Everywhere, on the coastal belt it is proved without doubt that the pristine ground water quality was severely deteriorated after the 26 December 2004 Indian Ocean Tsunami. But how far is more relevant, as it is decided by the so-called pre-tsunamic situation of the region. In water quality studies it is this reference finger print which earmarks regional ground water chemistry based on which the monthly variability could rationally be interpreted. This Ph D thesis comprises the testing and evaluation of the facts: whether there is any significant difference in the water quality parameters under study between stations and between months in Tsunami Affected Dug Wells (TADW). Whether the selected water quality parameters vary significantly from BIS and WHO standards. Whether the water quality index (WQI) differ significantly between Tsunami Affected Dug Wells (TADW) and Bore Wells (BW). Whether there is any significant difference in the water quality parameters during December 2005 and December 2008. Is there any significant change in the Water Quality Parameters before 2001 and after tsunami (2005) in TADW.

An experiment was designed to assess the occurrence of multiple antibiotic resistances in Vibrio sp from different (brackish and marine) environments. Water samples front nine marine landing sites and two coastal inland aquaculture farms were screened for the Vibrio spp and assessed their resistance to twenty-two different antibiotics, which arc commonly encountered in the aquatic ecosystem. Tissue samples (shrimp, mussel and sepia) were tested from the sampling site with highest antibiotic resistance. Of' the total 119 Vibrio isolates, 16. 8% were susceptible to all antibiotics. Of the resistant (83.19%) Vibrio strains, 30.3% were resistant against three antibiotics, 55.5% were resistant against 4-10 antibiotics, 14.14% were resistant against more than 10 antibiotics and 54% have shown multiple antibiotics resistance (MAR). Antibiotic resistance index was higher in Coastal 3, 6, Aqua farm 2 in isolates from water samples and all the tissues tested. Interestingly, incidence of antibiotic resistance in isolates from water samples was comparatively lower in aquaculture farms than that observed in coastal areas. Highest incidence of antibiotic resistance was evident against Amoxycillin, Ampicillin, Carbencillin and Cefuroxime followed by Rilanipicin and Streptomycin and lowest against Chloramphenicol, Tetracycline, Chlortetracycline, Furazolidone, Nalidixic acid, Gentamycin Sulphafurazole, Trimcthoprinr, Neomycin and Amikacin irrespective of the sampling sites. Results from various tissue samples collected from the sites of highest antibiotic resistance indicated that antibiotic resistance Vibrio spp collected from fish and tissue samples were higher than that of water samples. Overall results indicated that persistent use of antibiotics against diseases in human beings and other life forms may pollute the aquatic system and their impact on developing antibiotic resistant Vibrio sp may be a serious threat in addition to the use of antibiotics in aquaculture farms.

Low-lying coastal areas are more vulnerable to the impacts of climate change as they are highly prone for inundation to SLR (Sea-Level Rise). This study presents an appraisal of the impacts of SLR on the coastal natural resources and its dependent social communities in the low-lying area of VellareColeroon estuarine region of the Tamil Nadu coast, India. Digital Elevation Model (DEM) derived from SRTM 90M (Shuttle Radar Topographic Mission) data, along with GIS (Geographic Information System) techniques are used to identify an area of inundation in the study site. The vulnerability of coastal areas in Vellar-Coleroon estuarine region of Tamil Nadu coast to inundation was calculated based on the projected SLR scenarios of 0.5 m and 1 m. The results demonstrated that about 1570 ha of the LULC (Land use and Land cover) of the study area would be permanently inundated to 0.5 m and 2407 ha for 1 m SLR and has also resulted in the loss of three major coastal natural resources like coastal agriculture, mangroves and aquaculture. It has been identified that six hamlets of the social communities who depend on these resources are at high-risk and vulnerable to 0.5 m SLR and 12 hamlets for 1 m SLR. From the study, it has been emphasized that mainstreaming adaptation options to SLR should be embedded within a coastal zone management and planning effort, which includes all coastal natural resources (ecosystem-based adaptation), and its dependent social communities (community-based adaptation) involved through capacity building

The constructional activities in the coastal belt of our country often demand deep foundations because of the poor engineering properties and the related problems arising from weak soil at shallow depths.The soil profile in coastal area often consists of very loose sandy soils extending to a depth of 3 to 4 m from the ground level underlain by clayey soils of medium consistency.The very low shearing resistance of the foundation bed causes local as well as punching shear failure.Hence structures built on these soils may suffer from excessive settlements.This type of soil profile is very common in coastal areas of Kerala,especially in Cochin. Further,the high water table and limited depth of the top sandy layer in these areas restrict the depth of foundation thereby further reducing the safe bearing capacity.

Shrimp Aquaculture has provided tremendous opportunity for the economic and social upliftment of rural communities in the coastal areas of our country Over a hundred thousand farmers, of whom about 90% belong to the small and marginal category, are engaged in shrimp farming. Penaeus monodon is the most predominant cultured species in India which is mainly exported to highly sophisticated, quality and safety conscious world markets. Food safety has been of concem to humankind since the dawn of history and the concern about food safety resulted in the evolution of a cost effective, food safety assurance method, the Hazard Analysis Critical Control Point (HACCP). Considering the major contribution of cultured Penaeus monodon to the total shrimp production and the economic losses encountered due to disease outbreak and also because traditional methods of quality control and end point inspection cannot guarantee the safety of our cultured seafood products, it is essential that science based preventive approaches like HACCP and Pre requisite Programmes (PRP) be implemented in our shrimp farming operations. PRP is considered as a support system which provides a solid foundation for HACCP. The safety of postlarvae (PL) supplied for brackish water shrimp farming has also become an issue of concern over the past few years. The quality and safety of hatchery produced seeds have been deteriorating and disease outbreaks have become very common in hatcheries. It is in this context that the necessity for following strict quarantine measures with standards and code of practices becomes significant. Though there were a lot of hue and cry on the need for extending the focus of seafood safety assurance from processing and exporting to the pre-harvest and hatchery rearing phases, an experimental move in this direction has been rare or nil. An integrated management system only can assure the effective control of the quality, hygiene and safety related issues. This study therefore aims at designing a safety and quality management system model for implementation in shrimp farming and hatchery operations by linking the concepts of HACCP and PRP.

The metals present in the surface sediments have high demand on a global perspective, and the main reservoir of these elements is believed to be the ocean floor. A lot of studies on metals are going on throughout the world for its quantification and exploitation. Even though, some preliminary attempts have been made in selected areas for the quantitative study of metals in the western continental shelf of India, no comprehensive work has been reported so far. The importance of this study also lies on the fact that there has not been a proper evaluation of the impact of the Great Tsunami of 2004 on the coastal areas of the south India. In View of this, an attempt has been made to address the seasonal distribution, behavior and mechanisms which control the deposition of metals in the sediments of the western continental shelf and Cochin Estuary, an annex to this coastal marine region.Surface sediment samples were collected seasonally from two subenvironemnts of southwest coast of India, (continental shelf of Kerala and Cochin estuarine system), to estimate the seasonal distribution and geochemical behavior of non-transition, transition, rare-earth elements, Th and U. Bottom water samples were also taken from each station, and analysed for temperature, salinity and dissolved oxygen, hence the response of redox sensitive elements to oxygen minimum zone can be addressed. In addition, other sedimentary parameters such as sand, silt, clay fractions, CaCO3 and organic carbon content were also estimated to evaluate the control factors on level of metals present in the sediment. The study used different environmental data analysis techniques to evaluate the distribution and behavior of elements during different seasons. This includes environmental parameters such as elemental normalisation, enrichment factor, element excess, cerium and europium anomalies and authigenic uranium.

The distribution of the holothurian H. KH.) scabra indicated its availability all along coastal areas on Palk Bay from Rameswavam to Mallipattinam and along the Gulf of Hannah coast from Pamban to Ervadi and Tuticmhin,'at 4~2O m depth.The major fishing for holmthurians was done by skin diving at all the centres. The tallu valai was operated at Tuticorin and Vedalai and trawlevs were operated at Rameswaram.The fmod of H. KN.) scabra consists of ovganic matter which contains mud, sand, shell debris, bivalves and algae. Obsehvatinns indicated the species seems to be a n0n—se1ective feeder. The assimilation efficiency from sediment to faeces indicated that the faecal pellets of H. KH.) scabra are semidigested.A multiple relationship was fitted between total length, total weight, gutted weight, gonad weight and maturity stages were found significant.The fishing season for holothurians commences from October to March along Gulf of Manner coast and from March to October along Palk Bay coast.The percentage of catches recorded by skin diving, trawlere and tallu valai were 80.04%, 10.27% and 9.69% respectively. Skin diving contributes to maximum catch.The holothurians landed all along the Gulf of Manner and Palk Bay coasts constitute 25.6% and 74.4% respectively. This showed that Palk Bay coast is more productive.

Plankton community, drawn from a vary wide variety of animal phyla, formed the basic food supply of marine life and indicators of water mass. The term meroplankton generally referred to that portion of the zooplankton which is transient in nature, remaining rest of their lives in the nektonic or benthic environment. This group was selected for intensive studies, considering the role of meroplankton in the economy of the sea and the scarcity of literature on them from the Indian Ocean. The preser .udy besides providing information regarding the fixation and preservation !e _ iniques and biochemical aspects of tropical meroplankton, also consolidates information regarding their zoogeography in the Indian Ocean region, with a view to amplifying the limited information available from this area. The distribution studies are based on the collections made during the International Indian Ocean Expedition (1960-65), whereas the material for preservation and biochemical studies was collected from the coastal waters during 1968-1978. Salient features:- 2% of formaldehyde buffered with 2% borax, added to the plankton in the ratio of 9:1 was found the best fixative. On fixation the plankton underwent shrinkage due to loss of 15 to 87% water. Addition of antioxidants prevented colour fading. Narcotization by different specific reagents prior to fixation reduced distortions due to violent reaction and improved morphological conditions. One percent formaldehyde solution in sea water buffered with borax or neutralised with calcium carbonate perfectly preserved majority of meroplankton. Equally good was one percent propylene phenoxetol buffered with borax. Biochemical compostion of vaioous taxa showed variations according to their age class, size groups metamorphosing stage, feeding mechanism, type of organism fed and time of collection. General distribution studies of 4 meroplankton taxa - Anthozoan larvae, cirripedia larvae, sipunculoid larvae and gastropod larvae stowed abundance in the coastal areas especially during the SW monsoon period. Based on the larval distribution different zoo-geographical areas in the Indian Ocean are differentiated.

At present, there is paucity of information regarding the breeding biology, especially from southeastern region, which differs in its climatic features from northeastern coastal areas, where some attempts have been made to study the biology of the fish, though not documented in detail (Jhingran and Natarajan, I969; Patnaik and Jena, 1976; Kowtal, 1977; Roy e_t _a_l_., I977). The present work, first to undertake detailed investigation into reproductive aspects of the fish from India is not only confined to biology, but extend to the study on histological as well as physiological changes in the processes associated with natural reproductive cycle. The main objective of the study is to develop a background for the standardisation of subsequent artificial propagation techniques for the Sea Dass The thesis is presented in three chapters. The first chapter; general introduction, surveys the literature pertaining to the status of research on Lates calcarifer with specific reference to reproduction and acquaculture. The second chapter entitled, materials and methods gives description regarding collection of specimens as well as samples in addition to the various analytical methodologies employed during the study.

No serious attempt has so far been made in India to make use of the ‘Mussel watch’ concept as a useful tool for pollution monitoring of the marine and estuarine environment. The recently conducted 'National seminar on mussel watch’ by the Cochin University of Science and Technology (13-14 Feb, 1986) discussed the technical aspects related to mussel watch programme and the application of sentinel organism concept to the coastal areas of India. It is well known that the biological and physiological characteristics of the organism inhabiting tropical waters such as those prevailing in India, and the ecological as well as the environmental characteristics of temperate areas, where mussel watch programmes are already in existence differ greatly. So it is essential to adopt the techniques and standards developed for temperate species to the situations and conditions in India. In this context it is a prerequisite to collect information on physiology and other biological indices of stress of possible sentinel organisms like P.viridis. In consideration of the above, P. viridis which is a potential sentinel organism, is selected for the present study

Tropical cyclones genesis, movement and intensification are highly dependent on its environment both oceanic and atmospheric. This thesis has made a detailed study on the environmental factors related to tropical cyclones of North Indian Ocean basin. This ocean basin has produced only 6% of the global tropical cyclones annually but it has caused maximum loss of human life associated with the strong winds, heavy rain and particularly storm surges that accompany severe cyclones as they strike the heavily populated coastal areas. Atmospheric factors studied in the thesis are the moisture content of the atmosphere, instability of the atmosphere that produces thunderstorms which are the main source of energy for the tropical cyclone, vertical wind shear to which cyclones are highly sensitive and the Sub-Tropical westerly Jetsteram and its Asian high speed center. The oceanic parameters studied are sea surface temperature and heat storage in the top layer of the ocean. A major portion of the thesis has dealt with the three temporal variabilities of tropical cyclone frequency namely intra-seasonal (mainly the influence of Madden Julian Oscillation), inter- annual (the relation with El Nino Southern Oscillation) and decadal variabilities. Regarding decadal variability, a prominent four decade oscillation in the frequency of both tropical cyclones and monsoon depressions unique to the Indian Ocean basin has been brought out. The thesis consists of 9 chapters.

The present study is focused on the intensity distribution of rainfall in different classes and their contribution to the total seasonal rainfall. In addition, we studied the spatial and diurnal variation of the rainfall in the study areas. For the present study, we retrieved data from TRMM (Tropical Rain Measuring Mission) rain rate available in every 3 h temporal and 25 km spatial resolutions. Moreover, station rainfall data is used to validate the TRMM rain rate and found significant correlation between them (linear correlation coefficients are 0.96, 0.85, 0.75 and 0.63 for the stations Kota Bharu, Senai, Cameron highlands and KLIA, respectively). We selected four areas in the Peninsular Malaysia and they are south coastal, east coastal, west coastal and highland regions. Diurnal variation of frequency of rain occurrence is different for different locations. We noticed bimodal variation in the coastal areas in most of the seasons and unimodal variation in the highland/inland area. During the southwest monsoon period in the west coastal stations, there is no distinct diurnal variation. The resurgence of the enteric pathogen Vibrio cholerae, the causative organism of epidemic cholera, remains a major health problem in many developing countries like India. The southern Indian state of Kerala is endemic to cholera. The outbreaks of cholera follow a seasonal pattern in regions of endemicity. Marine aquaculture settings and mangrove environments of Kerala serve as reservoirs for V. cholerae. The non-O1/non-O139 environmental isolates of V. cholerae with incomplete ‘virulence casette’ are to be dealt with caution as they constitute a major reservoir of diverse virulence genes in the marine environment and play a crucial role in pathogenicity and horizontal gene transfer. The genes coding cholera toxin are borne on, and can be infectiously transmitted by CTXΦ, a filamentous lysogenic vibriophages. Temperate phages can provide crucial virulence and fitness factors affecting cell metabolism, bacterial adhesion, colonization, immunity, antibiotic resistance and serum resistance. The present study was an attempt to screen the marine environments like aquafarms and mangroves of coastal areas of Alappuzha and Cochin, Kerala for the presence of lysogenic V. cholerae, to study their pathogenicity and also gene transfer potential. Phenotypic and molecular methods were used for identification of isolates as V. cholerae. The thirty one isolates which were Gram negative, oxidase positive, fermentative, with or without gas production on MOF media and which showed yellow coloured colonies on TCBS (Thiosulfate Citrate Bile salt Sucrose) agar were segregated as vibrios. Twenty two environmental V. cholerae strains of both O1 and non- O1/non-O139 serogroups on induction with mitomycin C showed the presence of lysogenic phages. They produced characteristic turbid plaques in double agar overlay assay using the indicator strain V. cholerae El Tor MAK 757. PCR based molecular typing with primers targeting specific conserved sequences in the bacterial genome, demonstrated genetic diversity among these lysogen containing non-O1 V. cholerae . Polymerase chain reaction was also employed as a rapid screening method to verify the presence of 9 virulence genes namely, ctxA, ctxB, ace, hlyA, toxR, zot,tcpA, ninT and nanH, using gene specific primers. The presence of tcpA gene in ALPVC3 was alarming, as it indicates the possibility of an epidemic by accepting the cholera. Differential induction studies used ΦALPVC3, ΦALPVC11, ΦALPVC12 and ΦEKM14, underlining the possibility of prophage induction in natural ecosystems, due to abiotic factors like antibiotics, pollutants, temperature and UV. The efficiency of induction of prophages varied considerably in response to the different induction agents. The growth curve of lysogenic V. cholerae used in the study drastically varied in the presence of strong prophage inducers like antibiotics and UV. Bacterial cell lysis was directly proportional to increase in phage number due to induction. Morphological characterization of vibriophages by Transmission Electron Microscopy revealed hexagonal heads for all the four phages. Vibriophage ΦALPVC3 exhibited isometric and contractile tails characteristic of family Myoviridae, while phages ΦALPVC11 and ΦALPVC12 demonstrated the typical hexagonal head and non-contractile tail of family Siphoviridae. ΦEKM14, the podophage was distinguished by short non-contractile tail and icosahedral head. This work demonstrated that environmental parameters can influence the viability and cell adsorption rates of V. cholerae phages. Adsorption studies showed 100% adsorption of ΦALPVC3 ΦALPVC11, ΦALPVC12 and ΦEKM14 after 25, 30, 40 and 35 minutes respectively. Exposure to high temperatures ranging from 50ºC to 100ºC drastically reduced phage viability. The optimum concentration of NaCl required for survival of vibriophages except ΦEKM14 was 0.5 M and that for ΦEKM14 was 1M NaCl. Survival of phage particles was maximum at pH 7-8. V. cholerae is assumed to have existed long before their human host and so the pathogenic clones may have evolved from aquatic forms which later colonized the human intestine by progressive acquisition of genes. This is supported by the fact that the vast majority of V. cholerae strains are still part of the natural aquatic environment. CTXΦ has played a critical role in the evolution of the pathogenicity of V. cholerae as it can transmit the ctxAB gene. The unusual transformation of V. cholerae strains associated with epidemics and the emergence of V. cholera O139 demonstrates the evolutionary success of the organism in attaining greater fitness. Genetic changes in pathogenic V. cholerae constitute a natural process for developing immunity within an endemically infected population. The alternative hosts and lysogenic environmental V. cholerae strains may potentially act as cofactors in promoting cholera phage ‘‘blooms’’ within aquatic environments, thereby influencing transmission of phage sensitive, pathogenic V. cholerae strains by aquatic vehicles. Differential induction of the phages is a clear indication of the impact of environmental pollution and global changes on phage induction. The development of molecular biology techniques offered an accessible gateway for investigating the molecular events leading to genetic diversity in the marine environment. Using nucleic acids as targets, the methods of fingerprinting like ERIC PCR and BOX PCR, revealed that the marine environment harbours potentially pathogenic group of bacteria with genetic diversity. The distribution of virulence associated genes in the environmental isolates of V. cholerae provides tangible material for further investigation. Nucleotide and protein sequence analysis alongwith protein structure prediction aids in better understanding of the variation inalleles of same gene in different ecological niche and its impact on the protein structure for attaining greater fitness of pathogens. The evidences of the co-evolution of virulence genes in toxigenic V. cholerae O1 from different lineages of environmental non-O1 strains is alarming. Transduction studies would indicate that the phenomenon of acquisition of these virulence genes by lateral gene transfer, although rare, is not quite uncommon amongst non-O1/non-O139 V. cholerae and it has a key role in diversification. All these considerations justify the need for an integrated approach towards the development of an effective surveillance system to monitor evolution of V. cholerae strains with epidemic potential. Results presented in this study, if considered together with the mechanism proposed as above, would strongly suggest that the bacteriophage also intervenes as a variable in shaping the cholera bacterium, which cannot be ignored and hinting at imminent future epidemics.
