Existing method of culture were largely based on empirical knowledge. Lacking a scientific basis as such methods did, they were often wasteful and suffered severe limitation. Modern methods of fish and prawn culture based on scientific research, have revolutioned the industry in recent years and not only extended its scope to cover the whole country but led to increased fish and prawn production. An understanding of the biological capability of the water in the perennial and seasonal culture ponds, and the nature and extent of the influence of the abiotic factors on the production of organisms in the primary level of food chain would contribute to effectively implement management measures in the stocking strategies and in the evaluation of economics of production of prawns. It is against this background that the present topic of investigation "Studies on the ecology and production of algae in prawn culture systems” was selected.

Given the economic importance of Jatropha curcas, and its limited availability in the wild, it would be desirable to establish plantations ofthe tree so as to obtain assured supply of raw material for extraction of phytochemicals, and seeds for production of biodiesel. However both seed propagation as well as propagation by cuttings is unsatisfactory in this tree species. Seeds have poor viability and are genetically heterozygous leading to genetic variability in terms of growth, biomass, seed yield, and oil content. Stern cuttings have poor roots and the trees are easily uprooted. Tissue culture techniques could possibly be gainfully employed in the propagation of elite plants ofJaIropha. When plant tissue is passaged through in vitro culture, there is possibility of induction of variations. An estimation of somaclonal variability is useful in a determination of culture protocols. Molecular markers could be employed to estimate the amount of variations induced in callus and regenerants by different honnonal combinations used in culture. In this context the present study aims to develop an in vitro propagation protocol for the production of plantlets and to evaluate the variation induced in callus and regenerants in comparison with mother plant by the use of molecular markers and by studying phytochemicals and bio active compounds present in callus and regenerated plants

In India, directed research on penaeid prawn nutrition was taken up only recently when the aquaculture of prawns gained momentum. One of the important penaeid prawns sought for culture and has great potential is Penagus indiggs, H.Milne Edwards. The Central Marine Fisheries Research Institute working on different aspects of culture of this species over the past one and half decades, has developed a hatchery technology for mass production of its seed and has suggested several improvements on its farming in the grow-out systems. One of the areas of active research in this direction has been on the nutrition of the species with a view to develop suitable feed not only for hatchery production of seed, but also in the field culture. As part of this investigation, the present study, on the evaluation of different protein and carbohydrate sources and mineral requirements for the juvenile E, indicus was taken up and the results obtained are embodied in the thesis

The present investigation was envisaged to determine the prevalence and identify the different Salmonella serovar in seafood from Cochin area. Though, the distribution of Salmonella serovars in different seafood samples of Cochin has been well documented, the present attempt was made to identify the different Salmonella serovars and determine its prevalence in various seafoods. First pan of this investigation involved the isolation and identification of Salmonella strains with the help of different conventional culture methods. The identified isolates were used for the further investigation i.e. serotyping, this provides the information about the prevalent serovars in seafood. The prevalent Salmonella strains have been further characterized based on the utilization of different sugars and amino acids, to identify the different biovar of a serovar.A major research gap was observed in molecular characterization of Salmonella in seafood. Though, previous investigations reported the large number of Salmonella serovars from food sources in India, yet, very few work has been reported regarding genetic characterization of Salmonella serovars associated with food. Second part of this thesis deals with different molecular fingerprint profiles of the Salmonella serovars from seafood. Various molecular typing methods such as plasmid profiling, characterization of virulence genes, PFGE, PCR- ribotyping, and ERIC—PCR have been used for the genetic characterization of Salmonella serovars.The conventional culture methods are mainly used for the identification of Salmonella in seafood and most of the investigations from India and abroad showed the usage of culture method for detection of Salmonella in seafood. Hence, development of indigenous, rapid molecular method is most desirable for screening of Salmonella in large number of seafood samples at a shorter time period. Final part of this study attempted to develop alternative, rapid molecular detection method for the detection of Salmonella in seafood. Rapid eight—hour PCR assay has been developed for detection of Salmonella in seafood. The performance of three different methods viz., culture, ELISA and PCR assays were evaluated for detection of Salmonella in seafood and the results were statistically analyzed. Presence of Salmonella cells in food and enviromnental has been reported low in number, hence, more sensitive method for enumeration of Salmonella in food sample need to be developed. A quantitative realtime PCR has been developed for detection of Salmonella in seafood. This method would be useful for quantitative detection of Salmonella in seafood.

Success achieved in seed production and culture of prawns induced researchers to explore the feasibiltiy of culture of other important crustaceans like lobsters and crabs. The CMFRI initiated a project to study the prospects of spiny lobster’ culture and I was associated with this project from the beginning. Compared to shrimp farming, culture of spiny lobsters is an entirely different proposition due to inherent problems in producing their seeds. The emphasis on .obster culture, therefore, shifted to rearing of juvenile lobsters, which form a major portion in commercial lobster landings in India. Commendable success was achieved in enhancing growth rate of four important speices of spiny lobsters by bilateral eyestalk ablation. But due to strong reservations expressed against the "blinding" of lobsters by people from all walks of life, this technique could not be tested commercially for its economic viability. As in prawns, energetics of reproduction and egg development have not yet been documented in spiny lobsters and so these aspects were included in this study. Though some limited informations on food conversion efficiency are availble in spiny lobster, a detailed study on the effect of size and sexual maturity on food conversion was wanting and this also was included in this investigation. The most dominant spiny lobster in south east and south west coast of India, Panulirus homarus (Linnaeus) was selected for this study. Uptake, accumulation and utilization of minerals and trace elements are important events during embryogenesis of aquatic invertebrates. The scope of this study was widened to include these aspects also in order to understand how specific requirements of minerals and trace elements are met in the developing eggs of these two species of crustaceans.
