Résumé: Depuis plusieurs années, le thème des réseaux sociaux est au centre de l'intérêt des études historiques. Est-il possible de formaliser l'analyse de réseaux sociaux spécifiques - parenté, clientèle, solidarités locales, etc. - afin d'en analyser l'influence sur des événements historiques et des individus précis ? L'étude présentée prend en considération les luttes souvent violentes entre radicaux et conservateurs dans le Val de Bagnes, en Valais (Suisse), entre 1839 et 1900. La comparaison entre les généalogies des familles de la vallée et les informations sur la vie politique et sociale nous permet de relever l'influence de la parenté dans l'organisation des factions politiques. Les réseaux de parenté sont toutefois ouverts et souples, permettant des adaptations aux évolutions de la situation politique, économique et sociale. L'affaire autour du faux-monnayeur italien Joseph S. Farinet, dans les années 1870, nous permet par exemple de suivre l'évolution des réseaux de solidarité, à la suite d'une crise politique, ainsi que l'émergence de nouvelles activités économiques, notamment le tourisme, avec les hôteliers, les aubergistes et les guides de montagne souvent liés au milieu radical. L'analyse permet également de nuancer l'influence des réseaux de patronage : les collaborations horizontales, à l'intérieur des classes populaires, semblent mieux expliquer les solidarités politiques. Abstract: For several years, historians have been closely concerned with the question of social networks. Is it possible to conceptualize specific networks - like kinship, patronage or local solidarities - and to analyze their influence on concrete individuals or historical events? This paper considers the violent struggles between a radical political faction and a conservative one in a Swiss alpine valley, the Val de Bagnes (Valais) between 1839 and 1900. It compares information about political and social conflicts in the valley with genealogies of local families. By this way the eminent influence of kinship ties on political organizations becomes visible. But kinship networks are open and very supple, allowing adaptations to new political and social configurations. The trials against the Italian smuggler and counterfeiter Joseph S. Farinet in the Seventies allow to describe the evolution of local cooperation networks as a consequence of a political crisis in the canton of Valais and of new economic activities. The paper stresses the active role of a emerging group of hotel- or inn-owners and mountain guides, often closely tied with the radical milieu. The analysis of social transactions raises critical questions about the role of patronage in political mobilization: horizontal cooperation and kinship ties between peasants, small cattle owners and artisans seem to explain political solidarities better than patronage structures.

This essay focuses on how Spielberg's film engages with and contributes to the myth of Lincoln as a super-natural figure, a saint more than a hero or great statesman, while anchoring his moral authority in the sentimental rhetoric of the domestic sphere. It is this use of the melodramatic mode, linking the familial space with the national through the trope of the victim-hero, which is the essay's main concern. With Tony Kushner, author of Angels in America, as scriptwriter, it is perhaps not surprising that melodrama is the operative mode in the film. One of the issues that emerge from this analysis is how the film updates melodrama for a contemporary audience in order to minimize what could be perceived as manipulative sentimental devices, observing for most of the film an aesthetic of relative sobriety and realism. In the last hour, and especially the final minutes of the film, melodramatic conventions are deployed in full force and infused with hagiographic iconography to produce a series of emotionally charged moments that create a perfect union of American Civil Religion and classical melodrama. The cornerstone of both cultural paradigms, as deployed in this film, is death: Lincoln's at the hands of an assassin, and the Civil War soldiers', poignantly depicted at key moments of the film. Finally, the essay shows how film melodrama as a genre weaves together the private and the public, the domestic with the national, the familial with the military, and links pathos to politics in a carefully choreographed narrative of sentimentalized mythopoesis.

Machado-Joseph disease is the most frequently found dominantly-inherited cerebellar ataxia. Over-repetition of a CAG trinucleotide in the MJD1 gene translates into a polyglutamine tract within the ataxin 3 protein, which upon proteolysis may trigger Machado-Joseph disease. We investigated the role of calpains in the generation of toxic ataxin 3 fragments and pathogenesis of Machado-Joseph disease. For this purpose, we inhibited calpain activity in mouse models of Machado-Joseph disease by overexpressing the endogenous calpain-inhibitor calpastatin. Calpain blockage reduced the size and number of mutant ataxin 3 inclusions, neuronal dysfunction and neurodegeneration. By reducing fragmentation of ataxin 3, calpastatin overexpression modified the subcellular localization of mutant ataxin 3 restraining the protein in the cytoplasm, reducing aggregation and nuclear toxicity and overcoming calpastatin depletion observed upon mutant ataxin 3 expression. Our findings are the first in vivo proof that mutant ataxin 3 proteolysis by calpains mediates its translocation to the nucleus, aggregation and toxicity and that inhibition of calpains may provide an effective therapy for Machado-Joseph disease.

Machado-Joseph disease or Spinocerebellar ataxia type 3 is a progressive fatal neurodegenerative disorder caused by the polyglutamine-expanded protein ataxin-3. Recent studies demonstrate that RNA interference is a promising approach for the treatment of Machado-Joseph disease. However, whether gene silencing at an early time-point is able to prevent the appearance of motor behavior deficits typical of the disease when initiated before onset of the disease had not been explored. Here, using a lentiviral-mediated allele-specific silencing of mutant ataxin-3 in an early pre-symptomatic cerebellar mouse model of Machado-Joseph disease we show that this strategy hampers the development of the motor and neuropathological phenotypic characteristics of the disease. At the histological level, the RNA-specific silencing of mutant ataxin-3 decreased formation of mutant ataxin-3 aggregates, preserved Purkinje cell morphology and expression of neuronal markers while reducing cell death. Importantly, gene silencing prevented the development of impairments in balance, motor coordination, gait and hyperactivity observed in control mice. These data support the therapeutic potential of RNA interference for Machado-Joseph disease and constitute a proof of principle of the beneficial effects of early allele-specific silencing for therapy of this disease.

En février 1944, une soixantaine d'hommes, venus de différents pays d'Europe dominés par des dictatures, sont rassemblés dans un camp spécial situé dans le Jura. Le camp de Bassecourt réunit des réfugiés politiques. Sa création est le résultat d'une politique initiée dans les années 1930, qui se prolonge jusqu'à la fin de la guerre. Il constitue un terrain fécond pour examiner, d'une part, le traitement réservé par la Confédération à ces réfugiés, considérés comme dangereux et indésirables en raison de leur engagement politique, et, d'autre part, les stratégies mises en oeuvre par les internés pour défendre leurs intérêts et poursuivre leur lutte contre le fascisme. Les changements survenus dans l'équilibre des forces européennes au tournant des années 1944 et 1945 conduisent la Suisse à changer de politique, illustrant par là même le caractère improvisé de cette dernière ; ils offrent aussi aux réfugiés l'opportunité de participer à la reconstruction de leur pays et d'y jouer un rôle politique.
