The in situ nuclear matrix was obtained from HeLa cells. After permeabilization with nonionic detergent, the resulting structures were incubated for 1 h at 37 degrees C to determine whether or not such an incubation might result in the redistribution of nuclear polypeptides which resisted extraction with buffers of high-ionic strength (1.6 M NaCl or 0.25 M (NH4)2SO4 as well as DNase I digestion. Using indirect immunofluorescence experiments and monoclonal antibodies we show that heating to 37 degrees C changes the distribution of a 160 kDa protein previously shown to be a component of the inner matrix network. On the other hand, a 125 kDa polypeptide was not affected at all by the incubation. Our results clearly indicate that the inclusion of a 37 degrees C incubation (for example during digestion with DNase I) in the protocol to obtain the in situ nuclear matrix can result in the formation of in vitro artifacts.

Superantigens induce a vigorous immune response by stimulating T cells that express particular T-cell receptor V beta chains. Mouse mammary tumor virus is a milk-transmitted retrovirus that encodes such a superantigen. Paradoxically, as discussed by Werner Held and colleagues, the strong superantigen-induced immune response permits the survival of the virus via T-cell dependent clonal expansion of infected B cells.

Superantigens (SAgs) encoded by infectious mouse mammary tumor viruses (MMTVs) play a crucial role in the viral life cycle. Their expression by infected B cells induces a proliferative immune response by SAg-reactive T cells which amplifies MMTV infection. This response most likely ensures stable MMTV infection and transmission to the mammary gland. Since T cell reactivity to SAgs from endogenous Mtv loci depends on MHC class II molecules expressed by B cells, we have determined the ability of MMTV to infect various MHC congenic mice. We show that MHC class II I-E+ compared with I-E- mouse strains show higher levels of MMTV infection, most likely due to their ability to induce a vigorous SAg-dependent immune response following MMTV encounter. Inefficient infection is observed in MHC class II I-E- mice, which have been shown to present endogenous SAgs poorly. Therefore, during MMTV infection the differential ability of MHC class II molecules to form a functional complex with SAg determines the magnitude of the proliferative response of SAg-reactive T cells. This in turn influences the degree of T cell help provided to infected B cells and therefore the efficiency of amplification of MMTV infection.

Raman spectroscopy has been used by fluid inclusionists to: 1) identify and quantitatively determine the relative abundances of gaseous species within fluid inclusions; 2) identify solid phases precipitating from, or accidentally trapped, within fluid inclusions; and 3) determine the detection limits of the C-13/C-12 ratio in the CO2 bearing phase of fluid inclusions.

Spontaneous pneumothorax (PNO) is usually due to rupture of a small subpleural bleb into the pleural cavity and affects mainly young men. After simple drainage, recurrence occurs in about 50% of cases. The risk of recurrence increases after each new PNO. Secondary PNO complicates an underlying pulmonary disease, especially chronic obstructive pulmonary disease with emphysema. A new form of secondary PNO has emerged in the recent years in AIDS patients with pneumocystis carinii pneumonia. We have shifted to a thoracoscopic therapy of PNO since May 1991. 25 PNO in 24 patients (1 bilateral) have been treated since that time up to April 1993. 19 PNO were primary, whereas 6 were secondary, included 3 iatrogenic PNO. Resection of the leaking parenchymal area was performed in 20 patients, and parietal partial pleurectomy was done in 20 cases. In the remaining cases, fibrin glue was applied on the lesion and in 3 cases, chemical pleurodesis was attempted using silver nitrate or talc. 1 AIDS patient died of ARDS. 3 patients had recurrent PNO and had thoracotomy without complication. 21 patients did well. Partial PNO recurred in one of them 4 months later, and was treated by simple needle aspiration. Thoracoscopy is a useful method to treat recurrent or persistent spontaneous PNO. After only 25 cases, our success rate in primary PNO is 90%. There should be a learning curve. On the basis of our experience, we believe that recognition of the lesion and its resection as well as apical parietal pleurectomy are necessary to obtain good results and a low recurrence rate.

Karl Barth (1886-1968) est sans nul doute l'un des géants de la théologie chrétienne du XXe siècle, toutes confessions confondues. À l'instar de ses plus prestigieux modèles - un saint Augustin, un Luther notamment -, c'est dans le corps à corps avec les Écritures saintes, singulièrement avec l'épître de Paul aux Romains, qu'il s'est forgé une conception originale de la pensée chrétienne et qu'il a bouleversé la situation religieuse et théologique de son temps. « Révolutionnaire » dans sa manière de vouloir congédier les théologies libérales du XIXe siècle et de contester en son principe toute forme de complicité du christianisme avec la monde profane ou même avec la modernité, le jeune Barth, celui des années 20, s'est peu à peu transformé en un imposant professeur de dogmatique et en un auteur universellement admiré, étudié, discuté, critiqué. Sa monumentale « Kirchliche Dogmatik », publiée de 1932 à 1968, inachevée, est devenue une référence, aussi bien à l'intérieur du protestantisme que pour le catholicisme romain, comme l'a montré son influence croissante sur des auteurs comme Balthasar, Bouillard et Küng et sur un certain nombre de Pères conciliaires à Vatican II. Le propos de cet ouvrage est de rompre avec les clichés et les idées reçues qui entourent la plupart du temps l'oeuvre multiforme de Barth. Barth y apparaît en permanente recherche, ouvert aux enjeux de la culture et de la société civile, en dialogue critique, certes, mais également très réceptif avec les courants de la modernité et en prise avec l'actualité politique - contre la guerre en 1918, contre le nazisme en 1933-1945, contre la bombe atomique et l'impérialisme américain à la fin de sa vie. L'éthique - individuelle, mais aussi sociale et politique - joue une place centrale dans sa pensée, elle est loin de se réduire à un simple appendice de la dogmatique. Un survol très large de la réception internationale de Barth permet de mesurer l'écho intellectuel et oecuménique considérable de cette théologie en mouvement. Le choix de textes proposé à la fin de l'ouvrage mêle des passages devenus classiques à des aperçus dérangeants et détonnants de l'existence théologique, éthique, politique et culturelle de Barth.
