Contrary to common belief, aesthetics had an important function in ritual forms implemented by Reformed Calvinist Churches. The impact of aesthetics on Reformed piety rested less on images, considered to be a source of distraction, than on music. By reconsidering the evolution of Calvin's thoughts on the relationship between music and religious services between 1536 and 1543, this study reveals how Calvin came to consider that, by singing psalms, Christians in their devotion could conciliate both a cognitive process guided by the meaning of the words and an affective response triggered by the tune. For Calvin, the spiritual elevation to which religious services should lead was to emerge from the conjunction of these two impetuses.

Wilhelm Zielys ca. 1511-21 in Bern entstandene Prosaromane ‛Olwier und Artus' und ‛Valentin und Orsus' nach französischer Vorlage sind bisher kaum erschlossen. Lediglich ihre Paratexte werden seit jeher registriert: Zielys Vorrede zum 1521 als Doppelausgabe erschienenen Erstdruck beider Romane sowie ein erläuterndes Schlusskapitel zu ‛Olwier und Artus' gelten als innovative Lösungsversuche des im 16. Jahrhundert virulenten Wahrheitsproblems der Literatur und als frühe Zeugnisse für die Entwicklung modernen Fiktionsbewusstseins. Der Beitrag analysiert zunächst in systematischem Zugriff Verbindungspunkte zur neuzeitlichen Literaturästhetik und zeigt auf, daß diese Verbindungen nicht historisch sein können. In einem zweiten, historischen Zugriff untersucht er Zielys Modell im Kontext seines Gesamtwerks und stellt es in den kulturgeschichtlichen Zusammenhang der Jahre um 1520 in der alten Eidgenossenschaft. Er arbeitet die enge Anbindung der poetologischen Positionierung Zielys an den glaubensgeschichtlichen Diskurs am Vorabend der Berner Reformation heraus und wirft neue Fragen zur Auseinandersetzung mit dem Fiktionsproblem im Prosaroman, zugleich auch nach dem literaturwissenschaftlichen Zugang zu vormoderner Fiktionalität, auf.

Quantitative information from magnetic resonance imaging (MRI) may substantiate clinical findings and provide additional insight into the mechanism of clinical interventions in therapeutic stroke trials. The PERFORM study is exploring the efficacy of terutroban versus aspirin for secondary prevention in patients with a history of ischemic stroke. We report on the design of an exploratory longitudinal MRI follow-up study that was performed in a subgroup of the PERFORM trial. An international multi-centre longitudinal follow-up MRI study was designed for different MR systems employing safety and efficacy readouts: new T2 lesions, new DWI lesions, whole brain volume change, hippocampal volume change, changes in tissue microstructure as depicted by mean diffusivity and fractional anisotropy, vessel patency on MR angiography, and the presence of and development of new microbleeds. A total of 1,056 patients (men and women ≥ 55 years) were included. The data analysis included 3D reformation, image registration of different contrasts, tissue segmentation, and automated lesion detection. This large international multi-centre study demonstrates how new MRI readouts can be used to provide key information on the evolution of cerebral tissue lesions and within the macrovasculature after atherothrombotic stroke in a large sample of patients.

The lipid raft proteins reggie-1 and -2 (flotillins) are implicated in membrane protein trafficking but exactly how has been elusive. We find that reggie-1 and -2 associate with the Rab11a, SNX4, and EHD1-decorated tubulovesicular recycling compartment in HeLa cells and that reggie-1 directly interacts with Rab11a and SNX4. Short hairpin RNA-mediated down-regulation of reggie-1 (and -2) in HeLa cells reduces association of Rab11a with tubular structures and impairs recycling of the transferrin-transferrin receptor (TfR) complex to the plasma membrane. Overexpression of constitutively active Rab11a rescues TfR recycling in reggie-deficient HeLa cells. Similarly, in a Ca(2+) switch assay in reggie-depleted A431 cells, internalized E-cadherin is not efficiently recycled to the plasma membrane upon Ca(2+) repletion. E-cadherin recycling is rescued, however, by overexpression of constitutively active Rab11a or SNX4 in reggie-deficient A431 cells. This suggests that the function of reggie-1 in sorting and recycling occurs in association with Rab11a and SNX4. Of interest, impaired recycling in reggie-deficient cells leads to de novo E-cadherin biosynthesis and cell contact reformation, showing that cells have ways to compensate the loss of reggies. Together our results identify reggie-1 as a regulator of the Rab11a/SNX4-controlled sorting and recycling pathway, which is, like reggies, evolutionarily conserved.
