Airborne microbial products have been reported to promote immune responses that suppress asthma, yet how these beneficial effects take place remains controversial and poorly understood. We have found that pulmonary exposure with the bacterium Escherichia coli leads to a suppression of allergic airway inflammation, characterized by reduced airway-hyperresponsiveness, eosinophilia and cytokine production by T cells in the lung. This immune modulation was neither mediated by the induction of a Th1 response nor regulatory T cells; was dependent on TLR-4 but did not involve TLR-desensitization. Dendritic cell migration to the draining lymph nodes and subsequent activation of T cells was unaffected by prior exposure to E.coli indicating that the immunomodulation was limited to the lung environment. In non-treated control mice ovalbumin was primarily presented by airway CD11b+ CD11c+ DCs expressing high levels of MHC class II molecules whilst the DCs in E.coli-treated mice displayed a less activated phenotype and had impaired antigen presentation capacity. Consequently, in situ Th2 cytokine production by ovalbuminspecific effector T cells recruited to the airways was significantly reduced. The suppression of airways hyper responsiveness was mediated through the recruitment of IL-17-producing gd-T cells; however, the suppression of dendritic cells and T cells was mediated through a distinct mechanism that could not be overcome by the local administration of activated dendritic cells, or by the in vivo administration of TNF-alpha. Taken together, these data reveal a novel multi-component immunoregulatory pathway that acts to protect the airways from allergic inflammation.

There is a renewal of interest among psychotherapy researchers and psychotherapists towards psychotherapy case studies. This article presents two paradigms that have greatly influenced this increasing interest in psychotherapy case studies : the pragmatic case study and the theory-building case study paradigm. The origins, developments and key-concepts of both paradigms are presented, as well as their methodological and ethical specificities. Examples of case studies, along with models developed, are cited. The differential influence of the post-modern schools on both paradigms are presented, as well as their contribution to the field of methods of psychotherapy case studies discussed and assessed in terms of relevance for the researcher and the psychotherapist.

The research reported in this series of article aimed at (1) automating the search of questioned ink specimens in ink reference collections and (2) at evaluating the strength of ink evidence in a transparent and balanced manner. These aims require that ink samples are analysed in an accurate and reproducible way and that they are compared in an objective and automated way. This latter requirement is due to the large number of comparisons that are necessary in both scenarios. A research programme was designed to (a) develop a standard methodology for analysing ink samples in a reproducible way, (b) comparing automatically and objectively ink samples and (c) evaluate the proposed methodology in forensic contexts. This report focuses on the last of the three stages of the research programme. The calibration and acquisition process and the mathematical comparison algorithms were described in previous papers [C. Neumann, P. Margot, New perspectives in the use of ink evidence in forensic science-Part I: Development of a quality assurance process for forensic ink analysis by HPTLC, Forensic Sci. Int. 185 (2009) 29-37; C. Neumann, P. Margot, New perspectives in the use of ink evidence in forensic science-Part II: Development and testing of mathematical algorithms for the automatic comparison of ink samples analysed by HPTLC, Forensic Sci. Int. 185 (2009) 38-50]. In this paper, the benefits and challenges of the proposed concepts are tested in two forensic contexts: (1) ink identification and (2) ink evidential value assessment. The results show that different algorithms are better suited for different tasks. This research shows that it is possible to build digital ink libraries using the most commonly used ink analytical technique, i.e. high-performance thin layer chromatography, despite its reputation of lacking reproducibility. More importantly, it is possible to assign evidential value to ink evidence in a transparent way using a probabilistic model. It is therefore possible to move away from the traditional subjective approach, which is entirely based on experts' opinion, and which is usually not very informative. While there is room for the improvement, this report demonstrates the significant gains obtained over the traditional subjective approach for the search of ink specimens in ink databases, and the interpretation of their evidential value.

Dopaminergic, serotonergic and noradrenergic nuclei form the trimonoamine modulating system (TMMS). This system modulates emotional/motivational activities mediated by the limbic circuitry, where glutamate is the major excitatory neurotransmitter. Two main concepts are the basis of this review. First, since 1950 and the discovery of the antipsychotic activity of the dopamine D2 receptor antagonist chlorpromazine, it appears that drugs that can modulate the TMMS possess therapeutic psychiatric properties. Second, the concept of glutamate/trimonoamine imbalance in the cortico-striato-thalamo-cortical loop that has been so successful in explaining the pathophysiology of Parkinson disease has been applied in the pathophysiology of schizophrenia. This review will focus on the complex interactions between the fast synaptic glutamatergic transmission and the TMMS in specific parts of the limbic lobe and we will try to link these interactions to some psychiatric disorders, mainly depression, schizophrenia and drug addiction.

En matière de loteries et paris, presque tous les Etats européens ont mis en place un système plus ou moins similaire reposant sur un principe d'interdiction des grandes loteries et des paris, sauf exception. L'exception consiste à accorder l'autorisation d'organiser ces jeux à un organisateur unique qui dispose ainsi d'un monopole sur son territoire d'autorisation, et à imposer que les bénéfices soient intégralement reversés à l'utilité publique, à l'exemple de la Suisse, ou affectés au budget de l'Etat, à l'exemple de la France. Dans une perspective légistique, ce travail de recherche propose de tracer un état des lieux de la situation actuelle en Suisse, sous l'angle juridique et économique, et, sous l'éclairage des éléments remettant en cause le monopole, de discuter cinq scénarii envisageables pour l'avenir accompagnés des conséquences juridiques, économiques et sociales pour chacun d'eux. Ces réflexions conduisent à constater que le cadre juridique n'est plus adapté aux tendances actuelles. Die Regelung der Lotterie- und Wettmärkte ist in fast allen europäischen Staaten im Wesentlichen ähnlich: es besteht ein grundsätzliches Verbot der Durchführung von grossen Lotterien und Wetten, das durch eine Ausnahmeregelung zugunsten eines einzigen oder eines stark eingeschränkten Kreises von Spielveranstaltern relativiert wird. Die Spielveranstalter verfügen über ein Exklusivrecht zur Durchführung dieser Glücksspiele auf ihrem Vertriebsgebiet. Damit verbunden ist die Pflicht, die generierten Erträge vollumfänglich entweder - wie in der Schweiz - so genannt "gemeinnützigen Zwecken" oder - wie etwa in Frankreich - der Staatskasse zufliessen zu lassen. Im Rahmen der vorliegenden Forschungsarbeit wird zunächst aus legistischer Perspektive die aktuelle Situation im Lotterie- und Wettbereich in der Schweiz analysiert. Im Lichte der Elemente, die das erwähnte Exklusivrecht in Frage stellen, werden sodann fünf mögliche Zukunftsszenarien mit ihren juristischen, wirtschaftlichen und sozialen Konsequenzen skizziert. Die Untersuchung kommt zum Schluss, dass der heute in der Schweiz geltende rechtliche Rahmen nicht mehr den aktuellen Entwicklungen entspricht.
