regulating DMI in cattle fed fat requires further investigation.

Oxidized low-density lipoprotein (oxLDL) exhibits many atherogenic effects, including the promotion of monocyte recruitment to the arterial endothelium and the induction of scavenger receptor expression. However, while atherosclerosis involves chronic inflammation within the arterial intima, it is unclear whether oxLDL alone provides a direct inflammatory stimulus for monocyte-macrophages. Furthermore, oxLDL is not a single, well-defined entity, but has structural and physical properties which vary according to the degree of oxidation. We tested the hypothesis that the biological effects of oxLDL will vary according to its degree of oxidation and that some species of oxLDL will have atherogenic properties, while other species may be responsible for its inflammatory activity. The atherogenic and inflammatory properties of LDL oxidized to predetermined degrees (mild, moderate and extensive oxidation) were investigated in a single system using human monocyte-derived macrophages. Expression of CD36 mRNA was up-regulated by mildly- and moderately-oxLDL, but not highly-oxLDL. The expression of the transcription factor, proliferator-activated receptor-gamma (PPARgamma), which has been proposed to positively regulate the expression of CD36, was increased to the greatest degree by highly-oxLDL. However, the DNA binding activity of PPARgamma was increased only by mildly- and moderately-oxLDL. None of the oxLDL species appeared to be pro-inflammatory towards monocytes, either directly or indirectly through mediators derived from lymphocytes, regardless of the degree of oxidation. The effects on the intestinal microbiota of a short period of marginal over-eating, characteristic of holiday or festival periods, were investigated in a pilot study. Fourteen healthy male subjects consumed a diet rich in animal protein and fat for seven days. During this period, the subjects significantly increased their dietary energy, protein, carbohydrate and fat intakes by 56, 59, 53 and 58%, respectively (all P < 0.05). The mean weight gain of 0.27 kg was less than the expected 1 kg, but this was consistent with a degree of under-reporting on the baseline diet. Fluorescence in situ hybridisation analysis confirmed the relative stability of each individual’s faecal microbiota but showed considerable variations between them. The diet was associated with a significant increase in numbers of total faecal bacteria and the bacteroides group, as detected by the universal bacterial probe (DAPI) and Bacteroides probe (Bac 303), respectively. Overall, there was a decrease in numbers of the Lactobacillus/Enterococcus group (Lab 158 probe; 2.8 ± 3.0% to 1.8 ± 1.8%) and the Bifidobacterium group (Bif 164 probe; 3.0 ± 3.7% to 1.7 ± 1.2%), although there was considerable inter-individual variation. Analysis of the relative proportions of each bacterial group as a percentage of the subject’s total bacteria showed a trend for a change in the intestinal microbiota that might be considered potentially unhealthy.
