Visual object tracking has been one of the most popular research topics in the field of computer vision recently. Specifically, hand tracking has attracted significant attention since it would enable many useful practical applications. However, hand tracking is still a very challenging problem which cannot be considered solved. The fact that almost every aspect of hand appearance can change is the fundamental reason for this difficulty. This thesis focused on 2D-based hand tracking in high-speed camera videos. During the project, a toolbox for this purpose was collected which contains nine different tracking methods. In the experiments, these methods were tested and compared against each other with both high-speed videos recorded during the project and publicly available normal speed videos. The results revealed that tracking accuracies varied considerably depending on the video and the method. Therefore, no single method was clearly the best in all videos, but three methods, CT, HT, and TLD, performed better than the others overall. Moreover, the results provide insights about the suitability of each method to different types and situations of hand tracking.

This study examines the practice of supply chain management problems and the perceived demand information distortion’s (the bullwhip effect) reduction with the interfirm information system, which is delivered as a cloud service to a company operating in the telecommunications industry. The purpose is to shed light in practice that do the interfirm information system have impact on the performance of the supply chain and in particularly the reduction of bullwhip effect. In addition, a holistic case study of the global telecommunications company's supply chain is presented and also the challenges it’s facing, and this study also proposes some measures to improve the situation. The theoretical part consists of the supply chain and its management, as well as increasing the efficiency and introducing the theories and related previous research. In addition, study presents performance metrics for the bullwhip effect detection and tracking. The theoretical part ends in presenting cloud -based business intelligence theoretical framework used in the background of this study. The research strategy is a qualitative case study, supported by quantitative data, which is collected from a telecommunication sector company's databases. Qualitative data were gathered mainly with two open interviews and the e-mail exchange during the development project. In addition, other materials from the company were collected during the project and the company's web site information was also used as the source. The data was collected to a specific case study database in order to increase reliability. The results show that the bullwhip effect can be reduced with the interfirm information system and with the use of CPFR and S&OP models and in particularly combining them to an integrated business planning. According to this study the interfirm information system does not, however, solve all of the supply chain and their effectiveness -related problems, because also the company’s processes and human activities have a major impact.

The main objective of the study was to identify and evaluate criteria for international partner selection in university-university context. This study attempted at promoting better understanding of how universities should proceed in selecting partners for producing joint research publications. Thus, the aim of the study was to gain an understanding of how research collaborations can be developed and how partners can be selected. The choice of a right partner has been identified as a precondition for partnership success. In international research collaborations partnering scientists with different skills and backgrounds bring together complementary knowledge into research projects, which in most cases results in a higher quality output. Therefore, prior to selecting a partner, the set of criteria should be established. This research examined twelve Russian universities with the status of national research university as potential partners for Lappeenranta University of Technology, and selected the most appropriate universities based on established set of criteria. Potential partners’ evaluation was done using secondary sources by tracking partners’ academic success during the period 2005 – 2010. Based on established criteria, the study calculated the partnership index for each university. The results of the research reveal that among twelve examined universities there are four potential partners who have been rather active in publishing scientific articles during 2005 – 2010.

Cardiac troponin (cTn) I and T are the recommended biomarkers for the diagnosis and risk stratification of patients with suspected acute coronary syndrome (ACS), a major cause of cardiovascular death and disability worldwide. It has recently been demonstrated that cTn-specific autoantibodies (cTnAAb) can negatively interfere with cTnI detection by immunoassays to the extent that cTnAAb-positive patients may be falsely designated as cTnI-negative. The aim of this thesis was to develop and optimize immunoassays for the detection of both cTnI and cTnAAb, which would eventually enable exploring the clinical impact of these autoantibodies on cTnI testing and subsequent patient management. The extent of cTnAAb interference in different cTnI assay configurations and the molecular characteristics of cTnAAbs were investigated in publications I and II, respectively. The findings showed that cTnI midfragment targeting immunoassays used predominantly in clinical practice are affected by cTnAAb interference which can be circumvented by using a novel 3+1-type assay design with three capture antibodies against the N-terminus, midfragment and C-terminus and one tracer antibody against the C-terminus. The use of this assay configuration was further supported by the epitope specificity study, which showed that although the midfragment is most commonly targeted by cTnAAbs, the interference basically encompasses the whole molecule, and there may be remarkable individual variation at the affected sites. In publications III and IV, all the data obtained in previous studies were utilized to develop an improved version of an existing cTnAAb assay and a sensitive cTnI assay free of this specific analytical interference. The results of the thesis showed that approximately one in 10 patients with suspected ACS have detectable amounts of cTnAAbs in their circulation and that cTnAAbs can inhibit cTnI determination when targeted against the binding sites of assay antibodies used in its immunological detection. In the light of these observations, the risk of clinical misclassification caused by the presence of cTnAAbs remains a valid and reasonable concern. Because the titers, affinities and epitope specificities of cTnAAbs and the concentration of endogenous cTnI determine the final effect of circulating cTnAAbs, appropriately sized studies on their clinical significance are warranted. The new cTnI and cTnAAb assays could serve as analytical tools for establishing the impact of cTnAAbs on cTnI testing and also for unraveling the etiology of cTn-related autoimmune responses.

The problem of automatic recognition of the fish from the video sequences is discussed in this Master’s Thesis. This is a very urgent issue for many organizations engaged in fish farming in Finland and Russia because the process of automation control and counting of individual species is turning point in the industry. The difficulties and the specific features of the problem have been identified in order to find a solution and propose some recommendations for the components of the automated fish recognition system. Methods such as background subtraction, Kalman filtering and Viola-Jones method were implemented during this work for detection, tracking and estimation of fish parameters. Both the results of the experiments and the choice of the appropriate methods strongly depend on the quality and the type of a video which is used as an input data. Practical experiments have demonstrated that not all methods can produce good results for real data, whereas on synthetic data they operate satisfactorily.

Tutkimuskäyttöön tarkoitettujen rekombinanttiproteiinien tuottaminen fermentoimalla on yleinen menetelmä bioteollisuudessa. Mikrobit kasvatetaan fermentorissa, joka tarjoaa kontrolloidun kasvuympäristön ja sopivat tuotto-olosuhteet halutulle tuotteelle. Eräs fermentointimuodoista on korkeatuottoinen ja pitkäkestoinen panossyöttökasvatus, jossa saavutetaan panoskavatusta merkittävästi korkeampi solutiheys jatkamalla panosvaiheen jälkeen kasvua rajoittavan substraatin syöttöä. Laboratoriomittakaavassa fermentorikasvatusten tilavuudet vaihtelevat litrasta kymmeniin ja niissä kasvatusta seurataan sekä ohjataan joko fermentorista tai tietokoneesta. Tyypillisessä fermentointiprosessissa operaattori tarkkailee muun muassa vaahdonkorkeutta sekä käynnistää pumppuja olosuhteiden muuttuessa. Tällaiset tehtävät ovat teollisen mittakaavan laitteistoissa usein automatisoituja. Diplomityön tarkoituksena oli päivittää kahden Turun yliopiston biotekniikan laboratoriossa sijaitsevan BioFlo® -sarjan pöytäfermentorin MS-DOS -pohjainen tietokoneohjausohjelma nykyaikaiseksi ja lisätä siihen etäseuranta ja -ohjaus. Ohjelmaan oli tarkoitus liittää erillinen optinen solutiheysanturi, jonka lukemien häiriötä haluttiin myös vähentää signaalinkäsittelyllä. Lisäksi vaahdonestoaineen ja indusorin lisäykset haluttiin automatisoida panossyöttökasvatuksessa. Vaahdonkorkeuden havaitsemisen mahdollisuutta konenäön menetelmin haluttiin selvittää, jotta vaahdonestoaineen automaattiset lisäykset voitaisiin toteuttaa nettikameran syötteen perusteella. Koekasvatuksilla osoitettiin päivitetyn ohjausohjelman toimivan panos- ja panossyöttömuodoilla. Uuden käyttöliittymän avulla pystyttiin automatisoimaan panoskasvatuksen lisäykset ja syöttönopeuden muutokset sekä tunnistamaan kasvatusliuosten vaahdonkorkeutta vaahdonestoaineen lisäykseen riittävällä kahden senttimetrin tarkkuudella. Lisäksi käyttöliittymä mahdollisti kasvatuksen ohjauksen ja seurauksen myös etänä. Työssä kehitetty ohjausohjelma julkaistiin avoimena ohjelmana ilman etä- ja nettikameratoimintoja. Ohjelma toimii hyvin BioFlo® -sarjan fermentorien käyttöliittymänä, mutta avoimen lähdekoodin ansiosta kuka tahansa voi hyödyntää ohjelmaa pohjana myös uusissa projekteissa tai muissa fermentorimalleissa.

This MSc work was done in the project of BIOMECON financed by Tekes. The prime target of the research was, to develop methods for separation and determination of carbohydrates (sugars), sugar acids and alcohols, and some other organic acids in hydrolyzed pulp samples by capillary electrophoresis (CE) using UV detection. Aspen, spruce, and birch pulps are commonly used for production of papers in Finland. Feedstock components in pulp predominantly consist of carbohydrates, organic acids, lignin, extractives, and proteins. Here in this study, pulps have been hydrolyzed in analytical chemistry laboratories of UPM Company and Lappeenranta University in order to convert them into sugars, acids, alcohols, and organic acids. Foremost objective of this study was to quantify and identify the main and by-products in the pulp samples. For the method development and optimization, increased precision in capillary electrophoresis was accomplished by calculating calibration data of 16 analytes such as D-(-)-fructose, D(+)-xylose, D(+)-mannose, D(+)-cellobiose, D-(+)-glucose, D-(+)-raffinose, D(-)-mannitol, sorbitol, rhamnose, sucrose, xylitol, galactose, maltose, arabinose, ribose, and, α-lactose monohydratesugars and 16 organic acids such as D-glucuronic, oxalic, acetic, propionic, formic, glycolic, malonic, maleic, citric, L-glutamic, tartaric, succinic, adipic, ascorbic, galacturonic, and glyoxylic acid. In carbohydrate and polyalcohol analyses, the experiments with CE coupled to direct UV detection and positive separation polarity was performed in 36 mM disodium hydrogen phosphate electrolyte solution. For acid analyses, CE coupled indirect UV detection, using negative polarity, and electrolyte solution made of 2,3 pyridinedicarboxylic acid, Ca2+ salt, Mg2+ salts, and myristyltrimethylammonium hydroxide in water was used. Under optimized conditions, limits of detection, relative standard deviations and correlation coefficients of each compound were measured. The optimized conditions were used for the identification and quantification of carbohydrates and acids produced by hydrolyses of pulp. The concentrations of the analytes varied between 1 mg – 0.138 g in liter hydrolysate.

Food safety has always been a social issue that draws great public attention. With the rapid development of wireless communication technologies and intelligent devices, more and more Internet of Things (IoT) systems are applied in the food safety tracking field. However, connection between things and information system is usually established by pre-storing information of things into RFID Tag, which is inapplicable for on-field food safety detection. Therefore, considering pesticide residue is one of the severe threaten to food safety, a new portable, high-sensitivity, low-power, on-field organophosphorus (OP) compounds detection system is proposed in this thesis to realize the on-field food safety detection. The system is designed based on optical detection method by using a customized photo-detection sensor. A Micro Controller Unit (MCU) and a Bluetooth Low Energy (BLE) module are used to quantize and transmit detection result. An Android Application (APP) is also developed for the system to processing and display detection result as well as control the detection process. Besides, a quartzose sample container and black system box are also designed and made for the system demonstration. Several optimizations are made in wireless communication, circuit layout, Android APP and industrial design to realize the mobility, low power and intelligence.
