Ionic Liquids (ILs) belong to a class of compounds with unusual properties: very low vapour pressure; high chemical and thermal stability and the ability to dissolve a wide range of substances. A new field in research is evaluating the possibility to use natural chiral biomolecules for the preparation of chiral ionic liquids (CILs). This important challenge in synthetic chemistry can open new avenues of research in order to avoid some problems related with the intrinsic biodegradability and toxicity associated to conventional ILs. The research work developed aimed for the synthesis of CILs, their characterization and possible applications, based on biological moieties used either as chiral cations or anions, depending on the synthetic manipulation of the derivatives. Overall, a total of 28 organic salts, including CILs were synthesized: 9 based on L-cysteine derivatives, 12 based on L-proline, 3 based on nucleosides and 4 based on nucleotides. All these new CILs were completely characterized and their chemical and physical properties were evaluated. Some CILs based on L-cysteine have been applied for discrimination processes, including resolution of racemates and as a chiral catalyst for asymmetric Aldol condensation. L-proline derived CILs were also studied as chiral catalysts for Michael reaction. In parallel, the interactions of macrocyclic oligosugars called cyclodextrins (CDs) with several ILs were studied. It was possible to improve the solubility of CDs in water and serum. Additionally, fatty acids and steroids showed an increase in water solubility when ILs-CDs systems were used. The development of efficient and selective ILs-CDs systems is indispensable to expand the range of their applications in host-guest interactions, drug delivery systems or catalytic reactions. Novel salts derived from nucleobases were used in order to enhance the fluorescence in aqueous solution. Additionally, preliminary studies regarding ethyl lactate as an alternative solvent for asymmetric organocatalysis were performed.

Ligand K-edge XAS of an [Fe3S4]0 model complex is reported. The pre-edge can be resolved into contributions from the í2Ssulfide, í3Ssulfide, and Sthiolate ligands. The average ligand-metal bond covalencies obtained from these pre-edges are further distributed between Fe3+ and Fe2.5+ components using DFT calculations. The bridging ligand covalency in the [Fe2S2]+ subsite of the [Fe3S4]0 cluster is found to be significantly lower than its value in a reduced [Fe2S2] cluster (38% vs 61%, respectively). This lowered bridging ligand covalency reduces the superexchange coupling parameter J relative to its value in a reduced [Fe2S2]+ site (-146 cm-1 vs -360 cm-1, respectively). This decrease in J, along with estimates of the double exchange parameter B and vibronic coupling parameter ì2/k-, leads to an S ) 2 delocalized ground state in the [Fe3S4]0 cluster. The S K-edge XAS of the protein ferredoxin II (Fd II) from the D. gigas active site shows a decrease in covalency compared to the model complex, in the same oxidation state, which correlates with the number of H-bonding interactions to specific sulfur ligands present in the active site. The changes in ligand-metal bond covalencies upon redox compared with DFT calculations indicate that the redox reaction involves a two-electron change (one-electron ionization plus a spin change of a second electron) with significant electronic relaxation. The presence of the redox inactive Fe3+ center is found to decrease the barrier of the redox process in the [Fe3S4] cluster due to its strong antiferromagnetic coupling with the redox active Fe2S2 subsite.

As sulfonas são conhecidas pelos químicos orgânicos pela sua versatilidade sintética e actividade biológica, nomeadamente os fármacos conhecidos por sulfa drugs, extremamente importantes em química medicinal. As vinil-sulfonas constituem uma sub-família das sulfonas e têm demonstrado ao longo do tempo a sua capacidade de inibir diversos processos enzimáticos, introduzindo propriedades biológicas únicas. Do ponto de vista sintético, o método mais directo para obter estes compostos é via redução dos produtos de adição de Michael a vinil-di-sulfonas, o que não é favorável em termos de economia de átomos. Por outro lado, o mecanismo da reacção de adição de Michael de compostos carbonílicos a vinil-sulfonas tem levantado algumas dúvidas, devido à existência de dois possíveis mecanismos reacionais. O objectivo principal desta dissertação é racionalizar o mecanismo de adição de compostos carbonílicos a vinil-mono e di-sulfonas, recorrendo a estudos teóricos de DFT e experimentais, incluindo RMN. Foram considerados os mecanismos via intermediário enamina e via catálise básica, e através de estudos de RMN foi possível retirar conclusões sobre as velocidades relativas das reacções, bem como meio de comprovar a formação de alguns produtos. Os resultados teóricos obtidos revelam que, para as mono-sulfonas, o mecanismo via enamina é mais favorável energeticamente. No entanto, a existência de reacções competitivas de energia de activação mais baixa não permitem obter o produto desejado. Para eliminar estas reacções, utilizam-se aminas terciárias e a reacção segue um mecanismo via catálise básica. O estudo dos coeficientes da orbital HOMO dos compostos carbonílicos revelou que a localização do enol é um factor determinante para estas reacções. Por outro lado, os resultados obtidos revelaram que as di-sulfonas sendo extremamente reactivas, podem adoptar qualquer um dos caminhos reacionais, em função do catalisador escolhido, e não são sensíveis ao substrato. Neste caso, as reacções competitivas que ocorrem são reversíveis,e o produto de adição conjugada é sempre favorecido. Estudou-se ainda, teoricamente, a reacção de adição a mono-sulfonas de forma intramolecular, que apresenta claramente um mecanismo via enamina, cuja energia de activação é mais baixa que a respectiva adição competitiva.

Clayish earth-based mortars are been recognized, all over the world, as eco-efficient products for plastering. Apart from being a product with low embodied energy when compared to other types of plasters, their application on the interior surface of walls may give a strong contribution for the health and comfort of inhabitants. As part of an ongoing research regarding earth-based plasters this work assesses the influence of the addition of two types of natural fibres – oat straw and typha fiber-wool – on the characteristics of plastering mortars made with a clayish earth. Mechanical and physical characteristics were tested, showing that addition of these fibers contribute to decrease linear drying shrinkage and thermal conductivity, as well as promoting the adhesion strength of plaster to the substrate. The improvement of mechanical resistance reveal to be dependent on the type of fiber added while the hygroscopic capacity of the plaster is maintained regardless of the fiber additions.
