Pd-catalysed reactions of 2-Cl, 2-Br and 2-I derivatives of a 6-chloropurine nucleoside with benzamide have been compared, using Pd2dba3, Xantphos and Cs2CO3 in toluene, between 20 and 80 °C. The reactivity order was 2-I > 2-Br > 6-Cl ≫ 2-Cl. The 2-I substituent could be replaced even at 0 °C, under conditions disclosed here for the first time. On the other hand, the replacement of the chlorine atom at position 2 (2-Cl) required 110 °C.

6-Phosphofructo-2-kinase/fructose-2,6-bisphosphatase (PFKFB)catalyzes the synthesis and degradation of fructose-2,6-bisphosphate, a key modulator of glycolysis-gluconeogenesis. To gain insight into the molecular mechanism behind hormonal and nutritional regulation of PFKFB expression, we have cloned and characterized the proximal promoter region of the liver isoform of PFKFB (PFKFB1) from gilthead sea bream (Sparus aurata). Transient transfection of HepG2 cells with deleted gene promoter constructs and electrophoretic mobility shift assays allowed us to identify a sterol regulatory element (SRE) to which SRE binding protein-1a (SREBP-1a)binds and transactivates PFKFB1 gene transcription. Mutating the SRE box abolished SREBP-1a binding and transactivation. The in vivo binding of SREBP-1a to the SRE box in the S. aurata PFKFB1 promoter was confirmed by chromatin immunoprecipitation assays. There is a great deal of evidence for a postprandial rise of PFKB1 mRNA levels in fish and rats. Consistently, starved-to-fed transition and treatment with glucose or insulin increased SREBP-1 immunodetectable levels, SREBP-1 association to PFKFB1 promoter, and PFKFB1 mRNA levels in the piscine liver. Our findings demonstrate involvement of SREBP-1a in the transcriptional activation of PFKFB1, and we conclude that SREBP-1a may exert a key role mediating postprandial activation of PFKFB1 transcription.

Here we investigate the formation of superficial micro- and nanostructures in poly(ethylene-2,6-naphthalate) (PEN), with a view to their use in biomedical device applications, and compare its performance with a polymer commonly used for the fabrication of these devices, poly(methyl methacrylate) (PMMA). The PEN is found to replicate both micro- and nanostructures in its surface, albeit requiring more forceful replication conditions than PMMA, producing a slight increase in surface hydrophilicity. This ability to form micro/nanostructures, allied to biocompatibility and good optical transparency, suggests that PEN could be a useful material for production of, or for incorporation into, transparent devices for biomedical applications. Such devices will be able to be autoclaved, due to the polymer's high temperature stability, and will be useful for applications where forceful experimental conditions are required, due to a superior chemical resistance over PMMA.

Conjugates of a dicarba analogue of octreotide, a potent somatostatin agonist whose receptors are overexpressed on tumor cells, with [PtCl2(dap)] (dap = 1-(carboxylic acid)-1,2-diaminoethane) (3), [(η6-bip)Os(4-CO2-pico)Cl] (bip = biphenyl, pico = picolinate) (4), [(η6-p-cym)RuCl(dap)]+ (p-cym = p-cymene) (5), and [(η6-p-cym)RuCl(imidazole-CO2H)(PPh3)]+ (6), were synthesized by using a solid-phase approach. Conjugates 35 readily underwent hydrolysis and DNA binding, whereas conjugate 6 was inert to ligand substitution. NMR spectroscopy and molecular dynamics calculations showed that conjugate formation does not perturb the overall peptide structure. Only 6 exhibited antiproliferative activity in human tumor cells (IC50 = 63 ± 2 μM in MCF-7 cells and IC50 = 26 ± 3 μM in DU-145 cells) with active participation of somatostatin receptors in cellular uptake. Similar cytotoxic activity was found in a normal cell line (IC50 = 45 ± 2.6 μM in CHO cells), which can be attributed to a similar level of expression of somatostatin subtype-2 receptor. These studies provide new insights into the effect of receptor-binding peptide conjugation on the activity of metal-based anticancer drugs, and demonstrate the potential of such hybrid compounds to target tumor cells specifically.

TCF7L2 is the susceptibility gene for Type 2 diabetes (T2D) with the largest effect on disease risk that has been discovered to date. However, the mechanisms by which TCF7L2 contributes to the disease remain largely elusive. In addition, epigenetic mechanisms, such as changes in DNA methylation patterns, might have a role in the pathophysiology of T2D. This study aimed to investigate the differences in terms of DNA methylation profile of TCF7L2 promoter gene between type 2 diabetic patients and age- and Body Mass Index (BMI)- matched controls. We included 93 type 2 diabetic patients that were recently diagnosed for T2D and exclusively on diet (without any pharmacological treatment). DNA was extracted from whole blood and DNA methylation was assessed using the Sequenom EpiTYPER system. Type 2 diabetic patients were more insulin resistant than their matched controls (mean HOMA IR 2.6 vs 1.8 in controls, P<0.001) and had a poorer beta-cell function (mean HOMA B 75.7 vs. 113.6 in controls, P<0.001). Results showed that 59% of the CpGs analyzed in TCF7L2 promoter had significant differences between type 2 diabetic patients and matched controls. In addition, fasting glucose, HOMA-B, HOMA-IR, total cholesterol and LDL-cholesterol correlated with methylation in specific CpG sites of TCF7L2 promoter. After adjustment by age, BMI, gender, physical inactivity, waist circumference, smoking status and diabetes status uniquely fasting glucose, total cholesterol and LDL-cholesterol remained significant. Taken together, newly diagnosed, drug-naïve type 2 diabetic patients display specific epigenetic changes at the TCF7L2 promoter as compared to age- and BMI-matched controls. Methylation in TCF7L2 promoter is further correlated with fasting glucose in peripheral blood DNA, which sheds new light on the role of epigenetic regulation of TCF7L2 in T2D.

Estudi aproximatiu a la síntesi en fase sòlida de la part glicosídica de la digitoxina, essencial per l’activitat biològica com a reguladora de les propietats farmacocinètiques, i per tant molt important per l’avenç de la química mèdica i bioinorgànica. La metodologia que s’ha seguit inclou: síntesi de la pentosa de partença adequada, a partir de la qual, es van obtindre les unitats de 2,6- didesoxiglicósids; síntesi dels suports polimèrics necessaris per la síntesi en fase sòlida dels compostos objectiu; estudi de diferent tipus de glicosil dadors i activador de tioglicosids, lactols i tricloroacetimidats per la reacció de glicosilació amb el linker. S’ha assolit comprensió i coneixement de la tecnologia associada a la síntesi en fase sòlida, que requereix procediment diferenciat de la síntesi en solució convencional.

Aquest estudi permet conèixer les implicacions dels diferents subtipus de cáncer de mama: luminal A, luminal B, HER2+, triple negatiu en el desenvolupament y pronostic de la carcinomatosi leptomeníngea (CL). Es van identificar 38 pacients, major proporció: luminal B y HER2+, 53% va rebre quimioteràpia sistémica (QTS). La mitjana de supervivencia post CL: 2,6 messos. A l’anàlisis univariat: ECOG de 0-2 y tractament amb QT van ser variables pronòstiques i al multivariant nomès QTS. En conclussió el subtipus de cáncer de mama influiex en el temps d’aparició de la CL, no afectant la supervivencia desprès del diagnóstic.

Conegut el fenomen epidèmic associat a la Insuficiència Cardíaca, l’objectiu de l’estudi va ser comparar pacients de la Unitat d’Insuficiència Cardíaca i d’Atenció Primària, tres mesos després de l’alta hospitalària. Els 112 pacients de la UIC van presentar una freqüència de reingressos de 8,1% i mortalitat de 7,1%, davant els 69 pacients d’AP amb 24,2% i 29,9%, respectivament. Una anàlisi multivariada, va demostrar que el seguiment en AP és un factor independent de major mortalitat i reingrés (OR 2,8 (IC 95%:1,2-6,6) i que rebre betablocadors és un efecte protector de la IC (OR 0,2 (IC 95%: 0,1-0,6).

Se han sintetizado nueve alcoholes que incorporan diversos isómeros de los grupos piridil o quinolil y metil-o-carborano u o-carborano, 1-[n′-Piridil(hidroxi)metil]-2-R-1,2-dicarba-closo-dodecaboranos (n′ = 2, R = H (2a); n′ = 3, R = Me (1c) y H (2c)), 1-[2′-6-metil-piridil(hidroxi)metil]-2-R-1,2-dicarba-closo-dodecaboranos (R = Me (1b) y H (2b)), 1-[n′-Quinolil(hidroxi)metil]-2-R-1,2-dicarba-closo-dodecaboranos (n′ = 2, R = Me (1e) y H (2e); n′ = 4, R = Me (1f) y H (2f), mediante la adición de la sal de litio del metil-o-carborano u o-carborano a los aldehídos correspondientes. Los compuestos se han obtenido con rendimientos altos en la mayoría de los casos y han sido caracterizados mediante Resonancia Magnética Nuclear, por Espectroscopía Infrarroja y análisis elemental. Las estructuras cristalinas de siete de los nueve compuestos (1c, 1f, 2a, 2b, 2c, 2e y 2f) se han determinado mediante Cristalografía por Rayos X. Se han analizado las estructuras cristalinas de los todos los compuestos y se han comparado con las estructuras de los compuestos relacionados 1a, 1b, y 2a, obtenidas previamente en el grupo de Síntesis Inorgánica y Catálisis del ICMAB. Todas las estructuras muestran interacciones por puentes de hidrógeno O–H···N moderadamente fuertes. Posteriormente se ha sintetizado el derivado mesilado 4 (derivado del o-carborano) mediante la reacción del alcohol 2a con cloruro de mesitilo a temperatura ambiente, con rendimiento alto. El nuevo mesilato 4 se ha caracterizado mediante Resonancia Magnética Nuclear. El nuevo mesilato resulta un compuesto de partida muy versátil para la síntesis de nuevos derivados mediante la substitución del grupo mesilo por nucleófilos apropiados, como por ejemplo las aminas. El grupo ha publicado recientemente la síntesis de diaminas mediante la reacción de substitución del mesilato relacionado 3 (derivado del metil-o-carborano) con aminas, entre ellas la bencilamina. Se ha realizado por tanto la reacción de substitución del grupo mesilato en 4 por la bencilamina para obtener la diamina correspondiente. Los resultados preliminares de esta reacción muestran que si bien el grupo mesilato en 4 ha sido substituido, el clúster se degrada casi en su totalidad a un derivado tipo nido. Esto contrasta claramente con la reactividad del mesilato análogo 3 frente a la bencilamina, ya que en este caso se obtiene la diamina deseada y el clúster no se degrada apreciablemente bajo las mismas condiciones de reacción. Se han realizado estudios preliminares por RMN que muestran que la bencilamina substituye al grupo mesilo en 4 y posteriormente el clúster closo se degrada a una especie nido que no ha sido posible caracterizar totalmente hasta el momento.
