Surface chemistry is of great importance in plant biomass engineering and applications. The surface chemical composition of biomass which includes lignin, carbohydrates and extractives influences its interactions with chemical agents, such as pulp processing/papermaking chemicals, or enzymes for different purposes. In this thesis, the changes in the surface chemical composition of lignocellulosic biomass after physical modification for the improvement of resulting paper properties and chemical treatment for the enhancement of enzymatic hydrolysis were investigated. Low consistency (LC) refining was used as physical treatment of bleached softwood and hardwood pulp samples, and the surface chemistry of refined samples was investigated. The refined pulp was analysed as whole pulp while the fines-free fibre samples were characterized separately. The fines produced in LCrefining contributed to an enlarged surface specific area as well as the change of surface coverage by lignin and extractives, as investigated by X-ray photoelectron spectroscopy (XPS). The surface coverage by lignin of the whole pulp decreased after refining while the surface coverage by extractives increased both for pine and eucalyptus. In the case of pine, the removal of fines resulted in reduction of the surface coverage by extractives, while the surface coverage by lignin increased on fibre sample (without fines). In the case of eucalyptus, the surface coverage by lignin of fibre samples decreased after the removal of fines. In addition, the surface distribution of carbohydrates, lignin and extractives of pine and eucalyptus samples was determined by Time-of-Flight Secondary Ion Mass Spectrometry (ToF-SIMS). LC-refining increased the amounts of pentose, hexose and extractives on the surface of pine samples. ToF-SIMS also gave clear evidence about xylan deposition and reduction of surface lignin distribution on the fibre of eucalyptus. However, the changes in the surface chemical composition during the physical treatment has led to an increase in the adsorption of fluorescent whitening agents (FWAs) on fibres due to a combination of electro-static forces, specific surface area of fibres and hydrophobic interactions. Various physicochemical pretreatments were conducted on wood and non-wood biomass for enhancing enzymatic hydrolysis of polysaccharides, and the surface chemistry of the pretreated and enzymatically hydrolysed samples was investigated by field emission scanning electron microscopy (FE-SEM), XPS and ToF-SIMS. A hydrotrope was used as a relatively novel pretreatment technology both in the case of wood and non-wood biomass. For comparison, ionic liquid and hydrothermal pretreatments were applied on softwood and hardwood as well. Thus, XPS analysis showed that the surface lignin was more efficiently removed by hydrotropic pretreatment compared to ionic liquid or hydrothermal pretreatments. SEM analysis also found that already at room temperature the ionic liquid pretreatments were more effective in swelling the fibres compared with hydrotropic pretreatment at elevated temperatures. The enzymatic hydrolysis yield of hardwood was enhanced due to the decrease in surface coverage of lignin, which was induced by hydrotropic treatment. However, hydrotropic pretreatment was not appropriate for softwood because of the predominance of guaiacyl lignin structure in this material. In addition, the reduction of surface lignin and xylan during pretreatment and subsequent increase in cellulose hydrolysis by enzyme could be observed from ToF-SIMS results. The characterisation of the non-wood biomass (e.g. sugarcane bagasse and common reed) treated by hydrotropic method, alkaline and alkaline hydrogen peroxide pretreatments were carried out by XPS and ToF-SIMS. According to the results, the action for the removal of the surface lignin of non-wood biomass by hydrotropic pretreatment was more significant compared to alkaline and alkaline hydrogen peroxide pretreatments, although a higher total amount of lignin could be removed by alkaline and alkaline hydrogen peroxide pretreatment. Furthermore, xylan could be remarkably more efficiently removed by hydrotropic method. Therefore, the glucan yield achieved from hydrotropic treated sample was higher than that from samples treated with alkaline or alkaline hydrogen peroxide. Through the use of ToF-SIMS, the distribution and localization of lignin and carbohydrates on the surface of ignocelluloses during pretreatment and enzymatic hydrolysis could be detected, and xylan degradation during enzymatic hydrolysis could also be assessed. Thus, based on the results from XPS and ToF-SIMS, the mechanism of the hydrotropic pretreatment in improving the accessibility of enzymes to fibre and further ameliorating of the enzymatic saccharification could be better elucidated.

Rasvaisten jätevesien puhdistus on sitä tuottaville yrityksille kallista ja hankalaa. Nykyisten päästövaatimusten saavuttaminen on perinteisillä jätevedenkäsittelymenetelmillä vaikeaa tai lähes mahdotonta, riippuen käsiteltävän jäteveden ominaisuuksista. Rasvaisen jäteveden käsittelyssä on käytetty mm. laskeutusta, flotaatiota, hydrosykloneja, pisaroitusta, suodatusta sekä biologista käsittelyä. Lisäksi happohydrolyysiä voidaan soveltaa edellä mainittujen menetelmien esikäsittelynä. Useita näitä erotusmenetelmiä voidaan myös tehostaa kemikaalien lisäyksellä. Työn kirjallisuusosassa on käsitelty rasvaisten jätevesien ja emulsioiden kalvosuodatusta ja kalvojen käyttöä pisaroituselementtinä. Tavanomaisessa kalvosuodatuksessa tarkoituksena on erottaa kalvoa läpäisemätön rasvajae ja permeoituva vesijae toisistaan, kun taas pisaroittamisen tarkoituksena on saada dispergoituneen faasin pisarakoko kasvamaan joko kalvon pinnalla tai sen huokosissa. Pisarakoon kasvaessa emulsion stabiilisuus heikkenee ja faasit voidaan helpommin erottaa toisistaan. Työn kokeellisessa osassa tavoitteena oli tutkia kalvosuodatuksen ja erilaisten kalvojen toimivuutta esteröintilaitoksen rasvaisten jätevesien käsittelyssä. Tutkimuksessa käytettiin MW- (GE Osmonics), C30F- (Nadir Filtration), Teflon Typar- (Tetratex) sekä JX-kalvoa (Osmonics Desal). Haastetta työhön syntyi tutkittujen jätevesien ominaisuuksien suuresta vaihtelusta sitä tuottavan laitoksen panostyylisestä toiminnasta johtuen. Lisäksi tutkittiin, onko syöttöliuoksen pH-säädöllä ja laskeutuksella ennen suodatusta merkittävää etua itse kalvosuodatusprosessiin. Kalvotekniikkaa voidaan tämän tutkimuksen perusteella soveltaa myös esteröintilaitoksen rasvaisten jätevesien suodatukseen, ja erityistä etua saadaan jäteveden pH-säädöllä (pH 3) ja laskeutuksella ennen varsinaista suodatusta. Tällaiseen käsittelyyn soveltuu tutkituista kalvoista parhaiten hydrofiilinen regeneroidusta selluloosasta valmistettu C30F-kalvo, jonka etuna on vähäinen foulaantuminen muihin tutkittuihin kalvoihin verrattuna.

Työn tavoitteena oli tutkia kiertoveden puhdistamiseen käytetyn kalvosuodatuksen esikäsittelymenetelmien vaikutusta ultrasuodatukseen. Suodatettava vesi oli kierrätyskuitua käyttävän paperikoneen kiekkosuotimen kirkassuodosta. Koeajon loppuosassa käytettiin biokirkassuodosta. Paperitehtaassa oli suljettu vesikierto. Pilot-laitteistona käytettiin Metso PaperChemin OptiFilter CR550/40 ultrasuodatusyksikköä. Testatut kalvot olivat C30 kalvoja. Suodatuksissa seurattiin esikäsittelymenetelmien vaikutusta permeaatin määrään ja laatuun. Ultrasuodatus tehtiin sekä ilman esikäsittelyä että lamelliselkeytin esikäsittelynä. Esikäsittelykemikaalina käytettiin talkkia. Ultrasuodatuksissa havaittiin, että suodatuslämpötilalla oli suora vaikutus kapasiteettiin. Lämpötila vaikutti lineaarisesti kirkassuodoksen ja biokirkassuodoksen permeaattivuohon. Lämpötila nostettiin 55oC:een suodatuslämpötilasta 35 – 40oC. Korkeassa lämpötilassa kirkassuodos oli kapasiteetiltaan suurempi kuin biokirkassuodos. Konsentrointiajoista nähtiin, että ultrasuodatuksen syötön konsentraatioaste vaikutti kirkassuodoksen permeaattivuohon. Syötön konsentraatioastetta lisättäessä kapasiteetti laski. Konsentrointiajoja vertailtaessa havaitaan, että ilman esikäsittelyä vuo laski konsentroitaessa nopeammin kuin suodatuksissa, joissa käytettiin esikäsittelyä. Lamelliselkeyttimellä ja talkilla esikäsitelty suodatus laski kapasiteettia vähiten. Konsentraatioasteen nostaminen lisäsi konsentraatin kemiallista hapenkulutusta. Konsentraatin viskositeetti nousi konsentraatioastetta lisättäessä. Analyysitulosten perusteella saadaan ultrasuodatuskalvoille tyypilliset reduktiot. COD-reduktio oli 10 – 30 %, anionisuuden 40 – 70 % ja värin 80 – 90 %. Vuon kanssa korreloivat pH, uuteaine, ligniini, viskositeetti ja hemiselluloosat.

Tässä diplomityössä tutkittiin kalvosuodatuksen esikäsittelymenetelmiä ja kalvonpesua. Työn kirjallisuusosassa käsitellään vuon alenemiseen vaikuttavia tekijöitä, esikäsittelymenetelmiä ja kalvonpesua. Kokeellisessa osassa tutkittiin kemiallisten esikäsittelyjen vaikutusta vuon alenemiseen paperitehtaan happaman kiertoveden kirkkaan suodoksen kalvosuodatuksessa. Esikäsittelykemikaalit olivat ympäristöystävällisiä ja paperinvalmistusprosessiin soveltuvia. Lisäksi tutkittiin kalvonpesuaineiden pesutehokkuuksia. Tutkitut esikäsittelyaineet olivat mikrokiteinen kitosaani, karboksimetyyliselluloosa, selluloosa- ja puukuitu sekä kaupallinen antiskalantti. Pesuaineista tutkittiin kolmea kaupallista kalvonpesuainetta, yhtä kalvopesun tehostusainetta sekä peretikkahappoa. Kokeet tehtiin kahdella laboratoriomittakaavaisella kalvosuodattimella. Kalvoina käytettiin kahta nanosuodatus- ja yhtä ultrasuodatuskalvoa. Vuon alenemista tutkittiin suodatuksen aikaisena alenemisena ja vesivuohon verrattavana alenemisena. Esikäsittelyjen vaikutusta erotustehokkuuteen tutkittiin ioni-, johtokyky-, orgaanisen hiilen kokonaispitoisuus-, sokeri-, sameus- ja ligniinireduktioilla. Lisäksi määritettiin kalvon likaantuminen suodatuksen aikana vesivuon määrityksillä ennen ja jälkeen suodatuksen. Pesutehokkuus määritettiin vesivuon määrityksillä suodatuksen jälkeen ja pesun jälkeen. Kitosaani- ja karboksimetyyliselluloosakäsittelyillä oli vuon alenemista estävä vaikutus hydrofiilisellä nanosuodatuskalvolla suodatettaessa. Kitosaanikäsittelyn 5 g/dm3:n ja karboksimetyyliselluloosakäsittelyn 2 g/ dm3:n annostuksella vuot alentuivat suodatuksen aikana 8 %-yksikköä vähemmän kuin ilman esikäsittelyä. Puukuitukäsittely stabiloi 0,1 g/dm3:n annostuksella saman kalvon vuota, kun kiintoainetta ei poistettu syötöstä. Hydrofobisen nanosuodatuskalvon vuon alenemista ehkäisivät puu- ja selluloosakuitukäsittelyt sekä karboksimetyyliselluloosakäsittely. Karboksimetyyliselluloosakäsittely vähensi vuon alenemista 25 %-yksikköä ja puukuitukäsittely 13 %-yksikköä. Hydrofiilisellä ultrasuodatuskalvolla vuon aleneminen oli pientä ilman esikäsittelyä. Reduktioihin esikäsittelyt vaikuttivat parhaiten ultrasuodatuskalvolla. Kitosaanikäsittely nosti 1 g/dm3:n annostuksella alumiinireduktion 50 %:sta 96 %:iin ja 5 g/dm3:n annostuksella rautareduktion 30 %:sta 55 %:iin. Karboksyylimetyyliselluloosakäsittelyt vaikuttivat parantavasti mangaanin, magnesiumin, raudan ja kalsiumin reduktioihin. Optimi karboksyylimetyyliselluloosa-annostus oli 2 g/dm3. Merkittävin reduktion nousu oli kalsiumilla, jonka reduktio nousi esikäsittelyllä 4 %:sta 57 %:iin. Reduktiota nostava mekanismi oli kalvon pinnalle muodostuva sekundaarikerros. Pesuaineista tehokkain oli entsyymiä sisältävä kalvonpesuaine. Suurin vaikutus sillä oli hydrofobisen nanosuodatuskalvon pesussa. Optimiannostuksella (0,5 %) kalvon vesivuo pesun jälkeen oli 114 % pesua edeltäneestä vesivuosta. Muut kaupalliset pesuaneet oli tehokkaita hydrofiilisille kalvoille. Peretikkahappo oli yksittäisenä pesuaineena heikkotehoinen.

This thesis focuses on fibre coalescers whose efficiency is based on the surface properties/characteristics. They have the ability to preferentially wet or interact with one or more of the fluids to be separated. Thus, the interfacial phenomena governing the separation efficiency of the coalescers is investigated depending on physical factors such as flowrates, phase ratios and coalescer packing density. Design of process equipment to produce and separate of the emulsions was carried out.The experimentation was carried out to test the separation efficiency of the coalescing medias, namely fibreglass, polyester I and polyester II. The performances of the coalescing medias were assessed via droplet size information. In conclusion, the objectives (design of process equipment and experimentation) were achieved. Fibre glass was the best coalescing media, next was polyester I and then finally polyester II. Droplets sizes increased with decreased flowrates and increased packing density of the coalescer. Phase ratio had effect on the droplet sizes of the feed but had no effect on the coalescence of droplets of the feed.

In many industries, such as petroleum production, and the petrochemical, metal, food and cosmetics industries, wastewaters containing an emulsion of oil in water are often produced. The emulsions consist of water (up to 90%), oils (mineral, animal, vegetable and synthetic), surfactants and other contaminates. In view of its toxic nature and its deleterious effects on the surrounding environment (soil, water) such wastewater needs to be treated before release into natural water ways. Membrane-based processes have successfully been applied in industrial applications and are considered as possible candidates for the treatment of oily wastewaters. Easy operation, lower cost, and in some cases, the ability to reduce contaminants below existing pollution limits are the main advantages of these systems. The main drawback of membranes is flux decline due tofouling and concentration polarisation. The complexity of oil-containing systems demands complementary studies on issues related to the mitigation of fouling and concentration polarisation in membranebased ultrafiltration. In this thesis the effect of different operating conditions (factors) on ultrafiltration of oily water is studied. Important factors are normally correlated and, therefore, their effect should be studied simultaneously. This work uses a novel approach to study different operating conditions, like pressure, flow velocity, and temperature, and solution properties, like oil concentration (cutting oil, diesel, kerosene), pH, and salt concentration (CaCl2 and NaCl)) in the ultrafiltration of oily water, simultaneously and in a systematic way using an experimental design approach. A hypothesis is developed to describe the interaction between the oil drops, salt and the membrane surface. The optimum conditions for ultrafiltration and the contribution of each factor in the ultrafiltration of oily water are evaluated. It is found that the effect on permeate flux of the various factors studied strongly depended on the type of oil, the type of membrane and the amount of salts. The thesis demonstrates that a system containing oil is very complex, and that fouling and flux decline can be observed even at very low pressures. This means that only the weak form of the critical flux exists for such systems. The cleaning of the fouled membranes and the influence of different parameters (flow velocity, temperature, time, pressure, and chemical concentration (SDS, NaOH)) were evaluated in this study. It was observed that fouling, and consequently cleaning, behaved differently for the studied membranes. Of the membranes studied, the membrane with the lowest propensity for fouling and the most easily cleaned was the regenerated cellulose membrane (C100H). In order to get more information about the interaction between the membrane and the components of the emulsion, a streaming potential study was performed on the membrane. The experiments were carried out at different pH and oil concentration. It was seen that oily water changed the surface charge of the membrane significantly. The surface charge and the streaming potential during different stages of filtration were measured and analysed being a new method for fouling of oil in this thesis. The surface charge varied in different stages of filtration. It was found that the surface charge of a cleaned membrane was not the same as initially; however, the permeability was equal to that of a virgin membrane. The effect of filtration mode was studied by performing the filtration in both cross-flow and deadend mode. The effect of salt on performance was considered in both studies. It was found that salt decreased the permeate flux even at low concentration. To test the effect of hydrophilicity change, the commercial membranes used in this thesis were modified by grafting (PNIPAAm) on their surfaces. A new technique (corona treatment) was used for this modification. The effect of modification on permeate flux and retention was evaluated. The modified membranes changed their pore size around 33oC resulting in different retention and permeability. The obtained results in this thesis can be applied to optimise the operation of a membrane plant under normal or shock conditions or to modify the process such that it becomes more efficient or effective.

In the theory part the membrane emulsification was studied. Emulsions are used in many industrial areas. Traditionally emulsions are prepared by using high shear in rotor-stator systems or in high pressure homogenizer systems. In membrane emulsification two immiscible liquids are mixed by pressuring one liquid through the membrane into the other liquid. With this technique energy could be saved, more homogeneous droplets could be formed and the amount of surfactant could be decreased. Ziegler-Natta and single-site catalysts are used in olefin polymerization processes. Nowadays, these catalysts are prepared according to traditional mixing emulsification. More homogeneous catalyst particles that have narrower particle size distribution might be prepared with membrane emulsification. The aim of the experimental part was to examine the possibility to prepare single site polypropylene catalyst using membrane emulsification technique. Different membrane materials and solidification techniques of the emulsion were examined. Also the toluene-PFC phase diagram was successfully measured during this thesis work. This phase diagram was used for process optimization. The polytetrafluoroethylene membranes had the largest contact angles with toluene and also the biggest difference between the contact angles measured with PFC and toluene. Despite of the contact angle measurement results no significant difference was noticed between particles prepared using PTFE membrane or metal sinter. The particle size distributions of catalyst prepared in these tests were quite wide. This would probably be fixed by using a membrane with a more homogeneous pore size distribution. It is also possible that the solidification rate has an effect on the particle sizes and particle morphology. When polymeric membranes are compared PTFE is probably still the best material for the process as it had the best chemical durability.

Tässä diplomityössä tutkittiin alkoholilla ja orgaanisella hapolla tehtävien esikäsittelyiden vaikutusta nanosuodatuskalvon ominaisuuksiin. Työn tarkoituksena oli parantaa nanosuodatuskalvon fraktiointiominaisuuksia sekä kasvattaa monosakkaridien suotautuvuutta. Tarkasteluissa käytetty membraani oli GE Osmonicsin valmistamaa Desal-5 DL nanosuodatuskalvoa, jota modifioitiin erilaisilla maitohappo- ja isopropanoliesikäsittelyillä. Suodatukset tehtiin kahdella erilaisella laboratoriomittakaavan levysuotimella käyttäen malliaineina väkevää sokeriliuosta sekä laimeampaa sokeri-suola-liuosta. Happo- ja alkoholiesikäsiteltyjen kalvojen vuo- ja retentioarvoja verrattiin referensseinä käytettyjen vesiliotettujen kalvojen vastaaviin arvoihin. Puhtaat esikäsitellyt kalvot analysoitiin myös tarkemmin kalvoissa tapahtuneiden muutosten ymmärtämiseksi. Suodatusten ja analyysitulosten perusteella sekä alkoholi- että happoesikäsittelyt paransivat nanosuodatuskalvon ominaisuuksia parantaen sokerivuota, heikentämättä kuitenkaan kalvon fraktiointikykyä eri moolimassan omaavien sokereiden suhteen. Hapolla saavutettiin hieman alkoholikäsittelyä suotuisammat muutokset, mutta molemmilla käsittelyillä haluttujen komponenttien retentio monesti jopa parani referenssikalvoon verrattuna. Havaitut muutokset kalvoissa olivat pääosin fysikaalisia, mutta erityisesti happokäsittely muutti kalvon rakennetta myös kemiallisesti. Molemmat käsittelyt lisäsivät myös tarkasteltavan membraanin hydrofiilisuutta.

Tässä kandidaatintyössä tutkittiin, voidaanko puuhydrolysaatin esikäsittelyllä vähentää sen ultrasuodatuksen aikana esiintyvää kalvojen foulaantumista. Kokeellinen osa rajattiin koskemaan hydrolysaatin käsittelyä lakkaasientsyymillä, flokkulanteilla, kitosaanilla sekä adsorbentin ja lakkaasientsyymin yhdistelmällä. Esikäsittelyjen tavoitteena oli poistaa tai hajottaa hydrolysaatin sisältämää ligniiniä. Käsittelyjen teho testattiin suodattamalla eri tavoin käsiteltyä hydrolysaattia laboratoriokokeissa. Kokeissa adsorbentin ja lakkaasientsyymin yhdistelmäkäsittely osoittautui parhaaksi. Suodatuskapasiteetti parani ja foulaantuminen väheni verrattuna esikäsittelemättömän hydrolysaatin suodatukseen. Ligniiniä poistui käsittelyssä, kuten tavoitteena oli. Suodatuksessa kuitenkin edelleen tapahtui foulaantumista ja esikäsittelyn tehoa tulisi siksi vielä parantaa. Myös esikäsittely lakkaasientsyymillä paransi suodatuskapasiteettia ja vähensi foulaantumista, mutta vähemmän kuin entsyymi-adsorbenttikäsittely.

Wood contains only a very small amount of lipophilic extractives, commonly known as wood pitch. The pitch is known to cause severe problems in papermaking processes. The amount of pitch in process waters can be decreased by seasoning of the raw material prior to pulping, pulp washing, removal of pitch by flotation, adsorption of pitch onto various mineral surfaces, and retention of pitch to the fibre material by cationic polymers. The aim of this study was to determine the influence of pH on some of the methods used for pitch control. Experiments were performed using laboratory-made wood pitch emulsions with varying pH, salt concentration, hemicellulose concentration and pitch composition. These emulsions were used to study the phase distribution of resin and fatty acids, the colloidal stability of pitch with and without steric stabilisation by galactoglucomannans, and the interactions between wood pitch and mineral particles. Purification of unbleached and peroxidebleached mill process water was performed by froth flotation in combination with a foaming agent. The distribution of resin and fatty acids (RFAs) between colloidal pitch droplets and the water phase was very dependent on pH. At pH 3, almost all of the RFAs were attached to the pitch droplets, while increasing the pH led to increasing concentration of dissolved RFAs in the water phase. The presence of salt shifted the release of RFAs towards higher pH, while lower ratio of neutral pitch in the emulsion resulted in release of RFAs at lower pH. It was also seen that the dissolution and adsorption of RFAs at sudden pHchanges takes place very quickly. Colloidal pitch was more stable against electrolyte-induced aggregation at higher pH, due to its higher anionic charge. The concentration of cationic polymers needed to aggregate colloidal pitch also increased with increasing pH. The surface characteristics of solid particles, such as amount of charged groups, were very important for understanding their interactions with colloidal wood pitch. Water-soluble galactoglucomannans stabilised the colloidal pitch sterically against aggregation, but could not completely prevent interactions between wood pitch and hydrophilic particles. Froth flotation of unbleached and peroxidebleached process water showed that the pitch could be removed more effectively and selectively at low pH, compared to at neutral pH. The pitch was removed more effectively, using lower concentrations of foaming agent, from peroxide-bleached water than from unbleached water. The results show that pH has a major impact on various pulping and papermaking processes. It determines the anionic charge of the colloidal pitch and the solubility of certain pitch components. Because of this, the pH influences the effectiveness of pitch retention and removal of pitch. The results indicate that pitch problems could be diminished by acknowledging the importance of pH in various papermaking processes.

The present study introduce two pretreatment technologies which are torrefaction and steam explosion, and compare energy balance for both technologies to investigate and compare the use of these technologies to improve pelletization. In this research, torrefaction and steam explosion pretreatments were accomplished on the mixed small diameter wood (70%) with moisture content of 40 %, and logging residues (30%) with moisture content of 45 % at temperature 230 ̊C, and treatment duration 10 min. Competing methods were evaluated, and the results showed higher volumetric energy for steam explosion pellet than torrefied pellet.

In recent years, there have been studies that show a correlation between the hyperactivity of children and use of artificial food additives, including colorants. This has, in part, led to preference of natural products over products with artificial additives. Consumers have also become more aware of health issues. Natural food colorants have many bioactive functions, mainly vitamin A activity of carotenoids and antioxidativity, and therefore they could be more easily accepted by the consumers. However, natural colorant compounds are usually unstable, which restricts their usage. Microencapsulation could be one way to enhance the stability of natural colorant compounds and thus enable better usage for them as food colorants. Microencapsulation is a term used for processes in which the active material is totally enveloped in a coating or capsule, and thus it is separated and protected from the surrounding environment. In addition to protection by the capsule, microencapsulation can also be used to modify solubility and other properties of the encapsulated material, for example, to incorporate fat-soluble compounds into aqueous matrices. The aim of this thesis work was to study the stability of two natural pigments, lutein (carotenoid) and betanin (betalain), and to determine possible ways to enhance their stability with different microencapsulation techniques. Another aim was the extraction of pigments without the use of organic solvents and the development of previously used extraction methods. Stability of pigments in microencapsulated pigment preparations and model foods containing these were studied by measuring the pigment content after storage in different conditions. Preliminary studies on the bioavailability of microencapsulated pigments and sensory evaluation for consumer acceptance of model foods containing microencapsulated pigments were also carried out. Enzyme-assisted oil extraction was used to extract lutein from marigold (Tagetes erecta) flower without organic solvents, and the yield was comparable to solvent extraction of lutein from the same flowers. The effects of temperature, extraction time, and beet:water ratio on extraction efficiency of betanin from red beet (Beta vulgaris) were studied and the optimal conditions for maximum yield and maximum betanin concentration were determined. In both cases, extraction at 40 °C was better than extraction at 80 °C and the extraction for five minutes was as efficient as 15 or 30 minutes. For maximum betanin yield, the beet:water ratio of 1:2 was better, with possibly repeated extraction, but for maximum betanin concentration, a ratio of 1:1 was better. Lutein was incorporated into oil-in-water (o/w) emulsions with a polar oil fraction from oat (Avena sativa) as an emulsifier and mixtures of guar gum and xanthan gum or locust bean gum and xanthan gum as stabilizers to retard creaming. The stability of lutein in these emulsions was quite good, with 77 to 91 percent of lutein being left after storage in the dark at 20 to 22°C for 10 weeks whereas in spray dried emulsions the retention of lutein was 67 to 75 percent. The retention of lutein in oil was also good at 85 percent. Betanin was incorporated into the inner w1 water phase of a water1-in-oil-inwater2 (w1/o/w2) double emulsion with primary w1/o emulsion droplet size of 0.34 μm and secondary w1/o/w2 emulsion droplet size of 5.5 μm and encapsulation efficiency of betanin of 89 percent. In vitro intestinal lipid digestion was performed on the double emulsion, and during the first two hours, coalescence of the inner water phase droplets was observed, and the sizes of the double emulsion droplets increased quickly because of aggregation. This period also corresponded to gradual release of betanin, with a final release of 35 percent. The double emulsion structure was retained throughout the three-hour experiment. Betanin was also spray dried and incorporated into model juices with different pH and dry matter content. Model juices were stored in the dark at -20, 4, 20–24 or 60 °C (accelerated test) for several months. Betanin degraded quite rapidly in all of the samples and higher temperature and a lower pH accelerated degradation. Stability of betanin was much better in the spray dried powder, with practically no degradation during six months of storage in the dark at 20 to 24 °C and good stability also for six months in the dark at 60 °C with 60 percent retention. Consumer acceptance of model juices colored with spray dried betanin was compared with similar model juices colored with anthocyanins or beet extract. Consumers preferred beet extract and anthocyanin colored model juices over juices colored with spray dried betanin. However, spray dried betanin did not impart any off-odors or off-flavors into the model juices contrary to the beet extract. In conclusion, this thesis describes novel solvent-free extraction and encapsulation processes for lutein and betanin from plant sources. Lutein showed good stability in oil and in o/w emulsions, but slightly inferior in spray dried emulsions. In vitro intestinal lipid digestion showed a good stability of w1/o/w2 double emulsion and quite high retention of betanin during digestion. Consumer acceptance of model juices colored with spray dried betanin was not as good as model juices colored with anthocyanins, but addition of betanin to real berry juice could produce better results with mixture of added betanin and natural berry anthocyanins could produce a more acceptable color. Overall, further studies are needed to obtain natural colorants with good stability for the use in food products.

Koalesenssi on ilmiö, jossa dispergoidun faasin pisarat pyrkivät muodostamaan suurempia pisaroita kunnes erotettava faasi muodostuu. Koalesenssi tapahtuu kolmessa päävaiheessa, jotka ovat lähestyminen, kiinnittyminen ja irrotus. Lähestymiseen vaikuttavat mekanismit ovat muuan muassa sieppaus, diffuusio, törmäysvaikutus, sedimentaatio, sähköiset repul-siovoimat ja van der Waalsin voimat. Kiinnittymisvaiheessa dispergoidun faasin pisarat syrjäyttävät väliaineen nestekalvon samalla kostuttaen väliaineen pinnan. Irrotusvaiheessa pisaran hydrodynaaminen voima voittaa pisaran ja väliaineen välisen adheesiovoiman. Koalesenssin tehokkuuteen vaikuttavat useat eri parametrit kuten virtausnopeus, pedin ominaisuudet, väliaineen ominaisuudet sekä emulsion ominaisuudet. Nämä kaikki asiat tulee ottaa huomioon koalesenssisuodatuksen suunnittelussa. Koalesenssisuodatus lukeutuu syväsuodatusmenetelmiin, jotka on ollut käytössä jo yli 100 vuotta. Koalesenssisuodatusmenetelmä on tehokas menetelmä pienten pisaroiden erottami-seen. Menetelmää käytetään esimerkiksi öljyisten jätevesien puhdistuksessa. Teollisen öljyn syväsuodatuksen etuihin kuuluu muun muassa sen kompakti koko, alhaisemmat käyt-tökustannukset, korkea erotusaste, kyky erotella pienetkin pisarat sekä helppo operointi, automatisointi ja huolto. Suurin haittapuoli on kuitenkin väliaineen tukkeutuminen, joten prosessi vaatii puhdistuksen tai väliaineen uusimisen. Tämän kandidaatintyön tarkoituksena oli koota kirjallisuustyö öljyn koalesenssisuodatuk-sesta. Työssä kartoitettiin koalesenssisuodatuksen lähtökohdat, teoria, tärkeimmät teolli-suuden sovellukset sekä väliaineet.

Tämän kandidaatintyön tarkoituksena oli perehtyä bioetanolin eri valmistusvaiheisiin ja sen valmistuksessa käytettäviin raaka-aineisiin sekä myöskin tutkia eri esikäsittelymenetelmien vaikutusta sahajauhojen jauhautumiseen sekä energiankulutukseen, kun niitä jauhetaan vasaramyllyllä. Kirjallisuusosassa keskityttiin paljon aineksen hienonnukseen, sillä varsinkin biomassat tulee hienontaa, jotta halutut kemialliset reaktiot saataisiin tapahtumaan riittävän tehokkaasti. Kokeellisessa osassa tutkittiin eripituisien vesiliotuksien sekä emäs-käsittelyn vaikutusta sahajauhon jauhautumiseen sekä jauhautumisen energiankulutukseen, kun niitä jauhettiin vasaramyllyllä. Tutkimuskysymyksinä tässä työssä oli: Miten vesiliotus sekä liotus emäksisessä liuoksessa vaikuttaa eri sahajauhojen jauhautumiseen vasaramyllyllä?, Millaisia vaikutuksia esikäsittelyillä on myllyn energiankulutukseen?, Miten esikäsittelyt vaikuttavat jauhautumistuotteiden partikkelikokoihin?. Tuloksista käy ilmi, että 1. ja 3. tunnin vesiliotukset eivät olleet riittäviä sillä mylly ei jaksanut jauhaa minkään puulaadun sahajauhoja näillä ajoilla. Yleisesti tuloksista huomattiin, että haapajauhon jauhamiseen kului kaikkein eniten energiaa, riippumatta esikäsittelytekniikasta, mutta haapa myös jauhautui parhaiten pH-esikäsittelyn tuloksena.

Hemicelluloses are potential raw material for several items produced in future wood-based biorefineries. One possible method for recovering hemicelluloses from wood extracts is ultrafiltration (UF). However, low filtration capacities and severe fouling restrict the use of tight UF membranes in the treatment of wood extracts. The lack of suitable commercial membranes creates a need for pretreatment which would decrease fouling and increase the filtration capacity. This thesis focuses on the evaluation of the possibility to improve the filtration capacity and decrease fouling with the pretreatment of wood extracts. Methods which remove harmful compounds and methods which degrade them are studied, as well as combinations of the methods. The tested pretreatments have an influence on both the concentration of different compounds and the molecular mass distribution of the compounds in the extract. This study revealed that in addition to which kind of compounds were removed, also the change in molecular size distribution affected the filtration capacity significantly. It was shown that the most harmful compounds for the filtration capacity of the hydrophobic 5 kDa membrane were the ones capable of permeating the membrane and fouling also the inner membrane structure. Naturally, the size of the most harmful compounds depends on the used UF membrane and is thus case-specific. However, in the choice of the pretreatment method, the focus should be on the removal of harmful compound sizes rather than merely on the total amount of removed foulants. The results proved that filtration capacity can be increased with both adsorptive and oxidative pretreatments even by hundreds of per cents. For instance, the use of XAD7 and XAD16 adsorbents increased the average flux in the UF of a birch extract from nearly zero to 107 kg/(m2h) and 175 kg/(m2h), respectively. In the treatment of a spruce extract, oxidation by pulsed corona discharge (PCD) increased the flux in UF from 46 kg/(m2h) to 158 kg/(m2h). Moreover, when a birch extract batch was treated with laccase enzyme, the flux in UF increased from 15 kg/(m2h) to 36 kg/(m2h). However, fouling was decreased only by adsorptive pretreatment while oxidative methods had a negligible or even negative impact on it. This demonstrates that filtration capacity and fouling are affected by different compounds and mechanisms. The results of this thesis show that filtration capacity can be improved and fouling decreased through appropriate pretreatment. However, the choice of the best possible pretreatment is case-specific and depends on the wood extract and the membrane used. Finding the best option requires information on the extract content and membrane characteristics as well as on the filtration performance of the membrane in the prevailing conditions and a multivariate approach. On the basis of this study, it can be roughly concluded that adsorptive pretreatment improves the filtration capacity and decreases fouling rather reliably, but it may lead to significant hemicellulose losses. Oxidation reduces the loss of valuable hemicelluloses and could improve the filtration capacity, but fouling challenges may remain. Combining oxidation with adsorptive pretreatment was not a solution for avoiding hemicellulose losses in the tested cases.
