Neuropathic pain is a common form of chronic pain, and is unsuccessfully alleviated by usual medications. Mounting evidence strongly point at non-neuronal glial cells in the spinal cord as key actors behind the persistence of pain. In particular, a change in the astrocytic capacity to regulate extracellular concentrations of neurotransmitters might account for the strengthened spinal nociceptive neurotransmission. Therefore, we investigated whether spinal expressions of GABA (GAT) and glutamate (EAAT) transporters were affected in the spared nerve injury (SNI) rat model of neuropathic pain. SNI was induced in male Sprague-Dawley rats by a unilateral section of tibial and common peroneal branches of the sciatic nerve, leaving the sural branch untouched. Western-blot analysis was performed to study the expression of GAT-1 and GAT-3 as well as EAAT-1 and EAAT-2, the main astrocytic GABA and glutamate transporters respectively. Seven days post-surgery, a significant increase in GAT-1, GAT-3 and EAAT-1 expressions is detected in both ipsilateral and contralateral sides of lumbar spinal cord in comparison to sham animals. No change in EAAT-2 signal could be detected. Furthermore, the astrocytic reaction parallels the glutamate and GABA transporters changes as we found an increased GFAP expression compared to the sham condition, in both spinal sides. Together, our results indicate that modifications in GABA and glutamate transport may occur along with SNI-associated painful neuropathy and identify spinal neurotransmitter reuptake machinery as a putative pharmacological target in neuropathic pain.

Variables measured during static and dynamic pupillometry were factor-analyzed. Following factors were obtained regardless whether investigations were carried out in normals or in psychiatric patients: A static factor, a dynamic factor, a stimulus-specific factor and a restitution-dependent factor. Evaluation of reliability in normals demonstrated a high reliability for the static variables of pupillometry.

The complex relationship between structural and functional connectivity, as measured by noninvasive imaging of the human brain, poses many unresolved challenges and open questions. Here, we apply analytic measures of network communication to the structural connectivity of the human brain and explore the capacity of these measures to predict resting-state functional connectivity across three independently acquired datasets. We focus on the layout of shortest paths across the network and on two communication measures-search information and path transitivity-which account for how these paths are embedded in the rest of the network. Search information is an existing measure of information needed to access or trace shortest paths; we introduce path transitivity to measure the density of local detours along the shortest path. We find that both search information and path transitivity predict the strength of functional connectivity among both connected and unconnected node pairs. They do so at levels that match or significantly exceed path length measures, Euclidean distance, as well as computational models of neural dynamics. This capacity suggests that dynamic couplings due to interactions among neural elements in brain networks are substantially influenced by the broader network context adjacent to the shortest communication pathways.

Purpose: In Rd1 mice, a PDE6ß mutation is responsible for the rapid loss of photoreceptors. We observed re-expression of cell cycle proteins during early stages of retinal degeneration and the deletion of Bmi1 markedly delayed photoreceptor death in Rd1;Bmi1-/- mice. The present study characterizes the link between the expression of CDKs and the apoptotic process in Rd1 photoreceptors.Methods: CDK expression levels were evaluated by immunostaining of wild-type, Rd1 and Rd1;Bmi1-/- eye sections. The role of CDKs in retinal degeneration is currently being investigated by treating Rd1 retinal explants with CDK inhibitors, and by injecting roscovitine-containing micelles into the vitreous of P10 Rd1 mice.Results: We show that some Rd1 photoreceptors express CDK4 already at P9, and that the number of CDK4-positive cells increases more than 6-fold by P11. CDK2 and CDK6 are also expressed in the mutant outer nuclear layer (ONL), however to a lesser extent than CDK4. Concomitant with the expression of CDKs, the apoptotic process in Rd1 photoreceptors is detected by TUNEL staining. Co-localization analyses suggest that CDK expression precedes photoreceptor cell death since TUNEL-single-positive cells are rarely detected at P9, and double-positive as well as TUNEL- or CDK4-single-positive cells are all present in P11 Rd1 retinas. The wild-type ONL does not contain any TUNEL- or CDK4-positive cells. Interestingly, Bmi1 deletion downregulates CDK4 expression in P12 Rd1;Bmi1-/- retinas, and influences the accumulation of cGMP in Rd1 retinas. More cGMP is detected in the P11 Rd1;Bmi1-/- ONL than in the Rd1 ONL, while it is strongly reduced at P15. To better characterize the link between CDK expression and retinal degeneration, current experiments include the analysis of CDK inhibition in Rd1 retinal explants and in mouse eyes injected with roscovitine-containing micelles.Conclusions: The time-course of cell cycle protein expression may be related to early events of the apoptotic process in Rd1 photoreceptors. Moreover, the loss of Bmi1 seems to interfere with the first stages of retinal degeneration and to influence the expression of CDK4. Further experiments will determine whether the deletion of Bmi1 prevents cell death through a direct CDK inhibition.

Résumé Le trouble somatoforme douloureux persistant et le syndrome de fatigue chronique constituent des entités cliniques qui incarnent la position particulière du corps dans notre société actuelle. Tous les deux se situent dans un carrefour entre le psychique et le somatique, en interaction étroite avec l'entourage socioculturel, soulignant ainsi la nécessité de considérer l'être humain dans son intégralité. La société actuelle, pauvre en repères collectifs, où l'exaltation de l'individualité est un critère primordial et où le corps social réparateur n'existe pas, provoque une grand mal être. Le sentiment de ne pas être reconnu dans son intériorité est incorporé et après s'exprime ensuite sous la forme d'une plainte douloureuse. Le contexte de l'émigration et celui des systèmes de prévoyance sociale dans les pays occidentaux, sont des exemples de conditions qui favorisent le changement dans la dynamique de la maladie. En s'appuyant sur la représentation du corps dans un moment historique précis, l'homme cherche à exprimer une souffrance profonde dans le vécu de soi-même, qui n'a pas accès à d'autres voies d'expression. Le contexte social, culturel, médical, professionnel, lui facilite les moyens d'obtenir la reconnaissance de son affliction sans être exclu du groupe d'appartenance sociale. Les deux symptômes, douleur et fatigue, rassemblent les conditions nécessaires, dans la conception actuelle du corps, sociale et médicale, pour transporter le désir et la demande d'une partie de la société qui ne peut pas exprimer sa souffrance.

Retrouver le sens de la violence est une démarche certes essentielle, mais non suffisante. Il s'agit aussi d'aller au-delà du sens et de revoir sans cesse les réponses aux expressions nouvelles et inattendues des phénomènes violents, individuels ou collectifs.
