Because environmental conditions within a given basin are different for each season and at different water depth, knowledge of the life history and depth distribution of target species is important for environmental and palaeoenvironmental interpretations based on ostracod species assemblages and/or the geochemical compositions of their valves. In order to determine the distribution of species with depth as well as the life history of species from Lake Geneva, a one year sampling campaign of living ostracods was conducted at five sites (2, 5, 13, 33 and 70 m water depth) on a monthly basis in the Petit-Lac (western basin of Lake Geneva, Switzerland). Based on the results, the different species can be classified into three groups. Littoral taxa are found at 2 and 5 m water depth and include, in decreasing numbers of individuals, Cypridopsis vidua (O. F.Müller, 1776), Pseudocandona compressa (Koch, 1838), Limnocythere inopinata (Baird, 1843), Herpetocypris reptans (Baird, 1835), Potamocypris smaragdina (Vávra, 1891), Potamocypris similis (G. W. Müller, 1912), Plesiocypridopsis newtoni (Brady & Robertson, 1870), Prionocypris zenkeri (Chyzer & Toth, 1858) and Ilyocypris sp. Brady & Norman, 1889. Sublittoral species are found in a majority at 13 m water depth and to a lesser extend at 33 m water depth and include, in decreasing numbers of individuals, Fabaeformiscandona caudata (Kaufmann, 1900), Limnocytherina sanctipatricii, Candona candida (O. F. Müller, 1776) and Isocypris beauchampi (Paris, 1920). Profundal species are found equally at 13, 33 and 70 m water depth and includes, in decreasing numbers of individuals, Cytherissa lacustris (Sars, 1863), Candona neglecta Sars, 1887 and Cypria lacustris Lilljeborg, 1890. The occurrence of Limnocytherina sanctipatricii (Brady & Robertson, 1869) is restricted from late winter to late spring when temperatures are low, while C. vidua, L. inopinata, P. smaragdina, P. similis, P. newtoni and Ilyocypris sp. occur predominantly from spring to early autumn when temperatures are high. Individuals of C. neglecta, C. candida, F. caudata, P. compressa, C. lacustris, H. reptans and Cp. lacustris occur throughout the year with juveniles and adults occurring during the same period (C. neglecta at 70 m, C. lacustris at 13, 33 and 70 m, and H. reptans at 2, 5 and 13 m water depth) or with juveniles occurring during a different period of the year than adults (C. neglecta at 13 and 33 m and C. candida, F. caudata and P. compressa at their respective depth of occurrence). Among the environmental parameters investigated, an estimate of the relationship between ostracod autoecology and environmental parameters suggests that in the Petit-Lac: (i) water temperature and substrate characteristics are important factors controlling the distribution of species with depth, (ii) water temperature is also important for determining the timing of species development and, hence, its specific life history, and (iii) water oxygen and sedimentary organic matter content is less important compared to the other environmental parameter monitored.

The highly polymorphic so called "Trichia hispida group" is taxonomically problematic. According to different authors, one to about one hundred species are recognized in this group. Some recent publications admit 8 species for Central Euorpe. The present study gives arguments from biochemical data (12 enzymatic loci are electrophoretically analysed) for regrouping 5 morphological types into 2 species : Trichia sericea (Müller) var. sericea, plebeia, montana and striolata, and T.hispida(L.). A multivariate analysis of morphological measurments, and patterns of coexistence seem to corroborate this interpretation. T. sericea presents a wider ecological and morphological range, and both species need further investigations on their ecological and taxonomical relationship.

2. Plan du travail Pour atteindre nos objectifs, notre travail sera essentiellement divisé en trois grandes sections. La première partie nous sert principalement à esquisser notre démarche et les enjeux de notre entreprise, ainsi qu'à la situer au sein d'un cadre théorique - la linguistique interactionnelle - combinant l'analyse grammaticale et l'analyse conversationnelle. Nous procéderons d'abord à une brève description de l'analyse conversationnelle ethnométhodologique, ensuite à celle du courant Interaction and Grammar. Dans le cadre de ces descriptions, nous illustrerons par des exemples les liens entre la grammaire et l'interaction sociale tels qu'ils ont été élaborés dans de nombreuses recherches dans le domaine. Finalement, nous résumerons nos objectifs et nos procédures d'analyse. La deuxième partie consiste essentiellement en une discussion et une problématisation de l'état de la recherche dans la littérature. L'exposé montrera les principaux problèmes que la pseudo-clivée pose aux différents niveaux d'analyse. Nous y traiterons des problèmes de définition et de délimitation de la construction en question au niveau syntaxique et sémantique. Nous enchaînerons avec l'exposé de l'état de la recherche au niveau pragmatique et discursif Cette discussion nous permettra de problématiser à la fois la fonction principale dévolue à la pseudo-clivée et l'établissement de la relation entre forme et fonction. Finalement, nous aborderons certains aspects dont nous ne nous occuperons pas ou seulement partiellement dans notre travail. Le résumé de cette deuxième partie nous servira à récapituler les problèmes soulevés par la pseudo-clivée française. La troisième partie - empirique et analytique - constitue la partie centrale de ce travail. Elle exposera nos résultats à travers l'analyse d'exemples qui exhibent les propriétés typiques des occurrences retenues dans notre corpus. Cette partie est donc consacrée aux formes, aux fonctionnements syntaxico-séquentiels et aux fonctions effectivement rencontrées dans nos données pour aboutir à une description de ses propriétés interactives et à une typologie compatible avec les usages interactifs réels. Après l'exposé de ce qui nous semble le plus typique dans l'usage interactif effectif de la pseudo-clivée par les interlocuteurs, nous nous dirigerons plus particulièrement vers la délimitation et la localisation de notre objet d'analyse, d'une part, par rapport à d'autres constructions (clivées, pseudo-clivées inversées, dislocations à gauche), et d'autre part, par rapport à d'autres unions d'unités qui n'ont pas le statut de construction en usage. Finalement, nous tenterons d'intégrer nos observations dans une description cohérente du "phénomène pseudo-clivé" dans l'usage des locuteurs. De plus, nous présenterons - sur la base de nos résultats - des réponses ou des éléments de réponse aux problématiques soulevées par notre objet d'analyse telles que nous les avons élaborées et discutées dans la partie II de ce travail. Nous terminerons par une discussion sur les apports de notre démarche, ainsi qu'à une évaluation de notre conception spécifique des faits de grammaire, et plus particulièrement d'une construction syntaxique (partie IV). Nous indiquerons des pistes de recherche possibles à suivre en fonction de nos résultats. Parmi d'éventuelles investigations futures, un accent particulier sera mis sur les aspects prosodiques et la recherche comparative.

The project of articulating a theological ethics on the basis of liturgical anthropology is bound to fail if the necessary consequence is that one has to quit the forum of critical modern rationality. The risk of Engelhardt's approach is to limit rationality to a narrow vision of reason. Sin is not to be understood as the negation of human holiness, but as the negation of divine holiness. The only way to renew theological ethics is to understand sin as the anthropological and ethical expression of the biblical message of the justification by faith only. Sin is therefore a secondary category, which can only by interpreted in light of the positive manifestation of liberation, justification, and grace. The central issue of Christian ethics is not ritual purity or morality, but experience, confession and recognition of our own injustice in our dealing with God and men.

Combustion-derived and manufactured nanoparticles (NPs) are known to provoke oxidative stress and inflammatory responses in human lung cells; therefore, they play an important role during the development of adverse health effects. As the lungs are composed of more than 40 different cell types, it is of particular interest to perform toxicological studies with co-cultures systems, rather than with monocultures of only one cell type, to gain a better understanding of complex cellular reactions upon exposure to toxic substances. Monocultures of A549 human epithelial lung cells, human monocyte-derived macrophages and monocyte-derived dendritic cells (MDDCs) as well as triple cell co-cultures consisting of all three cell types were exposed to combustion-derived NPs (diesel exhaust particles) and to manufactured NPs (titanium dioxide and single-walled carbon nanotubes). The penetration of particles into cells was analysed by transmission electron microscopy. The amount of intracellular reactive oxygen species (ROS), the total antioxidant capacity (TAC) and the production of tumour necrosis factor (TNF)-a and interleukin (IL)-8 were quantified. The results of the monocultures were summed with an adjustment for the number of each single cell type in the triple cell co-culture. All three particle types were found in all cell and culture types. The production of ROS was induced by all particle types in all cell cultures except in monocultures of MDDCs. The TAC and the (pro-)inflammatory reactions were not statistically significantly increased by particle exposure in any of the cell cultures. Interestingly, in the triple cell co-cultures, the TAC and IL-8 concentrations were lower and the TNF-a concentrations were higher than the expected values calculated from the monocultures. The interplay of different lung cell types seems to substantially modulate the oxidative stress and the inflammatory responses after NP exposure. [Authors]

Antibody-drug conjugates (ADC) are emerging as powerful treatment strategies with outstanding target-specificity and high therapeutic activity in patients with cancer. Brentuximab vedotin represents a first-in-class ADC directed against CD30(+) malignancies. We hypothesized that its sustained clinical responses could be related to the stimulation of an anticancer immune response. In this study, we demonstrate that the dolastatin family of microtubule inhibitors, from which the cytotoxic component of brentuximab vedotin is derived, comprises potent inducers of phenotypic and functional dendritic cell (DC) maturation. In addition to the direct cytotoxic effect on tumor cells, dolastatins efficiently promoted antigen uptake and migration of tumor-resident DCs to the tumor-draining lymph nodes. Exposure of murine and human DCs to dolastatins significantly increased their capacity to prime T cells. Underlining the requirement of an intact host immune system for the full therapeutic benefit of dolastatins, the antitumor effect was far less pronounced in immunocompromised mice. We observed substantial therapeutic synergies when combining dolastatins with tumor antigen-specific vaccination or blockade of the PD-1-PD-L1 and CTLA-4 coinhibitory pathways. Ultimately, treatment with ADCs using dolastatins induces DC homing and activates cellular antitumor immune responses in patients. Our data reveal a novel mechanism of action for dolastatins and provide a strong rationale for clinical treatment regimens combining dolastatin-based therapies, such as brentuximab vedotin, with immune-based therapies. PURPOSE: Corticosteroids have recorded beneficial clinical effects and are widely used in medicine. In ophthalmology, besides their treatment benefits, side effects, including ocular toxicity have been observed especially when intraocular delivery is used. The mechanism of these toxic events remains, however, poorly understood. In our present study, we investigated the mechanisms and potential pathways of corticosteroid-induced retinal cell death. METHODS: Rats were sacrificed 24 h and 8 days after an intravitreous injection of 1 microl (40 microg) of Kenacort Retard. The eyes were processed for ultra structure analysis and detection of activated caspase-3, cytochrome-C, apoptosis-inducing factor (AIF), LEI-L-Dnase II, terminal transferase dUTP nick end labeling (TUNEL), and microtubule-associated protein 1-light chain 3 (MAP-LC3). In vitro, rat retinal pigment epithelial cells (RPE), retinal Müller glial cells (RMG) and human ARPE-19 cells were treated with triamcinolone acetonide (TA) or other glucocorticoids. Cell viability was quantified by 3-(4,5-dimethylthiazol-2-yl)-2,5 phenyltetrazolium bromide test (MTT) assay and cell counts. Nuclei staining, TUNEL assay, annexin-V binding, activated caspase-3 and lactate dehydrogenase (LDH) production characterized cell death. Localization of cytochrome-C, AIF, LEI-and L-Dnase II, and staining with MAP-LC3 or monodansylcadaverine were also carried out. Finally, ARPE-19 cells transfected with AIP-1/Alix were exposed to TA. RESULTS: In vitro incubation of retinal cell in the presence of corticosteroids induced a specific and dose-dependent reduction of cell viability. These toxic events were not associated with the anti-inflammatory activity of these compounds but depended on the hydro solubility of their formulation. Before cell death, extensive cytoplasmic vacuolization was observed in the retinal pigment epithelial (RPE) cells in vivo and in vitro. The cells however, did not show known caspase-dependent or caspase-independent apoptotic reactions. These intracellular vacuoles were negative for MAP-LC3 but some stained positive for monodansylcadaverine. Furthermore, over expression of AIP-1/Alix inhibited RPE cell death. CONCLUSIONS: These observations suggest that corticosteroid-induced retinal cell death may be carried out mainly through a paraptosis pathway.
