Cells from two melanoma cell lines, Me43 and GLL-19, were cloned in methylcellulose cultures and 20 randomly selected colonies from each line were picked up by micromanipulation, expanded in liquid cultures, and considered as clones of the original cell lines. The antigenic cell surface phenotype of these clones defined by panel of 12 monoclonal antibodies (MAb) was analyzed by flow microfluorometry (FMF) using a fluorescence-activated cell sorter (FACS II) and compared with the known stable phenotype of the parent cell line. The antibody panel consisted of eight MAb against melanoma-associated antigens, two MAb against monomorphic determinants of HLA-DR (la) and HLA-ABC, respectively, one MAb against the common acute lymphoblastic leukemia antigen (CALLA) and one MAb against carcinoembryonic antigen used as control. A remarkable heterogeneity in terms of qualitative and quantitative expression of the cell surface antigens studied was observed among and within the different clones. The single-cell origin of the clones was assessed by comparing the clonogenic cell frequency, determined by limiting dilutions in microculture plates, with the cloning efficiency observed in Petri dishes. Both techniques using methylcellulose medium gave the same percentages of growing colonies. Cells from four Me43 clones were recloned in methylcellulose and the phenotype of five randomly selected subclones from each clone was analysed using the same panel of monoclonal antibodies. Each subclone also displayed heterogeneity with individual phenotypes different from that of the original clone and from the parental Me43 cell line. The antigen expression by individual cells in situ within clones was analyzed on frozen sections from colonies using the same panel of MAb and a biotin-avidin immunoperoxidase method. The results confirmed the marked heterogeneity of antigen expression within and among colonies, as indicated by the FMF analysis.

In a mode of nude mice bearing a human colon carcinoma xenograft, the biodistribution and tumor localization of metatetrahydroxyphenylchlorin (m-THPC) coupled to polyethylene glycol (PEG) were compared with those of the free form of this photosensitizer used in photodynamic therapy (PDT). At different times after i.v. injection of both forms of 125I-labeled photosensitizer, m-THPC-PEG gave on average a 2-fold higher tumor uptake than free m-THPC. In addition, at early times after injection, m-THPC-PEG showed a 2-fold longer blood circulating half-life and a 4-fold lower liver uptake than free m-THPC. The tumor to normal tissue ratios of radioactivity concentrations were always higher for m-THPC-PEG than for free m-THPC at any time point studied from 2 to 96 hr post-injection. Significant coefficients of correlation between direct fluorescence measurements and radioactivity counting were obtained within each organ tested. Fluorescence microscopy studies showed that m-THPC-PEG was preferentially localized near the tumor vessels, whereas m-THPC was more diffusely distributed inside the tumor tissue. To verify whether m-THPC-PEG conjugate remained phototoxic in vivo, PDT experiments were performed 72 hr after injection and showed that m-THPC-PEG was as potent as free m-THPC in the induction of tumor regression provided that the irradiation does for m-THPC-PEG conjugate was adapted to a well-tolerated 2-fold higher level. The overall results demonstrate first the possibility of improving the in vivo tumor localization of a hydrophobic dye used for PDT by coupling it to PEG and second that a photosensitizer conjugated to a macromolecule can remain phototoxic in vivo.

Three out of five human endometrial carcinomas were successfully grafted into nude mice (BALB/c/nu/nu). Two of these tumors could be maintained by serial transplantation. The morphological characteristics displayed by the grafted tumors were comparable to those of the original carcinomas. Permanent cell lines were established from these two tumors. Reinjection of cells grown in vitro into nude mice produced nodules of identical histology as compared to original solid transplants. The influence of medroxyprogesterone acetate on tumor growth in vivo and cell proliferation in vitro was studied. This hormonal treatment did not produce any significant effect on tumor cells, either in vitro or in vivo, for the two endometrial carcinomas. After medroxyprogesterone administration, a slight but non-significant growth inhibition of the tumor cells in vitro was observed and the tumor transplants in vivo did not appear to be influenced. The experiments illustrate the possible use of this model for testing potential anti-cancer agents.

OBJECTIVE: To estimate the incremental cost-effectiveness of the first-line pharmacotherapies (nicotine gum, patch, spray, inhaler, and bupropion) for smoking cessation across six Western countries-Canada, France, Spain, Switzerland, the United States, and the United Kingdom. DESIGN AND STUDY POPULATION: A Markov-chain cohort model to simulate two cohorts of smokers: (1) a reference cohort given brief cessation counselling by a general practitioner (GP); (2) a treatment cohort given counselling plus pharmacotherapy. Effectiveness expressed as odds ratios for quitting associated with pharmacotherapies. Costs based on the additional physician time required and retail prices of the medications. INTERVENTIONS: Addition of each first-line pharmacotherapy to GP cessation counselling. MAIN OUTCOME MEASURES: Cost per life-year saved associated with pharmacotherapies. RESULTS: The cost per life-year saved for counselling only ranged from US190 dollars in Spain to 773 dollars in the UK for men, and from 288 dollars in Spain to 1168 dollars in the UK for women. The incremental cost per life-year saved for gum ranged from 2230 dollars for men in Spain to 7643 dollars for women in the US; for patch from 1758 dollars for men in Spain to 5131 dollars for women in the UK; for spray from 1935 dollars for men in Spain to 7969 dollars for women in the US; for inhaler from 3480 dollars for men in Switzerland to 8700 dollars for women in France; and for bupropion from 792 dollars for men in Canada to 2922 dollars for women in the US. In sensitivity analysis, changes in discount rate, treatment effectiveness, and natural quit rate had the strongest influences on cost-effectiveness. CONCLUSIONS: The cost-effectiveness of the pharmacotherapies varied significantly across the six study countries, however, in each case, the results would be considered favourable as compared to other common preventive pharmacotherapies.

Résumé Rôle du transporteur de glucose GLUT2 dans les mécanismes centraux de glucodétection impliqués dans le contrôle de la sécrétion du glucagon et de la prise alimentaire. Les mécanismes centraux de glucodétection jouent un rôle majeur dans le contrôle de l'homéostasie glucidique. Ces senseurs régulent principalement la sécrétion des hormones contre-régulatrices, la prise alimentaire et la dépense énergétique. Cependant, la nature cellulaire et le fonctionnement moléculaire de ces mécanismes ne sont encore que partiellement élucidés. Dans cette étude, nous avons tout d'abord mis en évidence une suppression de la stimulation de la sécrétion du glucagon et de la prise alimentaire en réponse à une injection intracérébroventriculaire (i.c.v.) de 2-déoxy-D-glucose (2-DG) chez les souris de fond génétique mixte et déficientes pour le gène glut2 (souris RIPG1xglut2-/-). De plus, chez ces souris, l'injection de 2-DG n'augmente pas l'activation neuronale dans l'hypothalamus et le complexe vagal dorsal. Nous avons ensuite montré que la ré-expression de GLUT2 dans les neurones des souris RIPG1xg1ut2-/- ne restaure pas la sécrétion du glucagon et la prise alimentaire en réponse à une injection i.c.v. de 2-DG. En revanche, l'injection de 2-DG réalisée chez les souris RIPG1xg1ut2-/- ré-exprimant le GLUT2 dans leurs astrocytes, stimule la sécrétion du glucagon et l'activation neuronale dans le complexe vagal dorsal mais n'augmente pas la prise alimentaire ni l'activation neuronale dans l'hypothalamus. L'ensemble de ces résultats démontre l'existence de différents mécanismes centraux de glucodétection dépendants de GLUT2. Les mécanismes régulant la sécrétion du glucagon sont dépendants de GLUT2 astrocytaire et pourraient être localisés dans le complexe vagal dorsal. L'implication des astrocytes dans ces mécanismes suggère un couplage fonctionnel entre les astrocytes et les neurones adjacents « sensibles au glucose ». Lors de cette étude, nous avons remarqué chez les souris RIPG1xg1ut2-/- de fond génétique pur C57B1/6, que seul le déclenchement de la prise alimentaire en réponse à l'injection i.p. ou i.c.v. de 2-DG est aboli. Ces données mettent en évidence que suivant le fond génétique de la souris, les mécanismes centraux de glucodétection impliqués dans la régulation de la sécrétion peuvent être indépendants de GLUT2. Summary. Role of transporter GLUT2 in central glucose sensing involved in the control of glucagon secretion and food intake. Central glucose sensors play an important role in the control of glucose homeostasis. These sensors regulate general physiological functions, including food intake, energy expenditure and hormones secretion. So far the cellular and molecular basis of central glucose detection are poorly understood. Hypoglycemia, or cellular glucoprivation by intraperitoneal injection of 2-deoxy¬glucose (2-DG) injection, elicit multiple glucoregulatory responses, in particular glucagon secretion and stimulation of feeding. We previously demonstrated that the normal glucagon response to insulin-induced hypoglycemia was suppressed in mice lacking GLUT2. This indicated the existence of extra-pancreatic, GLUT2-dependent, glucose sensors controllling glucagon secretion. Here, we have demonstrated that the normal glucagon and food intake responses to central glucoprivation, by intracerebroventricular (i.c.v.) injections of 2-DG, were suppressed in mice lacking GLUT2 (RIPG1xglut2-/- mice) indicating that GLUT2 plays a role in central glucose sensing units controlling secretion of glucagon and food intake. Whereas it is etablished that glucose responsive neurons change their firing rate in response to variations of glucose concentrations, the exact mechanism of glucose detection is not established. In particular, it has been suggested that astrocytic cells may be the primary site of glucose detection and that a signal is subsequently transmitted to neurons. To evaluate the respective role of glial and neuronal expression of GLUT2 in central glucodetection, we studied hypoglycemic and glucoprivic responses following cellular glucoprivation in RIPG1xglut2-/- mice reexpressing the transgenic GLUT2 specifially in their astrocytes (pGFAPG2xRIPG1xglut2-/- mice) or their neurons (pSynG2xRIPG1xglut2-/- mice). The increase of food intake after i.p. injection of 2-DG in control mice was not observed in the pGFAPG2xRIPG1xglut2-/- mice. Whereas a strong increase of glucagon secretion was observed in control and pGFAPG2xRIPG1xglut2-/- mice, not glucagonemic response was induced in pSynG2xRIPG1xglut2-/- mice. Our results show that GLUT2 reexpression in glial cells but not in neurons restored glucagon secretion and thus present a strong evidence that glucose detection and the control of glucagon secretion require a coupling between glial cells and neurons. Furthermore, these results show the existence of differents glucose sensors in CNS. Résumé tout public. Rôle du transporteur de glucose GLUT2 dans les mécanismes centraux de glucodétection impliqués dans le contrôle de la sécrétion du glucagon et de la prise alimentaire. Chez les mammifères, en dépit des grandes variations dans l'apport et l'utilisation du glucose, la glycémie est maintenue à une valeur relativement constante d'environ 1 g/l. Cette régulation est principalement sous le contrôle de deux hormones produites par le pancréas l'insuline et le glucagon. A la suite d'un repas, la détection de l'élévation de la glycémie par le pancréas permet la libération pancréatique de l'insuline dans le sang. Cette hormone va alors permettre le stockage dans le foie du glucose sanguin en excès et diminuer ainsi la glycémie. Sans insuline, le glucose s'accumule dans le sang. On parle alors d'hyperglycémie chronique. Cette situation est caractéristique du diabète et augmente les risques de maladies cardiovasculaires. A l'inverse, lors d'un jeûne, la détection de la diminution de la glycémie par le cerveau permet le déclenchement de la prise alimentaire et stimule la sécrétion de glucagon par le pancréas. Le glucagon va alors permettre la libération dans le sang du glucose stocké par le foie. Les effets du glucagon et de la prise de nourriture augmentent ainsi les concentrations sanguines de glucose pour empêcher une diminution trop importante de la glycémie. Une hypoglycémie sévère peut entraîner un mauvais fonctionnement du cerveau allant jusqu'à des lésions cérébrales. Contrairement aux mécanismes pancréatiques de détection du glucose, les mécanismes de glucodétection du cerveau ne sont encore que partiellement élucidés. Dans le laboratoire, nous avons observé, chez les souris transgéniques n'exprimant plus le transporteur de glucose GLUT2, une suppression de la stimulation de la sécrétion du glucagon et du déclenchement de la prise alimentaire en réponse à une hypoglycémie, induite uniquement dans le cerveau. Dans le cerveau, le GLUT2 est principalement exprimé par les astrocytes, cellules gliales connues pour soutenir, nourrir et protéger les neurones. Nous avons alors ré-exprimé spécifiquement le GLUT2 dans les astrocytes des souris transgéniques et nous avons observé que seule la stimulation de la sécrétion du glucagon en réponse à l'hypoglycémie est restaurée. Ces résultats mettent en évidence que la sécrétion du glucagon et la prise alimentaire sont contrôlées par différents mécanismes centraux de glucodétection dépendants de GLUT2.

After decades of management reforms in the public sector, questions on the impact of leader-ship behavior in public organizations have been attracting increasing attention. This paper investigates the relationship between transformational leadership behavior and organizational citizenship behavior as one major extra-role outcome of transformational leadership. Refer-ring to a growing body of research that shows the importance of public service values and employee identification in public administration research, we include public service motiva-tion and organizational goal clarification as mediating variables in our analysis. Structural equation modeling is applied as the method of analysis for a sample of 569 public managers at the local level of Switzerland. The findings of our study support the assumed indirect relation-ship between leadership and employee behavior and emphasize the relevance of public ser-vice values when analyzing leadership behavior in public sector organizations.

We report a case series of three children with solid pseudopapillary tumor of the pancreas (SPT) in which a complete radiological work-up, including ultrasound, computed tomography scans, and MRI, has been carried out. The aim of this article is to highlight the characteristic imaging findings of SPT in the pediatric age group and to establish a correlation with typical histopathological findings of the lesion.

Endoscopic subureteral collagen injection has become an accepted means for the treatment of vesicoureteral reflux in children. The aim of this study was to evaluate the histological behavior of glutaraldehyde cross-linked bovine collagen implants. The specimens were harvested from 29 patients who underwent reimplant surgery 2 to 30 months (mean 9.5) after unsuccessful subureteral injection therapy. In addition to routine hematoxylin and eosin staining, a new staining method (solophenyl red 3BL) able to demonstrate selectively neoformation of types I and III human collagen, was applied. Invasion of host fibroblasts into the bovine implant and the formation of endogenous types I and III collagen were demonstrated in all 29 cases. Adverse histological reactions were rare and, if present, they were predominantly of an inflammatory nature.

(English version below) Deux expériences littéraires, quasiment simultanées, ont été menées sur "les Maréchaux", les boulevards extérieurs parisiens, autour de l'an 2000. Cette coïncidence temporelle et spatiale est précieuse : elle permet de mesurer les écueils et les apports liés à une faculté de "voir la ville", aujourd'hui largement partagée, mais dont les écrivains sont les détenteurs historiques. En interrogeant la dimension sociale de ces voyages de proximité, c'est-à-dire la relation d'altérité que cette pratique institue avec les hommes qui sont dans la ville, une politique peut être mise au jour, et la précarité d'un horizon commun. Two almost simultaneous literary experiments were conducted around the year 2000 on the exterior Parisian boulevards, known as "les Maréchaux" ("the Marshals"). This temporal and spatial coincidence is especially valuable in that it serves to assess the pitfalls and the contributions relating to an ability to "see the city" that is now widely experienced, but of which writers are the historical custodians. Examining the social dimension of these "propinquitous travels" (journeys to nearby destinations), meaning the distancing that this practice institutes with the people inside the city, serves to reveal a policy and the precarious nature of a shared horizon.

Objectives: To compare the results of rapid PCR screening for MRSA using the GeneXpert system with those of cultures in an outbreak setting. Methods: GeneXpert was used for screening MRSA in nose, throat, groin, and other clinical samples during a 6-month period. Samples were performed using a double-swab transystem. When >1 sample was found positive in a screening set, all second swabs of the set were analysed by culture. Results: From June to October 2009, 7568 rapid tests were performed, among which 432 (5.7%) were positive (nose: 149/2090, 7.1%; throat: 98/2078, 4.7%; groin: 152/2080, 7.3%; urine: 14/1090, 1.3%; wounds: 18/150, 12%; and others:1/27, 3.7%), and 84 (1.1%) were invalid. A total of 1517 samples were analyzed by both rapid PCR and culture. Rapid tests had a sensitivity of 0.896 compared to cultures, a specificity of 0.769, a PPV of 0.763, and a NPV of 0.899. The rapid test was found to be less sensitive in throat samples (0.81) than in nose or inguinal samples (0.93 for both). In 32/192 (16%) patients a positive rapid PCR result was not confirmed by culture, despite several subsequent screening samples in some patients. Cycle threshold (Ct) for SCCmec of these PCR positive reactions were all >30. Conclusions: GeneXpert MRSA was found to be suitable for the rapid detection in nose, inguinal, and throat samples, however with a lower sensitivity in the later. Negative cultures in 16% of our PCR-positive patients raised the question of false positivity or higher sensitivity of GeneXpert. Further work is needed to investigate these cases.

We examined drivers of article citations using 776 articles that were published from 1990-2012 in a broad-based and high-impact social sciences journal, The Leadership Quarterly. These articles had 1,191 unique authors having published and received in total (at the time of their most recent article published in our dataset) 16,817 articles and 284,777 citations, respectively. Our models explained 66.6% of the variance in citations and showed that quantitative, review, method, and theory articles were significantly more cited than were qualitative articles or agent-based simulations. As concerns quantitative articles, which constituted the majority of the sample, our model explained 80.3% of the variance in citations; some methods (e.g., use of SEM) and designs (e.g., meta-analysis), as well as theoretical approaches (e.g., use of transformational, charismatic, or visionary type-leadership theories) predicted higher article citations. Regarding the statistical conclusion validity of quantitative articles, articles having endogeneity threats received significantly fewer citations than did those using a more robust design or an estimation procedure that ensured correct causal estimation. We make several general recommendations on how to improve research practice and article citations.

Resting metabolic rate (RMR) and the thermic effect of a meal (TEM) were measured in a group of 26 prepubertal children divided into three groups: (1) children with both parents obese (n = 8, group OB2); (2) children with no obese parents and without familial history of obesity (n = 8, OB0); and (3) normal body weight children (n = 10, C). Average RMR was similar in OB2 and OB0 children (4785 +/- 274 kJ/day vs 5091 +/- 543 kJ/day), but higher (P < 0.05) than in controls (4519 +/- 322 kJ/day). Adjusted for fat-free mass (FFM) mean RMRs were comparable in the three groups of children (4891 +/- 451 kJ/day vs 5031 +/- 451 kJ/day vs 4686 +/- 451 kJ/day in OB2, OB0, and C, respectively). The thermic response to the mixed meal was similar in OB2, OB0 and C groups. The TEM calculated as the percentage of RMR was lower (P < 0.05) in obese than in control children: 10.2% +/- 3.1% vs 10.9% +/- 4.3% vs 14.0% +/- 4.3% in OB2, OB0, and C, respectively. The similar RMR as absolute value as well as adjusted for FFM, and the comparable thermic effect of food in the obese children with or without familial history of obesity, failed to support the view that family history of obesity can greatly influence the RMR and the TEM of the obese child with obese parents.

OBJECTIVES: Genomewide association studies (GWAS) have identified clear evidence of genetic markers for nicotine dependence. Other smoking phenotypes have been tested, but the results are less consistent. The tendency to relapse versus the ability to maintain long-term abstinence has received little attention in genetic studies; thus, our aim was to provide a better biological understanding of this phenotype through the identification of genetic loci associated with smoking relapse. METHODS: We carried out a GWAS on data from two European population-based collections, including a total of 835 cases (relapsers) and 990 controls (abstainers). Top-ranked findings from the discovery phase were tested for replication in two additional independent European population-based cohorts. RESULTS: Of the seven top markers from the discovery phase, none were consistently associated with smoking relapse across all samples and none reached genomewide significance. A single-nucleotide polymorphism rs1008509, within the Xylosyltransferase II (XYLT2) gene, was suggestively associated with smoking relapse in the discovery phase (β=-0.504; P=5.6E-06) and in the first replication sample (ALSPAC) (β=-0.27; P=0.004; n=1932), but not in the second sample (KORA) (β=0.19; P=0.138; n=912). We failed to identify an association between loci implicated previously in other smoking phenotypes and smoking relapse. CONCLUSION: Although no genomewide significant findings emerged from this study, we found that loci implicated in other smoking phenotypes were not associated with smoking relapse, which suggests that the neurobiology of smoking relapse and long-term abstinence may be distinct from biological mechanisms implicated in the development of nicotine dependence.
