The cytokine macrophage migration inhibitory factor plays a central role in inflammation, cell proliferation and tumorigenesis. Moreover, macrophage migration inhibitory factor levels correlate with tumor aggressiveness and metastatic potential. Histone deacetylase inhibitors are potent antitumor agents recently introduced in the clinic. Therefore, we hypothesized that macrophage migration inhibitory factor would represent a target of histone deacetylase inhibitors. Confirming our hypothesis, we report that histone deacetylase inhibitors of various chemical classes strongly inhibited macrophage migration inhibitory factor expression in a broad range of cell lines, in primary cells and in vivo. Nuclear run on, transient transfection with macrophage migration inhibitory factor promoter reporter constructs and transduction with macrophage migration inhibitory factor expressing adenovirus demonstrated that trichostatin A (a prototypical histone deacetylase inhibitor) inhibited endogenous, but not episomal, MIF gene transcription. Interestingly, trichostatin A induced a local and specific deacetylation of macrophage migration inhibitory factor promoter-associated H3 and H4 histones which did not affect chromatin accessibility but was associated with an impaired recruitment of RNA polymerase II and Sp1 and CREB transcription factors required for basal MIF gene transcription. Altogether, this study describes a new molecular mechanism by which histone deacetylase inhibitors inhibit MIF gene expression, and suggests that macrophage migration inhibitory factor inhibition by histone deacetylase inhibitors may contribute to the antitumorigenic effects of histone deacetylase inhibitors.

Introduction : Le cancer de la vessie est la 4ème cause de cancer en Europe, chez l'homme. Dans 70% des¦cas, le diagnostic initial de cancer transitionnel de la vessie est non-musculo invasif mais jusqu'à 45%¦progresseront en tumeur musculo-invasive selon des facteurs de risques qui ont été scorés par¦l'Organisation européenne de recherche sur le traitement du cancer (EORTC). Après résection¦endoscopique transuréthrale vésicale (RTUV) de la tumeur, une instillation intravésicale (ives) d'agents¦chimiothérapeutiques ou d'immunomodulateurs tel que le Bacille Calmette Guérin (BCG) permet de¦réduire les récurrences/progressions. Cependant l'utilisation du BCG est limitée aux patients avec un risque¦de progression haut et intermédiaire au vu des effets secondaires occasionnés. Le système immunitaire¦joue un rôle certain dans l'évolution d'un processus néoplasique. Toutefois, les cancers adoptent différents¦mécanismes pour supprimer la réponse antitumorale. La variation de l'environnement immunologique¦tumoral pourrait avoir une valeur pronostique pour l'évolution naturelle de la maladie. Le but de notre¦étude est d'utiliser des coupes paraffines archivées de RTUV afin de définir l'environnement immunitaire¦des carcinomes urothéliaux non musculo-invasifs, en particulier les caractéristiques immunosuppressives,¦comme prédictif d'un comportement de progression néoplasique.¦Méthodologie : L'exploration d'une base de données de patients suivis pour le cancer de la vessie au¦CHUV afin de faire un choix des coupes de RTUV à examiner a été réalisée. Une approche transversale a été¦abordée en regroupant des patients de stades tumoraux différents ou en évaluant des foyers tumoraux¦multiples au sein de la vessie d'un même patient à un moment donné (i.e lors d'une même RTUV). Une¦approche longitudinale a également été adoptée avec comme objectif de comparer, chez le même patient,¦des tumeurs de stades, de grades et de score de risque de progression de l'EORTC différents au cours du¦temps. L'ARN des tissus de RTUV fixés au formol et enrobés en paraffine a été extrait et purifié. Un kit¦d'amplification en temps réel par réaction en chaîne par polymérase (RCP) pour 84 gènes impliqués dans¦l'anergie des cellules T et la tolérance immunitaire a été utilisé.¦Résultat : Nous avons réuni les informations cliniques de 157 sujets atteints de tumeur vésicale non¦musculo-invasive. 35% des sujets ont reçu une chimiothérapie ives et 40 % ont reçu du BCG ives. Les troisquarts¦de ces derniers ont reçu un cycle de BCG complet (6 semaines). Néanmoins, 38 % d'entre eux vont¦tout de même subir une progression de leur cancer. Les scores de progression de l'EORTC ont pu être¦calculés pour 94 sujets (39% haut risque, 42 % risque intermédiaire et 19% bas risque). 76% des patients à¦haut risque a été traité avec du BCG ives. Parmi les patients avec un risque intermédiaire de progression,¦seuls 15 % ont effectivement progressé incluant 2 patients avec des échantillons de RTUV disponibles pour¦analyse. L'analyse par RCP s'est focalisée sur une approche longitudinale incluant 6 patients suivis sur une¦longue période avec de multiples RTUV. 29 échantillons ont été sélectionnés, leur ARN purifiés, mais seuls¦16 ARN se sont révélés en quantité et qualité suffisante pour être analysé par RCP. L'analyse par RCP¦quantitative en temps réel a montré des problèmes dans la quantification de l'ADN génomique, ainsi que¦des gènes domestiques. Ceci a grandement handicapé nos analyses et n'a pas permis la mise en évidence¦convaincante de gènes immuno-modulateurs associés à la progression du cancer de la vessie.¦Discussion : Notre analyse du suivi des patients au CHUV montre que les chirurgiens façonnent leur prise¦en charge durant l'intervention selon des critères adaptés à la situation et tendent ainsi à une stratification¦des risques permettant un traitement adapté, de la même manière que le permet le score de l'EORTC, en¦tous cas en ce qui concerne les patients à haut risque de progression. Les nombreux facteurs impliqués¦dans le cancer de la vessie montrent néanmoins qu'il y aurait des avantages à rationaliser la prise en charge. L'archivage de tissus fixé au formol et enrobé en paraffine a l'avantage de représenter une source¦de matériel considérable et de grande valeur pour la recherche. Néanmoins, malgré l'évolution des¦techniques et la publicité des fabricants, il s'est avéré difficile d'exploiter ce matériel délicat pour en¦obtenir des résultats convaincants.

ABSTRACT: Transapical aortic valve replacement is an established technique performed in high-risk patients with symptomatic aortic valve stenosis and vascular disease contraindicating trans-vascular and trans-aortic procedures. The presence of a left ventricular apical diverticulum is a rare event and the treatment depends on dimensions and estimated risk of embolisation, rupture, or onset of ventricular arrhythmias. The diagnosis is based on standard cardiac imaging and symptoms are very rare. In this case report we illustrate our experience with a 81 years old female patient suffering from symptomatic aortic valve stenosis, respiratory disease, chronic renal failure and severe peripheral vascular disease (logistic euroscore: 42%), who successfully underwent a transapical 23 mm balloon-expandable stent-valve implantation through an apical diverticulum of the left ventricle. Intra-luminal thrombi were absent and during the same procedure were able to treat the valve disease and to successfully exclude the apical diverticulum without complications and through a mini thoracotomy. To the best of our knowledge, this is the first time that a transapical procedure is successfully performed through an apical diverticulum.

Aquaglyceroporin-9 (AQP9) facilitates diffusion of water and energy substrates such as glycerol and monocarboxylates. AQP9 is present in plasma membrane and mitochondria of astrocytes and catecholaminergic neurons, suggesting that it plays a role in the energetic status of these cells. Using specific small interference RNA directed against AQP9 in astrocyte cultures, we showed that glycerol uptake is decreased which is associated with an increase in glucose uptake and oxidative metabolism. Our results not only confirm the presence of AQP9 in astrocytes but also suggest that changes in AQP9 expression alter glial energy metabolism.

The expected therapeutic gain of a combined radioimmunotherapy (RIT) with conventional radiotherapy (RT) would be a synergy of tumor irradiation, provided that toxic, dose-limiting side effects concern different organs. We have shown in a model of subcutaneous human colon cancer transplants in nude mice that RIT with 131I-labeled anti-CEA antibody fragments combined with fractionated RT give an additive therapeutic effect without increase of side effects. A second study of different timing schedules of RIT and RT has shown that close association of both therapies without delay is more efficient than a therapy with a treatment-free interval of two weeks. In a new model of human colon cancer liver metastases in nude mice, early treatment with RIT and with RT has been curative, whereas therapies initiated later were less efficient, suggesting that the combined therapy is likely to be more efficient in an adjuvant situation after surgery. At the clinical level, six patients with limited liver metastatic disease from colorectal cancer were treated with RIT using 200 mCi 131I-labeled anti-CEA MAb F(ab')2 fragments combined with fractionated external beam RT of 20 Gy to the entire liver. As expected, spontaneously reversible bone marrow toxicity grade 3 to 4 and reversible liver toxicity grade 1 to 3 have been observed. By computerized tomography, three patients showed stable disease and one patient partial remission, whereas two patients had progressive disease. In conclusion, animal experiments have shown a clear advantage of combined RT and RIT, and the clinical study shows the feasibility of such a therapy in patients with colorectal cancer liver metastases.

Aim This study used data from temperate forest communities to assess: (1) five different stepwise selection methods with generalized additive models, (2) the effect of weighting absences to ensure a prevalence of 0.5, (3) the effect of limiting absences beyond the environmental envelope defined by presences, (4) four different methods for incorporating spatial autocorrelation, and (5) the effect of integrating an interaction factor defined by a regression tree on the residuals of an initial environmental model. Location State of Vaud, western Switzerland. Methods Generalized additive models (GAMs) were fitted using the grasp package (generalized regression analysis and spatial predictions, http://www.cscf.ch/grasp). Results Model selection based on cross-validation appeared to be the best compromise between model stability and performance (parsimony) among the five methods tested. Weighting absences returned models that perform better than models fitted with the original sample prevalence. This appeared to be mainly due to the impact of very low prevalence values on evaluation statistics. Removing zeroes beyond the range of presences on main environmental gradients changed the set of selected predictors, and potentially their response curve shape. Moreover, removing zeroes slightly improved model performance and stability when compared with the baseline model on the same data set. Incorporating a spatial trend predictor improved model performance and stability significantly. Even better models were obtained when including local spatial autocorrelation. A novel approach to include interactions proved to be an efficient way to account for interactions between all predictors at once. Main conclusions Models and spatial predictions of 18 forest communities were significantly improved by using either: (1) cross-validation as a model selection method, (2) weighted absences, (3) limited absences, (4) predictors accounting for spatial autocorrelation, or (5) a factor variable accounting for interactions between all predictors. The final choice of model strategy should depend on the nature of the available data and the specific study aims. Statistical evaluation is useful in searching for the best modelling practice. However, one should not neglect to consider the shapes and interpretability of response curves, as well as the resulting spatial predictions in the final assessment.

PPARs are nuclear hormone receptors which, like the retinoid, thyroid hormone, vitamin D, and steroid hormone receptors, are ligand-activated transcription factors mediating the hormonal control of gene expression. Two lines of evidence indicate that PPARs have an important function in fatty acid metabolism. First, PPARs are activated by hypolipidemic drugs and physiological concentrations of fatty acids, and second, PPARs control the peroxisomal beta-oxidation pathway of fatty acids through transcriptional induction of the gene encoding the acyl-CoA oxidase (ACO), which is the rate-limiting enzyme of the pathway. Furthermore, the PPAR signaling pathway appears to converge with the 9-cis retinoic acid receptor (RXR) signaling pathway in the regulation of the ACO gene because heterodimerization between PPAR and RXR is essential for in vitro binding to the PPRE and because the strongest stimulation of this gene is observed when both receptors are exposed simultaneously to their activators. Thus, it appears that PPARs are involved in the 9-cis retinoic acid signaling pathway and that they play a pivotal role in the hormonal control of lipid metabolism.

We introduce a microscopic method that determines quantitative optical properties beyond the optical diffraction limit and allows direct imaging of unstained living biological specimens. In established holographic microscopy, complex fields are measured using interferometric detection, allowing diffraction-limited phase measurements. Here, we show that non-invasive optical nanoscopy can achieve a lateral resolution of 90 nm by using a quasi-2 pi-holographic detection scheme and complex deconvolution. We record holograms from different illumination directions on the sample plane and observe subwavelength tomographic variations of the specimen. Nanoscale apertures serve to calibrate the tomographic reconstruction and to characterize the imaging system by means of the coherent transfer function. This gives rise to realistic inverse filtering and guarantees true complex field reconstruction. The observations are shown for nanoscopic porous cell frustule (diatoms), for the direct study of bacteria (Escherichia coil), and for a time-lapse approach to explore the dynamics of living dendritic spines (neurones).

Olfactory systems are evolutionarily ancient, underlying the common requirement for all animals to sense and respond to diverse volatile chemical signals in their environment. Odor detection is mediated by odorant receptors (ORs) that, in most olfactory systems, comprise large families of divergent G protein-coupled receptors. Here, I discuss our and others' recent investigations of ORs in the fruit fly, Drosophila melanogaster, which have revealed insights into the distinct evolutionary origin and molecular function of insect ORs. I also describe a bioinformatics strategy that we developed to identify molecules that function with these insect-specific receptors in odor detection.

Eosinophilic esophagitis (EoE) shows an increasing incidence and prevalence in western countries and is currently the main cause of dysphagia in adults. This disease is more prevalent in males and is frequently associated with allergies. Diagnosis is based on the presence of esophageal symptoms, dense eosinophilic esophageal infiltration, and the exclusion of other conditions associated with esophageal eosinophilia. Topical corticosteroids lead to a rapid clinical and histological improvement of active EoE. Especially in children, elimination diets can have similar efficacy as topical corticosteroids. Esophageal dilation of EoE-induced strictures can also be effective in improving symptoms, but this therapy has no effect on the underlying inflammation. Long-term therapeutic strategies have yet to be defined.

The aim of this contribution is to explore how the recent internationalization and the increasing importance of 'cosmopolitan capital' has impacted on the structure and character of the field of the Swiss business elite. For this purpose we will develop the notion of cosmopolitan capital and comparatively investigate the field of the Swiss business elite in 1980, 2000 and 2010 with multiple correspondence analysis. We can show that in this period international managers with transnational careers and networks not only grow in number, but come to conquer the apex of the biggest and highest capitalized Swiss firms. At the same time, national forms of capital decline in importance and Swiss managers themselves are differentiated increasingly into national and international fractions.

A new generation of microcapsules based on the use of oligomers which participate in polyelectrolyte complexation reactions has been developed. These freeze-thaw stable capsules have been applied as a bioartificial pancreas and have resulted in normoglycemia for periods of six months in concordant xenotransplantations. The new chemistry permits the control of permeability and mechanical properties over a wide range and can be adapted both to microcapsule and hollow fiber geometries rendering it a robust tool for encapsulation in general. Methods, and metrics, for the characterization of the mechanical properties and permeability of microcapsules are presented.

We carried out a cross-section study of the sex-specific relationship between bone mineral content and physical activity at sites with different loading in pre- and early pubertal girls and boys. There was significant sensitivity of bone mineral content of the hip to physical exercise in boys, but not in girls. BACKGROUND: Since little is known whether there are sex differences in sensitivity of bone to loading, we investigated sex differences in the cross-sectional association between measures of physical activity (PA) and bone mass and size in pre- and early pubertal children of both sexes. METHODS: We measured bone mineral content/density (BMC/BMD) and fat-free mass (FFM) in 269 6- to 13-year-old children from randomly selected schools by dual-energy X-ray absorptiometry. Physical activity (PA) was measured by accelerometers and lower extremity strength by a jump-and-reach test. RESULTS: Boys (n = 128) had higher hip and total body BMC and BMD, higher FFM, higher muscle strength and were more physically active than girls (n = 141). Total hip BMC was positively associated with time spent in total and vigorous PA in boys (r = 0.20-0.33, p < 0.01), but not in girls (r = 0.02-0.04, p = ns), even after adjusting for FFM and strength. While boys and girls in the lowest tertile of vigorous PA (22 min/day) did not differ in hip BMC (15.62 vs 15.52 g), boys in the highest tertile (72 min/day) had significantly higher values than the corresponding girls (16.84 vs 15.71 g, p < 0.05). CONCLUSIONS: Sex differences in BMC during pre- and early puberty may be related to a different sensitivity of bone to physical loading, irrespective of muscle mass.
