Tissue protein hypercatabolism (TPH) is a most important feature in cancer cachexia, particularly with regard to the skeletal muscle. The rat ascites hepatoma Yoshida AH-130 is a very suitable model system for studying the mechanisms involved in the processes that lead to tissue depletion, since it induces in the host a rapid and progressive muscle waste mainly due to TPH (Tessitore, L., G. Bonelli, and F. M. Baccino. 1987. Biochem. J. 241:153-159). Detectable plasma levels of tumor necrosis factor-alpha associated with marked perturbations in the hormonal homeostasis have been shown to concur in forcing metabolism into a catabolic setting (Tessitore, L., P. Costelli, and F. M. Baccino. 1993. Br. J. Cancer. 67:15-23). The present study was directed to investigate if beta 2-adrenergic agonists, which are known to favor skeletal muscle hypertrophy, could effectively antagonize the enhanced muscle protein breakdown in this cancer cachexia model. One such agent, i.e., clenbuterol, indeed largely prevented skeletal muscle waste in AH-130-bearing rats by restoring protein degradative rates close to control values. This normalization of protein breakdown rates was achieved through a decrease of the hyperactivation of the ATP-ubiquitin-dependent proteolytic pathway, as previously demonstrated in our laboratory (Llovera, M., C. García-Martínez, N. Agell, M. Marzábal, F. J. López-Soriano, and J. M. Argilés. 1994. FEBS (Fed. Eur. Biochem. Soc.) Lett. 338:311-318). By contrast, the drug did not exert any measurable effect on various parenchymal organs, nor did it modify the plasma level of corticosterone and insulin, which were increased and decreased, respectively, in the tumor hosts. The present data give new insights into the mechanisms by which clenbuterol exerts its preventive effect on muscle protein waste and seem to warrant the implementation of experimental protocols involving the use of clenbuterol or alike drugs in the treatment of pathological states involving TPH, particularly in skeletal muscle and heart, such as in the present model of cancer cachexia.

Sensor networks have many applications in monitoring and controlling of environmental properties such as sound, acceleration, vibration and temperature. Due to limitedresources in computation capability, memory and energy, they are vulnerable to many kinds of attacks. The ZigBee specification based on the 802.15.4 standard, defines a set of layers specifically suited to sensor networks. These layers support secure messaging using symmetric cryptographic. This paper presents two different ways for grabbing the cryptographic key in ZigBee: remote attack and physical attack. It also surveys and categorizes some additional attacks which can be performed on ZigBee networks: eavesdropping, spoofing, replay and DoS attacks at different layers. From this analysis, it is shown that some vulnerabilities still in the existing security schema in ZigBee technology.

Background: Toll-like receptors (TLRs) are critical components for host pathogen recognition and variants in genes participating in this response influence susceptibility to infections. Recently, TLR1 gene polymorphisms have been found correlated with whole blood hyper-inflammatory responses to pathogen-associated molecules and associated with sepsis-associated multiorgan dysfunction and acute lung injury (ALI). We examined the association of common variants of TLR1 gene with sepsis-derived complications in an independent study and with serum levels for four inflammatory biomarker among septic patients. Methodology/Principal Findings: Seven tagging single nucleotide polymorphisms of the TLR1 gene were genotyped in samples from a prospective multicenter case-only study of patients with severe sepsis admitted into a network of intensive care units followed for disease severity. Interleukin (IL)-1 b, IL-6, IL-10, and C-reactive protein (CRP) serum levels were measured at study entry, at 48 h and at 7th day. Alleles -7202G and 248Ser, and the 248Ser-602Ile haplotype were associated with circulatory dysfunction among severe septic patients (0.001<=p <= 0.022), and with reduced IL-10 (0.012<= p <=0.047) and elevated CRP (0.011<= p <=0.036) serum levels during the first week of sepsis development. Additionally, the -7202GG genotype was found to be associated with hospital mortality (p =0.017) and ALI (p =0.050) in a combined analysis with European Americans, suggesting common risk effects among studies Conclusions/Significance: These results partially replicate and extend previous findings, supporting that variants of TLR1 gene are determinants of severe complications during sepsis.

In the present study, we examined seawater biofiltration in terms of adenosine triphosphate (ATP) and turbidity. A pilot biofilter continuously fed with fresh seawater reduced both turbidity and biological activity measured by ATP. Experiments operated with an empty bed contact time (EBCT) of between 2 and 14 min resulted in cellular ATP removals of 32% to 60% and turbidity removals of 38% to 75%. Analysis of the water from backwashing the biofilter revealed that the first half of the biofilter concentrated around 80% of the active biomass and colloidal material that produces turbidity. By reducing the EBCT, the biological activity moved from the first part of the biofilter to the end. Balances of cellular ATP and turbidity between consecutive backwashings indicated that the biological activity generated in the biofilter represented more than 90% of the detached cellular ATP. In contrast, the trapped ATP was less than 10% of the overall cellular ATP detached during the backwashing process. Furthermore, the biological activity generated in the biofilter seemed to be more dependent on the elapsed time than the volume filtered. In contrast, the turbidity trapped in the biofilter was proportional to the volume filtered, although a slightly higher amount of turbidity was found in the backwashing water; this was probably due to attrition of the bed medium. Finally, no correlations were found between turbidity and ATP, indicating that the two parameters focus on different matter. This suggests that turbidity should not be used as an alternative to cellular concentration.

Under pathological conditions, microglia, the resident CNS immune cells, become reactive and release pro-inflammatory cytokines and neurotoxic factors. We investigated whether this phenotypic switch includes changes in the expression of the L-type voltage-gated calcium channel (VGCC) in a rat model of N-methyl-d-aspartate-induced hippocampal neurodegeneration. Double immunohistochemistry and confocal microscopy evidenced that activated microglia express the L-type VGCC. We then analyzed whether BV2 microglia express functional L-type VGCC, and investigated the latter's role in microglial cytokine release and phagocytic capacity. Activated BV2 microglia express the CaV1.2 and CaV1.3 subunits of the L-type VGCC determined by reverse transcription-polymerase chain reaction, Western blot and immunocytochemistry. Depolarization with KCl induced a Ca2+ entry facilitated by Bay k8644 and partially blocked with nifedipine, which also reduced TNF-α and NO release by 40%. However, no nifedipine effect on BV2 microglia viability or phagocytic capacity was observed. Our results suggest that in CNS inflammatory processes, the L-type VGCC plays a specific role in the control of microglial secretory activity.
