S'ha realitzat un estudi retrospectiu de 41 casos clínics de queratocon mesurant les variables abans i després de la intervenció (al primer mes postoperatori, als 3 mesos i als 6 mesos). El crosslinking corneal amb riboflavina i radiació ultraviolada sembla ser un procediment segur i efectiu, que ha assenyalat la parada a la progressió del queratocon, una reducció, encara que no estadísticament significativa de la curvatura corneal, de l'equivalent esfèric i de l'astigmatisme en ulleres a pacients que prèviament havien manifestat una progressió del queratocon. Conclusions que podem treure d'aquest estudi: 1) Es pot esperar una mínima millora de l'agudesa visual o una detecció la pèrdua d'aquesta en els casos de queratocon progressiu. Encara que aquesta millora no va ser estadísticament significativa. 2) No es demostra que el crosslinking freni la progressió del queratocon, però sí observem un augment de les mitjanes dels valors que ens indiquen el grau de duresa corneal comparant els valors abans de la intervenció, i als 3 i 6 mesos, encara que no hagi diferència estadísticament significativa. Podem constatar que el Crosslinking és una tècnica més que podem emprar en el tractament del queratocon. Possiblement el seu efecte en la visió no serà molt apreciable, però davant de casos amb alt risc de progressió pot ser factible el seu ús ja que no té efectes secundaris d'importància.

The inner ear is responsible for the perception of motion and sound in vertebrates. Its functional unit, the sensory patch, contains mechanosensory hair cells innervated by sensory neurons from the statoacoustic ganglion (SAG) that project to the corresponding nuclei in the brainstem. How hair cells develop at specific positions, and how otic neurons are sorted to specifically innervate each endorgan and to convey the extracted information to the hindbrain is not completely understood. In this work, we study the generation of macular sensory patches and investigate the role of Hedgehog (Hh) signaling in the production of their neurosensory elements. Using zebrafish transgenic lines to visualize the dynamics of hair cell and neuron production, we show that the development of the anterior and posterior maculae is asynchronic, suggesting they are independently regulated. Tracing experiments demonstrate the SAG is topologically organized in two different neuronal subpopulations, which are spatially segregated and innervate specifically each macula. Functional experiments identify the Hh pathway as crucial in coordinating the production of hair cells in the posterior macula, and the formation of its specific innervation. Finally, gene expression analyses suggest that Hh influences the balance between different SAG neuronal subpopulations. These results lead to a model in which Hh orients functionally the development of inner ear towards an auditory fate in all vertebrate species.

Genes of interest can be targeted specifically to respiratory epithelial cells in intact animals with high efficiency by exploiting the receptor-mediated endocytosis of the polymeric immunoglobulin receptor. A DNA carrier, consisting of the Fab portion of polyclonal antibodies raised against rat secretory component covalently linked to poly-L-lysine, was used to introduce plasmids containing different reporter genes into airway epithelial cells in vivo. We observed significant levels of luciferase enzyme activity in protein extracts from the liver and lung, achieving maximum values of 13,795 +/- 4,431 and 346,954 +/- 199,120 integrated light units (ILU) per milligram of protein extract, respectively. No luciferase activity was detected in spleen or heart, which do not express the receptor. Transfections using complexes consisting of an irrelevant plasmid (pCMV lacZ) bound to the bona fide carrier or the expression plasmid (pGEMluc) bound to a carrier based on an irrelevant Fab fragment resulted in background levels of luciferase activity in all tissues examined. Thus, only tissues that contain cells bearing the polymeric immunoglobulin receptor are transfected, and transfection cannot be attributed to the nonspecific uptake of an irrelevant carrier-DNA complex. Specific mRNA from the luciferase gene was also detected in the lungs of transfected animals. To determine which cells in the lungs are transfected by this method, DNA complexes were prepared containing expression plasmids with genes encoding the bacterial beta-galactosidase or the human interleukin 2 receptor. Expression of these genes was localized to the surface epithelium of the airways and the submucosal glands, and not the bronchioles and alveoli. Receptor-mediated endocytosis can be used to introduce functional genes into the respiratory epithelium of rats, and may be a useful technique for gene therapy targeting the lung.

Estudio elaborado a partir de una estancia en la Universidad de Rochester, Estados Unidos, de octubre del 2006 a enero del 2007. La estancia realizada en la Universidad de Rochester estuvo orientada al aprendizaje en profundidad del oftalmoscopio láser de barrido. El oftalmoscopio láser de barrido emplea una técnica confocal con la finalidad de visualizar diferentes estructuras retinianas en seres vivos. El instrumento diseñado y desarrollado en el Centro de Ciencias de la Visión incorpora un sistema de óptica adaptativa y fluorescencia. La óptica adaptativa aplicada en este oftalmoscopio tiene como objetivo corregir las aberraciones existentes en el ojo y así permitir observar detalles de la retina que de otra forma se verían emborronados. De esta forma se consigue alcanzar valores de resolución muy cercanos a los impuestos por difracción. Por otro lado el uso de fluorescencia tiene por objetivo el permitir la visualización de células y estructuras que, de no ser teñidas, son transparentes a la luz y visible. Esta técnica se ha estado utilizando principalmente en primates y ratas, aunque actualmente también se están llevando a cabo medidas de células de epitelio pigmentario en seres humanos ya que el pigmento contenido en estas células permite la aplicación de la fluorescencia sin necesidad de utilizar tinción.

La distribución de alimentos líquidos al ganado porcino es un sistema alternativo en el cual el alimento se prepara y distribuye a través de un equipo totalmente informatizado y automatizado. Además de materias primas y piensos permite también el uso de co-productos líquidos de la industria agroalimentaria o productos fermentados. Su utilización evita el coste de su transformación o eliminación permitiendo rebajar el coste de producción del cerdo porque su precio es altamente competitivo. La aplicación de alimentos líquidos fermentados tiene efectos benéficos sobre el epitelio intestinal, la microflora digestiva y el estado sanitario de los lechones, reduciendo la morbilidad y aumentando los resultados de crecimiento. Este sistema de alimentación facilita igualmente la aplicación de programas multifase ajustando diariamente el aporte a las necesidades en nutrientes del cerdo reduciendo la excreción (N, P, metales pesados) y con ello, el riesgo de contaminación medioambiental. The distribution of liquid diets for swine is an alternative system where feed is prepared and distributed by an automatic computerized device. In addition to feedstuffs and complementary dry feed, this system allows the use of fresh or fermented agri-food liquid by-products. These products are cheaper and its use as feed avoids the cost of its transformation or elimination, reducing the environmental load and the cost per kg of pig meat. The fermented liquid feed protects the intestinal epithelium, helps to control digestive microflora and animal health improving piglet growth performance after weaning. This feeding device also helps implementation of multiphase feeding programs, adjusting nutrient supplies to requirements daily, and consequently reducing the nutrient excretion (N, P, heavy metals) and the risk of environmental pollution.

La incidència d’hipertròfia benigna de pròstata és aproximadament d’un 50 % en pacients majors de 50 anys. La Tamsulosina és el medicament més prescrit per aquesta dolència i la principal causa del síndrome d’iris flàccid intraoperatori a la cirurgia de cataractes. Els nostres dos estudis han sigut; per una banda, avaluar la toxicitat corneal i afectació cardíaca de Fenilefrina intracamerular a una concentració de 2,5% respecte la pauta de dilatació tòpica estàndard al nostre centre, i per l’altra, comparar l’ efectivitat i diferència al recompte endotelial de Fenilefrina intracamerular a una concentració de 2,5% respecte a una altra concentració del 1,66 %, mesurant els seus efectes en diverses variables.

Objectiu: valorar l'índex de recidiva simple o recidiva clínicament significativa dels diferents tipus de distròfies estromals anteriors en empelts corneals després cirurgia de queratoplàstia. Material i mètodes: S'ha realitzat una revisió retrospectiva del 1954 al 2008 al Centre d'Oftalmologia Barraquer, identificant a tots els pacients diagnosticats de distròfia corneal estromal anterior i de la membrana de Bowman (Distròfia de Reis Bücklers, Distròfia Granular, Distròfia reticular, distròfia macular) que han estat intervinguts de queratoplàstia penetrant (QP) o queratoplàstia laminar (QL). S'han utilitzat estadístics descriptius i la funció de supervivència amb les corbes de Kaplan Meier per dur a terme l'anàlisi de la mostra. Resultats: La mostra total del nostre estudi ha estat de 109 ulls de 66 pacients amb distròfies estromals anteriors operats de queratoplàstia: 8 casos de distròfies de la capa de Bowman (6 ulls amb CDRB o CDB-I (distròfia de Reis Bücklers), i 2 ulls amb CDTB o CDB-I (distròfia de Thiel Behnke)), 19 casos de distròfies granulars (CDG), 53 casos de distròfia de lattice (CDL) i 29 casos de distròfia macular (CDM). Amb un llarg de temps de seguiment (75-180 mesos de mitjana), la recurrència simple ha estat del 33% dels casos de CDRB amb 46 mesos de temps mitjà de supervivència, del 58, 8% dels casos de CDG amb 74.6 mesos de supervivència, el 41'5% de les CDL amb 106 mesos de supervivència i en el 10% dels casos amb CDM amb 96 mesos de temps mitjà de supervivència. La recurrència clínicament significativa es va manifestar en la seva majoria en els casos de CDL i CDG amb un temps mitjà de supervivència de 127.6 mesos i 124.1 mesos respectivament. El símptoma principal d'aquestes manifestacions clíniques va ser la disminució de la AV. Conclusió: La queratoplàstia penetrant és un tractament clàssic eficaç per als casos de distròfies estromals anteriors simptomàtics, amb opacitats profundes en l'estroma o fracàs previ d'altres teràpies menys invasives com la PTK. Com és d'esperar, per la seva origen genètic, l'aparició de recurrència de la distròfia després del trasplantament existeix i s'incrementa amb el major temps de seguiment. La tendència d'aquesta recidiva és a presentar inicialment en el pla cornial epitelial i requereix un llarg temps d'evolució per fer-se simptomàtica, especialment en els casos de CDG, CDL i CDM.

La distròfia endotelial de Fuchs es caracteritza per la formació de guttas endotelials i en estadis avançats pot induir edema corneal i pèrdua d’agudesa visual. A diferència del microscopi especular, el microscopi confocal té un disseny que permet evitar la llum aberrant causada per l’edema corneal o opacitats estromals. Comparant ambdues probes en la valoració de l’endoteli corneal en pacients amb distròfia de Fuchs, s’observa millor qualitat d’imatge per microscòpia confocal tot i que no es podia valorar la fiabilitat de l’esmentada proba en la determinació de la densitat cel•lular endotelial en les còrnies afectades.

We performed complete eye exams on 50 eyes in 25 patients with congenital aniridia. Factors such as age, history of ocular surgery, dry eye score and aesthesiometry results correlated with the degree of aniridia-related keratopathy. Schirmer’s test I in 86.8%, Schirmer’s test II in 94.4% and TFBUT in 83.3% of cases were all normal. Corneal staining was altered in 54.2%, and conjunctival staining was altered in 45.7%. The tear ferning pattern was abnormal in 80%. Conjunctival metaplasia was present in 76.9%.Corneal endothelial cell density was normal. Ultrasonic pachymetry was higher than average in all eyes examined.

A study of the spermiogenesis and spermatozoa of Helicolenus dactylopterus was conducted. Females of this species have the capacity to store sperm within their ovaries, and male gametes have a considerable cytoplasmic mass surrounding their heads to survive the long period of intraovarian sperm storage. Our observations show that early spermatids are round-shaped cells and have a spherical nucleus with diffuse chromatin. The nuclear volume decreases as a result of progressive chromatin condensation during spermiogenesis, causing the nucleus to take on a U-shape. Flagellar insertion is not central to the nucleus but consistently occurs at an oblique angle towards one side of it. The flagellum is inserted into the nuclear fossa, without subsequent nuclear rotation. In mature spermatozoa, the flagellum is adjacent to the nucleus. A comparison of the spermatozoa in the testicular lobules and those in the intraovarian storage structures suggests that the increase in volume of the cytoplasmic mass may occur in the posterior region of the testis, in the testicular duct. Spermatozoa enter the ovary in groups that reach the ovarian lumen and are surrounded by the ovarian epithelium for storage in sperm storage crypts

BACKGROUND: Cytoskeletal changes after longterm exposure to ethanol have been described in a number of cell types in adult rat and humans. These changes can play a key part in the impairment of nutrient assimilation and postnatal growth retardation after prenatal damage of the intestinal epithelium produced by ethanol intake. AIMS: To determine, in the newborn rat, which cytoskeletal proteins are affected by longterm ethanol exposure in utero and to what extent. ANIMALS: The offspring of two experimental groups of female Wistar rats: ethanol treated group receiving up to 25% (w/v) of ethanol in the drinking fluid and control group receiving water as drinking fluid. METHODS: Single and double electron microscopy immunolocalisation and label density estimation of cytoskeletal proteins on sections of proximal small intestine incubated with monoclonal antibodies against actin, alpha-tubulin, cytokeratin (polypeptides 1, 5, 6, 7, 8, 10, 11, and 18), and with a polyclonal antibody anti-beta 1,4-galactosyl transferase as trans golgi (TG) or trans golgi network (TGN) marker, or both. SDS-PAGE technique was also performed on cytoskeletal enriched fractions from small intestine. Western blotting analysis was carried out by incubation with the same antibodies used for immunolocalisation. RESULTS: Intestinal epithelium of newborn rats from the ethanol treated group showed an overexpression of cytoskeletal polypeptides ranging from 39 to 54 kDa, affecting actin and some cytokeratins, but not tubulin. Furthermore, a cytokeratin related polypeptide of 28-29 kDa was identified together with an increase in free ubiquitin in the same group. It was noteworthy that actin and cytokeratin were abnormally located in the TG or the TGN, or both. CONCLUSIONS: Longterm exposure to ethanol in utero causes severe dysfunction in the cytoskeleton of the developing intestinal epithelium. Actin and cytokeratins, which are involved in cytoskeleton anchoring to plasma membrane and cell adhesion, are particularly affected, showing overexpression, impaired proteolysis, and mislocalisation.

Transforming growth factor alpha (TGF alpha) is a polypeptide, which binds to the epidermal growth factor receptor to carry out its function related to cell proliferation and differentiation. The ultrastructural localisation of TGF alpha was studied in both the proximal and the distal colon. The columnar cells, lining the surface epithelium of the proximal colon, showed a strong immunoreactivity in the polyribosomes and in the interdigitations of the lateral membrane. The columnar cells of the crypts and the goblet cells in both the proximal and the distal colon showed the immunostaining in the cis and trans cisternae of the Golgi apparatus. TGF alpha seems to be processed differently in the surface columnar cells and in the crypt columnar cells and goblet cells. Moreover, it probably has different roles in proliferation and differentiation.
