Using cellulosic reinforcement to produce plastic composites is a globally growing trend. One of such materials are wood-plastic composites, which are an extensively studied group of materials for which the global industry is looking for new applications. Issues such as bondability, durability and fire resistance still require development to improve the usability of the wood-plastic composite material. Improvement of the usability of wood-plastic composites is studied in this thesis through the effects of using selected modification technology in wood and plastic industry. The applied modification methods are surface by mechanical abrasion and plasma, chemical impregnation of wood flour, and structural modification by the co-extrusion process. The study shows that the properties of WPC can be influenced by the selected modification methods. The selected methods are also found to be able to result as improvement in the properties of the material. The may also affect other than just the targeted properties of the end-product, either in a positive or a negative manner. Therefore modification as performance improvement should be considered as a caseby- case study. Introducing WPC materials for new applications can be done by using modification technology. Structuralmodification can possibly be used to reduce material costs of the modified WPC material.

Diplomityö on osa YTI-tutkimuskeskuksessa vuosina 2002 - 2004 toteutettavaa Jätekompostit rakeiksi tuhkaseostuksella -käyttöarvon parantaminen -projektia. Työssä tutkittiin Etelä-Savon Energia Oy:n Pursialan voimalaitoksen lentotuhkan fraktioimista voimalaitoksen nykyisellä 3-kenttäisellä sähkösuodattimella ja pilot-mittakaavaisella Ion Blast -koelaitteistolla. Sähkösuodattimen koeajojen aikana muuteltiin sen ajotapaa mm. CBO -suhteen ja maksimijänniteasetuksen avulla. Ion Blast -koelaitteistolla tutkittiin mahdollisuuksia voimalaitoksen lentotuhkan puhdistamiseksi raskasmetalleista. Lentotuhkan hyötykäyttöä vaikeuttaa sen raskasmetallipitoisuuksien suuri vaihtelu. Ongelmallisin raskasmetalli puuperäisessä lentotuhkassa on kadmium, jonka lannoitelainsäädännön raja-arvo on tällä hetkellä 3 mg/kg. Sähkösuodattimella tehtyjen fraktiointikokeiden perusteella voidaan todeta raskasmetallipitoisuuksien olevan pienimmillään sähkösuodattimen 1-kentässä ja suurimmillaan 3-kentässä. Tämä johtuu siitä, että 1-kenttään kerääntyy hiukkaskooltaan suurimmat lentotuhkahiukkaset ja 3-kentässä on mukana enemmän pienhiukkasia sisältävää tuhkaa. Lannoitteeksi menevän tuhkan Cd-pitoisuutta voidaan vähentää parhaimmillaan jopa 70 % sähkösuodattimella fraktioimalla. Muiden raskasmetallien pitoisuudet eivät vähene aivan yhtä paljon. Sähkösuodattimella voidaan tulosten perusteella fraktioida lentotuhkaa. Sähkösuodattimella ei kuitenkaan voida varmasti saavuttaa alle 3 mg/kg Cd-pitoisuuksia polttoaineen laadunvaihtelun vuoksi. Ion Blast -koelaitteiston tulokset tukevat sähkösuodattimella tehtyjä kokeita. Erottimen jännitteen kasvaessa raskasmetalleja sisältävien hiukkasten erotusaste kasvaa. Ion Blast -laitteistolla tehdyissä kokeissa myös Cd-pitoisuus oli korkeimmillaan pienimmän raeluokan hiukkasissa ja laski sitten raeluokan suurentuessa. Ion Blast -laitteisto ei kuitenkaan sellaisenaan ole hyvä fraktiointiin. Se on liian tehokas, jolloin se puhdistaa tehokkaasti myös raskasmetalleja sisältävät pienhiukkaset. Jos laitetta aiotaan käyttää fraktiointiin, tulisi sen rakennetta muuttaa.

Diplomityössä selvitetään PWR PACTEL -koelaitteiston APROS-mallin ylätilan toimi¬vuut¬ta ja tutkitaan tapoja parantaa mallin simulointitarkkuutta. Työssä on esitelty PWR PACTEL -koelaitteiston sekä APROS-simulointiohjelman pääpiirteet ja tutustuttu simu¬loin¬ti¬ohjelmien kelpoistukseen. Simulointiosiossa tarkastellaan yksinkertaisen ja muokatun ylätilamallin toimivuutta ver¬taa¬mal¬la simulointia koelaitteistolla tehtyjen kokeiden mittaustuloksiin. Lisäksi tutkitaan nel¬¬jää tapaa parantaa ylätilamallin toimivuutta: virtausvastusten säätö, lämpöhäviöiden sä䬬tö, rakenteelliset muutokset ja laskentakorrelaatioiden vaihtaminen. Simulointituloksia ver¬¬ra¬¬taan kahteen koelaitteistolla suoritettuun kokeeseen, joista toinen on tasapainotilankoe ja toinen dynaamisen tilan koe. Työssä havaittiin, että nykyisin käytössä olevaa mallia on kehitetty jo varsin paljon, ver¬rat¬tu¬na ns. yksinkertaiseen malliin. Testatuilla parannusvaihtoehdoilla saatiin hieman paran¬net¬tua ylätilan lämpötilojen simulointia tasapainotilassa. Dynaamisen tilan osalta havaittiin pien¬tä parannusta mallin toiminnassa ylätilamallin rakennemuutosten jälkeen. Tulosten poh¬jalta arvioitiin, että mallia on mahdollista kehittää muuttamalla ylätilamallin raken¬net¬ta.

The main objective of this thesis is to study the impact of different mineral fillers and fire retardants on the reaction-to-fire properties of extruded/coextruded wood-plastic composites (WPCs). The impact of additives on the flammability properties of WPCs is studied by cone calorimetry. The studied properties are ignition time, peak heat release rate, total heat release, total smoke production, and mass loss rate. The effects of mineral fillers and fire retardants were found to vary with the type of additive, the type of additive combinations, the amount of additives, as well as the production method of the WPCs. The study shows that talc can be used to improve the properties of extruded WPCs. Especially ignition time, peak heat release rate and mass loss rate were found to be improved significantly by talc. The most significant improvement in the fire retardancy of coextruded WPCs was achieved in combinations of natural graphite and melamine. Ignition time, peak heat release rate and total smoke production were improved essentially. High increase in smoke production was found in samples where the amount of ammonium polyphosphate was 10% or higher. Coextrusion as a structural modification was found as a promising way to improve the flammability properties of composite materials in a cost-effective way.

Laser beam welding (LBW) is applicable for a wide range of industrial sectors and has a history of fifty years. However, it is considered an unusual method with applications typically limited to welding of thin sheet metal. With a new generation of high power lasers there has been a renewed interest in thick section LBW (also known as keyhole laser welding). There was a growing body of publications during 2001-2011 that indicates an increasing interest in laser welding for many industrial applications, and in last ten years, an increasing number of studies have examined the ways to increase the efficiency of the process. Expanding the thickness range and efficiency of LBW makes the process a possibility for industrial applications dealing with thick metal welding: shipbuilding, offshore structures, pipelines, power plants and other industries. The advantages provided by LBW, such as high process speed, high productivity, and low heat input, may revolutionize these industries and significantly reduce the process costs. The research to date has focused on either increasing the efficiency via optimizing process parameters, or on the process fundamentals, rather than on process and workpiece modifications. The argument of this thesis is that the efficiency of the laser beam process can be increased in a straightforward way in the workshop conditions. Throughout this dissertation, the term “efficiency” is used to refer to welding process efficiency, specifically, an increase in efficiency refers an increase in weld’s penetration depth without increasing laser power level or decreasing welding speed. These methods are: modifications of the workpiece – edge surface roughness and air gap between the joining plates; modification of the ambient conditions – local reduction of the pressure in the welding zone; modification of the welding process – preheating of the welding zone. Approaches to improve the efficiency are analyzed and compared both separately and combined. These experimentally proven methods confirm previous findings and contribute additional evidence which expand the opportunities for laser beam welding applications. The focus of this research was primarily on the effects of edge surface roughness preparation and pre-set air gap between the plates on weld quality and penetration depth. To date, there has been no reliable evidence that such modifications of the workpiece give a positive effect on the welding efficiency. Other methods were tested in combination with the two methods mentioned above. The most promising - combining with reduced pressure method - resulted in at least 100% increase in efficiency. The results of this thesis support the idea that joining those methods in one modified process will provide the modern engineering with a sufficient tool for many novel applications with potential benefits to a range of industries.

It is commonly observed that complex fabricated structures subject tofatigue loading fail at the welded joints. Some problems can be corrected by proper detail design but fatigue performance can also be improved using post-weld improvement methods. In general, improvement methods can be divided into two main groups: weld geometry modification methods and residual stress modification methods. The former remove weld toe defects and/or reduce the stress concentrationwhile the latter introduce compressive stress fields in the area where fatigue cracks are likely to initiate. Ultrasonic impact treatment (UIT) is a novel post-weld treatment method that influences both the residual stress distribution andimproves the local geometry of the weld. The structural fatigue strength of non-load carrying attachments in the as-welded condition has been experimentally compared to the structural fatigue strength of ultrasonic impact treated welds. Longitudinal attachment specimens made of two thicknesses of steel S355 J0 have been tested for determining the efficiency of ultrasonic impacttreatment. Treated welds were found to have about 50% greater structural fatigue strength, when the slope of the S-N-curve is three. High mean stress fatigue testing based on the Ohta-method decreased the degree of weld improvement only 19%. This indicated that the method could be also applied for large fabricated structures operating under high reactive residual stresses equilibrated within the volume of the structure. The thickness of specimens has no significant effect tothe structural fatigue strength. The fatigue class difference between 5 mm and 8 mm specimen was only 8%. It was hypothesized that the UIT method added a significant crack initiation period to the total fatigue life of the welded joints. Crack initiation life was estimated by a local strain approach. Material parameters were defined using a modified Uniform Material Law developed in Germany. Finite element analysis and X-ray diffraction were used to define, respectively, the stress concentration and mean stress. The theoretical fatigue life was found to have good accuracy comparing to experimental fatigue tests.The predictive behaviour of the local strain approach combined with the uniformmaterial law was excellent for the joint types and conditions studied in this work.

Fatigue life assessment of weldedstructures is commonly based on the nominal stress method, but more flexible and accurate methods have been introduced. In general, the assessment accuracy is improved as more localized information about the weld is incorporated. The structural hot spot stress method includes the influence of macro geometric effects and structural discontinuities on the design stress but excludes the local features of the weld. In this thesis, the limitations of the structural hot spot stress method are discussed and a modified structural stress method with improved accuracy is developed and verified for selected welded details. The fatigue life of structures in the as-welded state consists mainly of crack growth from pre-existing cracks or defects. Crack growth rate depends on crack geometry and the stress state on the crack face plane. This means that the stress level and shape of the stress distribution in the assumed crack path governs thetotal fatigue life. In many structural details the stress distribution is similar and adequate fatigue life estimates can be obtained just by adjusting the stress level based on a single stress value, i.e., the structural hot spot stress. There are, however, cases for which the structural stress approach is less appropriate because the stress distribution differs significantly from the more common cases. Plate edge attachments and plates on elastic foundations are some examples of structures with this type of stress distribution. The importance of fillet weld size and weld load variation on the stress distribution is another central topic in this thesis. Structural hot spot stress determination is generally based on a procedure that involves extrapolation of plate surface stresses. Other possibilities for determining the structural hot spot stress is to extrapolate stresses through the thickness at the weld toe or to use Dong's method which includes through-thickness extrapolation at some distance from the weld toe. Both of these latter methods are less sensitive to the FE mesh used. Structural stress based on surface extrapolation is sensitive to the extrapolation points selected and to the FE mesh used near these points. Rules for proper meshing, however, are well defined and not difficult to apply. To improve the accuracy of the traditional structural hot spot stress, a multi-linear stress distribution is introduced. The magnitude of the weld toe stress after linearization is dependent on the weld size, weld load and plate thickness. Simple equations have been derived by comparing assessment results based on the local linear stress distribution and LEFM based calculations. The proposed method is called the modified structural stress method (MSHS) since the structural hot spot stress (SHS) value is corrected using information on weld size andweld load. The correction procedure is verified using fatigue test results found in the literature. Also, a test case was conducted comparing the proposed method with other local fatigue assessment methods.

The Tandem-GMAW method is the latest development as the consequences of improvements in the welding methods. The twin-wire and then the Tandem-method with the separate power sources has got a remarkable place in the welding of many types of materials with different joint types. The biggest advantage of Tandem welding method is the flexibility of choosing both the electrodes of different types from each other according to the type of the parent material. This is possible because of the feasibility of setting the separate welding parameters for both the wires. In this thesis work the effect of the variation in three parameters on the weld bead in Tandem-GMA welding method is studied. Theses three parameters are the wire feed rate in the slave wire, the wire feed rate in the master wire and the voltage difference in both the wires. The results are then compared to study the behaviour of the weld bead with the change in these parameters.

Tässä työssä tutkittiin jälkiseurantana putkipalkkirakenteisen ristikkotornin rakennusprojektia. Tutkimuksen kohteena oli suomalaiselle öljynjalostamolle vuonna 1992 rakennettu vapaastiseisova 105 m korkea soihtutorni. Työn tarkoituksena oli tutkia teräsrakennekokonaisuutta toisaalta suunnittelun ja lujuuslaskennan, toisaalta valmistuksen, kuljetuksen ja asennuksen kannalta. Lisäksi työssä etsittiin soihtutornin vaihtoehtoisia rakenneratkaisuja. Esisuunnittelun tuloksena syntyneitä rakenteita verrattiin vuonna 1992 toteutettuun rakenteeseen. Toteutetun ristikkomaston lujuusominaisuuksia arvioitiin käymällä läpi tehdyt lujuuslaskelmat ja selvittämällä mm. rakenteen eri kohdista mitoituksen tasaisuus. Havaittiin, että rakenteen lujuuden käyttöaste vaihteli 0,7:n ja 0,9:n välillä 72 m:n korkeuteen asti. Tästä ylöspäin käyttöasteet olivat pienempiä. Konepajavalmistuksen, kuljetusten ja asennustöiden onnistumista tutkittiin haastattelemalla sekä konepajan että tilaajan edustajia. Ylöspäin kapeneva rakenne osoittautui lujuusopillisesti järkeväksi, mutta valmistusteknisesti vanhanaikaiseksi. Massiivisten ristikkotornien kuljetus- ja nostomahdollisuuksia tutkittiin selvittämällä lainsäädännön asettamat rajoitukset ja käsikirjojen antamat ohjeet. Lisäksi haastateltiin erikoiskuljetuksia ja -nostoja suorittavan yrityksen edustajaa. Uusien rakenneratkaisujen suunnittelussa korostettiin pitkälle vietyä osavalmistusta sekä kuljetus- ja nostomahdollisuuksia. Uusista tornivaihtoehdoista tutkittiin tarkemmin massiivisista pyöreistä putkipalkeista tehty torni, joka on rakenteeltaan Vierendeel-kehän sovellutus. Rakenne on 62 % painavampi kuin vuonna 1992 toteutettu soihtutorni, mutta silti 9 % halvempi. Uuden rakenteen valmistusystävällisyys on merkittävästi parempi kuin vanhan.

The goal of the thesis was to study fundamental structural and optical properties of InAs islands and In(Ga)As quantum rings. The research was carried out at the Department of Micro and Nanosciences of Helsinki University of Technology. A good surface quality can be essential for the potential applications in optoelectronic devices. For such device applications it is usually necessary to control size, density and arrangement of the islands. In order to study the dependence of the structural properties of the islands and the quantum rings on growth conditions, atomic force microscope was used. Obtained results reveal that the size and the density of the In(Ga)As quantum rings strongly depend on the growth temperature, the annealing time and the thickness of the partial capping layer. From obtained results it is possible to conclude that to get round shape islands and high density one has to use growth temperature of 500 ̊C. In the case of formation of In(Ga)As quantum rings the effect of mobility anisotropy is observed that so the shape of the rings is not symmetric. To exclude this effect it is preferable to use a higher annealing temperature of 570 ̊C. Optical properties were characterized by PL spectroscopy. PL emission was observed from buried InAs quantum dots and In(Ga)As quantum rings grown with different annealing time and temperature and covered with a various thickness of the partial capping layer.

Members of the bacterial genus Streptomyces are well known for their ability to produce an exceptionally wide selection of diverse secondary metabolites. These include natural bioactive chemical compounds which have potential applications in medicine, agriculture and other fields of commerce. The outstanding biosynthetic capacity derives from the characteristic genetic flexibility of Streptomyces secondary metabolism pathways: i) Clustering of the biosynthetic genes in chromosome regions redundant for vital primary functions, and ii) the presence of numerous genetic elements within these regions which facilitate DNA rearrangement and transfer between non-progeny species. Decades of intensive genetic research on the organization and function of the biosynthetic routes has led to a variety of molecular biology applications, which can be used to expand the diversity of compounds synthesized. These include techniques which, for example, allow modification and artificial construction of novel pathways, and enable gene-level detection of silent secondary metabolite clusters. Over the years the research has expanded to cover molecular-level analysis of the enzymes responsible for the individual catalytic reactions. In vitro studies of the enzymes provide a detailed insight into their catalytic functions, mechanisms, substrate specificities, interactions and stereochemical determinants. These are factors that are essential for the thorough understanding and rational design of novel biosynthetic routes. The current study is a part of a more extensive research project (Antibiotic Biosynthetic Enzymes; www.sci.utu.fi/projects/biokemia/abe), which focuses on the post-PKS tailoring enzymes involved in various type II aromatic polyketide biosynthetic pathways in Streptomyces bacteria. The initiative here was to investigate specific catalytic steps in anthracycline and angucycline biosynthesis through in vitro biochemical enzyme characterization and structural enzymology. The objectives were to elucidate detailed mechanisms and enzyme-level interactions which cannot be resolved by in vivo genetic studies alone. The first part of the experimental work concerns the homologous polyketide cyclases SnoaL and AknH. These catalyze the closure of the last carbon ring of the tetracyclic carbon frame common to all anthracycline-type compounds. The second part of the study primarily deals with tailoring enzymes PgaE (and its homolog CabE) and PgaM, which are responsible for a cascade of sequential modification reactions in angucycline biosynthesis. The results complemented earlier in vivo findings and confirmed the enzyme functions in vitro. Importantly, we were able to identify the amino acid -level determinants that influence AknH and SnoaL stereoselectivity and to determine the complex biosynthetic steps of the angucycline oxygenation cascade of PgaE and PgaM. In addition, the findings revealed interesting cases of enzyme-level adaptation, as some of the catalytic mechanisms did not coincide with those described for characterised homologs or enzymes of known function. Specifically, SnoaL and AknH were shown to employ a novel acid-base mechanism for aldol condenzation, whereas the hydroxylation reaction catalysed by PgaM involved unexpected oxygen chemistry. Owing to a gene-level fusion of two ancestral reading frames, PgaM was also shown to adopt an unusual quaternary sturucture, a non-covalent fusion complex of two alternative forms of the protein. Furthermore, the work highlighted some common themes encountered in polyketide biosynthetic pathways such as enzyme substrate specificity and intermediate reactivity. These are discussed in the final chapters of the work.
