The interpretation of 64 seismic reflection profiles in the Algarve continental platform (36º 20'-37º 00' paralels and 7º 20'-8º 40' meridians) calibrated with five petroleum exploration wells, with the identification of the geometric relations between six Cenozoic seismic units (B to G) and tectonic structures, allowed the construction of sucessive time-isopach maps (twt/s) and detailed interpretation of the geologic evolution. Two major tectonic structures were identified: a) the Portimão-Monchique fracture zone (striking N-S); b) an off-shore NW-SE fault zone, probably the S. Marcos-Quarteira fault. This accident separates two tectonic domains: the western domain (with N-S and E-W predominant structures and, secondarily, NW-SE and NE-SW) and the eastern domain (dominated by WSW-ENE, NW-SE, NE-SW, NNE-SSW and NNW-SSE structures). A persistent halokinetic activity had two major moments: a) sin-C unit; b) sin- and post-E unit. An increasing flexuration of the margin was identified, with spacial and temporal variation of the subsidence. The tectonic regime is considered as generally compressive, but the interpretation of the successíve stress-fields is rendered dificult by the existence of tectonic sub-domains and evaporitic structures.

São Tomé Príncipe é um dos países mais pobres do mundo totalmente dependente da ajuda externa. Desde a independência, a 12 de Julho de 1975, o país mergulhou num clima de instabilidade sociopolítica e de pobreza que vem afetando e condicionando o desenvolvimento socioeconómico da sociedade santomense. A fraca capacidade governativa dos dirigentes e a falta de consenso na classe política, as sucessivas quedas de governo, o elitismo e o favoritismo reinante tornam quase impossível que se encontre um caminho consensual para que o país possa efetivamente sair da situação de pobreza em que se encontra e começar a dar passos significativos rumo ao desenvolvimento e ao progresso. Sendo um país com imensos problemas estruturais, que se vão agravando com o passar dos anos, São Tomé e Príncipe é um país extremamente frágil, onde a pobreza atinge a maior parte da população. A instabilidade política e social que se vive no país impede que a classe política nacional seja capaz de chegar a um entendimento para o progresso socioeconómico da nação. São Tomé e Príncipe é um país que dispõe de inúmeras potencialidades naturais, muitas delas continuam a ser mal aproveitadas devido à ausência de uma estratégia concertada que conduza o país ao desenvolvimento. Mas para que aconteça é imprescindível que os órgãos decisores do país sejam capazes de primar por políticas credíveis, transparentes e viáveis, para que o progresso e desenvolvimento tão desejado pela sociedade santomense não seja apenas um sonho. Com a descoberta do petróleo no território santomense, a nação tem depositado todas as suas esperanças na exploração petrolífera e que com a arrecadação das receitas daí provenientes o país começará a trilhar o caminho do desenvolvimento e do progresso.

This thesis was focused on the production, extraction and characterization of chitin:β-glucan complex (CGC). In this process, glycerol byproduct from the biodiesel industry was used as carbon source. The selected CGC producing yeast was Komagataella pastoris (formerly known as Pichia pastoris), due the fact that to achieved high cell densities using as carbon source glycerol from the biodiesel industry. Firstly, a screening of K. pastoris strains was performed in shake flask assays, in order to select the strain of K. pastoris with better performance, in terms of growth, using glycerol as a carbon source. K. pastoris strain DSM 70877 achieved higher final cell densities (92-97 g/l), using pure glycerol (99%, w/v) and in glycerol from the biodiesel industry (86%, w/v), respectively, compared to DSM 70382 strain (74-82 g/l). Based on these shake flask assays results, the wild type DSM 70877 strain was selected to proceed for cultivation in a 2 l bioreactor, using glycerol byproduct (40 g/l), as sole carbon source. Biomass production by K. pastoris was performed under controlled temperature and pH (30.0 ºC and 5.0, respectively). More than 100 g/l biomass was obtained in less than 48 h. The yield of biomass on a glycerol basis was 0.55 g/g during the batch phase and 0.63 g/g during the fed-batch phase. In order to optimize the downstream process, by increasing extraction and purification efficiency of CGC from K. pastoris biomass, several assays were performed. It was found that extraction with 5 M NaOH at 65 ºC, during 2 hours, associated to neutralization with HCl, followed by successive washing steps with deionised water until conductivity of ≤20μS/cm, increased CGC purity. The obtained copolymer, CGCpure, had a chitin:glucan molar ratio of 25:75 mol% close to commercial CGC samples extracted from A. niger mycelium, kiOsmetine from Kitozyme (30:70 mol%). CGCpure was characterized by solid-state Nuclear Magnetic Resonance (NMR) spectroscopy and Differential Scanning Calorimetry (DCS), revealing a CGC with higher purity than a CGC commercial (kiOsmetine). In order to optimize CGC production, a set of batch cultivation experiments was performed to evaluate the effect of pH (3.5–6.5) and temperature (20–40 ºC) on the specific cell growth rate, CGC production and polymer composition. Statistical tools (response surface methodology and central composite design) were used. The CGC content in the biomass and the volumetric productivity (rp) were not significantly affected within the tested pH and temperature ranges. In contrast, the effect of pH and temperature on the CGC molar ratio was more pronounced. The highest chitin: β-glucan molar ratio (> 14:86) was obtained for the mid-range pH (4.5-5.8) and temperatures (26–33 ºC). The ability of K. pastoris to synthesize CGC with different molar ratios as a function of pH and temperature is a feature that can be exploited to obtain tailored polymer compositions.(...)

There is a rising demand for local food which traveled only short distances and is marketed directly by the producer. With growing importance of local food also the amount of literature in this field increased. Yet, literature is lacking to examine the challenges and burdens consumers face while trying to purchase local food. Evidence is shown that a gap exists between the intention of consumers who would like to purchase local food and their actual behavior. However, reasons for this gap are only discovered as byproducts of other research objectives. Consequently, this study investigates the specific reasons for the intention-behavior gap of consumers in the local food market. The study makes use of semi-structured face-to-face interviews with rural and urban respondents. The interviews are analyzed by qualitative content analysis based on Elo and Kyngäs (2008). Revealed key drivers for the intention-behavior gap are (1) the lack of awareness where to purchase local food products, (2) the limited reachability of local food producers, (3) the assortment of the local producer in regard to size and composition and (4) the lack in transparency of the prices of local food. Consequently, it is the producers turn to eliminate the revealed barriers by proactively bringing their products closer to the consumer.
