A study of the reproductive physiology of P. (P). semisulcatus wasundertaken as this information is an essential prerequisite for brood stock development for hatchery operations, and the results are embodied in this thesis. The thesis is presented in seven chapters. The protein, lipid and carbohydrate contents of ovary were estimated for the different vitellogenic phases. The protein, lipid, carbohydrate and cholesterol contents were estimated in testes and spermatophore. The thesis present the results of induced maturation experiments by eyestalk ablation, CNS extract injection and UV rays application on immature female prawns.

The present investigation has been carried out to understand the process and events leading to maturation of the ovary and testis in the Indian white prawn Penaeus indicus. The study includes the classification of the ovarian maturity stages based on its colour, gonadosomatic index, oocyte diameter and morphological changes in the oocyte. Further the process of oogenesis has been investigated using light and electron microscopic techniques. A histochemical study of the ovary has also been carried out to determine the sequence in which yolk substances are synthesized or sequestered in the oocytes and also to elicit the nature of the penaeid yolk material. The process of spermatogenesis and the development of the spermatophore has been studied in detail using light and electron microscopic methods. In addition a brief histochemical study on the testis was also made to understand the nature of the organic reserves in the sperm cells.

Aquaculture is the dynamic pursuit of production of organisms from water a process analogous to agriculture on land. The field of aquaculture is an emerging bioindustry, based upon the culture and husbandry of economically utilizable aquatic organisms. Of late, there has been a global upsurge for aquaculture, the main reasons for which include the requirement of protein source for the increasing world population, the decision by various world nations to increase the fish yield by developing unutilized or partially utilized water bodies and depletion of natural stock which is evident in recent years due to excessive exploitation .The present study has been taken up on the reproductive physiology of the female grey mullet, M. cephalus. The thesis is presented in seven chapters. In the present study, variations in the major biochemical parameters namely, moisture, proteins, lipids, carbohydrates cholesterol, carotenoid, ash, calcium and iron in four tissues E. muscle, liver, ovary and bloodserum of cephalus have been analysed at different maturity stages.

Rainbow sardines of the genus belonging to the family Dueenaieriidae. are small pelagic fishes forming a fairly good, though not abundant. seasonal fishery all along the coasts of India inhabiting the coastal waters. There have been some earlier reports on such individual aspects as their systematic, distribution, abundance. Ostecology and a few biological factors but no attempt has been made towards a comprehensive study on this group. Two species of rainbow sardines are known to occur in the Indian seas and while a knowledge about their biology would be useful from the fishery point of view. it was also thought a study of their systematic position, especially regarding the identity or the two species which had raised doubts among earlier workers would lead to a better understanding or the group as a whole. This thesis is mainly based on studies during the period from April 1969 to march 1971 with a continued investigation of fishhery aspects till December 1975. from the Gulf of manar: and the Palk Bay around mandapam area. on the south-east coast of India. Thus the work deals with the systemtics, biology and fishery of rainbow sardines of Indian seas.

A growth medium with Leibovitz-15 L-15.as the base, supplemented with foetal bovine serum 10% vrv., fish muscle extract 10% vrv., prawn muscle extract 10% vrv., lectin concanavalin A. 0.02 mg mly1., lipopolysaccharide 0.02 mg mly1., glucose D 0.2 mg mly1., ovary extract 0.5% vrv.and prawn haemolymph 0.5%. has been formulated with 354"10 mOsm for the development and maintenance of a cell culture system from the ovarian tissue of African catfish, Clarias gariepinus. For its subculturing, a cell dissociationrextracting solution, composed of equal portions of trypsin phosphate versene glucose TPVG. containing 0.0125% wrv.trypsin and 25% vrv.non-enzymatic cell dissociation solution 1 and 2, has also been developed with which the cell culture can be passaged 15 times after which they cease to multiply and consequently perish. The cell cultures can be maintained for 12–15 days without fluid change between the passages. This is the first report of a cell culture system from the ovarian tissues of African catfish

To demonstrate pathological changes due to white spot virus infection in Fenneropenaeus indicus, a batch of hatchery bred quarantined animals was experimentally infected with the virus. Organs such as gills, foregut, mid-gut, hindgut, nerve, eye, heart, ovary and integument were examined by light and electron microscopy. Histopathological analyses revealed changes hitherto not reported in F. indicus such as lesions to the internal folding of gut resulted in syncytial mass sloughed off into lumen, thickening of hepatopancreatic connective tissue with vacuolization of tubules and necrosis of rectal pads in hindgut. Virus replication was seen in the crystalline tract region of the compound eye and eosinophilic granules infiltrated from its base. In the gill arch, dilation and disintegration of median blood vessel was observed. In the nervous tissues, encapsulation and subsequent atrophy of hypertrophied nuclei of the neurosecretory cells were found. Transmission electron microscopy showed viral replication and morphogenesis in cells of infected tissue. De novo formed vesicles covered the capsid forming a bilayered envelop opened at one end inside the virogenic stroma. Circular vesicles containing nuclear material was found fused with the envelop. Subsequent thickening of the envelop resulted in the fully formed virus. In this study, a correlation was observed between the stages of viral multiplication and the corresponding pathological changes in the cells during the WSV infection. Accordingly, gill and foregut tissues were found highly infected during the onset of clinical signs itself, and are proposed to be used as the tissues for routine disease diagnosis.

Lack of a valid shrimp cell line has been hampering the progress of research on shrimp viruses. One of the reasons identified was the absence of an appropriate medium which would satisfy the requirements of the cells in vitro. We report the first attempt to formulate an exclusive shrimp cell culture medium (SCCM) based on the haemolymph components of Penaeus monodon prepared in isosmotic seawater having 27 % salinity. The SCCM is composed of 22 amino acids, 4 sugars, 6 vitamins, cholesterol, FBS, phenol red, three antibiotics, potassium dihydrogen phosphate and di-sodium hydrogen phosphate at pH 6.8–7.2. Osmolality was adjusted to 720 ± 10 mOsm kg-1 and temperature of incubation was 25 8C. The most appropriate composition was finally selected based on the extent of attachment of cells and their proliferation by visual observation. Metabolic activity of cultured cells was measured by MTT assay and compared with that in L-15 (29), modified L-15 and Grace’s insect medium, and found better performance in SCCM especially for lymphoid cells with 107 % increase in activity and 85 ± 9 days of longevity. The cells from ovary and lymphoid organs were passaged twice using the newly designed shrimp cell dissociation ‘‘cocktail’’.
