Suite à une question initiale du théologien catholique Adrian Holderegger, l'auteur s'interroge sur la question du rôle de la raison en théologie et en éthique théologique. Il présente et discute la critique de la raison sourde proposée par Maurice Bellet. Les relations entre la rationalité athétique, athématique et amodale conduisent à se demander si une conception négative et critique de la rationalité ne serait pas la condition d'un redéploiement positif de la raison de l'être humain singulier comme capacité d'écoute attentive de l'autre, du monde et de Dieu.

Septic arthritis of the pubic symphysis caused by Streptococcus mitis.

Septic arthritis of the pubic symphisis is distinguished from osteitis pubis by positive cultures. The symptoms, physical examination and laboratory findings of these two conditions are comparable. We present a case of 57-year-old woman with septic arthritis of pubic symphisis caused by Streptococcus mitis, a commensal oral flora that belongs to viridans group streptococci, which normally reside in the oral cavity, the gastrointestinal and the urogenital tract.

Specificity of psychosis, mania and major depression in a contemporary family study.

There has been increasing attention to the subgroups of mood disorders and their boundaries with other mental disorders, particularly psychoses. The goals of the present paper were (1) to assess the familial aggregation and co-aggregation patterns of the full spectrum of mood disorders (that is, bipolar, schizoaffective (SAF), major depression) based on contemporary diagnostic criteria; and (2) to evaluate the familial specificity of the major subgroups of mood disorders, including psychotic, manic and major depressive episodes (MDEs). The sample included 293 patients with a lifetime diagnosis of SAF disorder, bipolar disorder and major depressive disorder (MDD), 110 orthopedic controls, and 1734 adult first-degree relatives. The diagnostic assignment was based on all available information, including direct diagnostic interviews, family history reports and medical records. Our findings revealed specificity of the familial aggregation of psychosis (odds ratio (OR)=2.9, confidence interval (CI): 1.1-7.7), mania (OR=6.4, CI: 2.2-18.7) and MDEs (OR=2.0, CI: 1.5-2.7) but not hypomania (OR=1.3, CI: 0.5-3.6). There was no evidence for cross-transmission of mania and MDEs (OR=.7, CI:.5-1.1), psychosis and mania (OR=1.0, CI:.4-2.7) or psychosis and MDEs (OR=1.0, CI:.7-1.4). The strong familial specificity of psychotic, manic and MDEs in this largest controlled contemporary family study challenges the growing assertion that the major types of mood disorders are manifestations of a common underlying diathesis.

Expression of brain-derived neurotrophic factor is not modulated by chronic mild stress in the rat hippocampus and amygdala.

Accumulating evidence supports a role for brain-derived neurotrophic factor (BDNF) in depression. However, most of these studies have been performed in animal models that have a low face validity with regard to the human disease. Here, we examined the regulation of BDNF expression in the hippocampus and amygdala of rats subjected to the chronic mild stress (CMS) model of depression, a paradigm that induces anhedonia, a core symptom of depression. We found that exposure of rats to the CMS paradigm did not modulate BDNF mRNA expression in the hippocampus and amygdala. In addition, chronic administration of imipramine, which reversed CMS-induced anhedonia, did not alter BDNF mRNA expression in these limbic structures.

While the development of early psychosis intervention programs have improved outcome of such disorders, primary prevention strategies are still out of reach. The elaboration, over the last 15 years, of scales and criteria to identify populations at high risk for psychosis is a real progress, but their low specificity is still a major obstacle to their use outside of research projects. For this reason, even if "ultra high risk", subjects present with real psychiatric disorders and sometimes significant decrease in functioning level, the fact that only a small proportion will eventually develop full blown psychosis will probably lead to the rejection of a "psychosis risk syndrom" from the future DSM-V classification.

Lire Certeau en théologien, c'est consonner avec une articulation à l'histoire, vue dans ses discontinuités et comme lieu de productions, imaginaires et autres, et ainsi scène d'un désir et d'une exposition maximale au monde. On renvoie alors à un excès, jamais dit directement, mais en travail au coeur du présent et de l'immanence. Lire Certeau en théologien, c'est assumer ainsi une modernité signant la fin du rapport à un cosmos différencié et signifiant, pour entrer dans une passion de l'Unique, absent. Au creux d'une perte, d'un exil, d'une épreuve, qu'attestent les mystiques, eux qui disent un corps à corps avec le monde, une altération originaire, un avènement singulier. Et qui les écrivent. Or, la théologie est justement articulée à de la mémoire, textuelle et fictive, et aux jeux institutionnels à laquelle cette mémoire émarge, fût-ce sur mode d'écart ou de dissidence ; et son travail passe par l'anthropologie d'un croire, de dimension radicalement humaine et irréductible en l'humain. S'y dit une subversion, non un dépassement, du monde.

Averting inflammation by targeting the cytokine environment.

Cytokines are key instigators and regulators of immune responses and therefore hold great potential as targets for new therapeutic strategies. However, the selection of which cytokines to target, and in particular the identification of which cytokines regulate the rate-limiting steps of disease pathways, is crucial to the success of such strategies. Moreover, balancing the need for ablating pathological inflammatory responses and simultaneously maintaining the ability to control infectious agents is a key consideration. Recent advances in our understanding of cytokine networks, as well as technical progress in blocking cytokines in vivo, are likely to be a source for new drugs that can control chronic inflammatory diseases.

En finir avec le désir. Michel de Certeau et l'hétérologie des voix

Depuis plusieurs décennies la philosophie, comme la théologie, a trouvé dans la thématique du désir une tentative paradoxale de sauvegarder une pensée de la totalité. A partir de Michel de Certeau, une pensée du multiple pur qui puisse entendre le bris des voix mondaines est possible. Par un parcours en trois temps, les a. tentent d'exposer comment, au niveau des discursivités, l'histoire, opération du deuil et de l'écart, se défie de l'emmurement opéré par la philosophie ; puis, au niveau des singularités, comment la mystique, par l'exemple d'un poème de Jean de la Croix, ne s'exacerbe pas d'un désir porté vers l'absent, mais dévoile la présence sans rassemblement des multiplicités ; enfin, pourquoi cet effondrement de l'un ne verse pas, au niveau ontologique, dans une tautologie de l'être.

Predicting drug metabolism: experiment and/or computation?

Drug metabolism can produce metabolites with physicochemical and pharmacological properties that differ substantially from those of the parent drug, and consequently has important implications for both drug safety and efficacy. To reduce the risk of costly clinical-stage attrition due to the metabolic characteristics of drug candidates, there is a need for efficient and reliable ways to predict drug metabolism in vitro, in silico and in vivo. In this Perspective, we provide an overview of the state of the art of experimental and computational approaches for investigating drug metabolism. We highlight the scope and limitations of these methods, and indicate strategies to harvest the synergies that result from combining measurement and prediction of drug metabolism.

Multitude libre/Multitude serve. Pour une lecture du Traité politique de Spinoza

Cette étude se donne pour objet de dégager la visée poursuivie par Spinoza dans son Traité politique et d'en extraire l'enseignement politique. Après avoir brièvement reconstruit la théorie spinoziste du droit naturel et l'advenue nécessaire d'un corps politique, elle s'attache, en se basant principalement sur une lecture des chapitres 6 à 11, à déterminer les conditions de la position d'une multitude libre plutôt que serve. On montre que ces conditions se résument dans le principe d'un renforcement circulaire de la puissance du souverain et de celle de la multitude et que la possibilité de son instauration repose sur une juste compréhension de la dynamique passionnelle ; et qu'à défaut, s'installe un régime de servitude marqué par le clivage des gouvernants et de la multitude. On conclut par un regard sur le présent à la lumière de la leçon de ce traité.
