The objective of this contribution is to extend the models of cellular/composite material design to nonlinear material behaviour and apply them for design of materials for passive vibration control. As a first step a computational tool allowing determination of optimised one-dimensional isolator behaviour was developed. This model can serve as a representation for idealised macroscopic behaviour. Optimal isolator behaviour to a given set of loads is obtained by generic probabilistic metaalgorithm, simulated annealing. Cost functional involves minimization of maximum response amplitude in a set of predefined time intervals and maximization of total energy absorbed in the first loop. Dependence of the global optimum on several combinations of leading parameters of the simulated annealing procedure, like neighbourhood definition and annealing schedule, is also studied and analyzed. Obtained results facilitate the design of elastomeric cellular materials with improved behaviour in terms of dynamic stiffness for passive vibration control.

Aging is a long-standing biological question of tremendous social and cultural importance. Despite this, only in the last 15 years has biology started to make significant progress in understanding the underlying mechanisms that regulate aging. This progress stemmed mainly from the use of model organisms, which allowed the discovery of several genes directly modulating longevity. Interestingly, several of these longevity genes are necessary for normal mitochondrial function, and disruption of their activity delays the aging process. This is somewhat paradoxical, considering the importance of cellular respiration for energy production and viability of eukaryotic organisms. One possible rationalization for this is that by decreasing cellular respiration, reactive oxygen species (ROS) generation is also reduced, and in that way, cellular decay and aging are delayed.(...)

The objective of this paper is to analyze the forearm muscular contraction levels associated to the use of anti-vibration gloves, by comparing the contraction levels with gloves and without gloves. Two different vibration tools were used in a simulated work environment: (1) A compact Duty Multi-Cutter Bosch and (2) and a Percussion Drill with a drill bit Ø20 mm. Standard operations were performed by each subject in the following materials: (1) Performing cross- sectional cuts in 80x40 mm pine section and (2) performing 20 mm diameter holes in a concrete slab 2 x 2 m, 70 mm thick. The forearm contraction level were measured by surface electromyography in four different muscles: Flexor Digitorum Superficialis (FDS), Flexor Carpi Ulnaris (FCU), Extensor Carpi Radialis Longus (ECRL) e Extensor Carpi Ulnaris (ECU). For the flexor muscles (FDS, FCU), an increase tendency in muscular contraction was observed when the operations are performed without gloves (2-5% MVE increase in the FDS and 3-9% MVE increase in the FCU). For the extensor muscles ECU a decrease tendency in muscular contraction was observed when the operations are performed without gloves (1-10% MVE decrease). Any tendency was found in the ECRL muscle. ECU was the muscle with the highest contraction level for 79% and 71% of the operators, during the operations respectively with the multi-cutter (P50= 27-30%MVE) and with the percussion drill (P50=46-55%MVE). As a final conclusion from this study, anti- vibration gloves may increase the forearm fatigue in the posterior region of the forearm (ECU muscle) during operations with the mentioned tools

In this paper analytical transient solutions of dynamic response of one-dimensional systems with sudden change of foundation stiffness are derived. In more details, cantilever dynamic response, expressed in terms of vertical displacement, is extended to account for elastic foundation and then two cantilever solutions, corresponding to beams clamped on left and right hand side, with different value of Winkler constant are connected together by continuity conditions. The internal forces, as the unknowns, can be introduced by the same values in both clamped beam solutions and solved. Assumption about time variation of internal forces at the section of discontinuity must be adopted and originally analytical solution will have to include numerical procedure.

High speed trains, when crossing regions with abrupt changes in vertical stiffness of the track and/or subsoil, may generate excessive ground and track vibrations. There is an urgent need for specific analyses of this problem so as to allow reliable esimates of vibration amplitude. Full understanding of these phenomena will lead to new construction solutions and mitigation of undesirable features. In this paper analytical transient solutions of dynamic response of one-dimensional systems with sudden change of foundation stiffness are derived. Results are expressed in terms of vertical displacement. Sensitivity analysis of the response amplitude is also performed. The analytical expressions presented herein, to the authors’ knowledge, have not been published yet. Although related to one-dimensional cases, they can give useful insight into the problem. Nevertheless, in order to obtain realistic response, vehicle- rail interaction cannot be omitted. Results and conclusions are confirmed using general purpose commercial software ANSYS. In conclusion, this work contributes to a better understanding of the additional vibration phenomenon due to vertical stiffness variation, permitting better control of the train velocity and optimization of the track design.

Passage of high-speed trains may induce high ground and track vibrations, which, besides increasing wheel, rail and track deterioration, may have a negative impact on the vehicle stability and on the passengers comfort. In this paper two distinct analyses are presented. The first one is dedicated to efficient decoupling of rail and soil vibrations by suggesting new interface materials in rail-sleeper fixing system, i.e. in the part where damping efficiency can be directly controlled and tested. The second analysis concerns with an adequate model of soils damping. Proper understanding and correct numerical simulation of this behaviour can help in suggesting soil improvement techniques.

Critical Velocity obtained using Simplified Models of the Railway Track: Viability and Applicability

Increased demands on the capacity of the railway network gave rise to new issues related to the dynamic response of railway tracks subjected to moving vehicles. Thus, it becomes important to evaluate the applicability of traditionally used simplified models which have a closed form solution. Regarding simplified models, transversal vibrations of a beam on a visco-elastic foundation subjected to a moving load are considered. Governing equations are obtained by Hamilton’s principle. Shear distortion, rotary inertia and effect of axial force are accounted for. The load is introduced as a time varying force moving at a constant velocity. Transversal vibrations induced by the load are solved by the normal-mode analysis. Reflected waves at the extremities of the full beam are avoided by introduction of semi-infinite elements. Firstly, the critical velocity obtained from this model is compared with results of an undamped Euler- Bernoulli formulation with zero axial force. Secondly, a finite element model in ABAQUS is examined. The aim of this contribution is to extend the techniques of composite materials design to non-linear material behaviour and apply it for design of new materials for passive vibration control. As a first step a computational tool allowing determination of macroscopic optimized one-dimensional isolator behaviour was developed. Voigt, Maxwell, standard and more complex material models can be implemented. Objective function considers minimization of the initial reaction and/or displacement peak as well as minimization of the steady-state amplitude of reaction and/or displacement. The complex stiffness approach is used to formulate the governing equations in an efficient way. Material stiffness parameters are assumed as non-linear functions of the displacement. The numerical solution is performed in the complex space. The steady-state solution in the complex space is obtained by an iterative process based on the shooting method which imposes the conditions of periodicity with respect to the known value of the period. Extension of the shooting method to the complex space is presented and verified. Non-linear behaviour of material parameters is then optimized by generic probabilistic meta-algorithm, simulated annealing. Dependence of the global optimum on several combinations of leading parameters of the simulated annealing procedure, like neighbourhood definition and annealing schedule, is also studied and analyzed. Procedure is programmed in MATLAB environment.

Rute C. Félix PALAVRAS-CHAVE: Malária, mosquito vector, Anopheles gambiae, parasita, Plasmodium berghei, infecção, enzimas de detoxificação, citocromos P450, tubulinas A malária, uma das doenças mais devastadoras que ocorrem em África é causada por um parasita do género Plasmodium e é transmitida aos humanos por mosquitos vectores do género Anopheles durante a refeição de sangue. Apesar da resposta do mosquito à infecção por Plasmodium ter vindo a ser intensamente estudada nos últimos anos, as interacções entre o mosquito vector e o parasita são muito complexas e, estão longe de serem completamente compreendidas. Este estudo tem como objectivo principal contribuir para o conhecimento da resposta do mosquito à infecção por Plasmodium, focando-se no papel das enzimas de detoxificação. Para atingir este objectivo realizouse uma análise transcriptómica com microarrays, com o intuito de identificar alterações de transcrição de enzimas de detoxificação no mosquito Anopheles gambiae em resposta à infecção por Plasmodium. Esta análise permitiu identificar alterações na expressão de 254 genes de destoxificação no estômago e corpo gordo de A. gambiae durante a invasão do intestino médio pelos oocinetos e durante a libertação dos esporozoítos do oocisto. Os resultados mostraram que a invasão do intestino médio pelos oocinetos causou alterações num maior número de genes em ambos os tecidos estudados, sendo o intestino médio do mosquito o tecido mais afectado nas duas fases da infecção do parasita. De todos os genes de destoxificação com expressão alterada, as tubulinas e os citocromos P450 destacaram-se e foram escolhidos para continuar o estudo. As tubulinas foram seleccionadas porque estão associadas à invasão do epitélio do intestino médio e a sua função na resposta à invasão do Plasmodium ainda não está bem definida. Os citocromos P450 foram seleccionados porque já foram descritos como tendo a expressão alterada em resposta ao Plasmodium e a outras infecções. Para identificar e caracterizar o papel das tubulinas durante a infecção pelo parasita e a sua possível associação com os citocromos P450 foi utilizado o silenciamento génico por RNA de interferência e a injecção de inibidores químicos de tubulinas. O silenciamento e co-silenciamento das tubulinas causaram um aumento da taxa e intensidade da infecção. No entanto, apesar de o aumento ser consistente não foi significativo. Por outro lado, a injecção de paclitaxel, um inibidor de tubulinas, aumentou significativamente a taxa e intensidade da infecção, fortalecendo a hipótese do envolvimento das tubulinas na resposta à infecção por Plasmodium. Este trabalho também mostrou que o co-silenciamento da tubulina A e tubulina B e a injecção do inibidor de tubulinas colchicine causam alterações significativas na expressão da CYP6Z2, sendo este proposto como um possível elo de ligação entre as tubulinas e os citocromos P450. Finalmente, uma análise comparativa foi realizada para estudar as regiões promotoras dos citocromos P450: CYP6M2 e o CYP6Z1. Este estudo obteve novos dados sobre compostos que activam estes citocromos e quais os possíveis factores de transcrição envolvidos. Dos diferentes estímulos utilizados, a exposição a insecticidas e a bactérias foram os que mais afectaram estes citocromos. O conjunto total das diferentes abordagens utilizadas neste trabalho contribuiu para aumentar o conhecimento do papel das enzimas de destoxificação durante a passagem do parasita da malária pelo mosquito vector.
