Here we present a 30 000 years low-resolution climate record reconstructed from groundwater data. The investigated site is located in the Bohemian Cretaceous Basin, in the corridor between the Scandinavian ice sheet and the Alpine ice field. Noble gas temperatures (NGT), obtained from groundwater data, preserved multicentennial temperature variability and indicated a cooling of at least 5-7 °C during the last glacial maximum (LGM). This is further confirmed by the depleted δ18O and δ2H values at the LGM. High excess air (ΔNe) at the end of the Pleistocene is possibly related to abrupt changes in recharge dynamics due to progression and retreat of ice covers and permafrost. These results agree with the fact that during the LGM permafrost and small glaciers developed in the inner valleys of the Giant Mountains (located in the watershed of the aquifers). A temporal decrease of deuterium excess from the pre-industrial Holocene to present days is linked to an increase of the air temperatures, and probably also to an increase of water pressure at the source region of precipitation over the past few hundred years

When colonizing a new habitat, populations must adapt their sexual behaviour to new ecological constraints. Because caves display drastically different conditions from surface habitats and cave animals are deprived from visual information, hypogean populations are expected to have modified their mate preference and signalling behaviour after cave colonization. Here, we experimentally examined the female preference and the sexual behaviour of brook newts Calotriton asper from different cave and river populations, either in light or in darkness. Our results suggest that females prefer large individuals in both hypogean and epigean populations, but that this preference is only expressed in the light conditions of their native habitat. Hence, some mate choice criteria would be maintained across genetically divergent populations and throughout dissimilar habitats. However, this sexual behaviour is likely to be expressed via a different sensory pathway in the different habitats, suggesting that a sensory shift has occurred in cave populations, enabling animals to communicate through a non-visual channel.

Active personal dosemeters (APD) have been found to be very efficient tools to reduce occupational doses in many applications of ionizing radiation. In order to be used in interventional radiology and cardiology (IR/IC), APDs should be able to measure low energy photons and pulsed radiation with relatively high instantaneous personal dose equivalent rates. A study concerning the optimization of the use of APDs in IR/IC was performed in the framework of the ORAMED project, a Collaborative Project (2008-2011) supported by the European Commission within its 7th Framework Program. In particular, eight commercial APDs were tested in continuous and pulsed X-ray fields delivered by calibration laboratories in order to evaluate their performances. Most of APDs provide a response in pulsed mode more or less affected by the personal dose equivalent rate, which means they could be used in routine monitoring provided that correction factors are introduced. These results emphasize the importance of adding tests in pulsed mode in type-test procedures for APDs. Some general recommendations are proposed in the end of this paper for the selection and use of APDs at IR/IC workplaces.

Introduction: Therapeutic drug monitoring (TDM) of imatinib has been increasingly proposed for chronic myeloid leukaemia (CML) patients, as several studies have found a correlation between trough concentrations (Cmin) >=1000ng/ml and improved response. The pharmacological monitoring project of EUTOS (European Treatment and Outcome Study) was launched to increase the availability of imatinib TDM, standardize labs, and validate proposed Cmin thresholds. Using the collected data, the objective of this analysis was to characterize imatinib Population pharmacokinetics (Pop-PK) in a large cohort of European patients, to quantify its variability and the influence of demographic factors and comedications, and to derive individual exposure variables suitable for further concentration-effect analyses.¦Methods: 4095 PK samples from 2478 adult patients were analyzed between 2006 and 2010 by LC-MS-MS and considered for Pop-PK analysis by NONMEM®. Model building used data from 973 patients with >=2 samples available (2590 samples). A sensitivity analysis was performed using all data. Available comedications (27%) were classified into inducers or inhibitors of P-glycoprotein, CYP3A4/5 and organic-cation-transporter-1 (hOCT-1).¦Results: A one-compartment model with linear elimination, zero-order absorption fitted the data best. Estimated Pop-PK parameters (interindividual variability, IIV %CV) for a 40-year old male patient were: clearance CL = 17.3 L/h (37.7%), volume V = 429L (51.1%), duration of absorption D1 = 3.2h. Outliers, reflecting potential compliance and time recording errors, were taken into account by estimating an IIV on the residual error (35.4%). Intra-individual residuals were 29.1% (proportional) plus ± 84.6 ng/mL (additive). Female patients had a 15.2% lower CL (14.6 L/h). A piece-wise linear effect of age estimated a CL of 18.7 L/h at 20 years, 17.3 L/h at 40 and 13.8 L/h at 60 years. These covariates explained 2% (CL) and 4.5% (V) of IIV variability. No effect of comedication was found. The sensitivity analysis expectedly estimated increased IIV, but similar fixed effect parameters.¦Conclusion: Imatinib PK was well described in a large cohort of CML patients under field conditions and results were concordant with previous studies. Patient characteristics explain only little IIV, confirming limited utility of prior dosage adjustment. As intra-variability is smaller than inter-patient variability, dose adjustment guided by TDM could however be beneficial in order to bring Cmin into a given therapeutic target.

Interferon-gamma release assays for the diagnosis of tuberculosis (TB) can give indeterminate results. The prevalence of indeterminate test results (ITRs) among T-SPOT.TB tests was assessed. A retrospective analysis of samples processed in 2005 was performed. ITRs were assessed by age, sex, immunosuppression, distance to the laboratory and season. A subgroup of tests performed for specific indications (contact tracing, migrants with positive tuberculin skin test, TB suspects and immunosuppression) were analysed separately. Of a total of 1,429 tests, 49 (3.4%) were indeterminate. ITRs were significantly associated with old age (>75 versus 5-75 yrs; odds ratio (OR) 7.97, 95% confidence interval (CI) 3.968-15.438) and the season during which samples were transported (autumn and winter versus spring and summer; OR 3.47, 95% CI 1.753-7.514). The incidence of ITR was 302 (2.0%) among TB contacts, 75 (1.6%) among immigrants, 156 (3.0%) in TB suspects and 32 (3.0%) among immunosuppressed patients. Sex, young age and distance to the laboratory were not associated with the rate of ITR. Of the 13 tests with ITR that were repeated, 10 gave a clear positive or negative result. Indeterminate test results with T-SPOT.TB under routine conditions were infrequent and more common in individuals aged >75 yrs than in children and younger adults. The incidence of indeterminate test results was low and similar among healthy tuberculosis contacts, immigrants with a positive tuberculin skin test, tuberculosis suspects and the immunosuppressed. The conditions of transportation may influence the incidence of indeterminate test results.

Résumé large public Le glucose est une source d'énergie essentielle pour notre organisme, indispensable pour le bon fonctionnement des cellules de notre corps. Les cellules β du pancréas sont chargées de réguler l'utilisation du glucose et de maintenir la glycémie (taux de glucose dans le sang) à un niveau constant. Lorsque la glycémie augmente, ces dernières sécrètent l'insuline, une hormone favorisant l'absorption, l'utilisation et le stockage du glucose. Une sécrétion insuffisante d'insuline provoque une élévation anormale du taux de glucose dans le sang (hyperglycémie) et peut mener au développement du diabète sucré. L'insuline est sécrétée dans le sang par un mécanisme particulier appelé exocytose. Une meilleure compréhension de ce mécanisme est nécessaire dans l'espoir de trouver des nouvelles thérapies pour traiter les 170 millions de personnes atteintes de diabète sucré à travers le monde. L'implication de diverses protéines, comme les SNAREs ou Rabs a déjà été démontrée. Cependant leurs mécanismes d'action restent, à ce jour, peu compris. De plus, l'adaptation de la machinerie d'exocytose à des conditions physiopathologiques, comme l'hyperglycémie, est encore à élucider. Le but de mon travail de thèse a été de clarifier le rôle de deux protéines, Noc2 et Tomosyn, dans l'exocytose ; puis de déterminer les effets d'une exposition prolongée à un taux élevé de glucose sur l'ensemble des protéines de la machinerie d'exocytose. Noc2 est un partenaire potentiel de deux Rabs connues pour leur implication dans les dernières étapes de l'exocytose, Rab3 et Rab27. Grâce à l'étude de différents mutants de Noc2, j'ai montré que l'interaction avec Rab27 permet à la protéine de s'associer avec les organelles de la cellule β contenant l'insuline. De plus, en diminuant sélectivement l'expression de Noc2, j'ai déterminé l'importance de cette protéine pour le bon fonctionnement du processus d'exocytose et le relâchement de l'insuline. Quant à Tomosyn, une protéine interagissant avec les protéines SNAREs, j'ai démontré son importance dans la sécrétion d'insuline en diminuant de manière sélective son expression dans les cellules β. Ensuite, grâce à une combinaison d'approches moléculaires et de microscopie, j'ai mis en évidence le rôle de Tomosyn dans les dernières étapes de l'exocytose. Enfin, puisque la sécrétion d'insuline est diminuée lors d'une hyperglycémie prolongée, j'ai analysé l'adaptation de la machinerie d'exocytose à ces conditions. Ceci m'a permis de découvrir que l'expression de quatre protéines essentielles pour le processus d'exocytose, Noc2, Rab3, Rab27 et Granuphilin, est fortement diminuée lors d'une hyperglycémie chronique. L'ensemble de ces données met en évidence l'importance de Noc2 et Tomosyn dans la sécrétion d'insuline. L'inhibition, par un taux élevé de glucose, de l'expression de Noc2 et d'autres protéines indispensables pour l'exocytose suggère que ce phénomène pourrait contribuer au développement du diabète sucré. Résumé L'exocytose d'insuline, en réponse au glucose circulant dans le sang, est la fonction principale de la cellule β. Celle-ci permet de stabiliser le taux de glucose sanguin (glycémie). Le diabète de type 2 est caractérisé par une glycémie élevée due, principalement, à un défaut de sécrétion d'insuline en réponse au glucose. La compréhension des mécanismes qui contrôlent l'exocytose d'insuline est essentielle pour clarifier les causes du diabète sucré. Plusieurs composants impliqués dans ce processus ont été identifiés. Ceux-ci incluent les SNAREs Syntaxin-1, VAMP2 et SNAP25 et les GTPases Rab3 et Rab27 qui jouent un rôle dans les dernières étapes de l'exocytose. Pendant mon travail de thèse, j'ai étudié le rôle de Noc2, un des partenaires de Rab3 et Rab27, dans l'exocytose d'insuline. Nous avons déterminé que Noc2 s'associe aux granules de sécrétion d'insuline grâce à son interaction avec Rab27. La diminution de l'expression de Noc2 dans la lignée cellulaire β INS-1E, par ARN interférence, influence négativement la sécrétion d'insuline stimulée par différents sécrétagogues et prouve que cette protéine Noc2 est essentielle pour l'exocytose d'insuline. L'interaction avec Munc13, une protéine impliquée dans l'arrimage des vésicules, suggère que Noc2 participe au recrutement des granules d'insuline à la membrane plasmique. Ensuite, j'ai analysé l'adaptation de la machinerie d'exocytose à des concentrations supraphysiologiques de glucose. Le niveau d'expression de Rab3 et Rab27 et de leurs effecteurs Granuphilin/S1p4 et Noc2 est fortement diminué par une exposition prolongée des cellules β à haut glucose. L'effet observé est en relation avec l'induction de l'expression de ICER, un facteur de transcription surexprimé dans des conditions d'hyperglycémie et également dans des modèles génétiques de diabète de type 2. La surexpression de ICER dans des cellules INS-1E diminue l'expression de Rab3, Rab27, Granuphilin/Slp4 et Noc2 et par conséquent l'exocytose d'insuline. Ainsi, l'induction de ICER, après une exposition prolongée à haut glucose, régule négativement l'expression de protéines essentielles pour l'exocytose et altère la sécrétion d'insuline. Ce mécanisme pourrait contribuer au dysfonctionnement de l'exocytose d'insuline dans le diabète de type 2. Dans la dernière partie de ma thèse, j'ai investigué le rôle de la protéine Tomosyn-1 dans la formation du complexe SNARE. Cette protéine a une forte affinité pour Syntaxin-1 et contient un domaine SNARE. Tomosyn-1 est concentrée dans les régions cellulaires enrichies en granules de sécrétion. La diminution sélective de l'expression de Tomosyn-1 induit une réduction de l'exocytose stimulée par différents sécrétagogues. Cet effet est dû à un défaut de fusion des granules avec la membrane plasmique. Ceci nous indique que Tomosyn-1 intervient dans une phase importante de la préparation des vésicules à la fusion, qui est nécessaire à l'exocytose. Abstract: Insulin exocytosis from pancreatic β-cells plays a central role in blood glucose homeostasis. Diabetes mellitus is a complex metabolic disorder characterized by secretory dysfunctions in pancreatic β-cells and release of amounts of insulin that are inappropriate to maintain blood glucose concentration within normal physiological ranges. To define the causes of β-cell failure a basic understanding of the molecular mechanisms that control insulin exocytosis is essential. Some of the molecular components involved in this process have been identified, including the SNARE proteins VAMP2, Syntaxin-1 and SNAP25 and the two GTPases, Rab3 and Rab27, that regulate the final steps of insulin secretion. I first investigated the role of Noc2, a potential Rab3 and Rab27 partner, in insulin secretion. I found that Noc2 associates with Rab27 and is recruited by this GTPase on insulin- containing granules. Silencing of the Noc2 gene by RNA interference led to a strong impairment in the capacity of the β-cell line INS-1E to respond to secretagogues, indicating that appropriate levels of the protein are essential for insulin exocytosis. I also showed that Noc2 interacts with Munc13, a protein that controls vesicle priming, suggesting a possible involvement of Noc2 in the recruitment of secretory granules at the plasma membrane. In the second part of my thesis, I investigated the adaptation of the molecular machinery of exocytosis to physiopathological conditions. I found that the expression of Rab3, Rab27 and of their effectors Granuphilin/Slp4 and Noc2 is dramatically decreased by chronic exposure of β-ce1ls to supraphysiological glucose levels. The observed glucotoxic effect is a consequence of the induction of ICER, a transcriptional repressor that is increased by prolonged hyperglycemia and in genetic models of type 2 diabetes. Overexpression of ICER reduced Granuphilin, Noc2, Rab3 and Rab27 levels and inhibited exocytosis. These results suggest that the presence of inappropriate levels of ICER diminishes the expression of a group of proteins essential for exocytosis and contributes to defective insulin release in type 2 diabetes. In the last part of my thesis, I focused my attention on the role of Tomosyn-1, a Syntaxin-1 binding protein possessing a SNARE-like motif, in the control of SNARE complex assembly. I found that Tomosyn-1 is concentrated in cellular compartments enriched in insulin-containing secretory granules. Silencing of Tomosyn-1 did not affect the number of secretory granules docked at the plasma membrane but decreased their release probability, resulting in a reduction in stimulus-induced insulin exocytosis. These findings suggest that Tomosyn-1 is involved in a post-docking event that prepares secretory granules for fusion and is necessary to sustain exocytosis in response to insulin secretagogues.

Identifying the factors that mediate covariation between an ornament and other phenotypic attributes is important to determine the signaling function of ornaments. Sign and magnitude of a covariation may vary across environments if the expression of the ornament or of its linked genes regulating correlated phenotypes is condition-dependent. I investigated in the barn owl Tyto alba whether sign and magnitude of covariation between body mass and two heritable melanin-based plumage ornaments change with food supply, along the reproductive cycle and from the morning to the evening. Using a dataset of 1,848 measurements of body mass in 336 breeding females, I found that females displaying large black spots were heavier than conspecifics with smaller spots in the afternoon (i.e., a long time after the last feeding) but not in the morning (i.e., a short time after the last feeding). This is consistent with the recently proposed hypothesis that eumelanin-based ornaments are associated with the ability to maintain energy balance between food intake and energy expenditure. Thus, covariation between melanin-based coloration and body mass can be detected only under specific conditions potentially explaining why it has been reported in only ten out of 28 vertebrate species. The proposition that ornamented individuals achieve a higher fitness than drab conspecifics only in specific environments should be tested for other ornaments.

Isotopic and trace element compositions of Miocene and Pliocene phosphatic brachiopods (Lingulidae and Discinidae) from southern North Sea, the Central Paratethys and the Atlantic coast of Europe were investigated in order to trace past environmental conditions and marine connections between the northern boreal and the southern subtropical-tropical marine basins. The North Sea genus Glottidia yielded low epsilon(Nd) and high delta O-18(PO4) values through the Mio-Pliocene indicating cold habitat temperature where the local seawater was dominated by the Atlantic Ocean. In contrast, the Middle Miocene Lingulidae and Discinidae of the Paratethys inhabited warm subtropical seawater with the possible influence of the Indian Ocean via the Mediterranean, as supported by their average epsilon(Nd) value of -8.3. The combined geochemical data support a thermal and marine separation of the Paratethys from the North Sea with no direct connection or major exchange of water from the Miocene onwards. The temperature in the Paratethys was very similar to that inferred from brachiopods from the Middle Miocene of western France, but the seawater epsilon(Nd) value here is identical to that of contemporaneous Atlantic Ocean. A Late Miocene lingulid brachiopod from southern Portugal has a high delta O-18(PO4), similar to the specimens investigated from the North Sea, reflecting either a deep water habitat or formation after the onset of major global cooling that resulted in an increased delta O-18 value of seawater. The epsilon(Nd) value of -8.4 for this site is compatible with an influence of Mediterranean outflow. (C) 2012 Elsevier B.V. All rights reserved.

Objectif Le présent travail a consisté à identifier et comparer, au moyen d'un questionnaire auto-administré, les conditions ergonomiques, les nuisances environnementales et les facteurs de stress rencontrés par le personnel soignant (n = 35) et le personnel pénitentiaire (n = 240) travaillant dans les mêmes prisons. Quatre ans ont séparé la remise du questionnaire auprès des surveillants et des soignants, ce qui limite la portée des résultats. Malgré cela, la comparaison des données fournies montre des tendances intéressantes. Résultats Sur un plan ergonomique, les soignants rencontrent une plus faible pénibilité que les surveillants. Sur un plan environnemental, les soignants sont gênés par un nombre moins important de nuisances que les surveillants, ces derniers étant exposés à des conditions très diverses de travail (divisions de haute sécurité, ateliers de production, domaines agricoles). Par contre, concernant les facteurs de stress, les soignants éprouvent des tensions dans des proportions presque identiques aux surveillants, suite aux interventions d'urgences, à la surcharge de travail, aux relations avec les détenus et à la mauvaise ambiance au travail. En outre, les soignants et surveillants paraissent souffrir différemment d'éléments d'organisation tels que le sentiment de travailler sous pression, le manque d'écoute et le manque de considération. Enfin les soignants signalent un risque de violence plus élevé ainsi qu'un manque de reconnaissance plus important que les surveillants. Conclusion Les surveillants doivent continuer à faire l'objet d'un suivi attentif des services de santé au travail. En ce qui concerne les soignants, l'accent doit être mis sur l'amélioration de leur organisation interne de travail en approfondissant, entre autres, la question du manque de reconnaissance, le sentiment de travail sous pression, ainsi qu'en poursuivant les efforts sur la formation à la lutte contre la violence. OBJECTIVE: To compare the heart-rate monitoring with the doubly labelled water (2H2(18)O) method to estimate total daily energy expenditure in obese and non-obese children. DESIGN: Cross sectional study of obese and normal weight children. SUBJECTS: 13 prepubertal children: six obese (4M, 2F, 9.1 +/- 1.5 years, 47.3 +/- 9.7 kg) and seven non-obese (3M, 4F, 9.3 +/- 0.6 years, 31.8 +/- 3.2 kg). MEASUREMENTS: Total daily energy expenditure was assessed by means of the doubly labelled water method (TEEDLW) and of heart-rate monitoring (TEEHR). RESULTS: TEEHR was significantly (P < 0.05) higher than TEEDLW in obese children (9.47 +/- 0.84 MJ/d vs 8.99 +/- 0.63 MJ/d) whereas it was not different in non-obese children (8.43 +/- 2.02 MJ/d vs 8.42 +/- 2.30 MJ/d, P = NS). The difference of TEE assessed by HR monitoring in the obese group averaged 6.2 +/- 4.7%. At the individual level, the degree of agreement (difference between TEEHR and TEEDLW +/- 2s.d.) was low both in obese (-0.36, 1.32 MJ/d) and in non-obese children (-1.30, 1.34 MJ/d). At the group level, the agreement between the two methods was good in nonobese children (95% c.i. for the bias:-0.59, 0.63 MJ/d) but not in obese children (0.04, 0.92 MJ/d). Duration of sleep and energy expenditure during resting and physical activity were not significantly different in the two groups. Patterns of heart-rate (or derived energy expenditure) during the day-time were similar in obese and non-obese children. CONCLUSION: The HR monitoring technique provides an estimation of TEE close to that assessed by the DLW method in non-obese prepubertal children. In comparison with DLW, the HR monitoring method yields a greater TEE value in obese children.

The conditions for the analysis of selected doping substances by UHPSFC-MS/MS were optimized to ensure suitable peak shapes and maximized MS responses. A representative mixture of 31 acidic and basic doping agents was analyzed, in both ESI+ and ESI- modes. The best compromise for all compounds in terms of MS sensitivity and chromatographic performance was obtained when adding 2% water and 10mM ammonium formate in the CO2/MeOH mobile phase. Beside mobile phase, the nature of the make-up solvent added for interfacing UHPSFC with MS was also evaluated. Ethanol was found to be the best candidate as it was able to compensate for the negative effect of 2% water addition in ESI- mode and provided a suitable MS response for all doping agents. Sensitivity of the optimized UHPSFC-MS/MS method was finally assessed and compared to the results obtained in conventional UHPLC-MS/MS. Sensitivity was improved by 5-100-fold in UHPSFC-MS/MS vs. UHPLC-MS/MS for 56% of compounds, while only one compound (bumetanide) offered a significantly higher MS response (4-fold) under UHPLC-MS/MS conditions. In the second paper of this series, the optimal conditions for UHPSFC-MS/MS analysis will be employed to screen >100 doping agents in urine matrix and results will be compared to those obtained by conventional UHPLC-MS/MS.
